1.Experimental study of the effect of ropivacaine combined with sufentanil analgesic on neuropathic pain in rats

Objective To explore the effect of the ropivacaine combined with sufentanil on neuropathic pain in rats.Methods The constriction injury method was performed to establish rat neuropathic pain model and 40 neu-ropathic rats were produced in accordance with the above modeling methods,which were divided into group A(ropiva-caine and sufentanil combined administration,n =10),group B(ropivacaine administration,n =10),group C(sufen-tanil administration,n =10)and group D(no administration,n =10).A total of 10 healthy rats without any treatment, the normal feeding,named as group E.At 1,3,7,11,14,21d after operation von -Frey filaments determination of mechanical hyperalgesia(MWT),and the Hargreaves method for the determination of thermal hyperalgesia(TWL) were measured and the ELISA Kit (IL -6 and TNF -α) were used to detect serum IL -6 and TNF -αconcentrations.Results Before and at 1 day after the operation mechanical allodynia and thermal hyperalgesia and allergy treatment had no significant difference(all P >0.05).Mechanical allodynia and thermal hyperalgesia of group A,B and C were significantly lower than those before the treatment and at 1 day after operation(all P <0.05),and mechanical hyperalgesia and thermal hyperalgesia of group A was significantly lower than those of group B and C (all P <0.05).The serum IL -6 and TNF -αconcentrations of 7,14 and 21 days in group A were significantly lower than those of group B and C(t =7.10,5.84,9.11,all P <0.05).Conclusion The effects of ropivacaine combined with sufentanil analgesic on neuropathic pain of rats was significantly higher than that of ropivacaine or sufentanil alone in the treatment.
2.Posterior surface topography of early keratoconus patients

Objective To investigate the corneal posterior surface topography characteristics of keratoconus at early stages applying the Pentacam anterior segment analysis system. Methods The present study included 43 eyes of 43 patients with keratoconus at the subclinical stage (group A), 40 eyes of 40 suspected keratoconus patients (group B), and 143 normal eyes of 143 controls (group C). Based on an examination of the Pentacam anterior segment analysis system of each subject, a series of data, including the posterior surface refractive power, the posterior surface elevation were collected. The differences among the three groups, the correlation among indices, and the area under the receiver operating characteristic (ROC) curves were analyzed. Results The mean values of the posterior surface maximum refractive power in 3 groups were -6.2 D, Q=0.5;-5.6 D, Q=0.3;and -5.5 D, Q=0.3, respectively. The mean values of the posterior surface maximum elevation were 23 μm, Q=14;11 μm, Q=8.5;and 7 μm, Q=6, respectively. The posterior surface maximum refractive power and maximum elevation among three groups were statistically different. The area under ROC (AUR) of posterior maximum refractive power and the AUR of posterior maximum elevation were greater than that of other indices in the diagnosis of early keratoconus. Conclusions Based on the results from the Pentacam anterior segment analysis system, the changes of the posterior surface refractive power and elevation are important characteristics of early keratoconus.

4. Difference in the culture of dorsal root ganglion cells in serum-free medium and serum medium

BACKGROUND: Both serum-free and serum media have been used to culture dorsal root ganglion cells, but the difference between the two remains unclear. OBJECTIVE: To explore whether serum-free medium can completely replace serum medium for culture of dorsal root ganglion cells. METHODS: The dorsal root ganglion of ICR mice at 8-10 weeks was taken and treated with collagenase and trypsin. After that, the mice were divided into the electroporation + serum group, electroporation + serum-free group, non-electroporation + serum group and non-electroporation + serum-free group. In the electroporation groups, the dorsal root ganglion cells were transfected with electroporation buffer and enhanced green fluorescent protein particles. Cells were cultured for three days. After Tuj1 antibody staining, in the non-electroporation + serum group and non-electroporation + serum-free group, axon branches, axon regeneration length, number of cell survival and the expression of proteins related to axon regeneration were counted. In the electroporation + serum group and electroporation + serum-free group, axon branches, length of axon regeneration, number of cell survival, and electroporation efficiency were measured. This study was approved by the Laboratory Animal Ethics Committee of the First Affiliated Hospital of Soochow University. RESULTS AND CONCLUSION: (1) In the non-electroporation + serum group and non-electroporation + serum-free group, there was no significant difference in axon branches, axon regeneration length, number of cell survival and the expression of axon regeneration related proteins (P > 0.05). (2) In the electroporation + serum group and electroporation + serum-free group, there was no significant difference in axon branches, axon regeneration length and electroporation efficiency (P > 0.05). Compared with electroporation + serum group, the number of cell survival of the electroporation + serum-free group was significantly lower (P < 0.05). (3) There was no significant change in the length of cell axon regeneration between the electroporation + serum group and the nonelectroporation + serum group (P > 0.05). The number of cell survival of the non-electroporation + serum group was significantly higher than that of the electroporation + serum group (P < 0.05). (4) The results showed that, in the condition of non-electroporation, the absence of serum does not affect the culture of dorsal root ganglion in vitro, and serum-free medium can replace serum medium. However, under the condition of electroporation, the number of cell survival would be decreased without serum medium, suggesting that serum plays an important role in the culture of dorsal root ganglion in vitro under the condition of electroporation. Therefore, serum-free media cannot replace serum media.
5.The exploration and practice of emergency personnel with order training based on school-enterprise cooperation

