1.Study on the distribution of auto-antibodies against platelet in adults and children with primary immune thrombocytopenia in Nanning

Objective Study on the distribution of auto‐antibodies against platelet in adults and children with primary immune thrombocytopenia(ITP) .Methods Platelet auto‐antibodies were detected in 83 ITP patients and 58 non‐ITP patients by enzyme linked immunosorbent assay (PAKAUTO kit) .Anti‐GP Ⅱ b/Ⅲ a ,GP Ⅰ b/Ⅸ and GP Ⅰ a/Ⅱ a auto‐antibodies were analyzed for 46 a‐dults and 37 children with ITP .Results The positive rate of autoantibody was 66 .27% in ITP patients ,which was much higher than that of non‐ITP patients ,the difference was significant(P < 0 .05) .As for ITP patients ,female ITP patients in adult group were 28 ,while in children group were 24 ,the difference was not significant(P > 0 .05) ,but the morbidity of ITP in females was higher than that of males in both of the two groups ,the difference was significant(P< 0 .05) .There was no significant difference on the distribution of platelet auto‐antibodies in adult and children group(P> 0 .05) ,the main of them was resistance against GP Ⅱ b/Ⅲ a and GP Ⅰ b/Ⅸ antibodies .Conclusion The detection of platelet auto‐antibodies could distinguish ITP and non‐ITP patients ,and GP Ⅰ a/Ⅱ a has an important reference value about therapy .
2.Dynamical effect of fractures combined with brain injury on the bone healing and bone metabolism

BACKGROUND:Peri-fracture nerve injury can inhibit osteoclast activity and promote early fracture healing. OBJECTIVE:To investigate dynamical y the effects of traumatic brain injury on the bone mineral density, microstructure, biomechanics property and bone metabolism in rat models of fractures. METHODS:Sixty-three male rats were randomly divided into three groups:sham group, simple fracture group and fracture combined with brain injury group. After 3, 6, and 3 months, the animals were sacrificed in batches under anesthesia, and then, the bones and serum specimens were used to detect the bone mineral density, microstructure, biomechanics property, serum cross-linked N-telopeptide of col agen type I and osteocalcin levels. RESULTS AND CONCLUSION:Compared with the simple fracture group, the fracture combined brain injury group had significantly increased bone mineral density of the proximal tibia, bone volume fraction of the cancel ous bone, trabecular thickness, cross-sectional area of tibial cortical bone and total area of the bone marrow, ultimate load and stress of the tibia, serum cross-linked N-telopeptide of col agen type I and osteocalcin levels at 3 and 6 weeks after modeling (P<0.05), but no differences in the above-mentioned indexes were found among the three groups at 3 months after modeling. These findings indicate that traumatic brain injury can increase the bone mineral density at the fracture site, improve bone microstructure and enhance biomechanical properties, thereby promoting bone healing and bone metabolism at the fracture site.
3.The characteristic of gene polymorphism of Kell blood group in Zhuang population from Guangxi region in China

Objective To investigate gene polymorphism of Kell blood group in different Zhuang population from Guangxi region .Methods The genotypes of Kell blood group of 1025 non‐related individuals in different areas of Guangxi Zhuang popula‐tion were analyzed by PCR‐SSP .Results The Kell antigen in all individuals was homozygous ,the gene frequency of K and Jsa was 0 ,while that of k and Jsb was 1 .000 .Conclusion The distribution characteristic of Kell blood group in Guangxi Zhuang population was monomorphism ,which was similar to other Chinese population reported by literatures .
4.Effect of Acupuncture and Moxibustion on the Quality of Life in Patients with Osteoporotic Vertebral Body Compression ;Fractures

Objective To investigate the effect of meridian-warming and unblocking acupuncture and moxibustion on the quality of life in patients with osteoporotic vertebral body compression fractures. Method Two hundred and eighty patients meeting the criteria of osteoporotic vertebral body compression fractures were enrolled and randomly allocated to treatment and control groups. The treatment group received meridian-warming and unblocking acupuncture and moxibustion and the control group, complex treatment of Western medicine. The Visual Analogue Scale (0-10 marks) score and the SF-36 score were recorded before and after treatment. The clinical therapeutic effects were evaluated. Result The pain score was decreased, the SF-36 score was increased and the clinical therapeutic effect was improved more in the treatment group than in the control group (P<0.05). Conclusion Meridian-warming and unblocking acupuncture and moxibustion helps to relieve the pain and improve the quality of life in osteoporosis patients.
5.Determination of six catechins in Xinnaojian Capsules (Tablets) by QAMS

