1.The clinical effects of methylenetetrahydrofolate reductase C677T gene mutation on mild vascular cognitive impairment in patients with cerebral infarction

Objective To study the clinical effects of methylenetetrahydrofolate reductase(MTHFR) C677T gene mutation on mild vascular cognitive impairment(VCI)in patients with cerebral infarction.Methods The prospective study was conducted in 180 patients with atherosclerotic acute cerebral infarction hospitalized from June 2015 to May 2016.180 patients were divided into normal homocysteine(Hcy)group (Hcy 5.46~16.20 μmol / L,n=130)and high Hcy group(Hcy>16.20 μmol / L,n=50).MTHFR C677T genotypes were detected by polymerase chain reaction-restriction endonuclease fragment length polymorphism.The VCI state and risk factors for VCI were analyzed with Montreal Cognitive Assessment scale(MoCA)and Mini Mental State Examination(MMSE)score in patients with acute cerebral infarction on admission and 14 days,30 days and 90 days after admission.Results MMSE and MoCA scores at 14 days,30 days,90 days after admission were lower in high Hcy group than in normal Hcy group(P<0.05 or P<0.01).MMSE and MoCA scores were lower in T/T genotype than in C/C genotype(P<0.05)at 14 day after admission,and lower in C/T genotype than in C/C genotype at 30 day and 90 day follow up(P<0.05or P<0.01).Logistic regression analysis showed that Hcy may be independent risk factors for mild VCI(OR =1.274,95%CI:1.027~1.264,P=0.018).Conclusions MTHFR C677T gene mutations may cause hyper-homocysteinemia,and promote the emergence of VCI in patients with cerebral infarction.MTHFR C677T gene mutations may be a genetic susceptibility factor for VCL
2.Study on concentration of docosahexenoic acid in glyceride by hydrolysis of fish oil withlipases

In this paper,the method about hydrolysis of fish oil with lipases for concentrating DHA(docosahexenoic acid) in glycerides was studied.Six lipases were screened with comparing the activities of hydrolysis of fish oil.Candida lipalytica Lipase is resistant to generating DHA in fish oil.The Effects of the quantity of lipases,the emulgent,hydrolysis time,temperature in the process,the ratio of oil to water were studied.The optimum conditions for the hydrolysis of fish oil are 12 hours of hydrolysis under 45℃,with 300 u?g -1 .The emulgent was and the ratio of oil to water was 0.4 mL?g -1 .Under these conditions,the content of DHA in glyceride were concentrated from 18.7% to 34.0%.But the EPA content remained close to that of fish oil.The DHA glyceride containing triglyceride,diglyceride and monoglyceride was identified by IR spectrum combining with separation of HPLC.
3.Preparation and Quality Control of Loxoprofen Sodium Patches

Objective:To study the preparation and quality control of loxoprofen sodium patches and develop an HPLC method for the determination of loxoprofen sodium. Methods:Loxoprofen sodium patches were prepared with CMC-Na and PVP as the adjuvants, and an HPLC method was used to determine the content of loxoprofen sodium. Results:The linear range of loxoprofen sodium was 4-24μg·ml-1(r=0. 999 7), the average recovery was 99. 99%(RSD=0. 99%, n=6). Conclusion:The preparation method is reason-able, simple and stable. The developed HPLC method is specific to determine the content of loxoprofen.

Kinesin spindle protein ( KSP) inhibitors is an important direction for the discovery of anticancer agents. Several KSP in-hibitors have been studied in clinic trials. The discovery of ATP-competitive KSP inhibitors may be a new approach for searching novel a-gents to overcome the mutation-mediated resistance to the allosteric inhibitors. The progress in the discovery of ATP-competitive KSP in-hibitors was reviewed in the paper to provide reference for the further development of KSP inhibitors.
5.Study on antarctic yeast Rhodotorula sp.NJ-298 producing astaxanthin

A yeast strain Rhodotorula sp.NJ-298,which was isolated from antarctic sea-ice,could produce carotenoids pigment,and the total production was 1986?g?g~(-1).UV-visible spectrum and LC/MS analysis indicated astaxanthin was the major component of the carotenoids,and HPLC analysis showed its content accounted for 81.9 % of total carotenoids productoin.Maximum astaxanthin production 6506 ?g?L~(-1) was obtained when the yeast was grown at steady-state phase.Optimum temperature for the strain grow was at 8℃,and sodium acetate and peptone were the best carbon and nitrogen sources for the astaxanthin production.Moreover,the pigment had strong activity of scavenging free radicals,and the IC_(50) was 0.40 ?g?mL~(-1) by DPPH assay.
6.Effects of Jiawei Buyang Huanwu Decoction on vascular stenosis and oxidative stress after balloon injury of rabbit iliac artery

