1.Analysis of the correlative factors of fetal growth restriction

Objective To discuss the correlative factors of fetal growth reatriction(FGR)in order to earlier detect,earlier diagnose,earlier treat FOR and prevent the complications and subsequent syndrome of FGR.Methods 292 cases of FGR were retrospectively analyzed during the last several years in our hospital.Results Among 292 cases of FGR,87(29.8%)had maternal factor,18(6.2%)had fetal factor,17(5.8%)had placental factor,58 (19.9%)had umbilical cord factor,112(38.0%)had unknown.In vagina childbirth group,newborn asphyxia was 24(41.4%),and in c-section group,there was 24(10.3%).There was significant difference between them.Conclu- sion The main cause of FGR is maternal factor.Treating the complications and subsequent syndrome of pregnance timely and rightly,broadening the refers drafts of c-section properly would be advantageous in reducing the sequelas of FGR.
2.Changes of Mercury and ACh, AChE Contents in Rat Brains After Acute Methylmercury Chloride Exposure

Objective To study the neurotoxic effects of acute exposure of methylmercury at low dose and to provide some experimental data for deeply exploring the early mechanism of neurotoxicity of methylmercury. Methods SD rats were administered with methylmercury chloride by intraperitoneal injection with different doses of 0.05, 0.50 and 5.00 mg/kg and different exposure times of 20 min, 1 h, 4 h, 24 h. Mercury, ACh and AChE contents in rat brains were measured. Results Mercury contents in rat brains significantly increased after 20 min-exposure at both 0.50 and 5.00 mg/kg doses. Significant increase occurred after 4 h exposure at dose of 0.05 mg/kg. ACh and AChE in rat brains significantly changed after 20 min at every dose, showing certain dose-response and time-response relations. Conclusion Changes of ACh and AChE in rat brain after administration of low dose(0.05 mg/kg) and short time(20 min) exposure suggested certain modulation of CNS had been initiated. With the increases of exposure dose and time, methylmercury might begin to accumulate in rat brain and induce the significant changes of ACh and AChE.

Objective To establish a method for determination of carbendazim(MBC) in fruits by using novel activated carbon fiber SMPE coupled with GC.Methods Carbendazim(MBC) in the fruits was determined by homemade activated carbon fiber-solid phase microextraction(ACF-SPME) coupled with GC-ECD.The conditions were optimized.Results The linear range was 1-100 ?g/L.The detection limit was 0.002 ?g/L.The recovery rates were 89%-95% with the relative standard deviation(RSD) was 5.6%.This method had successfully been applied to the determination of MBC in the fruits.Conclusion This method reveals a lower detection limit,high accuracy and needs no organic solvent,it is suitable for the determination of MBC in fruit.
5.NMR-based analysis of water soluble extracts of different Astragali Radix.

Water soluble extract (WSE) is an important index for the quality evaluation of Astragali Radix (AR). In this study, the WSE of the wild AR from Shanxi province (SX) and the cultivated AR from Gansu Province (GS) were compared. The WSEs of two types of AR were determined according to the appendix of Chinese pharmacopoeia. Then the WSEs were subjected to NMR analysis, and the obtained data were analyzed using HCA, PCA, OPLS-DA, microarray analysis, and Spearman rank analysis. In addition, the Pearson correlation of differential metabolites were also calculated. The results showed that the WSE content of GS-AR (37.80%) was higher than that of SX-AR (32.13%). The main constituent of WSE was sucrose, and other 18 compounds, including amino acids, organic acids, were also detected. Multivariate analysis revealed that SX-AR contained more choline, succinic acid, citric acid, glutamate, taurine and aspartate, while GS samples contained more sucrose, arginine and fumaric acid. In addition, the Pearson correlations between different metabolites of the two types of AR also showed apparent differences. The results suggested that the WSE of two types of AR differs not only in the content, but also in the chemical compositions. Thus, the cultivation way is important to the quality ofAR. This study supplied a new method for the comparison of extract of herbal drugs.
6.Effect of exercise on expression of p38 after cerebral infarction in rats

ObjectiveTo study the effect of exercise on expression of p38 after cerebral infarction in rats.MethodsThe right middle cerebral artery was occluded(MCAO) with electric coagulation in Wistar rats,and then were divided into three groups at random 48 hours later: training group which were given wheel-running exercise everyday(n=20);control group(n=20) and sham group(n=10).The expression of p38 in cortex of the three groups were detected on 7th,14th,21th and 28th day after the MCAO.ResultsIn the cortex around the infarcted focus,the expressions of p38 elevated much significantly after cerebral infarction(P<0.05).The expression of p38 in training group were higher than those in control group(P<0.05) on the 21th and 28th day.ConclusionThe cerebral infarction could induce the expression of p38.The exercise could enhance its expression.
7.Effect of osthole on tricalcium phosphate particles-induced calvarial oste-olysis in a mouse model