AIM To establish a quantitative analysis of multi-components by single marker(QAMS) method for the content determination of six catechins in Xinnaojian Capsules (Tablets) (tea extract).METHODS The analysis of 50% methanol extract of this drug was performed on a 35 ℃ thermostatic Shimadzu Wonda Cract ODS-2 column (4.6 mm ×250 mm,5 μm),with the mobile phase comprising of 0.5% acetic acid (A)-acetonitrile (B) flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 280 nm.With epigallocatechin gallate as an internal standard,the relative correction factors of epigallocatechin,catechin,epicatechin,gallocatechin gallate and epicatechin gallate were calculated,from which the content determination was made.RESULTS Six constituents showed good linear relationships within their own ranges (r ≥ 0.999 8),whose average recoveries were 96.00%-98.47% with the RSDs of 2.09%-2.91%.The results obtained by QAMS method approximated those obtained by external standard method.CONCLUSION This simple and reliable method can be used for the quality control of Xinnaojian Capsules (Tablets).
6.The role of microRNA-219-2-3p in Gastric Cancer and the underlying mechanism

Objective To evaluate the role of miR219‐2‐3p in gastric cancer and the underlying mechanisms .Methods Real time RT‐PCR was employed to quantify the expression level of miR219‐2‐3p in matched tissues of 82 patients .In vitro cell proliferation , the expression of tumorgenesis related protein ERK1/2 was performed by overexpressing miR‐219‐2‐3p in gastric cancer cell line MGC803 .Results The expression level of miR‐219‐2‐3p significantly decreased in late stage gastric cancer (P< 0 .05) .The cell proliferation rate was significantly reduced in gastric cancer cell lines overexpressing miR‐219‐2‐3p .Furthermore ,the expression level of p‐ERK that activated form of ERK was significantly decreased whereas the total level of ERK was unchanged after overex‐pressing of miR‐219‐2‐3p in MGC803 cell lines .The expression level of p‐ERK was also increased in gastric cancer tissues .Conclu‐sion miR‐219‐2‐3p may acts as a tumor suppressor in gastric cancer by decreasing the activity of ERK 1/2 signaling pathway .
7.Investigation and analysis on ABO and Rh blood groups distribution among Zhuang polulation in Nanning area

Objective To investigate the distribution situation of ABO and Rh blood groups among Zhuang population in Nanning area to guide the scientific voluntary blood donor recruitment and rational storage of blood in this area.Methods 2 052 blood samples from Zhuang population in Nanning area were performed the identification of ABO and Rh blood groups.Whether the gene distributions conforming to the Hardy-Weinberg equilibrium principle was assessed by using the Chi-square test.The data was compared with the distribution characteristics in other areas or other minorities.Results The distribution characteristics of ABO blood groups in Nanning area were O > B> A > AB.Blood group O was highest (46.78%)and blood group AB was lowest (4.34%).The gene frequencies of A,B and O were 0.131 9,0.181 5 and 0.686 6,respectively.The blood groups distribution con-formed to the Hardy-Weinberg equilibrium principle;the RhD positive proportion(RhD+ )in Zhuang population was 99.90%,while the RhD negative (RhD- )proportion was 0.10%,which was lower than that in Han population.No phenotypes of ccdE,CcdE, CCdee,CCdE,CCDEE and ccDee were observed in this study.Conclusion The distribution of the ABO and Rh blood groups among Zhuang population in Nanning still maintains their own exclusive characteristics.The extremely low proportion of RhD- will bring about serious difficulties to the patients with long term blood transfusion.
8.Effects of weightlessness on the apoptosis of hepatocytes in rats