BACKGROUND:Modern pharmacological studies have confirmed that Buyang Huanwu Decoction could expand blood vessels,improve microcirculation,decrease oxidative stress,Inflammation and platelet activation,so protect and improve the function of endothelial cells.But its certain mechanism is still unclear,especially for its impact on the restenosis of the coronary artery after percutaneous transluminal coronary angioplasty(PTCA).OBJECTIVE:To investigate the protective effects of Jiawei Buyang Huanwu Decoction on vascular stenosis and oxidative stress after balloon injury of rabbit iliac artery.METHODS:New Zealand rabbits,were randomized and divided into three groups,control group,model group and drug group respectively.Rabbits of control group were fed with common forage,but model group and drug group fed with high fat diet.Two weeks later,the iliac arteries were injured by balloon for model group and drug group.Meanwhile drug group were fed with Jiawei Buyang Huanwu Decoction after the operation,2 mL/kg per day.Control group underwent sham operation control.At the end of4 weeks,serum samples were stored to assay the levels of cholesterol,activity of superoxide dismutase(SOD)and levels of malondialdehyde(MDA).Injured iliac artery was fixed by neural formalin to observe the endothelial hyperplasia by light microscope,and the results were analyzed by picture analysis system.Differences of measurement were compared with one-way analysis of variance as well as repetitive measurements analysis of variance.RESULTS AND CONCLUSION:Iliac artery intima was thin,with Intact structure,without arteriosclerosis In the control group.The vascular lumina were narrower,intima was thicker and there were more enormous arteriosclerosis plaques in model group rabbits.The plaque thickness was reduced and stenosis was mild in drug group.TotaI cholesterol,triglyceridemia,low-density lipoprotein of cholesterol(LDL-C)and MDA levels were significantly lower in the drug group compared with the model group.whereas high-density lipoprotein of cholesterol(HDL-C)and serum SOD levels were significantly greater than the model group(P<0.05).The results showed that Jiawei Buyang Huanwu Decoction has significant preventive effect on intimal hyperplasia and the development of artherosclerosis in rabbits with iliac artery injury,and the mechanism of which may be related to modifying lipid metabolism and cleaning oxygen free radical anti-oxidative stress.
7.One-stage treatment of Morel-Lavallée injury complicated with pelvic fracture by lacunar closure-assisted internal fixation

Objective:To investigate the clinical effects of lacunar closure-assisted internal fixation in the one-stage treatment of Morel-Lavallée injury complicated with pelvic fracture.Methods:The 32 patients were retrospectively analyzed who had been treated for Morel-Lavallée injury complicated with pelvic fracture at Department of Orthopedic Trauma, Honghui Hospital Affiliated to Xi'an Jiaotong University from May 2018 to November 2020. They were 21 males and 11 females, aged from 18 to 58 years (average, 40.5 years). The injury was located at a unilateral hip in 20 cases, at bilateral hips in 6 cases, at low back in 4 cases, and at posterior thigh in 2 cases. The pelvic fractures were treated by open reduction and internal fixation while the Morel-Lavallée injuries by lacunar closure at the same time. Their wound healing, pelvic function and complications were observed regularly.Results:The hospital stay of 32 patients ranged from 14 to 28 days, averaging 19.2 days. The patients were followed up for 6 to 18 months (mean, 9.3 months). The area of Morel-Lavallée injury healed after one lacunar closure in 23 patients who obtained fine skin survival and no soft tissue necrosis or other complications; the wounds in the Morel-Lavallée injury area healed well after secondary lacunae closure in 4 patients. Superficial sensation of the skin decreased around the Morel-Lavallée injury area after wound healing in 3 patients. Wound fat liquefaction was found in the Morel-Lavallée injury area but responded to symptomatic treatment in one patient. A small amount of wound secretion found in the Morel-Lavallée injury area was cured also by symptomatic treatment in another patient whose bacterial culture was negative. All the fractures healed after 3 to 6 months (average, 3.9 months). At the last follow-up, the Majeed scores for the pelvic function ranged from 65 to 100 points, averaging 84.5 points.Conclusion:For Morel-Lavallée injury complicated with pelvic fracture, lacunar closure-assisted internal fixation can result in a satisfactory one-stage treatment.
8.Interpretation of laboratory test results in early and late onset severe preeclampsia

