1.An comparative study of the selective killing action of antibacterial protein from the embryonic cells of the musca domestica on human myeloid leukemia cells K562

Objective To stimulate the embryonic cells of the musca domestica with lipopolysaccharide(LPS) for production of antibacterial protein and to extract antibacterial protein ,then research the inhibitory action of the antibacterial protein and homohar‐ringtonine on human myeloid leukemia cells K562 and normal human cells .Methods The logarithmic growth phase′s embryonic cells of the musca domestica were stimulated using no serum M3 insect medium which contained 20 mg/L LPS for sixteen hours . The antibacterial protein was extracted from supernatant fluid .The antibacterial protein was prepared in 40 ,80 ,160 ,320 and 640μg/mL five density groups ;the MTT experiments were used to test the inhibition of antibacterial protein on K562 cells and the hu‐man umbilical vein vascular endothelial cells .The K562 cells and human umbilical vein vascular endothelial cells were prepared HTT and antibacterial protein of the embryonic cells of the musca domestica groups ,the normal control group was established by cells itself .Effective killing rate of K562 cells and the human umbilical vein vascular endothelial cells were measured .Results The effective inhibition ratio of homoharringtonine and the antibacterial protein on K562 cells and human umbilical vein vascular endo‐thelial cells on three density groups were detected by flow cytometry .The MTT examination demonstrated that all density antibac‐terial peptides had inhibition activities on K562 cells ,but no inhibition activities on human umbilical vein vascular endothelial cell . The effective killing and wound ratio of the control group ,the homoharringtonine group and of the antibacterial protein group from the embryonic cells of the musca domestica on the K562 cells were(28 .16 ± 2 .14)% ,(81 .41 ± 1 .95)% and (82 .90 ± 3 .03)% ,re‐spectively ;the effective killing rate on human umbilical vein vascular endothelial cells were(41 .13 ± 2 .51)% ,(82 .20 ± 2 .57)% and (36 .68 ± 1 .86)% ,respectively .Conclusion Compared with the common chemotherapeutics medicine ,the merit of the antibacterial protein from the embryonic cells of the musca domestica is that it can kill the tumor cells effectively ,but would not damage the nor‐mal person cells .
2.Clinical observation of cervial intraepithellal neoplasla managed by loop electrosurgical excision procedure

Objective To evaluate the clinical value of cervical intraepithelial neoplasia treated by loop elec-trosurgical excision procedure. Methods A retrospective analysis was performed in 192 patients with CIN which were diagnosed after LCT, oolpoccopy and multiple cervical biopsies. Results The satisfactory rate was 99. 5% (191/192) ,the LCT and digital colposcopy were all negative after operation 4 -6 months, 12 months, 18 months and 24 months. Conclusion LEEP is an effective treatment for CIN, with the advantages of easy, safe and quick procedure, minimal damage and blood loss. LEEP helps to avoid misdiagnosis of earlier cervical cancer.
3.Effect of sympathetic activity on peripheral immune function of middle cerebral artery occlusion in rats

Objective To investigate the relationship between the changes of the sympathetic nervous system activity and the peripheral cellular immunodepression after cerebral infarction.Methods An adult male Sprague-Dawley rat model of middle cerebral artery occlusion (MCAO)was induced.A double-antibody sandwich enzyme-linked immunosorbent assay was used to detect the levels of serum proinflammatory cytokines interleukin(IL)-1β,interferon-γ (INF-γ)and anti-inflamatory cytokines IL-10.A competitive enzyme-linked immunosorbent assay was used to detect the levels of serum metanephrine(MN)and normetanephrine(NMN).After blocking sympathetic activity by injecting β -blocker propranolol intraperitoneally,the sympathetic activity and the levels of pro/anti-inflammatory cytokines were detected.Results The levels of serum MN and NMN at 6 hours after MCAO began to increase in the cerebral infarction group compared with the sham operation group.They reached the peak at 72 hours,and then decreased slowly.However,they were still higher than those in the sham operation group 2 weeks after MCAO(P < 0.01 ).At 72 hours after MCAO,the levels of MN and NMN in the propranolol intervention group were decreased significantly(P <0.01 ),the levels of proinflammatory cytokines IL-1β and INF-γwere increased significantly(P <0.01 ),and the levels of anti-inflammatory cytokine IL-10 were decreased significantly(P <0.01).Conclusions When sympathetic nervous system is overactivated after cerebral infarction in rats,the peripheral cellular immunity is in a state of depression;the peripheral immunodepression will be fundamentally reversed after blocking the sympathetic function,and this suggests that the overactivation of sympathetic nervous system may play a important role in the process of immunodepression after cerebral infarction.
4.Clinical features and misdiagnosis analysis of six children MELAS syndrome patients

