1.Effect of long non-coding RNA MALAT1 on growth and invasion of thyroid cancer cells and its mechanism

Objective To evaluate the effect of long non-coding RNA(lncRNA) MALAT1 on the growth and invasion of thyroid cancer cells and explore its mechanism.Methods The relative expression levels of MALAT1 and miR-211-5p genes were detected by real-time quantitative PCR in cancer tissues and adjacent tissues of 44 patients with thyroid cancer,and the relative expression levels of MALAT1 gene in SW1736,TC1,Hth7 and Nthy-ori 3-1 cells were detected by the same method.MTT assay was used to detect the effect of MALAT1 on the survival of thyroid cancer cells(SW1736 cells were transfected with pcDNA3.1-MALAT1,pcDNA3.1,si-MALAT1 and si-NC,respectively),using which the effect of overexpression of miR-211-5p on the survival of thyroid cancer cells induced by MALAT1(SW1736 cells were transfected with pcDNA3.1-MALAT1,miR-211-5p mimics,Scramble and pcDNA3.1-MALAT1+miR-211-5p mimics,respectively) was detected.Transwell chamber assay was used to detect the effect of MALAT1 on the invasive ability of thyroid cancer cells(SW1736 cells were transfected with si-NC,si-MALAT1, pcDNA3.1,pcDNA3.1-MALA T1, miR-211-5pmimics and miR-211-5p mimics+pcDNA-MALAT1,respectively).The regulation of miR-211-3p expression by MALAT1 was detected by real-time quantitative PCR(SW1736 cells were transfected with pcDNA3.1-MALAT1,pcDNA3.1,si-MALAT1 and si-NC,respectively).Results In 44 thyroid cancer tissue samples,compared to adjacent tissues,MALAT1 gene was highly expressed in37 samples(84.1%),while miR-211-5p was down-regulated in 39 samples(88.6%).The expression of MALA T1 was negatively correlated with that of miR-211-5p in thyroid cancer samples(r~2=0.168,P=0.005 7).Compared with Nthy-ori 3-1 cells,the relative expression level of MALAT1 gene in SW1736,TC1 and Hth7 cells increased significantly(t=10.230,5.813and 74.310,P=0.009,0.004 and <0.001,respectively).At 72 h after transfection,the growth ability of SW1736 cells in si-MALAT1 group decreased significantly(t=7.780,P=0.001),while that in pcDNA3.1-MALAT1 group increased significantly(t=-7.410,P=0.002) compared with that in si-NC group;Compared with pcDNA3.1-MALAT1 group,the growth ability of SW1736 cells in pcDNA3.1-MALAT1+miR-211-5p mimics group decreased significantly(t=-5.640,P=0.005).Compared with si-NC group,the number of invasive SW1736 cells in si-MALAT1 and miR-211-5p mimics group decreased significantly(t=-6.710 and-6.290,respectively,each P=0.003),while increased significantly in pcDNA3.1-MALAT1 group(t=5.740,P=0.005).Compared with pcDNA3.1 group,the relative expression level of miR-211-5p gene in pcDNA3.1-MALAT1 group decreased significantly(t=-7.510,P=0.002);Compared with si-NC group,miR-211-5p gene expression in si-MALAT1 group increased significantly(t=7.960,P=0.001).Conclusion Interfering the expression of lncRNA MALAT1 can effectively inhibit the proliferation and migration of thyroid cancer cells,which might be achieved by up-regulating the expression level of miR-211-5p.MALAT1 is expected to be a potential molecular target for the treatment of thyroid cancer.
3.Treatment of advanced and metastatic colorectal cancer with combined chemotherapy based on oxaliplatin,irinotecan and fluorouracil

0.05),the overall response rate was 56.0% .Symptoms(18/25) related to cancer improved and side-reactions were tolerable. Conclusions:Treatment of relapse and metastatic colorectal cancer with combined chemotherapy based on oxaliplatin,irinotecan and 5-Fu by arterial intervention and iv drip is safe and effective,with only slight side-effects. This treatment can improve the symptoms and life quality of patients.
4.CONSTRUCTION OF EUKARYOTIC EXPRESSION VECTORS WITH HOT SPOT MUTATIONS IN HBV SIGNAL PEPTIDE REGION

