1.Determination of 39 Polybrominated Diphenyl Ethers in Coastal Sediments by Ultrasonic Extraction Coupled with Gas Chromatography-Mass Spectrometry

A method was developed for the determination of 39 polybrominated diphenyl ethers(PBDEs) from mono- to hepta-brominated in coastal sediment by gas chromatography/mass spectrometry. The sample was extracted with a solution of dichloromethane: n-hexane( 1:1,V/V) ,followed by ultrasonic extraction for 60 min at 25℃. The clean-up step was optimized using silica gel and alumina. The detection limit of the method was in the range of 0.003 - 0.10μg/kg. The results indicate that the average recovery of the internal standard was from 66.2% to 118. 6%. The relative standard deviation RSD( % ) was from 0. 8 % to 18. 2%. The method for the detection of 39 PBDE congeners in coastal sediment was sensitive,high reproducible with satisfactory recoveries.
2.Prevalence of hepatitis E virus infection among blood donors

【Objective】 To investigate the prevalence of hepatitis E virus (HEV) among voluntary blood donors, and the viability and practicability of nucleic acid testing (NAT) in Jinhua area. 【Methods】 Blood samples from 1 017 voluntary blood donors at Jinhua Blood Center from April to May 2021 were collected, and HEV RNA was detected by CHURAS BSS1200 blood nucleic acid detection system. The reactive samples were re-tested for IgM and IgG antibodies by ELISA, and HEV genotyping by gene sequencing. 【Results】 The yield rate of HEV RNA out of voluntary blood donors in Jinhua was 0.98 ‰ (1 / 017). The individual was negative for IgM and IgG antibodies, but was confirmed to be HEV RNA genotype 4 by sequencing. 【Conclusion】 HEV RNA positive donors have been found in this study. HEV RNA blood screening can effectively enhance the safety of blood transfusion.
3.Application of improved comparability verification method in performance verification of instruments and equipment

【Objective】 To explore the viability of improved comparability verification method for the performance verification of automatic biochemical analyzer in blood centers. 【Methods】 According to the relevant requirements of User Verification of Performance for Precision and Trueness; Approved Guideline-Second Edition (EP15-A2) and Guideline for Comparability Verification of Quantitative Results with One Health Care System(WS/T 407-2012), the performance of the newly purchased Beckman Coulter AU480 automatic biochemical analyzer was confirmed by means of comparability verification, and compared with the current Olympus AU400 automatic biochemical analyzer. 【Results】 The detection results given by Beckman AU480 biochemical analyzer on 5 samples for ALT detection performance test were significantly correlated with the target value (R2=0.998), and the trueness verification results met the requirements.The CV of medium and high ALT levels was 2.36% and 1.20%, respectively, within the acceptable standard range(<5%), and the precision verification results meet the requirement.The comparison deviation of two devices for medium and high ALT levels was 0.47% and 1.62%, respectively, within the acceptable standard range (<5%). The detection results of ALT from 40 blood donors given by two devices were significantly correlated (R2=0.956), and the precision verification results met the requirements.In the validation of linear evaluation, there was a good linear relationship between the expected and measured results (r=0.997)among samples with 6 levels of concentrations. 【Conclusion】 The comparability verification method is suitable for the performance verification of automatic biochemical analyzer in blood centers.
4.Relationship between the erythrocyte CR1 genomic density polymorphism and erythrocyte immune function in children with asthma

Objective To explore the hereditary susceptibility of children with asthma through studying the relationship between erythroeyte CR1 genomic density polymorphism and erythrocyte immune function. Methods The rates of RBC-C3_(3b)RR and RBC-ICRR were detected to the asthma group consisted of 65 children with asthma and the control group consisted of 28 normal children. The CR1 activity and genomic density polymorphism of erythrocyte from the two groups were detected by Hind Ⅲ restriction enzyme digestion, polymerase chain reaction restriction fragment length polymorphism. Results Frequencies of high expression gene (HH), mid expression gene (HL) and low expression gene (LL) genotypes were 43.08% (28/65), 36.92% (24/65) and 20.00% (13/65) in asthma group, and 78.57% (22/28), 17.86% (5/28) and 3.57%(1/28) in control group respectively. A significant difference was found in the distribution frequency of CR1 genotype between the two groups(P< 0.01).The rates of RBC-C_(3b)RR were significant lower and the rates of RBC-ICRR were significant higher in asthma group than those in control group (P < 0.01). The rates of RBC-C_(3b) RR in HH, HL and LL were decreased in order (P < 0.01),while the rates of RBC-ICRR in HH,HL and LL were increased in order (P < 0.01). Conclusion It suggests that CR1 gene polymorphism may play an important role in determining susceptibility to asthma.
5.The significance of lymphocyte and cytokine expression in laryngeal tuberculosis