Objective To investigate the effects of weightlessness on the apoptosis of hepatocytes in rats.Methods Eighty-four adult male Wistar rats were randomly divided into weightlessness group and control group according to the random number table.Tail-suspension was used to simulate the weightlessness model.Six rats were sacrificed in each group at the end of day 1-7.Hepatocyte apoptosis and p53 expression were detected by TUNEL assay and immunohistochemistry PV-6001,respectively.All data were analyzed by variance analysis.Results An increase of apoptosis index and up-regulated expression of p53 were detected in weightlessness group.The level of apoptotic index Was significantly higher in weightlessness group than in control group from day 1 to day 5(F=77.608,P<0.05),and the apoptotic indexes of the 2 groups were close at day 6 and day 7.The p53 proteins with positive expression were mainly detected in the nucleus.The level of positive expression of p53 in liver tissue in weightlessness group was higher than that in control group at the early stage(F=113.063,P<0.05).Few hepatocytes with positive expression were detected in control group and weightlessness group at a later stage.Conclusion There is a close relationship between hepatocyte apoptosis,changes of P53 expression and stress response and tolerance to weightlessness.
9.Contents Determination of Catechins Active Components in Lipid-lowering Slimming Health Products by Quantitative Analysis of Multi-Components via Single Marker

OBJECTIVE:To establish the method for contents determination of catechins active components in lipid-lowering slimming health products. METHODS:HPLC method was adopted. Using epigallocatechin gallate(EGCG)as a reference,relative correction factor(RCF)of EGCG to gallocatechin(EGC),catechin(C),epicatechin(EC),gallocatechin gallate(GCG)and gal-loylepicatechin(ECG)were calculated. The contents of EGC,C,EC,GCG and ECG in 5 batches of samples were calculated through RCF. The contents of EGC,C,EC,GCG and ECG determined by external standard method were used as measured value. The similarity of the value determined by external standard method with the value calculated by quantitative analysis of multi-com-ponents via single marker method(QAMS)was evaluated with vector included angle cosine method. RESULTS:The linear ranges of EGCG,EGC,C,EC,GCG and ECG were 0.0065-0.1305 mg/mL(r=0.9998)、0.0005-0.0107 mg/mL(r=0.9997)、0.0020-0.0400 mg/mL(r=0.9999)、0.0153-0.3053 mg/mL(r=0.9998)、0.0008-0.0155 mg/mL(r=0.9998)、0.0040-0.0792 mg/mL (r=0.9999);RSDs of precision,stability and reproducibility tests were all lower than 2.0%.The recoveries were 95.07%-100.35%(RSD=1.94%,n=6)、95.24%-101.87%(RSD=2.79%,n=6)、96.08%-103.86%(RSD=3.01%,n=6)、97.51%-101.06%(RSD=1.45%,n=6)、96.01%-101.66%(RSD=2.27%,n=6)、96.20%-102.89%(RSD=2.71%,n=6),respectively. There was no signifi-cant difference between measured value and calculated value. CONCLUSIONS:The method is simple,precise,stable and repro-ducible,and can be used for contents determination of catechins active components in lipid-lowering slimming health products.
10.Establishment of platelet antigen panel cells and application in platelet alloantibody detection

Objective To establish the platelet antigen panel cells and to apply them in the detection and identification of platelet alloantibody.Methods Human platelet antigen(HPA)1-16 genotyping from 1 500 un-related blood donors in Nanning area were performed by the polymerase chain reaction-sequence specific primers(PCR-SSP)technique,platelet antigen cells with O blood type were chosen to establish the panel cells of platelet antigen.The phenotype of the panel cells were verified by the reference sera from the 14th platelet immunology workshop of the International Society of Blood Transfusion(ISBT).And then these panel cells were used in clinic to detect the platelet alloantibody and the samples from the 14th and 15th platelet immunology workshop of ISBT.Re-sults Six platelet cells with consistent phenotype and genotypes and covering the HPA 1-5 and 1 5 systems were selected to estab-lish the platelet panel cells and successfully applied them in the clinical and scientific sample detection and identification.Conclusion Platelet antigen panel cells are established successfully,which provides the experimental basis for the diagnosis and research of platelet allogenic abnormal immunity diseases.