Molecular biological methods were used to construct the eukaryotic expression vectors of precore and core gene named EBO PreC/C. Then T1862, A1896, A1899 and A1896+A1899 variants were constructed by site mutagenesis in vitro. By PCR RFLP and sequencing ,T1862,A1896,A1899 and A1896+A1899 variants were obtained. Wild type and variants were transfected to HepG2 cells, and HBeAg was tested to observe the difference of HBeAg expression between wild type and variants. After stable expression in HepG2 cells, HBeAg was detected to be positive in cells transfected wild type and A1899 variants, and negative in cells transfected with T1862, A1896,A1896+A1899 variants. The construction of these variants will play an important pole in studying the relation of PreC/C mutations and HBV expression and replication of HBV genome.
5.YMDD mutations of HBV in HBeAg positive and HBeAg negative chronic hepatitis B

Objective To compare the occurrence of YMDD mutations in patients with HBeAg positve and HBeAg negative chronic hepatitis. Methods Two hundred and forty seven chronic hepatitis B patients who received lamivudine treatment were followed up.Liver function tests and hepatitis B virological tests including YMDD mutant analysis,HBV DNA quantitation and HBV serological marker analysis were performed regularly. Results YMDD mutants were detected in forty two patients.The cumulative rate of occurrence of YMDD mutation increased with the duration of lamivudine treatment.The mutation rate was significantly higher in pretreatment HBeAg positive patients than that in HBeAg negative patients.The ALT flare was more evident after emergence of YMDD mutants in pretreatment HBeAg positive patients than that in HBeAg negative patients.Conclusions The cumulative rate of occurrence of YMDD mutation was higher in pretreatment HBeAg positive patients than that in pretreatment HBeAg negative patients.
6.Photocytotoxic and apoptosis of hypericin-mediated on nasopharyngeal carcinoma cell line of human in vitro

AIM: To study effects of hypericin associated with light emitting diode irradiation on nasopharyngeal carcinoma cell line of human in vitro. METHODS: CNE-2 cells were exposed respectively to different concentration of hypericin,and compared with hematoporphyrin as positive control group,and incubated for 6 h in the dark, then accumulative radiated energe of yellow and red light irradiation equivalent to 5. 67 J/cm2 were given throughout 90 min,killing effect and apoptosis of CNE-2 cells were detected by MTT assay after incubation of 24 h and flow cytometry. RESULTS: MTT assay showed that cytotoxicity of hypericin and hematoporphyrin with light irradiation presented in dose-dependent way,their IC50 values were 0. 049 and 0. 650 ?g/mL,respectively. Flow cytometry showed that hypericin with light irradiation could block cell growth at S phase and G2 phase,when CNE-2 cells were exposed to hypericin at 0. 20 ?g/mL for 18,28 and 48 h,the apoptosis rate reached at 9. 97% ,72. 19% and 92. 24% . CONCLUSION: Hypericin with light irradiation could obviously inhibit the growth of CNE-2 cells and induce apoptosis.
7.Influence of problem-based learning on theoretical knowledge of Chinese students majoring in stomatology: a meta-analysis

ObjectiveTo evaluate the theoretical knowledge level of Chinese students majoring in stomatology based on the problem-based learning (PBL).MethodsThe Cochrane reviewer's handbook was followed by.Domestic published articles from 1989 -2010 were selected by computer and handwork.All these retrieved studies were controlled clinical trials related to oral theoretical education and were conducted by PBL and the traditional teaching methods.The quality of included studies was critically evaluated and the data were analyzed by the RevMan 5.0.1 software.ResultsA total of 138 articles were selected,but only 8 were included.Significant differences were found when the data were pooled(P < 0.01,I2 =80% ) and random effect model was used for meta-analysis.The results showed that comparing with the traditional teaching methods,PBL can improve the theoretical score of dental students [ MD 5.39,95% CI(2.20,8.58)],and sensitivity analysis showed that the result was stable.ConclusionPBL can improve the theoretical score of Chinese students majoring in stomatology.However,as part of the studies are not high quality literature,the above conclusion needs to be verified by more large sample-sized,rigorously designed randomized controlled clinical trials.