This article introduced the practice of emergency personnel cultivation according to order form based on the cooperation between Cang Zhou Medical College and the Beijing Red Cross emergency rescue center. The implementation process: First, we tried and built cooperative relation with the enterprises and public institutions which had strong cooperative intention, high demand and advanced techniques by field investigation. Second, we signed order training agreement, made clear the rights and obligations of both sides. Then, we selected the best students and formed the ordered class. Both sides organized the professional construction committee together. After much argument we thrashed practical talent training projects and professional development plans and implemented them strictly. We also built up a high quality team which involved full-time teachers as well as part-time teachers with balanced age and education. Besides, we adopted two plus one training pattern and em-ployed the best (the rate of employment beyond 80%). Thus, the win-win situation of students, schools and hospitals was realized.
6.Follow-up Study on the One-month's and One-year's Effect of 2006' Fujian International "Quit & Win" Competition

Objective To study the one-month and one-year effect of 2006' Fujian International "Quit&Win" Competition,and to explore the influencing factors for smoking-quitting behaviors.Methods A equidistant sampling survey was conducted among the 4619 participants at the end of one-month and one-year after the competition by telephone,mailing and face to face interview.The survey contents include personal information,smoking-quitting and relapse information.The data were input with EPIDA3.1 software,?2 test and logistic stepwise regression were done with SPSS 11.5.Result The rate of smoking-quitting sustained for one-month was 51.9%,and the rate of smoking-quitting sustained for one-year was 38.0%.With one-way Chi-square test,the influencial factor on smoking-quitting succession for one-month was their intention of joining in the competition(?2=75.799,P=0.000);and the influencing factors on smoking-quitting succession for one-year were cigarette consumption per day(?2=9.953,P=0.019),their intention of joining in the competition(?2=52.227,P=0.000) and the method of smoking-quitting(?2=7.882,P=0.019).With logistic regression analysis,the influencial factors on smoking-quitting succession for one-month were age,their intention of joining in the competition and the method of smoking-quitting,the OR values were 0.98(0.964~0.997),1.198(1.195~3.080) and 2.812(2.152~3.675),respectively;the influencial factors on smoking-quitting succession for one-year were marriage,cigarette consumption per day,their intention of joining in the competition and the method of smoking-quitting,the OR values were 1.915(1.085~3.381),0.720(0.546~0.951),0.529(0.330~0.847),3.171(2.233~4.501) and 1.901(1.294~2.793),respectively.The major factors for relapse were the surrounding people smoking,lack of support,et al.Conclusion The 2006' Fujian International "Quit & Win" Competition gained a great success and has important significance for smoking-control work.
7.Biliometric analysis about periphere venous catheter cannulation and maintenance in Chinese literature from 1990 to 2013

Objective To provide a reference for local best practice information sheet by way of analyzing the status quo of adult peripheral venous catheters nursing and maintenance.Methods Systematically analyzing the related 487 documents met the inclusion criteria in CBMdisc,CNKI Database and WanFang Database from the year of 1990 to 2013 by biliometric analysis methods.Results The number of documents,rising year after year,stood mainly in nursing journals,of which the majority authors are from Guangdong [10.3%(50/487)],Anhui [9.0%(44/487)] and Hubei [7.6%(37/487)].50.3% (245/487)of the documents did not refer in particular to the object.The three major research concerns are the prevention and treatment of complications[80.9%(394/487)],cannulation [23.6%(115/487)] and maintenance [19.9% (97/487)].Conclusions Peripheral venous catheter cannulation and maintenance has gained widespread attention among the domestic clinical staffs,but the overall quality of articles concerning these aspects is not high.High-quality research papers are expected to provide better evidence for local best practice information sheet.
8.Assessment the methodological and ethical quality of randomized controlled trials of non-small cell lung cancer in China

