1.Effects of Dihuang Yizhi Extract on the behavior and cholinergic system in the rats with Alzheimer's disease

AIM: To investigate the effects of Dihuang Yizhi Extract(DHYZE) on learning-memory ability and the central cholinergic system of the rats with Alzheimer's disease(AD) and to explore the possible mechanism of action. METHODS: Aggregated A?25-35 was injected into the right hippocampus of the rats to make experimental rat model of AD.The behavioral abnormalities were investigated by shuttle box.The contents of acetylcholine(Ach)、cholinesterase(ChE) activity、cyclic AMP response element-binding protein(CREB) and phosphorylated CREB serine 133(pCREB Ser133) in hippocampus were measured. RESULTS: DHYZE could markedly improve the function of learning and memory of rats in AD model in a dose-dependent manner.The contents of Ach and AchE activity in hippocampaus increased obviously in DHYZE medium dose group and high dose group compared with AD group.Such promotion was dose-dependent.The contents of pCREB Ser133 in hippocampaus increased obviously in DHYZE high dose group compared with AD group. CONCLUSION: The results indicate DHYZE could improve the ability of spatial learning and memory in AD rats.The mechanism might be associated with improving the impairment of the central cholinergic neurons induced by A?25-35 and activating CREB signal pathway in hippocampus of AD rats.
2. Early effectiveness of oblique lateral interbody fusion combined with pedicle screw fixation via small incision Wiltse approach for lumbar spondylolisthesis

Objective: To investigate the early effctiveness of oblique lateral interbody fusion (OLIF) combined with pedicle screw fixation via small incision Wiltse approach for the treatment of lumbar spondylolisthesis. Methods: Between January 2016 and December 2016, 21 patients with lumbar spondylolisthesis were treated with OLIF and pedicle screw fixation via small incision Wiltse approach. There were 9 males and 12 females, aged 57-73 years, with an average age of 64.5 years. The disease duration was 24-60 months, with an average of 34.6 months. All cases were spondylolisthesis at L 4 (15 cases of degreeⅠ, 6 cases of degreeⅡ); 1 case had vertebral arch isthmus, and 20 cases had spinal stenosis. Japanese Orthopaedic Association (JOA) scoring system was used to evaluate the effectiveness before operation and at last follow-up. Before operation and at 2 days after operation, anteroposterior and lateral X-ray films and CT were taken to measure the sagittal diameter and cross-sectional area of the spinal canal, and calculate the intervertebral height and degree of spondylolisthesis. At 6 months after operation, the intervertebral fusion was evaluated by CT. Results: The operation time was 120-180 minutes, with an average of 155 minutes; the intraoperative blood loss was 100-340 mL, with an average of 225.5 mL. One patient had slight injury of lower endplate, 1 patient had numbness of thigh and weakness of hip flexion after operation, 1 patient had sympathetic nerve trunk injury. All the cases were followed up 12-18 months, with an average of 14.3 months. The symptoms of low back pain, leg pain, and numbness of lower limbs significantly relieved after operation, and there was no complication such as protrusion of fusion cage, screw breakage, and endplate collapse. At 2 days after operation, the intervertebral height, degree of spondylolisthesis, sagittal diameter of spinal canal, and cross-sectional area of spinal canal significantly improved compared with preoperative ones ( P<0.05). At 6 months after operation, CT showed that 1 patient had poor interbody fusion (grade Ⅲ), the other 20 patients had good interbody fusion (grade Ⅰ and Ⅱ), and the interbody fusion rate was 95.2%. At last follow-up, JOA score of lumbar spine significantly increased compared with that before operation ( t=24.980, P=0.000). Conclusion: OLIF combined with pedicle screw fixation via small incision Wiltse approach for the lumbar spondylolisthesis has minimally invasive features, such as less trauma, fewer complications, and higher intervertebral fusion rate. It is a safe and effective method.
3.Study on the effect of chlorine disinfectants on the destruction of New coronavirus nucleic acid

Objective To observe the destructive effects of chlorine disinfectants on the nucleic acid of novel coronavirus, and to assess the feasibility of real-time fluorescent quantitative PCR technology for the evaluation of the disinfection effect of novel coronavirus. Methods A suspension quantitative inactivation test was used to observe the inactivation effects of different concentrations of chlorine containing disinfectants and different action times on the novel coronavirus. RT-PCR method was used to detect novel coronavirus nucleic acid for the disinfection effect evaluation. Results The chlorine disinfectants of 1 000 mg/L and 2 000 mg/L could destroy the nucleic acid of ew coronavirusafter application for over 30s. The chlorine disinfectant of 750 mg/L could destroy the nucleic acid of new coronavirusafter application for over 15min. Chlorine disinfectant of 500 mg/L could not completely destroy the nucleic acid of new coronavirusafter 30 min action. Conclusion The disinfection method stipulated in the national prevention and control plan using 1 000mg / L effective chlorine for the epidemic sites of new coronavirus for 30 minutes could completely destroy the nucleic acid of new coronavirus. The Real-time quantitative PCR can be used to evaluate the disinfection effect of new coronavirus.
4.Analysis of influenza surveillance results in Jingzhou, Hubei Province from 2016 to 2021

