1.The cAMP levels in the tissues of different brain regions in comparison with that of csf, plasma during ET-induced fever in rabbits

In order to determine whether the cAMP is a central madiator and participates in the mechanism of the ET-induced fever, in the following experiments, we have compared the cAMP levels in csf, plasma to the cAMP levels in tissues of hypothalamus, brain-stem and brain-cortex during the ET-induced fever. The results obtained demonstrate: the cAMP Ievels in tissues of hypothalamus and in csf are increased markedly during the fever Compared with the normal control. These changes of the cAMP levels are paralleled and correlated apparently with the body temperature. However, there isn't any obvious change of the cAMP levels in the tissues of brain-stem, brain-cortex and in plasma during the fever. This demonstrats that the cAMP increased in csf may indirectly reflect cAMP level in hypothalamus during the fever in rabbits and the main source of cAMP increased in csf may be synthesized and released by hypothalamus.
2.Real-time three plane ultrasonic imaging for evaluation of left atrial function in the general population

Objective:To evaluate the clinical efficacy of left left atrial function for the healthy by using real-time three plane ultrasonic imaging in.Methods: The three plane real-time ultrasound imaging was used for evaluation healthy people of left atrial function in 32 adults (including storage volume of left atrium, left atrial pipeline volume, left ventricular ejection fraction). At the same time, the two-dimensional echocardiography was also used to detect the parameters which were to compare two methods of correlation and consistency.Results:Two methods of ultrasonic examination in LARV, LACV and LVEF indicators have no obvious difference; LARV, LACV and LAEF in three plane image and two-dimensional ultrasound images show good correlation, the correlation coefficient rLARV=0.82, rLACV=0.85, rLVEF=0.88,P<0.05).Conclusion:Real-time three plane ultrasound imaging and two-dimensional echocardiography in the assessment of left atrial function of normal people have a good correlation.
3.Lumbar spinal pedicle subtraction osteotomy on single segment for correction of kyphosis in ankylosing spondylitis

[Objective] To assess the outcomes of lumbar spinal pedicle subtraction osteotomy on single segment for correction of kyphosis in ankylosing spindylitis.[Methods]15 patients were treated with L2 or L3 pedicle subtraction osteotomy and internal fixation by pedicle screw system.All the patients underwent X-ray examinations in standing position before and after the operations.The angles in different part of the sagittal planes were measured and the preoperative and postoperative differences were compared.[Results]All the operations were well complete.The mean time of operations was 3h and the mean bleeding was 840 ml.The lumbar lordosis angle increase from(14.5?15.5)?to(48.4?11.9)?,the total spinal kyphosis angle and the thoracolumbar kyphosis angle improve from(36.1?14.7)?and(30.7?9.3)?to(0.2?14.2)?and(23.7?12.3)? respectively,the sacral slope increase from(12.0?12.7)?to(28.9?8.8)?,whereas thoracic kyphosis angle remained relative stable.[Conclusion]Lumbar spinal pedicle subtraction osteotomy on single segment is a satisfactory and reliable technique for correction of kyphosis in ankylosing spindylitis and the average correction of lumbar lordosis was 33.9?.
4.Optimization the methodology of isolating human ubilical cord mesenchymal stromal cells from Wharton's jelly and examination of their passage effect on immune phenotype using flow cytometry

Objective To observe the effects of different collagenase digestions on isolating human umbilical cord mesenchymal stromal cells (MSC) from Wharton’s jelly, to exam their differentiation ability and to investigate their passage effect on the immune phenotype. Methods Human umbilical cord samples were digested by collagenaseⅠorⅡorⅣfor 4-18 hours then were passed through sieves . Cells were collected by centrifugation then inoculated in DMEM/F12 medium at concentration within range of 4.8×103-1×104/cm2 to compare the effect of different digestions on MSC. Von kossa staining and tetracycline fluorescence was used to label the osteogenic differentiation capacity of MSC. Also RT-PCR was employed to identify the differentiate capacity of MSC into myocardial-like cells. The immunophenotype of MSCs were detected by flow cytometry after subculture. Results Using collagenaseⅠdigestion, the number of MSCs isolated from human umbilical cord in Wharton’s jelly and their vitality were much higher while the period to show cell extension and primary culture time were shorter than those using collagenaseⅡorⅣdigestions. The analysis of surface marker revealed that the expression of positive markers include CD29, CD44, CD73, CD90 and CD105 did not change with passages while the negative markers such as CD31, CD34 and HLA-DR increased significantly with passages;Differential experiments induced in vitro show that human umbilical cord MSC in wharton’s jelly had the ability to differentiate into osteoblasts and myocardial-like cells. Con?clusion The human umbilical cord MSC in Wharton’s jelly was successfully isolated by collagenaseⅠdigestion. This meth?od was simple with a high success rate while cell loss and damage were minimum. This makes large-scale cultivation possi? ble. Negative markers increased with cell passages. This phenomenon revealed that MSC showed directional differentiation.

