2.Accuracy of tunnel-type template assisting mini-implant anchorage surgery based on multislice spiral computed tomography

BACKGROUND: Preplant diagnosis and rigorous implant strategy based on multislice spiral computed tomography (MSCT) imaging can improve success rate of mini-implants as anchorage. Effective application of imaging information in implant anchorage requires reliable tools and methods.OBJECTIVE: To investigate the accuracy of the radiographic and tunnel-type surgical templates assisting implant anchorage surgery and evaluate the clinical significance.METHODS: The transparent radiographic templates were made with occlusal rest to assist MSCT to scan two pigs' skulls locatively. After designed the implant anchorage angles in MSCT images accurately, we made the tunnel-type surgical template by angle data. A total of 32 miniscrews were implanted by surgical template, and the two pigs skulls were rescanned by MSCT. After dissected the pig alveolar process, the MSCT preoperative planning angles, the MSCT postoperative measurement angles and the measurement angles of anatomic bone samples were analyzed statistically. RESULT AND CONCLUSION: The 32 miniscrews were implanted successfully. They did not damage dental roots or surrounding important structures. Compared MSCT preoperative planning angles with MSCT postoperative measurement angles and anatomic bone samples measurement angles, there were no significant differences on axial sections or cross-sections and the r concordance correlations were very good. Results show that the radiographic and the tunnel-type surgical templates are accurate and reliable for operation and will help to improve the success rate of implant anchorage.
3.Nursing cooperation of implantation of modified capsular tension ring and foldable posterior chamber intraocular lens combined with phacoemusification in treatment of Marfan syndrome accompahied with lens subluxation

Objective To explore the nursing care of patients with Marfan syndrome accompanied with lens subluxation, and the clinical efficacy of phacoemusification combined with the implantation of modified capsular tension ring and foldable posterior chamber intraocular lens. Methods 11 patients (11 eyes) were performed the surgery. Uncorrected visual acuity preoperatively was (0.13±0.09). Best corrected visual acuity preoperatively was (0.20±0.17). Preoperative preparation, intraoperative observation and postoperative guidance were executed. The prognosis of these patients were observed. Results The operations were performed successfully. No intraoperative complications were founded. Mean uncorrected visual acuity at 2 year postoperatively was (0.60±0.28), and mean best corrected visual acuity was (0.70±0.28). Conclusions Careful preparation before surgery, close cooperation during surgery and the correct management after surgery were the important measures for the success of surgery.
4.Involvement of miR-106b in endothelial cells-mediated angiogenesis

Objective To study whether miR-106b is involved in endothelial cells-mediated angio-genesis .Methods miR-106b cultured and transfected with endothelial cells was divided into miR-106b group ,blank control group and positive control group .RNA was extracted from miR-106b .The transfection efficiency was confirmed by inverse transcription .Endothelial cell tubes formed in matrigel were observed .Apoptosis of transfected miR-106b was assayed by TUNEL assay .Target genes of miR-106b were detected .Expressions of miR-106b and candidate genes were detected by RT-PCR and Western blot ,respectively .Results The number of endothe-lial cell tubes formed in matrigel was significantly less in miR-106b group than in blank control group and positive control group .The ratio of formed tubes ,signal transduction and mRNA ex-pression level were significantly lower in miR-106b group than in positive control group ( P<0.05 ,P<0.01) .No significant difference was found in apoptosis of transfected miR-106b among the 3 groups (1.19% vs 3 .39% ,P>0 .05) .Conclusion miR-106b inhibits endothelial cells-medi-ated angiogenesis by down-regulating the signal transduction and STAT 3 ,which is not directly re-lated with VEGFA .
5.Statins decreases expression of five inflammation-associated microRNAs in the plasma of patients with unstable angina

Objective:To explore the influence of treatment with HMG-CoA reductase inhibitors ( sta-tins) on the expression profile of microRNAs ( miRNAs) in the plasma of patients with unstable angina ( UA) .Methods:The Taqman low-density miRNA array ( TLDA) and significance analysis of microar-rays ( SAM) were used to identify distinct miRNA expression profiles in the plasma of UA patients treated with long-term and regular statins ( UA receiving statins , n=6 ) compared with UA patients who had not received statins therapy before ( UA received no statins , n=6 ) .These differentially expressed miRNAs discovered in the profiling were further validated by real-time PCR in another 20 controls with non-cardiac chest pain , 26 UA patients received no statins , and 19 UA patients received statins .Results: By using TLDA and SAM , significantly decreased expression levels of 21 miRNAs were observed in the UA pa-tients receiving statins compared with those who received no statins ( fold change >3 and false discovery rate<0 .0001%) .The unsupervised hierarchical clustering based on miRNA expression clearly separa-ted the UA patients receiving statins from those who received no statins .Consistent with the profiling da-ta, the levels of 5 inflammation-associated miRNAs (miR-106b, miR-21, miR-25, miR-451, and miR-92a) were down regulated (P<0.05) in the UA patients receiving statins compared with those who re-ceived no statins.Conclusion: A group of inflammation-associated miRNAs, consisting of miR-106b, miR-21, miR-25, miR-451, and miR-92a, could be decreased by treatment with statins and may be used as a novel biomarker for effectiveness of statins therapy in patients with UA .