ObjectiveTo understand the epidemiological characteristics of influenza in Jingzhou from 2016 to 2021, so as to provide scientific evidence for the formulation of influenza prevention and control policies in this region, and effectively protect people's health. MethodsData of influenza-like illness (ILI) and pathogen surveillance in Jingzhou during 2016‒2021 were collected and statistically analyzed. ResultsA total of 46 272 ILI cases were reported from two hospitals in Jingzhou City from 2016 to 2021. The difference in the constituent ratio of ILI was statistically significant among different age groups (P<0.05). A total of 12 812 specimens were collected from two hospitals for influenza surveillance. A total of 1 513 cases were RNA positive,and the positive rate of influenza virus nucleic acid detection was 11.81%. The RNA positive specimens were mainly B (Victoria), accounting for 39.33%. There were statistically significant differences in the positive rate of influenza virus nucleic acids and different types of influenza virus nucleic acids among different years (P<0.05). ConclusionThe influenza epidemic in Jingzhou peaks in winter and spring, and the new A (H1),A (H3), B (Victoria) and B (Yamagata) types alternate and mixed epidemics dominate.
5.Protective effect of astragalus-containing serum on cobalt chloride-induced hypoxia injury in ARPE-19 cells

AIM: To study the protective effect of astragalus-containing serum on cobalt chloride(CoCl2)-induced hypoxia injury of human retinal pigment epithelial cells(ARPE-19), so as to explore whether astragalus can improve diabetic retinopathy(DR)by anti-oxidative stress.
METHODS: The ARPE-19 hypoxia model induced by CoCl2 was established and divided into the following 5 groups: normal group(cells were cultured normally without any treatment), hypoxia model group(200μmol/L CoCl2), blank serum group(200μmol/L CoCl2+blank serum), low-dose drug-containing serum group(200μmol/L CoCl2+10% medicated serum)and high-dose drug-containing serum group(200μmol/L CoCl2+20% medicated serum); CCK-8 detects cell viability; Detect the levels of reduced glutathione(GSH)and malondialdehyde(MDA)in the cell supernatant with a kit; ELISA was used to detect the content of hypoxia-inducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF)in cell culture medium; Real-time quantitative PCR(qPCR)to detect the mRNA levels of VEGF, HIF-1α and Prolyl hydroxylase-2(PHD-2); The expressions of VEGF, HIF-1α and PHD-2 were detected by Western Blot.
RESULTS: Hypoxia model of ARPE-19 can successfully establish by CoCl2 at 200μmol/L. Low-dose and high-dose astragalus-containing serum could inhibit hypoxia-induced ARPE-19 proliferation(P<0.05), increase the GSH level and reduce the MDA content in ARPE-19 with hypoxic injury(P<0.05). Low-dose and high-dose astragalus-containing serum could inhibit the expression of HIF-1α and VEGF in ARPE-19 hypoxic injury supernatant(P<0.05), as well as the mRNA and protein expressions of VEGF, HIF-1α and PHD-2 in ARPE-19(P<0.05).
CONCLUSION: Low-dose and high-dose astragalus-containing serum alleviates the hypoxia injury of ARPE-19 induced by CoCl2 through anti-oxidant effect.
6.Analysis of the status and influence factors of road traffic injuries in children of Shashi District, Jingzhou City

Objective To investigate the status and influencing factors of road traffic injuries among children aged 0-17 in Shashi District, Jingzhou City, and to provide a basis for formulating strategies and measures to prevent road traffic injuries in children. Methods A staged random sampling method was used to select preschool children from 8 communities, and students from 8 primary schools, 8 junior high schools and 4 senior high schools, respectively, in Shashi District of Jingzhou. A questionnaire survey was conducted on the children on road traffic safety, and the incidence of children's traffic injuries and their cognition and behavior were analyzed. Results A total of 8 299 children completed the survey. The incidence rate of non-fatal road traffic injuries was 1.07%. Children aged 11-15 years had the highest incidence of road traffic injuries among all age groups, accounting for 66.29% of the total injuries, followed by the 16-20 years old group. The main risk factors of road traffic injuries in children were not taking the special lane when riding a bicycle, running red lights, using mobile phones when walking, and not wearing a helmet when riding a motorcycle. Walking was the main way for students to have road traffic injuries. Conclusion Children's road traffic injuries became a social problem that should not be ignored. Students above fourth grade were the key population for children's road traffic injury intervention in Shashi District. Road traffic safety publicity and intervention should be carried out for children of different ages and genders.
7.Application of pulsed-field gel electrophoresis in a food-borne outbreak of Salmonella serotype Muenchen infection.

