1.Effects of Aripiprazole on PC12 Cell Injury Induced by Aβ25-35 and Its Mechanism

OBJECTIVE:To study the effects of aripiprazole on PC12 cell injury induced by amyloid β-protein(Aβ25-35)and its mechanism. METHODS:PC12 cells were randomized into normal control group,model group (20 μmol/L Aβ25-35),aripiprazole low-concentration,medium-concentration and high-concentration groups(5,10,20 μmol/L aripiprazole+20 μmol/L Aβ25-35). These groups were cultured with culture medium containing relevant medicine for 48 h,with 6 wells in each group. The viability(optical density value)of PC12 cell was measured by MTT assay,and PC12 cell apoptosis was measured by Hoechst staining. The activi-ties of Caspase-3 and Caspase-9 were determined by spectrophotometry. The protein expression of Bcl-2,Bax and PI3K and the phosphorylation of Akt were assayed by Western blot assay. RESULTS:Compared with normal control group,optical density value of model group was decreased while apoptotic rate was increased;the activities of Caspase-3 and Caspase-9,and the protein expres-sion of Bax were increased;the protein expression of Bcl-2 and PI3K,the phosphorylation of Akt were decreased(P<0.01). Com-pared with model group,optical density value of aripiprazole low-concentration,medium-concentration and high-concentration groups were increased,while apoptotic rate and the activities of Caspase-3 and Caspase-9 were decreased;the protein expression of Bcl-2 and PI3K and the phosphorylation of Akt were enhanced;while the protein expression of Bax were decreased in aripiprazole medium-concentration and high-concentration groups(P<0.05 or P<0.01). CONCLUSIONS:Aripiprazole can suppress cell apop-tosis of PC12 cell induced by Aβ25-35,which is related to activating PI3K/Akt signal pathway.
2.The expression of DNA methyltransferase,demethylase and tumor-associated genes in human gastric cancer

Objective To investigate the relationship of the expression of DNA methyltransferase,demethylase(mbd2) and tumor-associated genes in human gastric cancer. Methods Tissue samples of cancerous,para-cancerous and normal gastric mucosa were obtained surgically from 28 primary gastric cancer patients. The transcription level of DNA methyltransferase 1 (DNMT1),mbd2,methyl-CpG binding protein (MeCP2),p16 INK4A and c-myc were determined by using real-time RT-PCR and RT-PCR. The relationship between the expression of DNA methylation-associated genes and tumor-associated genes was analyzed. Results The mRNA level of DNMT1 was higher and the mRNA level of mbd2 gene was lower in cancerous tissue than that in normal tissue. The expression of c-myc instead of p16 INK4A and MeCP2 was increased in cancer tissues. The mRNA level of c-myc related negatively to mbd2 when gastric cancer developed. However,there was no any close relation between the transcription level of all above genes and tumor biological behavior in human gastric cancer. Conclusion This study indicates that MeCP2 but not DNMT1 may contribute to the regulation of tumor-associated genes expression in human gastric cancer.
3.Meta-analysis of photodynamic therapy and intravitreal injection of anti-vascular endothelial growth factor agent in the treatment of polypoid choroidal vasculopathy

Objective To systematically review the efficacy and safety of photodynamic therapy (PDT) and intravitreal vascular endothelial growth factor (VEGF) inhibitors in the treatment of polypoidal choroidal vasculopathy (PCV),and to investigate the primary treatment tentatively.Methods A systematic search of Pubmed,Embase,the Cochrane Library and the Wanfang Data was performed to identify all comparative studies that compared the outcomes of PDT alone,intravitreal VEGF inhibitors alone and combined intravitreal VEGF inhibitors and photodynamic therapy.Outcomes of interest included the regression and recurrence rate of polypoidal lesions,best corrected visual acuity (BCVA),central retinal thickness (CRT),therapeutic times,and the occurrence rate of adverse events.2 randomized controlled trials (RCT) and 19 non-RTCs were identified.According to treatment methods,the data extracted was classified to 3 groups,analyzed with odds ratio (OR),weighted mean difference (WMD) and 95%confidence interval (95%CI).Results Meta-analysis suggests that the regression rate of polypoidal lesions (OR=0.34,0.07;95%CI=0.13-0.88,0.02-0.36) and BCVA (WMD=0.25,0.11;95%CI=0.14-0.36,0.01-0.21) in combined therapy group were significantly better than those in PDT group and intravitreal VEGF inhibitors group (P<0.05).The recurrence rate of polypoidal lesions in PDT group was significantly lower than intravitreal VEGF inhibitors group (OR=0.35,95%CI=0.16-0.74,P=0.006).BCVA (P=0.025) and the occurrence rate of adverse events (OR=60.36,95%CI=6.04-603.50,P=0.000 5) in intravitreal VEGF inhibitors group were significant better than PDT group.Conclusions Combined treatment appeared to be superior to PDT alone or intravitreal VEGF inhibitors alone.Combined treatment takes priority over all others in the primary treatment of PCV.
4.Hemivertebra resection only via posterior approach and short-segment transpedicular instrumentation for congenital scoliosis