Objective To explore clinical features and misdiagnosis reasons in mitochondrial encephalomyopathy,lactic acidosis,and stroke-like episodes (MELAS) syndrome.Methods The results of clinical data,brain magnetic resonance imaging (MRI),and the course of diagnosis were analyzed in 6 patients with MELAS.Results (1) Clinical features:headache and vomiting were the starting symptoms in 4 of 6 cases,and developmental delay was initial symptoms in 2 of 6 cases.Marasmus occurred in 6 cases,seizure in 5 cases,fever in 3 cases,and hirsutism and visual impairment in 2 cases.(2) Experimental results:blood lactic acid was higher in 6 (4.28 ~ 10.3 mmol/L).(3) Brain MRI:6 patients had abnormal signals in parietal,occipital,temporal lobe,which were not in accordance with vascular distribution.(4) Molecular genetics:All the 6 patients had A3243G gene mutation.(5) Three patients were misdiagnosed for viral encephalitis,and 2 developmental retardation.Conclusions MELAS is characterized with developmental retardation,and repeated encephalitis attack.It is also misdiagnosed because of its variety of clinical features.If patients have high level of lactic acid and multiple MRI signal abnormalities of brain which are not in accordance with vascular distribution,MELAS should be suspected of.Genetic examination and muscle biopsy are especially important in the diagnosis of MELAS.
5.Expansion of cord blood primitive and committed progenitors cells in vitro

Objective To establish a method for ex vivo cord blood stem cell expansion,increase the number of the stem/progenitor,especially megakaryocyte progenitors so as to decrease the thrombocytopenia period after CBT.Methods Mononuclear cells (MNC) from 20 cord blood samples were cultured in presence of different cytokines:group 1:IL 3,IL 6,group 2:IL 3,IL 6,TPO,group 3:IL 3,IL 6,TPO and cord blood plasma.The final concentration of the cytokines was 50ng/ml.The control group had all medium except the cytokines and cord blood plasma.The cells were cultured for 21 days.At every 7 days,tests were done for nucleate cells number,CFU GM,CFU Mk,and CFU GEMM.Results At different times and in different groups,there were different changes.For NC,maximal increase were obtained in group 3 on day 21,which was 239.65?153.48.For CFU GM,the biggest expansion occurred on day 21 in group 3,the number being 184.17?140.40.For CFU Mk,the biggest expansion was obtained on day 14 in group 3,with the number of 135.34?105.20. The immature progenitor CFU GEMM achieved the max increase in group 3 on day 14,the number was 49.39?36.68. Conclusion The expansion of CFU Mk progenitor cells can be obtained in the presence of IL 3,IL 6,TPO.This combination also enhances the expansion of CFU GM and CFU GEMM.Addition of cord blood plasma can promote the expansion of mature and immature progenitor cells.
6.The influence of injection rate and time resolution on dynamic contrast-enhanced MR imaging of glioma

Objective To explore the influence of contrast agent injection rate and sequence time resolution on the dynamic contrast-enhanced MR iamging of brain glioma.Methods Fifty eight cases with suspected cerebral glioma were scanned on a Verio 3.0T MRI with the regular head sequences and DCE sequences.All the cases were divided into two groups.There were 41 cases in the regular group in which the injection rate was 3 mL/s and the time resolution was above 5.36 s.Another 1 7 cases were in the optimal group in which the injection rate was 4 mL/s and the time resolution was about 4 s.The DCE images of two groups were Postprocessed by the Tofts model analysis.The curve shape of AIF were evaluated and divided in to the excellent or the secondary level.Chi-square test was used to compare the results of two groups.Results The curve shape of optimal group was much better than that of regular group(P<0.05).Conclusion Speeding up the injection rate and the time resolution of DCE sequences would improve the image quality of DCE MRI of glioma.
7. A system review: the effects of T-type CCB or ACEI /ARB for hypertensive patients with chronic kidney disease