7.Effect of gender matching on the outcomes of living-donor renal transplantation

Objective To evaluate the effect of gender matching on the outcomes of livingdonor renal transplantation.Methods A total of 419 cases of living-donor renal transplantation in our center were divided into male-donor-male-recipient (MDMR) group,male-donor-female-recipient (MDFR) group,female-donor-male-recipient (FDMR) group,female-donor-female-recipient (FDFR)group.The outcomes including graft and patient survival,acute rejection and renal function were analyzed retrospectively.Results Compared to MDMR group,MDFR group and FDFR group had lower Scr [(80.7±17.9),(87.4±21.9) μmol/L vs (120.3±72.5) μmol/L,all P < 0.05] and uric acid (UA) [(318.1 ± 86.4),(303.5 ± 66.9) μmol/L vs (358.4 ± 77.8) μmol/L,P < 0.05] 6 months after operation.Compared to MDFR group,FDMR group had higher Scr[(117.7±27.4) μmol/L vs (80.7±17.9) μmol/L,P < 0.01],UA [(371.0±92.4) μmol/L vs (318.1±86.4) μmol/L,P < 0.05] and lower glomerular filtration rate (GFR) [(70.4± 17.8) ml/min vs (79.6± 18.9) ml/min,P < 0.05].Compared to FDMR group,FDFR group had lower Scr [(87.4±21.9) μmol/L vs (117.7±27.4) μmol/L,P < 0.01] and UA [(303.5±66.9)μmol/L vs (371.092.4) μmol/L,P< 0.01].Compared to MDFR group,FDFR group showed lower GFR [(72.4±25.3) ml/min vs (82.7± 18.7) ml/min,P < 0.05] 1 year after operation.Compared to MDMR group,FDFR group showed lower UA [(322.9±69.7) μmol/L vs (376.0±66.2) μmol/L,P < 0.05] 2 years after operation.Compared to FDMR group,FDFR group showed lower Scr [(88.7 ±27.0) μmol/L vs (112.7±27.8) μmol/L,P < 0.05] and UA [(318.3 ±61.2) μmol/L vs (396.2± 100.3) μmol/L,P < 0.05] 3 years after operation.5 years after operation,there were no significant differences in above indexes,the incidence of slow graft function,acute rejection and survival of graft and patient among groups.Conclusions Male recipients of female donors have the worst renal function while female recipients have better outcomes after operation.
8.Effects of fraction of inspired oxygen and positive end-expiratory pressure on gradient between arterial and end-tidal carbon dioxide in patients undergoing gynecological laparoscopic surgery

Objective To evaluate the effects of fraction of inspired oxygen (FiO2) and positive end-expiratory pressure (PEEP) on the gradient between arterial and end-tidal carbon dioxide (D(a-ET)CO2).Methods Sixty ASA Ⅰ or Ⅱ patients (body mass index < 30 kg/m2),aged 25-50 yr,weighing 45-75 kg,scheduled for elective gynecological laparoscopic surgery,were randomized into A,B and C groups (n =20 each).The patients were mechanically ventilated with pure oxygen in group A,and with air and O2 (FiO2 =50%) in groups B (PEEP =0) and C (PEEP =5 cm H2O).PETCO2 was monitored during mechanically ventilation.Blood samples were obtained from the radial artery immediately after tracheal intubation (T1) and at 1 h of pneumoperitoneum (T2) for blood gas analysis.D(a-ET) CO2 and Qs/Qt were calculated.Results D(a-ET) CO2 and Qs/Qt were significantly lower at T2 in groups B and C than in group A (P < 0.05).Compared with group B,D(a-ET)CO2 was significantly decreased at T2 (P < 0.05),and no significant change was found in Qs/Qt in group C (P > 0.05).Conclusion Decreased FiO2 and PEEP of 5 cm H2O can decrease D(a-ET)CO2 and increase the accuracy of PErCO2 for reflection of PaCO2,which is due to reduced intrapulmonary shunt.

Mitochondria play a central role in the regulation of cellular energy metabolism, bio-synthesis and cell death. Dysfunction of mitochondria can lead to many diseases. Mitochondrial proteomics provides important theoretical foundation for a systematic understanding of the biological functions of mitochondria, studying the mechanisms of mitochondria-related diseases, and promoting the research and development of mitochondria-targeting drugs. The methodologies, recent technology development, and characteristics and applications of mitochondrial proteomics were reviewed and the challenges and prospects were also discussed.
10.Effects of Cu/Zn SOD mediated by cell penetrating peptide 4 on hypoxia/reoxygenation injury in myocardial cells

Objective To evaluate the effect of cell-penetrating peptide (protein transduction domain 4,PTD4) mediated copper-zinc superoxide dismutase (Cu/Zn SOD) on hypoxia/reoxygenation injury (HRI) in rat myocardial cells.Methods Rat myocardial cell H9C2 HRI model was prepared by using the anaerobic incubator (85% N2,10% H2,5% CO2).The HRI group (without adding any treating factor in HRI cell culture fluid),HRI+Cu/Zn SOD group (adding 10 μmol/L Cu/Zn SOD) and HRI+PTD4-Cu/Zn SOD group (10 μmol/L PTD4-Cu/Zn SOD) were set up.In addition,normally cultured myocardial cells served as the normal control group.After incubating for 30 min,the ultra microstructure of mitochondria was observed under transmission electron microscope.The mitochondrial membrane potential was detected by JC-1 kit.The myocardial cell apoptosis was detected by TdT mediated dUTP nick end labeling TUNEL technique.Results The mitochondria injury degree after 30 min incubation in the PTD4-Cu/Zn SOD group was significantly improved compared with the HRI group.Compared with the normal control group,the mitochondrial membrane potential in the HRI group was significantly decreased,while the mitochondrial membrane potential in the PTD4-Cu/Zn SOD group was lower than that in the normal control group,but compared with the HRI group,which was obviously recovered.The cardiomyocyte apoptosis in the HRI+PTD4-Cu/Zn SOD group was (10.20±2.77)%,which was significantly decreased compared with (28.40±2.41)% in the HRI group,the difference was statistically significant (P<0.05).Conclusion PTD4 mediated Cu/Zn SOD can attenuate HRI in rat myocardial cells.