It undertakes the functions of early warning, emergency response and publicness maintenance, so that it values in improving the effectiveness of public health emergency management. The main paths to realize the value of public participation in the public health emergency management system are to be the gatekeepers of public health early warning and surveillance, the participants of the management and support of public health emergency prevention and control. It is a complex system to establish and improve the national public health system. Our society should pay attention to and give full play to the value of public participation.
9.Effects of protective transverse colostomy on the incidence of postoperative anastomotic leakage and stricture after laparoscopic resection for rectal cancer

Objective To investigate the value of protective transverse colostomy in decreasing postoperative anastomotic leakage and stricture after laparoscopic resection for rectal cancer.Methods The clinical data of 128 patients with rectal cancer who were admitted to the Jinhua Hospital of Zhejiang University from March 2008 to February 2012 were retrospectively analyzed.All the patients received laparoscopic anterior resection of rectal cancer after neoadjuvant chemoradiotherapy.Sixty-one patients received protective transverse colostomy (colostomy group) and 67 patients did not receive protective transverse colostomy (non-colostomy group).The incidences of postoperative anastomotic leakage and stricture,condition of the patients and the prognosis of the patients in the 2 groups were compared.Patients were followed up via out-patient examination,in-patient chemotherapy or phone call till May 2013.All data were analyzed using the t test,chi-square test or Fisher exact probability.Results The overall incidence of postoperative anastomotic leakage was 7.03% (9/128).The incidences of postoperative anastomotic leakage in the colostomy group and non-colostomy group were 6.56% (4/61) and 7.46% (5/67),with no significant difference between the 2 groups (x2 =0.000,P > 0.05).The systemic condition of 4 patients who were complicated with anastomotic leakage in the colostomy group was comparatively better,and all of them were cured by conservative treatment.The condition of 5 patients who were complicated with anastomotic leakage in the non-colostomy group was severe,and 4 of them received reoperation,with the reoperation rate of 4/5 ; the other patient received conservative treatment.There was significant difference in the reoperation rate between the 2 groups (P <0.05).The overall incidence of postoperative anastomotic stricture was 13.28% (17/128).The incidences of postoperative anastomotic stricture in the colostomy group and the non-colostomy group were 19.67% (12/61) and 7.46% (5/67),with significant difference between the 2 groups (x2=4.133,P < 0.05).The incidences of severe anastomotic stricture of the colostomy group and the non-colostomy group were 8.20 (5/61) and 0,respectively.Conclusion Protective transverse colostomy could not reduce the incidence of postoperative anastomotic leakage,but could mitigate the symptoms followed anastomotic leakage and reduce the reoperation rate.Protective transverse colostomy may improve the incidence of anastomotic stricture,therefore it should be applied cautiously.
10.The Effect of the Glutamate/Aspartate Transporter Antibody on Auditory Brainstem Response and Morphology of Hair Cells in Guinea Pig's Cochlea

Objective The purpose of this investigation was to observe the effects of the glutamate/aspartate transporter(GLAST) antibody to auditory brainstem response(ABR) and the pathologic morphology of hair cells in the guinea pig's cochlea. Methods 20 guinea pigs were randomly divided into the experimental group and control group. By perfusing the antibody to GLAST into tympanic canal in the cochlea of guinea pigs in experimental group and artificial perilymph into the guinea's cochlea in control group,the results of ABR ,basilar membrane stretched preparation and transmission electron microscope were observed. Results After antibody perfusion,the ABR was not induced from the third day to the ninth day.In control group,the ABR was nearly absent on the third day,but could be evolved,on the sixth day and the ninth day in all guinea pigs with the average threshold of 62.50?5.25 dB SPL and 47.50?6.18 dB SPL,respectively.