Objective To investigate the clinical significance of the expression of lymphocytes and cytokines in patients with laryngeal tuberculosis.Methods 96 cases of laryngeal tuberculosis were selected as the research objects.Of which 42 cases of infiltrating type,31 cases of ulcer type,mass type in 23 cases,the other 50 cases of healthy persons were selected as the healthy control group,all the research objects,extracting the morning fasting venous peripheral blood for detection of T lymphocyte subsets and cytokines were detected,and the results were compared.Results Invasive laryngeal tuberculosis CD3+ index (63.10 ± 5.33) % (t =1.922),CD4+ (40.32 ± 5.06) % (t =1.758),CD8+ (24.41 ± 4.59) % (t =1.710),compared with the control group,CD4+,CD3+ index significantly reduced,CD8+ index increased significantly,the differences had statistical significance (t =1.922,1.758,1.710,all P < 0.05),and the ulcer type and mass type results compared with invasive had significant difference(all P <0.05).In the analysis of laryngeal tuberculosis pathological types,reduced the mass type CD3+,CD4+ T index and CD8+ index of lymphocyte increased obviously the most significant;all laryngeal tuberculosis in patients with IL-4,IL-10 and TNF-alpha significantly increased compared with the control group,while the IFN-gamma was significantly reduced,and the mass in the type of change was the most significant,the differences had statistical significance(all P < 0.05).Conclusion The use of lymphocytes and cytokines detection can improve the laryngeal tuberculosis patients with the clinical diagnosis rate,with the detection results of different degree and different pathological types of laryngeal tuberculosis patients,can be used for preliminary assessment of laryngeal tuberculosis disease and pathological type,the choice of treatment modality for patients with guidance value,with in-depth clinical study and promotion application of value.

8.Tympanostomy tube under the ear endoscopy combined with adenoidectomy in children with ssecretory otitis media

Objective:To evaluate efficacy of tympanic membrane PE tube placement combined with oral endoscopic adenoidectomy in treating otitis media with effusion(OME) in children. Methods:Selected 87 cases(102ears)patients of secretory otitis media as research subjects between January 2012 to August 2014, in department of Otolaryngology in Heilongjiang Province, randomly divided into observation group(45cases, 55ears) and control group(42cases, 47ears),observation group used tympanostomy tube siphon drainage method combined with adenoidectomy for treatment, control group alone tympanostomy tube siphon drainage therapy, postoperative complications recorded data of patients later from the hospital after six and twelve months. Results:Two groups of patients after six months treatment and after twelve months(x2=5.705, x2=7.460;P<0.05), the results were statistically significant(P<0.05);in the middle ear effusion time, recurrent ear number, the number of ear infections, the results were also statistically significant(t=7.662, x2=4.197, x2=4.752;P<0.05). Conclusion:Under the ear drum using endoscopic drainage siphon tube method combined with adenoidectomy can effectively reduce postoperative complications in patients with quick, injury, etc, with high short-term and long-term efficacy, worthy application.
9.Dorsal plate assisted fixation of dorsal lunate fossa fracture of distal radius.

OBJECTIVE:
To explore clinical efficacy of dorsal plate assisted fixation of dorsal lunate fossa fracture block of distal radius.
METHODS:
From January 2019 to January 2022, 30 patients were treated with dorsal plate assisted fixation of dorsal lunate fossa fracture of distal radius, including 13 males and 17 females, aged from 42 to 68 years old with an average of (48.7±5.6) years old;According to Doi fracture classification, 24 patients were type 3 blocks and 6 patients were type 4 blocks. The degree of palmar angle of anterior and posterior distal radius was fixed by dorsal steel plate during operation. Fracture healing and functional recovery of wrist were observed after operation. Functional evaluation was performed by Gartland and Werley scoring system at 12 months after operation.
RESULTS:
All patients were followed up from 12 to 13 months with an average of (11.3±0.9) months. All fractures healed for 4 to 5 months with an average of(4.7±0.8) months. Median palpal inclination of anterior and posterior distal radius fixed by dorsal plate was 5.30°(4.85°, 6.03°), 12.45°(11.98°, 13.43°) respectively, and had statistical difference( P<0.01). Gartland and Werley scores was (1.1±0.4) at 12 months afteropertaion, and 27 patients got excellent result and 3 good.
CONCLUSION
Dorsal plate assisted fixation of dorsal lunate fossa fractures is beneficial to reduction and stabilization of displaced dorsal fractures and restoration of palmar inclination.

10.Analysis of HIV infection among voluntary blood donors in Jinhua, 2013~2020

【Objective】 To investigate the HIV infection among voluntary blood donors in Jinhua area through the analysis of HIV antibody detection and confirmation results, so as to guarantee the safety of clinical blood use. 【Methods】 419 823 samples of voluntary blood donors in Jinhua Central Blood Station from 2013 to 2020 were detedcted for HIV twice. Samples reactive with one assay were double-well retested. Samples initially reactive with two assays or positive with one retest assay were subjected to the CDC for confirmatory test. The HIV antibody infection and epidemiological characteristics of voluntary blood donors were retrospectively analyzed. 【Results】 A total of 419 823 samples of blood donors were dectected, among which 335 were reactive for HIV screening, 48 confirmed positive, with the positive rate at 0.11‰(48/419 823), of which 31 were registered residence in Jinhua and 17 in other places; 46 males and 2 females; 35~45 years old accounted for 28.23%; freelancers 43.61%; coinfection 4.16%.The positive rates differed significantly by gender (male 0.18 ‰ vs female 0.01 ‰, P<0.05 ), age groups (‰) (0.17 vs 0.07 vs 0.18 vs 0.01 vs 0.20 in 18~24, 25~34, 35~44, 45~54, 55~59 years old group, P<0.05), but not by registered residence( 0.10‰ of Jinhua area vs 0.14‰ of other areas) or occupation (‰)( 0.09 vs 0.24 vs 0.11 vs 0.03 vs 0.09 vs 0.11, and 0.13 for students, workers, farmers, civil servants, medical personnel, employees and freelancers, P>0.05). 【Conclusion】 The positive rate of HIV antibody in Jinhua area was lower than that in other areas. Blood stations should continue to carry out ELISA and NAT, strengthen the publicity and education of AIDS prevention and control, and ensure the safety and effectiveness of clinical blood use.
