1.Clinical application value of combined detection of serum miRNA-210 and miRNA-21 in non small cell lung cancer

Objective To investigate the clinical value of combined detection of serum miRNA-210 and miRNA-21 in non small cell lung cancer (NSCLC).Methods Forty-eight patients with NSCLC in the Bazhong Municipal Central Hospital were selected as the research subjects.Forty patients with benign lung diseases and 40 individuals undergoing healthy physical examination were selected as the control groups.The levels of serum miRNA-210 and miRNA-21were detected by RT-qPCR.Results The levels of serum miRNA-210 and miRNA-21 in the patients with NSCLC were significantly higher than those in the patients with benign lung diseases and healthy controls(P<0.05);serum miRNA-210 was highly expressed in adenocarcinoma and T3-4 stage patients(P<0.05) and miRNA-210 was highly expressed in adenocarcinoma,T3-4 stage and smoking patients(P<0.05);the area under the ROC curve(AUC) of miRNA-210 and miRNA-21 for diagnosing NSCLC was 0.845 and 0.725 respectively,but AUC of their combination was 0.928 (P =0.021);COX multivariate regression analysis showed that serum miRNA-210 (RR:1.977,95 % CI:0.985-3.018,P =0.009) and miRNA-21 (RR:1.853,95% CI:0.921-2.776,P =0.018) had some predictive value for the prognosis in the patients with NSCLC.Conclusion MiRNA-210 and miRNA-21 are highly expressed in serum of the patients with NSCLC.Their combined detection can have a certain diagnostic efficiency for NSCLC and have a certain prompt action on the prognosis.
2.The Establishment of ELISA Method for Detecting Babesia gibsoni by Recombinant Antigen BgAMA-1

In order to improve diagnostic method to Babesia gibsoni infection,gene recombination technique was used in our study.By this technique,we constructed and expressed recombinant apical membrane antigen-1(BgAMA-1),and recombinant BgAMA-1 was used in ELISA experiment as diagnostic antigen.The results showed the recombinant expressed plasmid was triumphantly constructed and the recombinant antigen BgAMA-1 had a high specificity in ELISA test.These results demonstrate that the recombinant BgAMA-1 might be a useful diagnostic reagent for detection of antibodies to B.gibsoni in dogs.
3.Determination of Main Component and the Related Substances in Pirenoxinum Natricum Eye Drops by HPLC

OBJECTIVE:To establish a HPLC method for the determination of main component and the related substances in pirenoxinum natricum eye drops. METHODS: A Inertsil C8-3 column was used with pH2.2 perchloric acid-methanol (35∶65) as mobile phase at a flow of 1.0 mL?min-1. The detection wavelength was 230 nm and the sample size was 10 ?L. RESULTS: Pirenoxinum natricum and the related substances could be baseline separated. The calibration curve was linear in the range of 49.5~990 ng(r=0.999 9). The detection limits was 89.3 pg, and average recovery was 99.1%, RSD was 0.42%. CONCLUSIONS: This method is rapid, accurate sensitive and can be used for the quality control of pirenoxinum natricum eye drops.
4.Effects of glutamine on immune function of patients with severe craniocerebral trauma

Objective:To evaluate the effects of nutrition supplemented with glutamine on immune function and systemic inflammatory response in patients with severe craniocerebral trauma.Methods: Forty postoperative patients with severe craniocerebral trauma were randomized into experiment group(n=20) and control group(n=20).All patients received similar nitrogen and calorie for one week.Patients in experiment group were additionally supplemented with glutamine at dose of 0.50 g/(kg?d).Blood samples were obtained for measurement of IgM,IgG,IgA,CD3,CD4,CD8,CD4/CD8 before and after nutrition support the systemic inflammatory response syndrome(SIRS)was observed daily,and the serum albumin,transferin,hemoglobin and GCS were detected before and after nutrition support.Results: IgG,IgA,CD4,CD4/CD8,GCS and transferin in experiment group increased significantly after nutrition than in control group.Conclusion: Nutrition supplemented with glutamine may improve immune function,GCS and transferin in patients with severe cranial trauma.
5.Determination of Phenylhydrazine Residues in Edaravone by HPLC

OBJECTIVE: To establish an HPLC method for the determination of phenylhydrazine residues in edaravone.METHODS: The determination was performed on Diamonsil C18 column with 0.05 mol?L-1 ammonium acetate-acetonitrile(80∶20) as mobile phase at a flow of 1.0 mL?min-1.The wavelength of UV detector was set at 233 nm and the sample size was 20 ?L.RESULTS: The linear range of phenylhydrazine was 22~550 ng?mL-1(r=0.999 9);The average recovery was 98.10%(RSD=0.85%),and the detection limit was 44 pg.CONCLUSION: The analytical method is simple,reliable and sensitive,and suitable for the determination of phenylhydrazine residues in edaravone preparation.
6.Dynamic enhancement MRI and multirow-detector helical CT for detection of small hepatocellular carcinoma with receiver operating characteristic analysis