OBJECTIVETo investigate the application of pulsed-field gel electrophoresis (PFGE) in food-borne outbreak.
METHODSPathogens were isolated and further characteristics identified by traditional methods. The strains isolated were carried out with molecular typing with using PFGE. PFGE was performed by Laboratory Directions for molecular subtyping of Salmonella by PFGE (CDC, USA) and the results of PFGE were analyzed by BioNumerics soft.
RESULTSTotally 14 Salmonella serotype Muenchen strains were isolated from 19 patients, 3 of 9 suspicious foods were positive for S. muenchen and 7 strains were isolated from 18 cooks. The biochemistry characterization and antimicrobial susceptibility of all the strains isolated were the same. 23 S. muenchen isolates were all shown indistinguishable by PFGE.
CONCLUSIONPFGE should play a key role in identifying the outbreak-associated isolates and distinguishing them from unrelated sporadic isolates. It might also demonstrate that the genetic fingerprints of serotype Muenchen isolates derived from patients were indistinguishable from those derived from drinks. PFGE might provide precise information on bacterial food-borne pathogens, promptly identify the source of infection, and effectively prevent from spreading. It should be one of the early warning method on controlling outbreak of the food-borne disease.

8.Research progress on the application of resting-state functional magnetic resonance imaging in glaucoma

Glaucoma is the world's No.1 irreversible disease causing blindness. It is one of the neuroophthalmic diseases characterized by an optic nerve injury and visual field defect. Numerous studies have shown that the damage caused by glaucoma is not only limited to the apoptosis of local visual ganglion cells in the eye, but also involves the entire visual pathway of the cerebral visual cortex, which is a neurodegenerative disease of the entire brain. Recently, with the rapid development of functional magnetic resonance imaging(fMRI), anatomy and function of nervous system are combined, so that the damage of local brain tissue corresponds to the lack of brain function, which helped in achieving the observation of anatomical structure and functional changes of the central nervous system under non-invasive conditions, thus providing the effective evidence for the pathological mechanism in glaucoma's research. Being a popular research technology in neuroimaging, resting-state functional magnetic resonance imaging(rs-fMRI)provides important evidences for exploring the anatomical structure, metabolism and functional changes of vision-related brain regions in patients with glaucoma. This review mainly discusses the principle of rs-fMRI and some common analysis methods such as amplitude of low-frequency fluctuation(ALFF), regional homogeneity(ReHo), degree centrality(DC)and functional connectivity(FC)in the application and research progress in glaucoma.
9.New progress on the pathogenesis research of primary angle-closure glaucoma

Primary angle-closure glaucoma(PACG)is the most common type of glaucoma in clinical. It is characterized by progressive and irreversible development with high global incidence and high possibility of blindness. The pathogenesis of PACG is complicated. It is not only associated with the abnormality of the ocular anatomy, such as the small cornea, short eye axis and shallow anterior chamber, but also found that the abnormality of the lens, iris, ciliary body and choroid are related to the pathogenesis of PACG in recent years. Moreover, the occurrence of PACG is also closely related to genetic and psychological stress factors. Our study summarized some literatures and made a brief discussion on the pathogenesis of PACG from the three aspects of anatomical characteristics, genetic factors, and psychological stress. We hope to provide useful theoretical support for the early diagnosis and treatment of glaucoma.
10.Establishment and evaluation of clinical nursing pathway after glossectomy and forearm flap reconstruction in patients undertaking tongue cancer surgery

Objective To research the significance of application of clinical nursing pathway in patients undertaking glossectomy and forearm flap reconstruction.Methods Sixty patients with tongue cancer who undertook glossectomy and forearm flap reconstruction were enrolled.The patients were randomly divided into the experimental group and the control group (30 cases in each group).The patients of the experimental group received nursing following clinical nursing pathway,and the patients in the control group received routine nursing service.The patients and doctors satisfaction evaluation were compared between the two groups with a self-designed nursing quality evaluation form.The qualities of care achieved in 2 groups were compared.Results It took less in-ward time and expenditure in the experimental group compared with the control group.The quality of care,the satisfactory degree of patients and doctors in the exipermental group were significantly higher than those of the control group.Conclusions The establishment and application of the clinical nursing pathway in patients undertaking glossectomy and forearm flap reconstrucition contributes to improve the satisfaction of patients and doctors and the quality of nursing service.
