1.Pigmented Villonodular Synovitis of the Knee Joint:Imaging Manifestations

Objective To explore the imaging characteristics of pigmented villonodular synovitis(PVNS) of knee joint.Methods X-ray,CT and MR imaging features of PVNS of knee confirmed by operation,arthroscope and pathology were retrospectively analysed with literatures review.Results X-ray could only show the swelling of the joint cpasules,the destruction,defect and sclerosis of the bone under the articular surface.CT could show the soft tissue nodules in joint cavity projected from joint capsule.MRI was sensitive in showing the thickened synovium of joint and joint effusion,the soft tissue nodules and thickened synovium of joint were iso-or low signal intensity on T1WI,isointensity in one and low or slight hyperintensity in 4 on T2WI.Multiple haemosiderin deposit nodules were seen in the thickened synovium,which were low signal intensity on T1WI and T2WI in 4 cases,and the nodules were enhanced obviously on contrast-enhanced MR images.Conclusion MRI possesses obviously in diagnosis of PVNS of knee joint than X-ray and CT,which is helpful in diagnosis and differential diagnosis of PVNS of knee joint.
2.Clinical analysis of eosinophilic gastroenteritis in two children and review of literatur e

Objective To explore the clinical-pathological characteristics, treatment and prognosis of eosinophilic gastro-enteritis in children. Methods The data of clinical manifestation, laboratory examination, endoscopy, biopsy, management and prognosis from 2 pediatric patients with eosinophilic gastroenteritis were retrospectively analyzed. Related articles were reviewed. Results The age of two patients was 13 and 14 years old. Case 1 was male and had an onset with acute pancreatitis. Case 2 was female with a history of food allergy and had an onset with unexplained ascites. Both patients presented with signiifcantly increased peripheral eosinophil count (45.9%-64.8%) and serum IgE (246-393 IU/ml). Bone marrow cytology showed increased proportion of eosinophils in both patients. Gastroscopy was performed in case 1 while gastroscopy and colonoscopy endoscopy were performed in Case 2 and the results indicated eosinophilic inifltration in duodenum. Duodenal mucosa biopsy showed eosinophilic inifltration in case 1 while gastrointestinal pathology biopsy showed multiple mucosal eosinophil inifltration in case 2. After the therapy of food avoidance, steroid and anti-allergic drugs, both patients had complete remission 1 week later. Meanwhile the peripheral eosinophil count was decreased to normal. Case 1 was followed up for 24 months, and case 2 was followed up for 2 months. Both patients showed no evidence of relapse. Conclusions The clinical manifestation and endoscopic examination of eosinophilic gastroenteritis in children are protean and lack of speciifcity. Unexplained gastrointestinal symptoms accompanied with eosinophilia may suggest the possibility of eosinophilic gastroenteritis. Eosinophilic inifltration in ascites and gastrointestinal mucosa strongly indicate the di-agnosis of eosinophilic gastroenteritis. After exclusion of other possible diseases, the deifnite diagnosis can be made.
3.Comparison of proliferative and adipogenetic potentials between three human mesenchymal stem cells

Objective To isolate and culture mesenchymal stem cells (MSCs)from human term umbilical cord and placenta,and to study the proliferative and adipogenic capacities of human fetus tissue MSCs (FT-MSCs ), umbilical cord MSCs (UC-MSCs)and placenta MSCs (PL-MSCs), and to provide an experimental basis for its clinical use.Methods The human term umbilical cord and placenta were obtained in sterile condition. The UC-MSCs and PL-MSCs were isolated by digestion using collagenaseⅠ,the FT-MSCs were provided by Zhongke Bio-engineering Co.,Ltd.The 3th or 4th passages of MSCs were selected in this study.The cell morphology was observed by inverted microscope.The proliferative ability was detected by living cell number counting method.The cell cycle and the positive expression rates of the surface markers of three kinds of MSCs were detected by flow cytometry (FCM).The 3th generation of UC-MSCs,PL-MSCs,and FT-MSCs were induced to differentiate into adipogenic cells.18 d after being induced,the differentiated cells were stained with Oil red O and the positive cells were counted. The adipogenic differentiation abilities of MSCs from the different tissues were assessed. Results The MSCs from different tissues were spindle-shaped adherent cells.The FCM results showed the surface marker of MSCs were positive for CD44,CD73,CD90,and CD105,and negative for CD14,CD34,and CD45. The FT-MSCs displayed higher proliferative ability than UC-MSCs and PL-MSCs (P<0.01).The proliferative index (PI)of FT-MSCs (36.66%±1.30%)was significantly higher than those of UC-MSCs (18.23%±1.10%) and PL-MSCs (10.03%±1.20%) (P<0.01).The lipid droplets were found after induction,and the results of Oil red O staining were positive in all kinds of MSCs. However the adipogenic capacity of UC-MSCs was significantly higher than those of PL-MSCs and FT-MSCs (P<0.01).Conclusion UC-MSCs,PL-MSCs and FT-MSCs have similar biological characteristics and cell surface markers. But FT-MSCs have the strongest proliferative ability and activity,UC-MSCs have the strongest adipogenic capacity invitro.
4.Reform and practice of teaching methods for culture of medicinal plant.

