1.Progress in the tumorigenic potential of iPSCs and methods to reduce it.

Since Yamanaka successfully reprogrammed murine fibroblasts into iPSCs in 2006, iPSCs technology has drawn much attention worldwide. Although iPSCs provides tremendous possibilities for both basic research and regenerative medicine, it has meanwhile potential risks, e.g. tumorigenicity. Scientists, therefore, have made efforts in clarifying the mechanism of the cause for iPSCs tumorigenicity and the way how to reduce the risk. The results of some researches reveal some of tumorigenic factors, e.g. the partial similarity of gene expression profiles between cancer cells and iPSCs, the accumulation of the genetic damages in the course of reprogramming process, and mutation in the cellular culture. As a consequence, numerous methods for reducing iPSCs tumorigenicity have been explored, such as minimized use of the reprogramming factors at the controlled manner, and the selection of the expression vector or parental cells. In this paper, the cause of iPSCs tumorigenicity and the current achievements on preventing iPSCs tumorigenesis are reviewed.

2.Analysis of drug resistance on extended spectrum β-lactamases-producing Escherichia coli and Klebsiella pneumoniae

Objective Analysis of clinical distribution and susceptibility to antimicrobial agents in producing extended spectrumβ-lactamases (ESBLs)of Escherichia coli(ECO)and Klebsiella pneumoniae (KPN).Methods The samples from 2013 April to 2014 December in Changji Prefecture People′s Hospital were cultured and identified,drug sensitivity test did by VITEK2compat au-tomatic bacteria analysis,phenotypic confirmatory test was did on producing ESBLs ECO and KPN,and the clinical distribution and drug resistance was analyzed.Results 1 576 strains of ECO and KPN isolated 306 strains of producing ESBLs strains and 343 strains of multi drug resistant bacteria,the isolation rates were 1 9.4% and 21.8% respectively.In 576 strains of ECO,there were 1 77 strains of producing ESBLs strains and 202 strains of multi drug resistant bacteria were isolated,the separation rates were 30.7% and 35.1% respectively,generic drug resistant strains were not found,and those producing ESBLs strains were isolated mainly from the urine specimens (39.5%),and mainly from the ICU (37.3%)and (29.9%)the surgical ward.There were 129 strains of producing ESBLs strains,141 strains of multi drug resistant bacteria and 4 generic drug resistant strains were isolated from 1 000 strains of KPN,the separation rates were 12.9%,14.1% and 0.4% respectively,and those producing ESBLs strains were isolated mainly from sputum specimens (83.7%),and mainly from the ICU (42.6%)and(29.5%)in the department of inter-nal medicine.Isolation of producing ESBLs ECO and multi drug resistant bacteria were higher than KPN,the difference were statis-tically significant (P <0.05).The sensitivities of ECO and KPN ESBLs strains to carbapenems imipenem andaminoglycosides ami-kacin and compound preparation piperacillin/tazobactam were good,the other antimicrobial drug resistance rates were more than 50.0%.Conclusion ESBLs ECO mainly causes urinary tract infections,KPN mainly causes respiratory tract infection.Detection rates of producing ESBLs ECO and KPN were high,and the multi drug resistance is serious,should strengthen the monitoring of drug resistance on producing ESBLs strains,and according to the results of drug susceptibility,standardize the rational use of anti-microbial agents,and to strengthen the management of antimicrobial agents,delay the occurrence of bacterial resistance.
3.Imaging Diagnosis of Primary Brain Iymphoma

Objective:To describe the imaging findings of primary brain lymphoma in immunologically normal patients.Methods:The clinical manifestations and imaging findings of 5 patients operated with pathological proved primary brain lymphoma were reviewed.Results:In 5 cases,4 patients were men and one was woman.The median age was 47(range 34 to 63 years).Pathological diagnosis was compatible with primary non-Hodgkins lymphoma of brain for all five patients.Primary brain lymphoma may be soiltary or mutiple and locate at supratentorial or infratentorial.Lesions presented as an isodense to hyperdense on CT scan,hypointense on T1-weighted images and isointense or hyperintense to gray matter on T2-weighted images.Most lesions showed dense homogenous enhancement on CT and MR.Tumors associated with different degrees of mass effect.One patient received whole-body 18 FDG PET examination that presented as an intracranial hypermetabolic lesion.Conclusion:Primary brain lymphoma is a rare tumor that imaging findings overlap with those of other intracranial mass lesions,we can only make a suggestive diagnosis of primary brain lymphoma and pathological confirmation is meeded.
4.Transitional care based on the Omaha system in postoperative rehabilitation of patients with breast cancer