Objective The purpose of this study was to compare gadolinium-enhanced multiphase dynamic MR imaging and multirow-detector helical CT (MDCT) multiphase scanning for detection of small hepatocellular carcinoma. Methods Multirow-detector helical CT multiphase scanning and MRI with SE sequence combined with FMPSPGR sequence were performed in 37 patients with 43 small HCCs. ROC curves were established to analyze the results for each modality. Results There was no statistical difference between MDCT and MRI in the areas below the receive operating characteristic curve (Az). The detection of small HCC on MDCT showed lesser false-positive lesions than that on MRI, however the difference was not statistically different. Sensitivity of detecting small HCC on MDCT with multiphase scanning was higher than that on dynamic MRI. The sensitivity of detection for small HCC (≤1 cm) was 90.0%, 95.0% on MDCT and 70.0%, 85.0% on MRI, respectively. The positive predictive values for small HCC were higher on dynamic MDCT (97.5%, 97.6%) than on dynamic MRI (90.7%, 94.7%), respectively. Conclusion For early detection of small HCC, multiphase hepatic CT scan using MDCT is highly recommended for the patients with chronic hepatitis and cirrhosis.
7.Preliminary study of fertilizer effect on yield and quality of Fritillaria ussuriensis.

OBJECTIVETo preliminarily study the effect of fertilizer methods on the yield and quality of Fritillaria ussuriensis.
METHODThrough the investigation of the available nitrogen, phosphorus, potassium in soil fertility of F. ussuriensis planting field as well as the measurement of total nitrogen, phosphorus, potassium contents in bulbus of F. ussuriensis, the balanced fertilization plan for cultivation of F. ussuriensis was made.
RESULTThe optimal fertilizing amount was 13.0 kg x m(-2) pig dung, or 5.5 kg x m(-2) deer dung and 6.5 kg x m(-2) pig dung, or 11.0 kg x m(-2) deer dung.
CONCLUSIONEffective fertilizing may increase significantly the yield and quality of F. ussuriensis.

8.Antigrowth effects of chitosan and its derivatives on human hepatocellular carcinoma cell line SMMC7721

BACKGROUND: Different derivatives of chitosan with different molecular weights or degrees of deacetylation show different anti-tumor effects.OBJECTIVE: To study the inhibition effect of water-soluble chitosan and its derivatives, such as sulfonated chitosan, carboxymethyl chitosan and chitooligosaccharides for the growth of hepatocellular carcinoma cell line SMMC7721.DESIGN, TIME AND SETTING: Controlled experiments based on observation were carried out in Jiangxi Institute of Digestive Disease (Nanchang, Jiangxi, China) from January 2004 to December 2006.MATERIALS: Hepatoma cell line SMMC7721 was provided by Jiangxi Institute of Digestive Disease (China). 85.5% deacetylated chitooligosaccharides and 85% deacetylated water-soluble chitosan were produced by Jinan Haidebei Ocean Biological Engineering Co., Ltd (China); Carboxymethyl chitosan and 88.5% deacetylated chitosan were the products of Shanghai Qisheng Biological Products Co., Ltd (China).METHODS: Sulfonated chitosan was prepared using 88.5% deacetylated chitosan and chlorosulfonic acid-formamide, and then was detected with infrared spectroscopy in the Detection Analysis and Test Center, East China University of Science and Technology. SMMC7721 cells in the log phase were inoculated into 96-well culture plates, which were then added with water-soluble chitosan, sulfonated chitosan, carboxymethyl chitosan and chitooligosaccharides with the final concentrations of 25, 50, 100, 200, 400 and 800mg/L. This test was repeated for 3 times, while the control group was also set each time. After 72 hours of routine culture, MTT solution was added into each well and inoculated for another 4 hours. After the culture was terminated, dimethyl sulfoxid was added. The absorbance value of each well was measured at 490nm wavelength on a microplate reader. Three tests were measured to obtain the mean value. Also the inhibition rate was calculated.MAIN OUTCOME MEASURES: Growth inhibition effect of chitosan and its derivatives on the hepatoma cell line SMMC7721.RESULTS: Among the chitosan and its derivates at four kinds of concentrations, water-soluble chitosan and sulfonated chitosan could significantly inhibit the growth of SMMC7721 cells (P<0.001), and the effect was the most significant in the case of sulfonated chitosan. Treatment with water-soluble chitosan and sulfonated chitosan at the concentration of 50mg/L could inhibit the growth of SMMC7721 cells in a dose-dependent manner, and reached a peak at the concentration of 400mg/L and 800mg/L, respectively. Carboxymethyl chitosan and chitooligosaccharides showed no growth inhibition effect (P>0.05).CONCLUSION: Water-soluble chitosan and sulfonated chitosan have significant antigrowth effects on hepatoma carcinoma cells, while carboxymethyl chitosan and chitooligosaccharides are ineffective.
9.Identification of two newly discovered subgenotypes of hepatitis B virus genotype C

Objective To analyze the genotypic characteristics of hepatitis B virus (HBV)C/D recombinant of two types of newly discovered HBV genotypea found in Western China.Methods The whole genomes of 17 HBV strains isolated from Western China were amplified by polymerase chain reaction (PCR). Bioinformatic softwares were used for the analysis of full genome structure,genetic distances and recombination points.Results The heterogenicity of the HBV C/D recombinant was more than 8% compared with genotype A,B,D,E or F,but 3.8% 0A-8.O% compared with genotype C Based on phylogenetie analysis, a11 C/D recombinant strains clustered within genotype C.but were rouped into two other clusters within the genotype C independently from C1-C5 subgenotypes,which were two kinds of new HBV/C genotypea.Condusion The HBV C/D recombinant could he considered as tWO kinds of new subgenotypea of HBV genotype C which are different from subgenotype C1-C5 based on the genetic distances analysis.