Silk fibroin is a naturally occurring protein polymer with excellent biocompatibility, remarkable mechanical properties, biodegradability and outstanding processability. Due to its unique properties, silk fibroin has become a favorable carrier material for the incorporation and delivery of a range of therapeutic agents. Based on the structure and characteristics of silk fibroin, this article provides an overview of the recent research progress of silk fibroin used as drug delivery materials.

7.Effects of Sini decoction against pulmonary injury induced by ex vivo ischemia-reperfusion in rats

AIM:To observe the effects of Sini decoction against pulmonary injury induced by ex vivo ischemia-reperfusion in rats. METHODS: The model of ischemia-reperfusion was established. Twenty-four Sprague-Dawley rats were randomly divided into Sham, I/R, and SND groups. Wet to dry lung weight ratio (W/D), mean pulmonary artery pressure (MPAP), SOD activity and MDA contents in pulmonary perfusate and tissue, NOS activity and NO contents in pulmonary tissue were detected. The pathologic changes in pulmonary tissue were also observed by light microscope. RESULTS: The morphological changes of pulmonary injury were alleviated in SND group. Wet/dry ratio, MPAP and MDA contents in pulmonary perfusate and tissue were significantly lower in SND group after ischemic/reperfusion. SOD activity in pulmonary perfusate and tissue, and NO contents in pulmonary tissue were significantly higher in SND group than those in I/R group. No significant difference in NOS activity in pulmonary tissue among three groups was observed. CONCLUSION: These results indicate that SND may have a protective effect on ischemia-reperfusion injured lung by its antioxidant activity and by adjusting NO level.
8.Nursing and study on correlation among cognitive dysfunction and post-stroke constipation

Objective To understand the status of cognitive dysfunction among new onset constipation after stroke and to explore the relationship between them.Methods With a self-designed questionnaire,the general information of 723 inpatients from 10 hospitals in Guangzhou were collected,including general demographic information,stroke situation,defecation situation and Mini-mental State Examination (MMSE)etc.Results The incidence of new-onset constipation,cognitive dysfunction(PSCD)after stroke was respectively 34.6% and 62.4%.Among whom 31.0% in ischemic stroke,44.8% in hemorrhagic stroke,47.4% in cerebral hemorrhage and infarction.There was statistical significance between stroke type and constipation,and ischemic stroke had statistical significance compared to other two types.During the acute phase reached 41.6%,the recovery phase 31.5% and the sequelae phase 22.6%.The rates of constipation patients with cognitive dysfunction were higher than patients without cognitive dysfunction,and the difference in directional force,memory,attention and calculation force,reading and expression was statistically significant respectively,among which expression was the most relevant.The difference in the incidence of constipation among different cognitive dysfunction groups was statistically significant,and they were positively correlated,compared with normal cognitive function,the light,medium and severe cognitive dysfunction was 1.519,2.879,3.064 times respectively.Conclusions Medical staff need to be alerted to the possibility that patients with impared cognitive function may be at greater risk of constipation.The preventive and treatment measures should be emphasized in order to improve patients' cognitive function.

10.Prenatal diagnosis of Aicardi-Goutières syndrome type 3: report of two pedigrees

Two pedigrees are reported here including two siblings and a boy who were diagnosed with Aicardi-Goutières syndrome type 3 (AGS3) caused by compound heterozygous variation of RNASEH2C gene. Prenatal gene diagnosis was performed when their mothers were pregnant again. All three cases presented with epilepsy, microcephaly, muscular hypertonia and severe language, motor and mental retardation. In pedigree 1, genetic analysis showed compound heterozygous variants of c.194G>A (p.Gly65Asp) and c.434G>T (p.Arg145Leu) in the RNASEH2C gene of proband 1 and her younger brother, which were inherited from their mother and father respectively. While in pedigree 2, c.194G>A(p.Gly65Asp) and c.227C>T(p.Pro76Leu) compound heterozygous variants in the RNASEH2C gene were found in proband 2, which were inherited from his father and mother, respectively. Diagnosis of AGS3 was confirmed in these three cases based on their medical history and the testing results. The mothers from the two families underwent prenatal diagnosis in their subsequent pregnancy, and the variation only inherited from the mothers was detected, suggesting that the two fetuses are carriers. Both families chose to continue the pregnancy and delivered at full-term. No growth or development abnormalities were reported in the children during a one-year follow-up.