Objective To explore the application of transitional care based on the Omaha system in postoperative rehabilitation of patients with breast cancer.Methods Seventy-two patients after radical mastectomy were assigned into two groups according to admission time,patients from June to December 2011 were set as the control group,patients from February to December 2012 were assigned to the intervention group,each group contained 36 patients.The patients in the control group received routine nursing,while patients in the intervention group were implemented transitional care program with Omaha system as framework from before discharge to 6-week follow-up after discharge from hospital.Nursing effect was compared between two groups from three aspects such as cognition,behavior and status.Results Twenty-three problems were found in the 6-week follow-up for the intervention group,including environmental domain,psychosocial domain,physiological domain and health-related behavior domain.Knowledge,behavior and status scores of the intervention group 6 weeks after discharge were significantly higher than those of the control group [(3.92 ± 0.48) scores vs.(2.41 ± 0.17) scores,(4.11 ± 0.49) scores vs.(2.83 ± 0.25) scores,(3.09 ±0.52) scores vs.(2.32 ±0.39) scores,t=14.81,15.36,9.42,P<0.01 or 0.05].Conclusions Transitional care based on the Omaha system shows common problems in rehabilitation of patients with breast cancer after operation,and provides standardized program for rehabilitation nursing.It can achieve better nursing effect.
5.Progress in improvement of lentiviral vectors' transcriptional read-through.

Four out of 10 patients of X-linked severe combined immunodeficiency (X-SCID) were finally developed leukemia after receiving the treatment of gene therapy delivered by gamma-retroviral vectors. This is due to the vector integrated to the proximity of lmo2 etc proto-oncogene promoters, leading to the activation of onco-gene expression, which raises the concern of the bio-safety of gene therapy vectors. Lentiviral vectors, especially self-inactivating lentiviral vectors, are considered to be much safer than gamma-retroviral vectors. However self-inactivating lentiviral vectors also have encountered with some unsafe factors and one of them is the problem of transcriptional "read-through" . During the past years, achievements have been made to reduce lentiviral vector transcriptional read-through, which are reviewed herein.

6.Effects of Ehanol Extract of Rhizoma Phragmitis on Liver Glycogen Content and Glycogen Synthetase in Diabetic Mice

Objective To study the effects of ethanol extract of rhizoma phragmitis on liver glycogen content and glycogen synthetase (GS) in streptozotocin (STZ) induced diabetic mice. Methods The diabetic model mice were divided in-to model control group, high-dose group and low-dose group, 10 mice for each group. Another 10 normal mice were used as control group. The liver glycogen content was detected by histochemical staining of glycogen (PAS) method. The expression of GS mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blot assays. Results After PAS staining the hepatic glycogen content decreased significantly in model control group, and which was significantly increased in low-dose group and high-dose group compared with that of model control group (P<0.01). The hepatic glyco-gen content was the highest in high-dose group compared with that of other three groups. The levels of GS mRNA and GS protein were significantly lower in model control group than those of other three groups, which were significantly lower in low-dose group than those of high-dose group (P<0.05). Conclusion There is a dose-dependent effect of ethanol extract of rhizoma phragmitis on liver glycogen in STZ induced diabetic mice, which may be related with the increased expression of liver glycogen synthetase.
7.Analysis of distribution and drug resistance of Enterococcus faecium

Objective To investigate the distribution and drug resistance ofEnterococcus faecium,provide evidence for clinical rational use of antimicrobial agents.Methods Retrospective analysis of drug resistance of Enterococcus faecium,summarize the specimen type,distribution of departments and so on,to explore the trend of drug resistance.Results Enterococcus faecium to vancomycin resistance rate from 3.54% (4/113) in 2010,rose to 8.45% (18/213) in 2013.One strains of hnezolid resistant Enterococcus faecium 717 was found,the minimum inhibitory concentration of 7.9 mg/L,not tigecycline resistance in Enterococcus faecium found.Conclusion Vancomycin resistant Enterococcus faecium for linezolid and tigecycline susceptibility is still high,the two kinds of antimicrobial agents can be used for the treatment of vancomycin resistant Enterococcus faecium.
8.Prevalence of anemia among primary school students from Kazakh ethnic groups in agricultural and pastoral areas of Altay Prefecture, Xinjiang Uygur Autonomous Region

