3.64 mmol/L)received 8-week double blind comparisons treatment. After treatment we randomly selected 15 patients for each group. The fasting serum lipid was measured at the end of 4-week and 8-week,and the serum hs-CRP and IL-6 were measured too.Meanwhile seven hypercholesterolemic patients from rosuvastatin group and seven ortholiposis out-patients were included. These patients were treated with statins for 4 weeks. The erythrocytes before and after treatment were kept to be measured the FA.Results (1)Compared with the pretreatment, the level of LDL-C,TC,hs-CRP remarkably reduced in two treatment groups at the end of 4-week therapy. These inflammatory parameters did not further reduced, but hs-CRP reduced further at the end of 8-week therapy.(2)The percent age of SFA was obviously higher in hypercholesterolemic group than ortholiposis group. On the other hand,PUFA and ?-3 FA were obviously lower. After 4-week treatment the percent age of SFA reduced, while PUFA and ?-3 FA increased significantly in hypercholesterolemic group. Conclusion (1)Statins reducing inflammatory factors is independent of lipid-modifying ability.(2)Rosuvastatin treatment can effectively reduce the level of LDL-C and TC,and no significant difference in the effect is observed between two treatment groups.(3)The erythrocyte membrane FA proportion was obviously different in hypercholesterolemic group from ortholiposis group.Statins treatment could make the proportion of FA in erythrocyte membrane restore normal.

In order to establish more simple and effective rat orthotopic lung transplantation models, 20 rats were divided into donor and recipient groups. Rat lung transplantation models were established by using improved cuff technique. All the 10 operations were accomplished successfully. The mean operative time of recipients was 45 +/- 4 min. The survival time was over 30 days after lung transplantation. The checks of X-ray were almost normal. There was no significant difference in the blood gas analysis before and after clipping the right hilum (P > .05). This method is more simple, applicable and requires less time.

Microbial nitrilases have attracted increasing attention in nitrile hydrolysis for carboxylic acid production in recent years. A bacterium with nitrilase activity was isolated and identified as Pseudomonas putida CGMCC3830 based on its morphology, physiological and biochemical characteristics, as well as 16S rRNA gene sequence. The nitrilase production was optimized by varying culture conditions using the one-factor-at-a-time method and response surface methodology. Glycerol 13.54 g/L, tryptone 11.59 g/L, yeast extract 5.21 g/L, KH2PO4 1 g/L, NaCl 1 g/L, urea 1 g/L, initial pH 6.0 and culture temperature 30 degrees C were proved to be the optimal culture conditions. It resulted in the maximal nitrilase production of 36.12 U/mL from 2.02 U/mL. Investigations on substrate specificity demonstrate P. putida nitrilase preferentially hydrolyze aromatic nitriles. When applied in nicotinic acid synthesis, 2 mg/mL P. putida cells completely hydrolyzed 20.8 g/L 3-cyanopyridine into nicotinic acid in 90 min. The results indicated P. putida CGMCC3830 displayed potential for industrial production of nicotinic acid.

Objective To investigate the effect and its significance of low-dose insulin on Nmethyl-D-aspartate receptor (NMDAR) level and its downstream signaling of c-Jun N-terminal kinase (JNK) and the extracellular signal-regulated protein kinase (ERK,p44/42MAPK) in streptozotocininduced diabetic rats. Methods Ten-week-old male SD rats were randomly divided into 3 groups:control group, diabetic group and insulin-treated diabetes group.Diabetes was induced by streptozocin (STZ,60mg/kg) injected intraperitoneally. Two weeks after STZ injection, insulin glargine (92u/day for 8 weeks) was subcutaneously injected in the insulin-treated diabetes group. Then, the rat lumbar spinal cords were collected. The protein levels of phospho-Insulin receptor substrate 1 (P-IRS1),phospho-NMDAR NR1 subunit (P-NR1),P-JNK and P-p44/42MAPK were evaluated by Western blotting. One week before the terminal of the study, paw thermal response latency was measured in all groups. Results Blood glucose levels were tremendously high in both the diabetic group and insulintreated diabetes group.Compared with the control group, paw thermal response latency was markedly shortened in the diabetic group (P< 0.001) and the insulin-treated diabetes group (P< 0.001),and the alteration was more pronounced in the diabetic group (P<0.05).Compared with the control group, the protein levels of P-IRS1,P-NR1,P-JNK and P-p44/42MAPK were increased by 79.2%,35.1%,47.6 %,64.3 % and 87.6 %,respectively in diabetic group and 49.4 %,19.1%,16.5 %,31.8% and 39.9%,respectively in insulin-treated diabetes group (all P<0.001 or 0.05).In comparison with diabetic group, the increased amplitudes of above 4 parameters were decreased by 29.8%,16.0%,31.2%,32.5% and 47.7% respectively in the insulin-treated diabetes group (all P<0.05).Conclusions NMDAR and its downstream signals, such as JNK and p42/44MARK,are involved in the pathogenesis of painful diabetic neuropathy.Although it could not efficiently control the blood glucose level,low-dose insulin treatment may partly inhibit the occurrence of thermal hyperalgesia through inhibiting NMDAR signal pathway.

In order to provide us new clues to induce some endogenous protective molecular mechanisms, the changes in gene expression profile induced by ischemia-reperfusion in pulmonary tissues of rats were investigated and the dynamic mechanism of pulmonary ischemia-reperfusion injury was elucidated. Thirty male Wistar rats were randomly divided into 6 groups: 5 ischemia-reperfusion (I/R) groups (I/R 0-h, I/R 1-h, I/R 3-h, I/R 6-h, I/R 24-h) and control group (n=5 in each). An in situ ischemia-reperfusion lung injury rat model was established by occluded hilus of lung. The RatRef-12 Expression Beadchip (22 226 gene probes per array) was used to analyze the pattern of gene expression in all groups. The results showed that 648, 340, 711, 1279 and 641 genes were differentially expressed in I/R 0-, 1-, 3-, 6-and 24-h groups respectively. The differentially expressed genes were classified as following 7 functional categories: cytokine, adhesion molecule, growth factor and apoptosis-related factor, oxidation and antioxidation molecule, metabolic enzyme, ion channel and aquaporin, signal transduction molecule. It was suggested that gene chip technology was an effective and quick method for screening differentially expressed genes. Many differentially expressed genes with different functions interacted each other to result in pulmonary ischemia-reperfusion injury.