AIM: To observe the effect of osthole on tricalcium phosphate (TCP) particles-induced calvarial osteolysis in vivo.METHODS:Male ICR mice were randomly divided into sham group , TCP group and osthole group .A mouse calvarial model of osteolysis was established by TCP particles .On the second postoperative day , osthole (20 mg/kg) was locally injected into the calvarium under the periosteum 3 times a week.Two weeks after osthole treatment , blood and calvaria were collected to determine the level of bone turnover markers such as alkaline phosphatase ( ALP) , osteocalcin and tartrate-resistant acid phosphatase ( TRACP) .The periosteum was performed to examine the release of tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6) and IL-1βby ELISA.The calvaria was obtained for histological and molecular analyses.RESULTS:Data from HE and TRACP staining revealed that osthole prevented TCP particles-induced obvious increase in osteoclastogenesis and resorption area in the metaphysis of mouse calvaria .Osthole treatment increased ALP ac-tivity and osteocalcin level , and dncreased the activity of TRACP in the mouse serum compared with TCP group .Further-more, TCP particles-induced the releases of TNF-α, IL-6 and IL-1βwere significantly suppressed by osthole treatment .In addition, Western blot demonstrated that endoplasmic reticulum ( ER) stress markers such as glucose-regulated protein 78 (GRP78) and CAAT/enhancer binding protein homologous protein (CHOP) were significantly up-regulated in TCP parti-cles-implanted calvarial mice , indicating that TCP particles triggered an ER stress response in the mouse calvarial osteolysis model , which obviously attenuated by osthole .CONCLUSION:Osthole inhibits TCP particles-induced calvarial osteolysis in mice, which is mediated by inhibition of ER stress signaling pathway .
8.Ad5-enhanced green fluorescent protein versus rAAV2-enhanced green fluorescent protein transfecting adipose-derived mesenchymal stem cells

BACKGROUND:There are reports concerning differentiation of adipose-derived mesenchymal stem cells(ADSCs)into chondrocytes using gene transfection technique.However,the transfection of adenovirus and adeno-associated virus into ADSCs is various.It is controversial whether adeno-associated virus(AAV)can transfect ADSCs.OBJECTIVE:To observe the enhanced green fluorescent protein(EGFP)expression following Ad5-EGFP and rAAV2-EGFP transfection into ADSCs,and investigate the cell proliferation ability following transfection.METHODS:ADSCs were isolated from the adipose tissue,which was from 6-month-old New Zealand albino rabbit back and neck by mechanical digestion and enzyme digestion,then ADSCs were cultured and amplified in vitro.ADSCs were infected with Ad5-EGFP and rAAV2-EGFP,and the EGFP expression was observed.A total of 2 μL sodium butyrate(1 mol/L)was added into the medium after rAAV2-EGFP transfection.MTT assay was used to detect the gene transfection effects on reproductive activity of ADSCs.RESULTS AND CONCLUSION:ADSCs isolated and cultured in vitro were flat,long-spindle and amplified stabry.The cell morphology was uniform.Many green fluorescent cells were observed in Ad5-EGFP and rAAV2-EGFP groups.Transfection efficiencies were about 88% and 83%.Adenovirus and adeno-associated virus vector can be transfected with ADSCs,and transfection efficiency is high.Adeno-associated virus needs sodium butyrate to increase its level of gene expression.
9.Diagnostic Evaluation of Articulations of Foot and Hand by 64-slice VCT Reconstruction

Objective To evaluate the diagnostic value of 64-slice VCT reconstruction techniques in the diseases of joints of foot and hand.Methods 50 patients with diseases of joints of foot and hand underwent MSCT scans,the data were reconstructed,including multiplanar reconstruction(MPR),maximum intensity projection(MIP),shaded surface display(SSD)and volume rendering(VR),the results were analysed.Results 33 patients with fractures,40 and 51 fractures were found by X-ray and MPR,VR,respectively.The joint surface involved in 7 fractures,which was best with VR and MPR to show the whole fractures.SSD was better to demonstrate the soft tissue and the vessels of the skin.3D bone reconstruction with MIP was better than plain film in showing the fractures in 4 patients with plaster external fixation.Conclusion 64-slice VCT reconstruction techniques are effective in diagnosis of the diseases of joints of foot and hand.
10.Imaging features of rare mastitis

Objective To summarise the imaging features of rare mastitis and explore the diagnostic value of ultrasound, mammography and MRI for rare mastitis. Methods The record of 24 patients diagnosed as rare mastitis in our hospital from Jan. 2000 to Jun. 2009 was reviewed, including clinical manifestations, pathological results, imaging diagnosis and diagnostic accurate rate. Results Of the 24 patients, 14 patients were ductal ectasia with chronic mastitis, 3 granulomatous mastitis, 6 chronic abscess and 1 mammary tuberculosis. 13 patients underwent ultrasonic scan, 12 patients underwent mammography and 3 patients underwent MRI, with the diagnostic accurate rate 77%, 25% and 100% respectively. Conclusions There are no special imaging manifestations for most rare mastitis, however, some differential characteristics still exist. MRI has a higher accuracy compared to ultrasound and mammography. The combination of multiple imaging methods can improve diagnostic accuracy.