OBJECTIVE: To evaluate whether T-type CCBs are equivalent with or superior to ACEIs/ARBs on renal outcomes in hupertensive patients with chronic kidney disease. METHODS: Cochrane Library, Pubmed, EMbase and CNKI were searched for relevant randomized controlled trials (RCTs) from inception to May 2012. The meta-analysis was performed by Revman 5.1 software. RESULTS: Five RCTs (563 subjects) were included in the present study. T-type CCBs performed a pooled improvement in creatinine clearance and glomerular filtration rate similar to ACEIs/ARBs but were inferior to ACEIs/ARBs on reducing proteinuria excretion (three RCTs, 389 subjects, WMD 0.26 g·d-1, 95% CI 0.10 to 0.43), although T-type CCBs and ACEIs/ARBs showed stable anti-hypertensive effect. CONCLUSION: Our findings suggest that despite T-type CCBs do offer salutary effects on kidney outcomes and hypertension can be applied to treat hypertensive patients with chronic kidney disease, ACEIs /ARBs might be better choice for pressure control in this target population especially when proteinuria is the main issue of renal dysfunction.
8.Correction of plasma inflammatory cytokines with the spleen weight index following cerebral ischemia-reperfusion in rats

Objective To investigate the relationship between the changes of peripheral cellular immune function and the spleen weight index and the spleen histopathological changes following cerebral ischemiareperfusion in rats.Methods A model of middle cerebral artery occlusion (MCAO) of adult male rats was induced.The peripheral blood cytokine levels,spleen weight index and the spleen histopathological changes were evaluated.Results The serum proinflammatory cytokine interleukin (IL) -1β and interferon (INF) -γ levels in rats increased at 6 and 12 hours respectively after modeling.They began to decrease at 24 hours and decreased to the lowest at 72 hours.Compared to the sham operation group,there were significant differences (P < 0.01 );on the contrary,the anti-inflammatory cytokine IL-10 level decreased at 6 hours after modeling It began to increase at 12 hours and reached the peak at 72 hours.Compared to the sham operation group,there were significant differences (P<0.01).The spleen weight index in a MCAO group decreased significantly at 6 hours after modeling(P<0.01) and it began to increase at 12 hours,but it was still lower than that in the sham operation group (P<0.01),and then it decreased gradually and decreased to the lowest at 72 hours (P<0.01).HE staining showed that the spleen germinal centers of rats reduced significantly at 72 hours after MCAO and the contours did not show very clear.The correlation analysis showed that the proinflammatory cytokine IL-1 β(r =0.304,P =0.002) and INF-γ (r =0.644,P =0.000) levels were positive correlated with the spleen weight index,and the anti-inflammatory cytokine IL-10 level was negatively correlated with the spleen weight index (r =0.492,P =0.000).Conclusions The rat peripheral cellular immune function after MCAO was in an inhibitory state.The changes of spleen may play an important role in the process of immunosuppression after stroke.

Ecotropic viral integration site-1 (Evi-1) gene is first identified as a binding site for retrovirus in murine myeloid leukemia.The human homologue with the murine in leukemia oncogene Evi-1 is about 91% of nuclear sequence and 94% of amino acid.Evi-1 gene is high expressed in myeloid malignancies and other tumors.The main tumorigenic mechanisms of Evi-1 gene include inhibiting TGF-β signal path by depressing Smad3,adjusting cell cycle,promoting angiogenesis,inhibiting JNK.activating AP-1,and so on.
10.The Unbalance of the Mechanism for Coordinating Interests among Interest Groups in Medicine Circula-tion and the Rectification Channel

OBJECTIVE:To put forward suggestions for rectifying the unbalance of the mechanism for coordinating interests in medicine circulation. METHODS:From three aspects of the expression,game and balance of interests,the interest group theory was used to analyze the unbalance of the mechanism for coordinating interests in medicine circulation as well as the cause of the un-balance,and rectification suggestions were put forward. RESULTS&CONCLUSIONS:At present,the mechanism in medicine cir-culation in China is unbalanced,including mechanism for interest expression,for interest game,for interest balance,etc. Corre-sponding rectifications should include perfecting legislation,effectively giving play to the role of media,defining the legal status of the interest group,strengthening organizational building of the vulnerable group,adjusting the interest pattern of the strong and weak groups and strengthening legal and social morality construction.
