Objective To explore the interventional effect of two different methods in the prevention of chemotherapy phlebitis caused by 5-fluorouracil.Methods Eighty cases of patients treated with cisplatin+5-fluorouracil (PF) chemotherapy in the First Affiliated Hospital of Guangxi Medical University from May 2015 to July 2016 were selected and divided into the observation group and control group according to admission time,40 cases in each group. The observation group was smeared with 2% anisodamine along the direction of vein puncture after peripheral venous puncture success, while the control group was smeared with mucopolysaccharide polysulphate emulsion paste. The occurrence of phlebitis and degree of local pain after intervention were compared between the two groups. The blood biochemical indexes and inflammatory factors levels before and after intervention were compared too. Results After the intervention, the distribution of phlebitis level and local pain degree had statistical difference between the two groups (Z=2.57,2.53;P=0.01);the observation group was superior to the control group in the aspects of phlebitis lesion degree and local pain alleviation;WBC,MPV and PDW in the observation group had statistical difference between before and after intervention;WBC,PLT and PCT in the control group had statistical difference between before and after intervention (P<0.05);PLT count was positively correlated with PCT in the two groups (r=0.952,P<0.05);the CRP level and WBC count after intervention had no statistical difference between the two groups(P>0.05).Conclusion 2% anisodamine can effectively prevent and alleviate the occurrence of 5-FU caused chemotherapy phlebitis and local pain.At the same time,PLT can provide a basis for preventing chemotherapy phlebitis.

Objective To investigate the effect of prophylaxis against respiratory and circulation depression caused by propofol during painless colonoscopy by intravenous doxpram with small dose. Methods 80 patients undergoing painless colonoscopy were randomly divided into the control group(A group) and the doxpram group(B group). All patients used propofol, and then used propofol after induction. In group B,doxpram was injected before anesthesia. The degree and incidence rate of respiratory and circulotion depression, the dose of propofol, the awake time, the examination time, the complication during anesthesia and awake were monitored. Results The SBP,DBP,SpO2,RR during anesthesia were obviously lower in group A than in group B(P

Objective To investigate the effect and clinical valve of target-controlled infusion of remifentanil for conscious intubation of patients under fibrobronchoscopy. Methods Forty patients were randomly divided into two groups(F and R),each group contained 20 cases. All of the patients were intubated on regional anaesthesia,as the first step, then the patients in F group were given fentanyl 2?g/kg by vein.In remifentanil target-controlled infusion group(R group),the patients were kept infusing remifentanil 2ng/ml.Results (1)Haemodynamics:in R group,SBP and HR kept steadily; In F group, while the two indices increased significantly.(2)Static pulmonary function:in R group,RR,VT deceased remarkablely. There were no difference between two groups in SpO_2 and P_ ET CO_2.(3)Tracheal intubating condition and Ramsay score in R group were better than in F group;Cardiovascular response in R group was lower. Conclusion Target-controlled infusion of remifentanil for conscious intubation can achieve excellent result, with less cardiovascular response.

Objective To conduct eugenics inspections for preparing pregnant couples,to find out risk factors causing fetal birth defects,to provide the scientific basis of eugenic and superior nurture. Methods A total of 2 1 63 preparing pregnant couples were conducted eugenics detection before childbirth to assess pregnant risk. Results A total of 1 547 cases were checked out with preg-nancy risk with the positive rate as 35.76%(1 547/4 326).Among them, the positive rate of male was 26.81% (580/2 1 63).the positive rate of women was 44.71% (967/2 1 63).Women positive rate was significantly higher than the male. Conclusion The eu-genics detection before pregnant could found risk of pregnancy as soon as possible,in order to take corresponding preventive meas-ures in time, has important significance for prevention of birth defects.

Objective To analyze the death status and main causes of death among children under 5 years old in Changsha from 2016 to 2021, and to provide a scientific basis for formulating preventive measures for children's health care. Methods The data of 1 761 deaths of children under 5 years old in Changsha City from 2016 to 2021 were collected, and the mortality trend, the order of causes of death and the utilization of pre-death medical care services were retrospectively analyzed. Results The 7-day neonatal mortality, 28-day neonatal mortality, 0-1-year-old neonatal mortality, and the mortality rate of children under 5 years old (U5MR) in Changsha City from 2016 to 2021 were 0.76‰, 1.28‰, 2.41‰, and 3.86‰, respectively. All the mortality rates showed a decreasing trend (P<0.05). U5MR in males was significantly higher than that in females (P<0.05), and U5MR in rural areas was significantly higher than that in urban areas (P<0.05). The top five causes of U5MR were drowning, premature delivery or low birth weight, pneumonia, other congenital anomalies, and accidental asphyxia, respectively. The death places of children under 5 years old were mainly medical and health institutions, and 81.72% of them were treated in hospitals before death. Conclusion From 2016 to 2021, the mortality rate of children under the age of 5 in Changsha City has gradually decreased. Preventing congenital malformations, reducing preterm birth or low birth weight, improving the treatment level of pneumonia, and preventing accidents such as drowning and accidental suffocation are the key to reducing the mortality rate of children under 5 years old.

Conjunctival melanoma (CM) is a rare and fatal malignant eye tumor. In this study, we deciphered a novel anti-CM mechanism of a natural tetracyclic compound named as cucurbitacin B (CuB). We found that CuB remarkably inhibited the proliferation of CM cells including CM-AS16, CRMM1, CRMM2 and CM2005.1, without toxicity to normal cells. CuB can also induce CM cells G2/M cell cycle arrest. RNA-seq screening identified KIF20A, a key downstream effector of FOXM1 pathway, was abolished by CuB treatment. Further target identification by activity-based protein profiling chemoproteomic approach revealed that GRP78 is a potential target of CuB. Several lines of evidence demonstrated that CuB interacted with GRP78 and bound with a K d value of 0.11 μmol/L. Furthermore, ATPase activity evaluation showed that CuB suppressed GRP78 both in human recombinant GRP78 protein and cellular lysates. Knockdown of the GRP78 gene significantly induced the downregulation of FOXM1 and related pathway proteins including KIF20A, underlying an interesting therapeutic perspective. Finally, CuB significantly inhibited tumor progression in NCG mice without causing obvious side effects in vivo. Taken together, our current work proved that GRP78-FOXM1-KIF20A as a promising pathway for CM therapy, and the traditional medicine CuB as a candidate drug to hinder this pathway.
