3′-deoxy-3′-18F-fluorothymidine has been described recently as a new positron emission tomography (PET) tracer for imaging tumor cell proliferation. In recent years, more and more clinical research demonstrate that 18F-FLT can image tumor cell proliferation during treatment, and can reflect accelerated reproliferation earlier. it can define tumor subvolumes with high proliferative activity preceding volumetric tumor response, and can predict relapse and prognosis according to the changing status during the treatment.

With the development of image technology,functional imaging was gradually applied to contouring the gross target volume,which generated the concept of biological target volume and biological intensity-modulated radiation therapy.In this review,we introduced our work and the current status of ~(18)F-fluorodeoxyglucose imaging,amino acids imaging,nucleic acid imaging,hypoxic imaging,gene imaging and molecular imaging.The future of functional imaging and it application in biological intensity-modulated radiation therapy were discussed in prospect.
4.The value of combined determination of serum hepatic fibrosis index for evaluation on the degree of liver fibrosis in patients with chronic hepatitis B

Objective To explore the value of combined determination of serum hepatic fibrosis index for evaluation on the degree of liver fibrosis in patients with chronic hepatitis B(HBV).Methods 275 patients with HBV were selected as chronic hepatitis group,which were divided into the mild chronic hepatitis group(75 cases),chronic moderate hepatitis group (72 cases),chronic severe hepatitis group (65 cases),liver cirrhosis group (63 cases) according to the severity of disease.In addition,50 healthy population were selected as the healthy control group.The four liver fibrosis indicators included serum hyaluronic acid(HA),laminin (LN),collagen type Ⅲ (PC Ⅲ),type IV collagen(IV-C) were determined by chemiluminescence immunoassay and compared among the groups,and the diagnostic results of combined different indicators were compared.Results The indexes of HA,PC Ⅲ,IV-C,LN were significantly higher than those of the healthy control group (t =2.567,2.687,2.811,2.741,all P < 0.01).The hepatic fibrosis indexes were increased by with the severity and duration increased gradually (t =2.665,2.787,2.755,2.811,all P < 0.01).The concentration-time curves area (AUC) of PC Ⅲ,Ⅳ-C,LN,HA were 0.680,0.825,0.716,0.826,respectively.Diagnosis value of three combination indicators in HA + PC Ⅲl + Ⅳ-C was highest.Conclusion The chemiluminescence dynamic detection of four liver fibrosis indicators is a good non-invasive detection method for guiding clinical diagnosis on the degree of liver fibrosis.
5.Tiotropium and salmeterol xinafoate and fluticasone propionate powder for inhalation combined with non-invasive ventilation in respiratory failure

Objective To explore the clinical efficacy of tiotropium and salmeterol xinafoate and fluticasone propionate powder for inhalation combined with noninvasive ventilator in the treatment of respiratory failure, and to provide reference for clinical treatment.Methods 72 patients with respiratory failure were randomly divided into treatment group (n=36) and control group (n=36).The patients were randomly divided into treatment group and control group.Patients in the control group were treated with conventional noninvasive ventilators and treatment group were treated with tiotropium and salmeterol xinafoate and fluticasone propionate powder for inhalation on the basis of control group.Then compare the exercise tolerance, arterial carbon dioxide partial pressure, heart rate, pH, respiratory rate, blood oxygen pressure between two groups.Results After the treatment, condition of the patients in the two groups have different degrees of improvement.The final results showed that the average hospital stay was significantly reduced, respiratory frequency was(28 ±5) times/min in treatment group, heart rate was(98 ±16) times per minute, PaO2 also increased to(72 ±3) mmHg, PaCO2 was(57 ±17) mmHg, pH value increased to (7.42 ±0.03), compared with the control group, with statistically significant difference (P<0.05).Conclusion In patients with clinical implementation of tiotropium and seretide combined with non-invasive ventilator treatment of respiratory failure, can significantly improve the patient's condition, so that patients with respiratory function was significantly improved, so that the indicators of patients tend to normal.Its can effectively reduce PaCO2 , increased PaO2 and speed up its cure rate.

AIM: To explore the effects of ethanol on replication and expression of HBV, so as to effectively guide the anti HBV treatment in clinic. METHODS: 2.2.15 cells were cultured in vitro. Ethanol of different concentrations was added into the supernatant and 3TC was used as the positive control. And there was no drug added in the negative control group. Effect of ethanol on HBV expression and replication was examined and the expression of antiviral gene MxA was also tested. RESULTS: Ethanol promoted the expression of intracellular HBsAg, HBeAg and intracellular HBcAg. But 3TC revealed no significant effect on all of them. Southern blotting showed that ethanol at 200 mmol?L -1 enhance the replication of HBV DNA, but 3TC inhibited the replication of HBV DNA at 2 ?mol?L -1 .Ethanol at 40 mmol?L -1 promoted the expression of MxA, while 3TC at 2 ?mol?L -1 also stimulated the expression of MxA. CONCLUSION: Ethanol can induce the overexpression of HBsAg, HBeAg and HBcAg, and enhance the expression of antiviral gene MxA which may increase the activity of enzymes of synthetic metabolism. Ethanol can promote replication of HBV DNA, but the mechanism is unclear and need to be studied.
8.Preparation of recombinant firefly luciferase by a simple and rapid expression and purification method and its application in bacterial detection

A simple and rapid expression and purification method of recombinant firefly luciferase was developed for bacteria detection. A modified luciferase gene from North American firefly Photinus pyralis was cloned into pET28a expression vector and the recombinant protein was produced in Escherichia coli BL21. The recombinant luciferase, equipped with a polyhistidine affinity tag, was purified by immobilized metal ion affinity chromatography (IMAC). The approach generated an abundant expression and an efficient purification of a recombinant luciferase with final yield 1.995mg/L of cell culture. Experiments on the recombinant luciferase also showed that the relative light units (RUL) of the enzyme were 5.8×108, and the specific activity was 2.9×1010 RLU/mg. By applying adenosine triphosphate (ATP) bioluminescence to detection of the coin bacteria using the recombinant protein, the ATP content of bacteria was 9.48×10-16mol/mL, and was identical to the bacteria counts (4500CFU/mL) in order of magnitude. Taken together, our results provided a simple and efficacious method of the preparation of recombinant luciferase, which could be applied in the determination of bacteria via ATP bioluminescence.

Objective To analyze the clinical features of intrahepatic biliary cystadenoma and cystadenocarcinoma.Methods Sixteen patients from Dec.2008 to Dec.2013 were retrospectively reviewed to investigate their clinical manifestations,image features,treatment and prognosis.Results Intrahepatic biliary systadenoma and occured occurred primarily in middle-aged women with no typical clinical manifestations.Image examination is the key method in diagnosis.Surgery is the only method to cure the disease.The procedure depends on its location,and its prognosis is related to complete surgical resection.Conclusion Intrahepatic biliary cystadenoma and cystadeno-carcinoma is a rare tumor occuring mostly in middle-aged women.Image examination is the key method in diagnosis.Complete surgical resection contributes to its better prognosis.