5.Improved synthesis and pharmacological evaluation of racemic 11-demethylcalanolide A

An improved and practical synthesis of racemic 11-demethylcalanolide A [(±)-1] was developed. This improved process involved Pechmann reaction on phloroglucinol with ethyl butyrylacetate to give 5, 7,-dihydroxy-4-n-propylcoumarin(3). Poly phosphoric acid (PPA) catalyzed acylation of compound(3) with crotonic acid, then intramolecular cyclization was achieved simultaneously in one step to afford the key intermediate chromanone(4). A microwave assisted synthetic method preparing chromene(6) using chromenynation of chromanone(4) with 1, 1-diethoxy-methyl-2-butene was conducted. Luche reduction of chromene(6) using NaBH<,4> with CeCl3·7H2O preferably gave (±)-1. The overall yield of this four step synthesis of (±)-1 was around 32% increasing one fold more than that of the previous method. An in vitro investigation showed that (±)-1 exhibited inhibitory activities against both wild-type and drug-resistant HIV-1 in HIV-1 RT and cell culture assay, and significant synergistic effects in combination with AZT, T-20, and indinavir. Its LD50 of acute toxicity in mice by intragastric administration and by intraperitoneal injection were 735.65mg·kg-1 and 525.10mg·kg-1, respectively. The C<,max> and AUC<,0-∞> were 0.54μg·mL-1 and 1.08(μg·mL-1)·h, respectively. The dynamics study of the inhibition of mice sera on HIV-1 RT showed that mice treated with 100mg·kg-1 (±)-1 once intraperitoneally were similar to that of 5mg·kg-1 of known clinical effective anti-HIV-1 drug neverapine. The results suggested that further investigation of the anti-HIV candidate (±)-1 was warranted.
6.Synchronized feature patterns of local field potentials in hippocampal-prefrontal cortex during working memory based on time-varying spectral coherence

Objective:To investigate the synchronized feature patterns of local field potentials in the hippocampus (HPC) and prefrontal cortex (PFC) during working memory based on time-varying spectral coherence so as to support the study of information processing mechanisms in working memory.Methods:The local field potentials (LFPs) signals of the ventral hippocampus (vHPC) and medial prefrontal cortex (mPFC) were collected from six SD rats during the performance of a spatial working memory task in the Y-maze, and the time-frequency distributions of vHPC and mPFC LFPs were calculated by applying the short-time Fourier transform (STFT) to determine the characteristic frequency bands of the working memory and then to investigate the synchronized patterns of vHPC and mPFC LFPs based on the coherent of the time-varying frequency spectrum. Finally, support vector machines were applied to explore the feasibility of applying spectral coherence values to predict working memory.Results:When rats performed working memory tasks correctly, the energy of the theta band (4 - 12 Hz) of the HPC and PFC increased (all P < 0.01), and the spectral coherence value of the theta band of the HPC-PFC increased ( P < 0.05). Support vector machine training and prediction using the average peak spectral coherence and the difference between the peak and the onset when correctly and incorrectly executing the working memory as features resulted in 89% accuracy, 90% precision, 88% recall, and 88% F1 scores, all of which were statistically significant differences compared to the results of the randomly disrupted labeled data rearranging (all P < 0.05). Conclusions:Synchronized synergy in the HPC-PFC theta band is one of the potential mechanisms for correctly performing information processing in working memory.
7.Effect of dronedarone versus amiodarone on the risk of bleeding in non-valvular atrial fibrillation patients undergoing rivaroxaban anticoagulation