Objective:
To investigate the prevalence of anemia and its influencing factors among primary school students from Kazakh ethnic groups in agricultural and pastoral areas of Altay Prefecture, Xinjiang Uygur Autonomous Region, so as to provide insights into improvements of nutrition and prevention of anemia among students.
Methods:
Grade 1 to 3 Kazakh ethnic students were sampled using a cluster sampling method from 7 primary schools in agricultural and pastoral areas of Altay Prefecture, Xinjiang Uygur Autonomous Region from April to June, 2022. Demographics and dietary behaviors were collected using questionnaire surveys, and the height and body weight were measured, while peripheral blood hemoglobin was detected. Factors affecting anemia were identified using a multivariable logistic regression model.
Results:
A total of 2 378 questionnaires were allocated, and 2 214 valid questionnaires were recovered, with an effective rate of 93.10%. The respondents included 1 123 men (50.72%) and 1 091 women (49.28%), and had a mean age of (8.12±1.41) years. The prevalence of anemia was 22.49%, and the detection rates of mild and moderate anemia were 15.31% and 7.18%, with no severe anemia detected. Multivariable logistic regression analysis identified frequency of eating eggs (0, OR=2.532, 95%CI: 2.114-3.015; 1 to 3 times/week, OR=1.926, 95%CI: 1.648-2.216), frequency of eating meat (1 to 3 times/week, OR=1.468, 95%CI: 1.256-1.689; 4 to 7 times/week, OR=2.212, 95%CI: 1.823-2.641), frequency of healthy check-up (0, OR=2.872, 95%CI: 2.523-3.231; 1 time/year, OR=1.826, 95%CI: 1.546-2.108), monthly mean household income (1 500 Yuan and lower per month, OR=1.324, 95%CI: 1.071-1.582; 1 501 to 3 000 Yuan per month, OR=1.124, 95%CI: 1.029-1.221) and caregiver (grandparents, OR=1.175, 95%CI: 1.038-1.324; others, OR=1.241, 95%CI: 1.032-1.466) as factors affecting anemia.
Conclusions
The prevalence of anemia is 22.49% among primary school students from Kazakh ethnic groups in agricultural and pastoral areas of Altay Prefecture, Xinjiang Uygur Autonomous Region, with mild anemia as the predominant type. Dietary behaviors, frequency of healthy check-up, economic status and caregiver may affect the development of anemia.

10.EIectroacupuncture effects on bladder capacity and tissue morphology of the neurogenical bladder after sacral spinal cord injury

BACKGROUND:Spinal cord injuries induced by sacral segment and lumbar lesions can damage the primary sacral micturition center or its surrounding nerves (parasympathetic and somatic nerve) to cause detrusor areflexia, thereby leading to urinary retention and further pathological changes in bladder tissue morphology.
OBJECTIVE:To observe the effects of electroacupuncture at the Ciliao (BL32), Zhongji (RN3), Sanyinjiao (SP6) acupoints on bladder capacity and bladder tissue morphology of rats with urinary retention after sacral spinal cord injury.
METHODS:Ten of 40 female Sprague-Dawley rats were randomly selected as blank group, and other 30 rats were randomly divided into model group, acupoint group and non-acupoint group. In the model group, the rats received no electroacupuncture. In the non-acupoint group, acupuncture and electroacupuncture at non-acupoint points were performed, respectively, for 20 minutes. In the acupoint group, acupuncture and electroacupuncture were performed at the Ciliao, Zhongji, and Sanyinjiao acupoints. Bladder capacity was measured at days 14 and 22 after treatment. After treatment, bladder tissues were extracted for morphological observation with hematoxylin-eosin staining.
RESULTS AND CONCLUSION:(1) The maximum bladder capacity in the acupoint group was obviously reduced after treatment (P<0.01);the curative effect in the acupoint group was more obvious than the non-acupoint group (P<0.05);the difference between preoperative and postoperative bladder capacity (d value) was more significant in the acupoint group than the non-acupoint group (P<0.05). (2) Compared with the model and non-acupoint groups, morphological changes in the bladder tissue were improved more significantly in the acupoint group. These findings indicate that electroacupuncture at the Ciliao, Zhongji, and Sanyinjiao acupoints can effectively reduce the maximum bladder capacity and repair the damaged bladder tissue after sacral spinal cord injury.