[Objective]To evaluate surgical outcomes of hemivertebra resection only via posterior approaches followed by short-segment transpedicular instrumentation for correction of the congenital scoliosos. [Method]A group of patients with a single hemivertebra between the ages 5-16 years who underwent operative treatment were evaluated.Hemivertebra resection through posterior approach and short-segment transpedicular instrumentation were used for correction of the scoliosis and kyphosis deformities.Radiographic evaluation were conducted before and after operation and at follow-up visits.Cobb's angles of the segmental curve,total main curve,cranial and caudal compensatory curves and kyphosis were measured on the posteroanterior and lateral radiographs.[Result]The cases in this study showed satisfied results.The mean Cobb's angle of segment curve was 41.5? before surgery and 15.1? after surgery with a 63.6% correction,and 14.7? at the lastest follow-up assessment with a 64.6% correction;total main curves improved from 46.9?to 18.4?,with a 60.8% correction;17.5?at last follow-up,with a 62.7% correction;the correction ratio for kyphosis was from 15.4?kyphosis to normal physical profile;cranial and caudal compensartory curves were obviously improved.[Conclusion]Hemivertebra resection and short-segment transpedicular instrumentation could be performed only through posterior approach,which had exerted satisfactory correction on congenital scoliosis.If the operation is performed before the maturity of the bone,it will effectively prevent the formation of secondly spinal deformities.
5.Transcutaneous Injection of MIIGX3 Artificial Bone and Methylprednisolone for Treatment of Recurrent Bone Cysts

Objective To investigate the efficacy of transcutaneous injection of MIIGX3 artificial bone and methylprednisolone for recurrent bone cysts.Methods From January 2004 to March 2006,a total of 13 children with recurrent bone cysts received transcutaneous injection of methylprednisolone and MIIGX3 in our hospital.X-ray was employed to detect the degradation of MIIGX3 and formation of new bones.Results The operation time ranged from 30 to 95 minutes(mean 42 minutes).The patients were followed up for 1 to 3 years.None of them had recurrence of bone cysts during this period.The artificial bones were completely degraded and replaced by new bones in 1.5 years after the injection.Conclusions Transcutaneous injection of methylprednisolone and MIIGX3 is effective for recurrent bone cysts.Patients have shorter operation time and hospital stay after this treatment because the procedure is microinvasive.
6.Prevalence of Mild Cognitive Impairment Among Old People in Urban and Rural Areas of Guizhou Province

Objective:To investigate the prevalence of mild cognitive impairment (MCI) among elderly people aged 60 and over in urban and rural areas of Guizhou Province. Methods:A total of 4535 residents aged 60 and over was selected by random cluster sampling in Giuyang City and Zheng'an County. All participants were interviewed with questionnaires, they also received physical examination for related information, and their cognitive function was assessed by the mini-mental state examination (MMSE). Frisoni's criteria for MCI, the score of MMSE below one standard deviation matching age and education was used to this investigation. Chi-square test and u test were used in data analysis. Results: The total prevalence of MCI among all elderly people aged 60 years and over was 15.4%. Prevalence for urban community was higher than that of rural area (16.3% vs. 13.2%,?2=6.71, P=0.01). Objective To evaluation the comprehensive acquired immunodeficiency syndrome (AIDS) intervention program conducting in men who have sex with men (MSM) population living in Inner Mongolia Autonomous region. Methods A comprehensive intervention program was carried out among MSM groups in Hohhot and Baotou City. Seven hundred and six and 767 MSM were enrolled for investigation through the snowball method during May 2008 and May to June 2009, respectively.At the same time, 5 mL intravenous blood sample was collected from each subject for human immunodeficiency virus (HIV) detection and intervention effect evaluation. Data were analyzed by x2test. Results After intervention, the awareness rate of HIV/AIDS knowledge increased from 70.7%(499/706) to 81.7%(627/767) (x2 =25. 004, P<0.01 ). The rate of consistent condom use in the last six months increased from 38.0%(254/668) to 45.3% (346/764) (x2=8. 269, P<0. 05). The percentage of subjects who had more than 5 different male sexual partner during the last 6 months significantly decreased from 42.8% to 24.6% (x2 = 55. 348, P<0.01). The median number of sexual partners in MSM decreased from 3 in 2008 to 2 in 2009. The HIV infection rate was 1.7 % in both 2008 and 2009. Conclusion The comprehensive intervention program among MSM population shows positive influence after one year implementation, which is helpful for increasing HIV/AIDS awareness and safe sex behavior in this population.

9.Treatment of floating knee injury in children.

The necessity and superiority of the surgical operation on children with floating knee injury and the fracture union and complications were investigated. Twenty-eight children with floating knee injury were subjected to open reduction and internal fixation or external fixator. The patients were followed up for 18 months to 7 years. The curative effectiveness was scored by Karlstrom criteria. The results showed that no nonunion or deformity was found. The affected limb was 1.2 cm to 1.5 cm longer in 2 cases, 0.8 to 1.2 cm shorter in 3 cases than the contralateral. No severe dysfunction of knee joint occurred. The excellent-good rate was 92.8% and the curative rate 71.4% respectively. So for children whose age is older than 5 years, it's a good way to treat the fractures of femur and tibia with open reduction and internal fixation or external fixator. The method can be advantageous for the nursing care, early function recovery, shortening of the hospital stay and avoidance of severe complications.

10.Injectable bone graft combined with percutaneous reduction by leverage for treatment of calcaneal fracture: 8 cases report

A retrospective analysis was performed among 8 patients with unilateral intraarticular calcaneal fracture, who were selected from the Department of Orthopaedics in Yangpu District Central Hospital of Shanghai. They were treated with the injectable bone graft MIIG[R] X3 combined with percutaneous reduction by leverage and axial fixation from June 2005 to November 2006. After treatment, the function of affected feet was evaluated according to the ankle-hindfoot score of the American Orthopaedic Foot & Ankle Society (totally 100 points). All the patients were rechecked with X-ray films periodically to observe the reaction between the materials and the host, the healing of fracture, the pain of limbs and joint motion. Eight patients were all followed up, the complication rate reached 12.5% (1/8), the improvement of Bohler angle excelled 18° to 25°, and that of Gissane angle exceeded 30° to 35°. Among the included patients, 6 cases achieved excellent effect and 2 cases good. All developed the bone union. None of all had complications, such as necrosis of skin, hematoma and deep infection.