Objective To assess the methodological and ethical quality of randomized controlled trials (RCT) of non-small cell lung cancer ( NSCLC ) in China. Methods The methodological and ethical quality on RCT of NSCLC, covered in CBM, CNKI, VIP and Wanfang was assessed according to the Jadad Scale and Berdeu Scale respectively. Results The average methodological and ethical quality was 62. 24% according to the Jadad Scale and was 18. 79% according to the Berdeu Scale. Correlation analysis showed that the methodological quality was closely correlated with the ethical quality, except that of curative effect of invasive therapy (OR=1. 064, 95%CI:0. 832-1. 361, P>0. 05). Conclusion The methodological and ethical quality of RCT of NSCLC in China is low. The methodological quality is positively correlated with the ethical quality.
9.Risk factors for early cognitive dysfunction after cardiac valve surgery

Objective Postoperative cognitive dysfunction (POCD) is a common complication of cardiac surgery, which seriously affects the prognosis of the patient.This study aimed to explore the risk factors for early POCD in patients undergoing cardiac valve surgery and the correlation between early POCD and the serum S100B protein level.Methods Eighty patients underwent mitral valve replacement surgery in combination with tricuspid plasty.At 1 day before and 5 days after surgery, we assessed the cognitive function of the patients and divided them into a POCD and a non-POCD group.We obtained such data as the age, sex, education, New EuroSCORE Ⅱ, and preoperative NYHA cardiac function grades and left ventricle ejection fraction (LVEF) of the patients, collected the venous blood to determine serum S100B protein concentration by ELISA, and analyzed the independent risk factors of early POCD using single-factor and binary logistic regression analyses.Results POCD was found in 20 (25%) of the patients, , Logistic regression analysis showed the independent risk factors for early POCD to be hyperglycemia (OR=6.038, 95% CI: 1.202-30.337), operation time (OR=6.423, 95% CI: 1.276-32.332), and aspartate aminotransferase (AST, 2 times higher than normal) (OR=12.878, 95% CI: 2.289-72.445).The serum S100B protein concentrations in the POCD group were (1.9±0.3) μg/L and (1.7±0.4) μg/L at 48 and 72 hours after cardiopulmonary bypass, significantly lower than (2.4±0.4) μg/L and (2.1±0.3) μg/L at 30 minutes and 24 hours (P<0.05), and so was it in the non-POCD group at 72 than at 48 hours postoperatively ([1.4±0.4]) vs [1.5±0.4] μg/L, P<0.05).Conclusion Long operation time, perioperative hyperglycemia and high AST are independent predictors and the serum S100B protein level is a significant marker of early POCD.
10.Human umbilical cord blood stem cells differentiate into nasal ciliated epithelial cells

BACKGROUND:Damage to nasal ciliated epithelial cels can lead to a severe injury in nasal biological function. Compared with other adult stem cels, human umbilical cord blood stem cels have better differentiation potential.
OBJECTIVE: To explore the feasibility of human umbilical cord blood stem cels differentiating into nasal ciliated epithelial cels through in vitro culture and induction techniques.
METHODS:Normal and healthy umbilical cord blood samples were colected to isolate human umbilical cord blood stem cels, folowed by identification and subculturein vitro. Umbilical cord blood stem cels at passage 3 were infected with recombinant adeno-associated virus carrying enhanced green fluorescent protein and cultured using air liquid interface culture method. Thereafter, PCR assay was employed for detecting MUCS expression in cultured stem cels at 1 and 2 weeks after induction, and immunofluorescent staining for FOXJ1 was performed at 3 weeks.
RESULTS AND CONCLUSION:After subculture, passage 3 umbilical cord blood stem cels that could express stem cel surface markers were visible in a uniform shape and had good refraction. After 3 hours of gene transfection, green fluorescence issued from the passage 3 cels were visible, and the cel positive rate was up to 96.2% until 48 hours, indicating good transfection efficiency. RT-PCR findings showed that MUC8 mRNA had no expression in the umbilical cord blood stem cels, but expressed strongly in the nasal ciliated epithelial cels, whose expression was weak at 1 week of culture and increased at 2 weeks. Additionaly, the positive expression of FOXJ1 red fluorescence was observed under the transfection of green fluorescent protein. These results suggest that human umbilical cord blood stem cels could differentiate into nasal epithelial cels under suitable conditions.