Objective To analyze the difference of laboratory test results between early-onset and late-onset severe preeclampsia and to investigate their clinical application values.Methods Totally 108 blood samples were collected from patients with severe preeclampsia who were diagnosed according to the Diagnostic Standard of Obstetrics and Gynecology(7th Edition) published by People′s Medical Publishing House,in Shandong Provincial Hospital affiliated to Shandong University from March to November 2016,which consisted of 64 early-onset severe preeclampsia before 34 weeks gestation(early onset group) and 44 late-onset severe preeclampsia after 34 weeks gestation(late onset group).In addition,42 women with normal pregnancies as the control group were selected.General clinical data were collected,and the blood sample was analyzed through detecting Hb,PLT,fibrinogen (FIB),D-dimer,AST,ALT,urea,creatinine (Cr),uric acid,CRP,urine protein.The tested results were analyzed and compared.Flow cytometry was used to analyze the proportion of T helper 1 cells(Th1) and T helper 2 cells(Th2),and the ratio of Th1/Th2 was also calculated.All data and F test were performed by use of statistical software SPSS19.0.Results The pre-pregnancy body mass index(29.55±4.49,30.66±5.13,26.62±3.17,F=9.829,P<0.05),diastolic blood pressure[(105.17±14.46)mmHg(1 mmHg=0.133 kPa),(99.80±12.56)mmHg,(74.36±8.42)mmHg,F=82.088,P<0.05],Hb[(123.22±14.38)g/L,(117.03±16.48)g/L,(112.62±11.24)g/L,F=7.133,P<0.05],urea[(6.56±2.36)mmol/L,(4.51±1.35)mmol/L,(3.04±0.87)mmol/L,F=51.733,P<0.05],Cr[(68.47±18.05)μmol/L,(61.37±14.37)μmol/L,(48.54±8.73)μmol/L,F=23.737,P<0.05],CRP[(7.68±8.76)mg/L,(5.88±6.03)mg/L,(3.56±2.41)mg/L,F=4.735,P<0.05],urine protein[(3.66±0.76)g/L,(2.20±1.05)g/L,(0.19±0.40)g/L,F=249.714,P<0.05]had a statistically significant difference among the early-onset,late-onset and control groups.The flow cytometry results demonstrated that the proportion of Th1 in early-onset group(19.83±3.04)was higher than that in both late-onset (14.49±2.79)and control groups(11.78±1.17),on the contrary,the result of Th2 was much lower(early-onset:1.02±0.12,late-onset: 1.11±0.12,control: 1.56±0.11),there was statistical significance among these three groups(Th1: F=135.110,P<0.05;Th2: F=293.687,P<0.05).Conclusions It′s necessary to real-time monitor the laboratory indicators,such as liver and kidney function,especially the immunologic function indicators for evaluating the disease of early-onset and late-onset severe preeclampsia and personal treatment,and for ensuring the health of mother and fetus and improving the prognostic of mother and fetus.
9.Ultrasonic radio frequency analysis technique for noninvasive evaluation of carotid artery intima-media thickness and elasticity in patients with dyslipidemia.

OBJECTIVETo assess the value of ultrasonic radio frequency analysis technique for noninvasively evaluation of carotid artery intima-media thickness (IMT) and elasticity in patients with dyslipidemia.
METHODSRadio frequency quality intima-media thickness (RFQIMT) technique and radio frequency quality arterial stiffness ((RF)QAS) technique were used for measurement of IMT and pulse wave velocity (PWV) of the left carotid artery in 86 patients with dyslipidemia and 60 healthy volunteers.
RESULTSApart from the patients' age, body mass index, systolic blood pressure, diastolic blood pressure, pulse pressure, total cholesterol, triglyceride, low density lipoprotein, high density lipoprotein, fasting blood glucose, IMT and PWV differed significantly between the control and case groups. IMT was positively correlated with PWV (r=0.521, P<0.05). In patients with IMT<1.0 mm (n=81), PWV differed significantly between patients with different number of other atherogenic risk factors (P=0.004), but IMT showed no such variation (P=0.079). In patients with more than 3 and those with 2 other risk factors, PWV increased significantly as compared with those with one risk factor (P=0.002). Multiple linear regression analysis showed that systolic blood pressure, age and fasting blood glucose were independently associated with PWV.
CONCLUSIONUltrasonic radio frequency analysis technique allows accurate measurement of IMT and arterial elasticity, and may thus become a valuable method for evaluating early structural and arterial functional change of the carotid artery.

10.Determination of tetrahydropulmatine content in treatment of winter's diseases in summer path by HPLC.

OBJECTIVETo establish a high performance liquid chromatography method for the determination of tetrahydropulmatine content in treatment of winter's diseases in summer path.
METHODThe analysis was performed on the Hypersil ODS2 column (4.6 mm x 250 mm, 5 microm) with methanol and 0.1% phosphoric acid solution (adjusted pH to 6.28 with triethylamine) as mobile phase at 63:37. The flow rate was 1.0 mL x min(-1) and the UV detection wavelength was 280 nm.
RESULTThe linear range of tetrahydropulmatine was obtained from the range of 2.08 - 49.92 mg x L(-1) (r = 0.999 8). The average recovery was 97.85% with RSD 2.49%.
CONCLUSIONThis method is simple, reliable and can be used for the determination of tetrahydropulmatine in this preparation.