Objective:To compare the influence of dronedarone and amiodarone on the bleeding risk of patients with atrial fibrillation treated with rivaroxaban anticoagulation.Methods:Clinical data of 81 patients with atrial fibrillation treated with rivaroxaban anticoagulation at Xi'an International Medical Center Hospital from January 2020 to July 2023, including 36 patients treated with dronedarone and 45 patients treated with amiodarone, were retrospectively analyzed. The effects of dronedarone and amiodarone on the anticoagulation of rivaroxaban were compared using the incidence of bleeding events, thrombosis events, and adverse reactions as outcome measures.Results:The total bleeding in the dronedarone group [22.22% (8/36)] was significantly higher than that in the amiodarone group [6.67% (3/45)] ( χ2 = 4.12, P < 0.05). The total bleeding of conventional-dose rivaroxaban in the dronedarone group was 30.00% (6/20), while the total bleeding of low-dose rivaroxaban was 12.50% (2/16), with no statistical significance ( χ2 = 1.58, P > 0.05). No thrombotic events or adverse reactions to dronedarone or amiodarone were observed in all patients. Conclusion:Compared with amiodarone, dronedarone significantly increases the bleeding risk of rivaroxaban anticoagulation in patients with non-valvular atrial fibrillation, and reducing the dose of rivaroxaban in patients using dronedarone does not reduce the bleeding risk.
8.Variation of monosacchride composition of polysacchrides in Dendrobium officinale by pre-column derivatization HPLC method.

The monosacchride composition of polysacchrides in Dendrobium officinal of different germplasms, physiological ages and closely related species were determined by pre-column derivatization HPLC. The results showed that the absolute and relative volumes of all monosacchrides were significantly different between D. officinale and its closely related species, different germplasms and physiological ages of D. officinale. Absolute peak areas of mannose ranged from 0.854 x 10(7) to 10.340 x 10(7) in closely related species of D. officinale, ranged from 1.467 x 10(7) to 8.475 x 10(7) in different germplasms of D. officinale and were 4.411 x 10(7) (2.577 x 10(7)-6.516 x 10(7)), 5.528 x 10(7) (3.179 x 10(7)-8.475 x 10(7)) and 3.601 x 10(7) (1.467 x 10(7)-5.888 x 10(7)), respectively, in one to three physiological ages of D. officinale. The ratio of mannose to glucose peak areas (relative peak area) ranged from 0.976 to 16.599 in closely related species of D. officinale and from 2.679 to 7.831 in different germplasms of D. officinale. Only the relative peak areas of D. pendulum and D. primulinum were in the range of different germplasms of D. officinale in all tested samples. The results revealed the variation of monosacchride composition of polysacchrides in D. officinale. Monosacchride composition of D. officinale could be altered by breeding new varieties and controlling harvesting season. Most adulterants of D. officinale could be ruled out according to the relative peak areas of D. officinale, providing a basis for quality control and resources training of D. officinale.

9.Dahuang Fuzi decoction enhances killing effect of NK cells on Lewis lung carcino-ma cells

The aim of this study is to investigate the effect of Dahuang Fuzi decoction(DHFZT)on the killing effect of natural killer(NK)cells and the growth of lung cancer xenografts in mice.Lewis lung cancer xenograft model was constructed,and the mice were randomly divided to the normal saline group,low and high concentrations of DHFZT groups.The growth of lung cancer xenografts in mice in the control group,low and high concentration of DHFZT treatment groups was detected.The levels of IFN-y,IL-2 and IL-10 were detected by ELISA.The killing effect of NK cells was detected by calcein release assay.The release of CD107α was detected by flow cytometry.The expression of activated receptors and the tumor cell surface ligands were detected in the con-trol group and DHFZT treatment groups.The results showed that the xenografts of Lewis mice in the DHFZT treatment groups were grown slower than that in the control group in a dose depend-ent manner(P<0.05).Compared with the control groups,the levels of IL-2 and IFN-y in tumor tissues were increased in DHFZT treatment groups(P<0.05),but no significant changes in IL-10.Calcein release assay showed that the killing efficiency of NK cells in the DHFZT treatment groups was increased(P<0.05).The secretion level of IFN-y in the culture supernatant was increased by DHFZT treatment(P<0.05).DHFZT treatment increased the expression level of CD107α of NK cells.The expression of MIC A/B on the surface of A549 and H1299 cells was up-regulated by DHFZT treatment,but not the expression of inhibitory ligand HLA-ABC.The results showed that DHFZT enhanced the killing effect of NK cells on mouse lung cancer cells by up-reg-ulating the expression of MIC A/B,increasing the secretion of IFN-y and CD107α.Hence,these re-sults indicated that DHFZT suppresses the lung cancer growth potential through regulating the killing effect of NK cells.
10.Research progress on interleukin-6 in lung cancer.

As the core of cellular immunotherapy, T cells are important aspects of research and treatment of lung cancer. IL-6 is a costimulatory signal factor of T cells that is directly targeted by lung cancer stem cells. As a highly expressed cytokine in lung cancer cells, IL-6 plays an important role in variety of biological activities such as tumor occurrence, development, invasion and metastasis. This article reviews the research progress on IL-6 in lung cancer, including cancer development and progression, and the therapeutic sensitivity of lung cancer.
