Object To study the effective portion in Cornus officinalis Sieb. et Zucc. extract (COE) for the treatment of arrhythmia. Methods Effect of COE on chloroform induced ventricular fibrillation in mice and electrophysiology of isolated guinea pig papillary muscle were studied. Results Antiarrhythmic effect of COE may be related to its prolongation of action potential duration, increase of the absolute value of resting potential and a decrease of autonomy of sinus node. The effective portion in COE may be its total organic acid and a certain yet unknown trace substance, whereas its total glycosides were devoid of such activities. Conclusion Pharmacodynamic and myocardial electrophysiologic studies showed that the total organic acid and a certain unknown trace substance possessed the obvious antiarrhythmic activity.

Mutation of ras gene family was studied in 206 cases of gastric cancer and precancerous lesions by PCR/RFLP, PCR/SSCP and DNA suquencmg. The results showed: the mutation rates of H-ras 12 codon in metaplasia, atypical dysplasia, early cancer and advanced cancer were 16.7%(6/36), 31.2%(15/48), 50.0% (5/10), 32.2% (20/60), respectively. In the groups of superficial gastritis and normal controls, no mutations were found. Mutations of H-ras 61 codon and N-ras 12 codon in various groups were the same as that of normal controls. Only 2 cases of K-ras 12 codon mutation were detected in gastric cancer group by PCR-SSCP, but it was not identified by DNA sequencing. It might be due to polymorphism. Objective To explore the value of endoscopic retrograde cholangiopancreatography (ERCP)in the treatment of chronic pancre-atitis (CP).Methods Thirty patients with CP who underwent ERCP from January 2008 to December 2012 were retrospectively analyzed. Serum amylase levels were determined before and at 24 hours after surgery.The reduction in abdominal pain was dynamically evaluated be-fore and after treatment.A follow -up of patients with steatorrhea and anxiety was performed.Regular reexamination using abdominal ima-ging or ERCP was performed to identify pancreatic lesions and conditions of pancreatic duct stents.Results All patients were successfully treated by ERCP and had pancreatic duct stents placed.The remission rate of abdominal pain reached 83.3% at 72 hours after surgery.One patient was assigned to surgery due to insignificant improvement in symptoms,and the case -fatality rate after surgery was 0.Conclusion ERCP is an effective method in the treatment of CP,which has the advantages of being safe,effective,and minimally invasive.

p53 gene mutation (exon4 , 5 , 6 . 7 . 8 and intron6) in gastric cancer and precancerous le- sions and p53 gene (exon4 and intron6) .APC gene deletion in gastric carcinomas were studied by PCR/ SSCP and PCR/RFLP. Results showed: mutation rate of p53 in intestinal metaplasia ,dysplasia and gas- tric carcinoma was 37. 5% (3/8) ,42. 1 % (8/19) , 53. 3% (16/30) ,respectively. There was significant dif- ference between groups of metaplasia、dysplasia、cancer and that of normal control. We found there were no exon8 mutation in metaplasia and dysplasia , but 4 cases in cancer group. lt is suggestted that exon8 mutation occurs at the late stage of gastric cancer , but exon 5 , 6 , 7 mutation occur in the course of pre- cancerous lesions to cancer. Loss of heterozygosity (LOH) of exon4 . intron6 . APC was 47. 4 % ( 9/19) . 8. 7%C2/23).16. 7%(3/18) ,respectively. There are some relationship between LOH of exon4 and poor- ly differentiation , lymph node metastas , depth of invasion. LOH of exon4 may be one of prognostic marker of gastric cancer. We concluded that p53 gene mutation is an early event and perhaps has syner- gism with ras oncogene in gastric carcinogenesis

BACKGROUND:Tissue inhibitor of matrix metaloproteinase 1 (TIMP-1) is the corresponding antagonist of matrix metaloproteinase 13 (MMP-13), and their balance between expression and functional activity exerts an important role in the metabolic state of the extracelular matrix. During the development of osteoarthritis, however, TIMP-1 and MMP-13 expressions and their expression ratio show unclear changes in DH guinea pigs.
OBJECTIVE:To explore the expression levels of MMP-13 and TIMP-1 in DH guinea pigs with different ages, and to analyze the relationship between the ratio of MMP-13 to TIMP-1 and the age-dependent degenerative changes in the articular cartilage.
METHODS:Twenty-four female Dunkin Hartley guinea pigs were sacrificed at age of 2, 4, 8, 12 months separately, with six animals at each time point. The knee joints were colected and gross visual appearance of the articular cartilage was observed, then were decalcified and prepared for paraffin sections. VG staining and Mankin score were used to analyze the histological changes. Immunohistochemistry was conducted to assess the expression levels of MMP-13 and TIMP-1 in the cartilage. Integrated absorbance values were used as the quantitive analysis calculated by Image pro-Plus 6.0. Linear regression analysis was done to analysis the relationship between Mankin score and the ratio of MMP-13/TIMP-1. RESULTS AND CONCLUSION:Normal appearance in the articular cartilage was observed in 2-month-old DH guinea pigs, while degenerative changes in the articular cartilage were shown in 4-month-old animals, which became severer with age. Significant difference was found in Mankin score between any two groups (P < 0.05). The ratio of MMP-13 to TIMP-1 increased with age, and the ratio was positively correlated to the Mankin score (P < 0.05). Age-related articular cartilage degeneration occurred in Dunkin Hartley guinea pigs at 4 months of age, and devoloped with age, which is related with the imbanlance of the expression ratio of MMP-13 to TIMP-1.

Objective To investigate the value of contrast enhanced ultrasonography in evaluating cystic renal mass. Methods According to the imaging diagnosing standard, 29 cystic renal mass lesions were studied with contrast enhanced ultrasonography(CEUS), unenhanced ultrasonography (US), contrast enhanced CT(CECT) and then correlated their results with final histopathological results. The data were tested by receiver operating characteristic(ROC) curve. Results The area under ROC curve of CEUS and US was 0. 721, 0. 997, respectively. There was significant difference between the 2 groups (P=0. 003). The area under ROC curve of CEUS and CECT was 0. 997, 0. 997,respectively. There was no significant difference between the 2 groups (P= 1. 000). Conclusions CEUS is better than US and similar to CECT in evaluating cystic renal mass. CEUS with the Bosniak classification is useful for evaluating cystic renal mass.

The plasmid containing HPV16E6 gene was transfected into DC with lipofectamine. The morphology of DC was observed dynamically, and the expression of surface markers of DC vaccine could be detected using immuno-cytochemical staining and flow cytometry. MTT assay was applied to detect the activity of CTL in vitro.Results:The transfected DC had typical morphologic and phenotypic characteristics, and expressed E6 protein 47.3%, CD80 82.5%, CD86 79.8% and CD83 85.7%.
