1.Relationship between the T3394C mutation of the mitochondrial DNA ND1 gene and type 2 diabetes mellitns in the elderly Chinese Han people in Zhejiang

ObjectiveTo explore the relationship between the T3394C mutation of the mitoehondrial DNA ND1 gene and type 2 diabetes mellitus (T2DM) in the elderly. MethodsDirect sequencing of PCR product was applied to study the T3394C mutation of mitochondrial DNA in 340 unrelated individuals in T2DM patients (90 cases of elderly patients, 250 cases of non-elderly patients) and 265 normal controls (130 cases of elderly controls, 135 cases of non-elderly controls) from Zhejiang province. The mutation was analyzed by DNASTAR and Antheprot 5.0 softwares. ResultsThe T3394C mutation of mitochondrial DNA was found in 5 of 90 elderly T2DM patients (5.6%),1 of 130 elderly controls(0.8%), and 2 of 250 non-elderly T2DM patients (0.8%). The incidence of the mutation was significantly different between the elderly T2DM patients and elderly controls(P<0.05), as well as between the elderly T2DM patients and non-elderly T2DM patients(P<0.05). The T3394C mutation caused the change of the ND1 protein secondary structure. ConclusionsThe T3394C mutation of the mitochondrial DNA ND1 gene may contribute to the pathogenesis of T2DM in the elderly with other genetic and environment factors.
2.Clinical observation on acupuncture for 80 patients with abdominal obesity in Germany: based on the theory of unblocking and regulating the Belt Vessel

Objective: To observe the efficacy of unblocking and regulating the Belt Vessel acupuncture method for patients with abdominal obesity in Germany and its influence on the body fat parameters and glycolipids metabolism. Methods: A total of 82 cases of abdominal obesity in Germany were selected. There were two dropout cases in the treatment process. All of the patients received acupuncture at Daimai (GB26), Daheng (SP15), Tianshu (ST25), Zhongwan (CV12), Shuidao (ST28), Waiguan (TE5), and Zulinqi (GB41). The patients received acupuncture therapy once every 2-3 d, 3 times a week. The treatment duration lasted 4 weeks. Before and after treatment, the body mass (BM), waist circumference (WC), abdominal circumference (AC), hip circumference (HC), total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and fasting blood glucose (GLU) were measured respectively, and the body mass index (BMI), waist-to-hip ratio (WHR), waist-to-height ratio (WHtR), and body fat rate (BFR) were calculated. Results: After treatment, the total effective rate was 72.5%; the levels of BM, WC, AC, HC, BMI, WHR, WHtR, BFR, TC, LDL-C, and GLU declined significantly compared with those before treatment (P<0.01), while the TG and HDL-C slightly declined, but the differences were statistically insignificant (P>0.05). Conclusion: The unblocking and regulating the Belt Vessel acupuncture method can significantly reduce body fat parameters such as BM, AC, and BFR, and correct the disorders of glucose and lipid metabolism in patients with abdominal obesity in Germany.
4.Expression of influence of continuous blood purification on the endothelin-1 and prognosis value in the neurogenic pulmonary edema

Objective To explore the influnce on the endothelin-1 (ET-1) and clinical application value of continuous blood purification (CBP) on the treatment of severe craniocerebral injury with the neurogenic pulmonary edema (NPE).Methods All data about sixty patients with NPE were prospectively studied.These 60 patients were randomly (random number) divided into control group (n =30) and treatment group (n =30).In control group,patients were rapidly given with lowering intracranial pressure,mechanical ventilation,calming,antibiotic therapy and so on.In the treatment group,patients received CBP integrated with routine treatment.On admission and 72 h posttreatment,ET-1,static lung compliance and oxygenation index were observed.Time of mechanical ventilation support,incidence rates of multipal organ dysfunction syndrome (MODS) were compared between two groups.The paired t-test was used for the amount data within the group.Chi-square was used for the constitute ratio and incidence ratio of the each relevant information.P < 0.05 was considered statistically significant.Results Compared to the control group,the level of ET-1 was decreased significantly in the treatment group [(48 ± 10) ng/L vs.(85 ± 14) ng/L] after 72 h post-treatment,while static lung compliance [(60.9 ± 2.3) mL/cmH2O vs.(31.4 ±4.8) mL/cmH2O] and oxygenation index [(317 ± 11) mmHg vs.(192 ± 14) mmHg] increased significantly (P < 0.05).In treatment group and control group,the time of respirator intervention were [(6.0 ± 2.1) d vs.(11 ± 3.2) d],and the statistical significance was shown (P < 0.05).Compared to the control group [56.7% (17/30)],incidence rate of MODS [20.0% (6/30)] was lower in treatment group (P < 0.05).Conclusions CBP combined with routine treatment,which can remove ET-1 effectively,improve oxygenation,reduce the time of mechanical ventilation support and incidence rate of MODS.
5.Roles of HOXC gene in tumors

The protein encoded by the HOXC gene regulates multiple processes during embryonic development,including cell growth,differentiation,apoptosis,migration and angiogenesis.The HOXC gene is also involved in the occurrence and development of various human solid tumors,such as colorectal cancer,gastric cancer,liver cancer,breast cancer,endometrial cancer,etc.In addition,the HOXC gene is closely related to the clinical stage and distant metastasis of the tumor.Thus,the HOXC gene can be used as a biological predictor of tumor metastasis and prognosis.

Objective To explore the influence of parenteral nutrition (PN)supportive therapy on the nutritional status and immune function in patients with advanced gastrointestinal tu-mor in peri-chemotherapy period.Methods 90 patients with advanced gastrointestinal tumor were randomly divided into control group and observation group.Efficacy was compared between two groups by detecting the changes of various nutritional and immune indexes before and after chemotherapy.Results There were no significant differences of the nutritional and immune func-tion indexes between two groups before chemotherapy and after the first-second cycle of chemothera-py.All nutritional and immune function indexes after the third cycle of chemotherapy in the obser-vation group were significantly better than those in the control group.The incidence rate of compli-cations such as leucopenia,thrombocytopenia,nausea,vomiting and diarrhea in the observation group was significantly lower than the control group.Conclusion PN supportive therapy can not only ameliorate the nutritional status and immune function of patients with advanced gastrointestinal tumor,but also alleviate the chemotherapy-induced adverse reactions,so it is very important for safety in peri-chemotherapy period.
