1.Transferrin C Subtypes and D Variants in Chinese by Ultrathin Layer Isoelectric Focusing

By ultrathin-layar isoelectric focusing, the transferrin (Tf) C subtypes and D variants of 474 Chinese blood donors without kinship in Chengdu were identified. The allele frequencies of Tf~(C1), Tf~(C2), Tf~(C3) and Tf~(Dchi) respectively were 0.73312, 0.24789, 0.00422 and 0.01266. In addition, two C_2DChengdu were observed, and the allele frequency of Tf DChengdu was 0.00211. This paper is the first report of Tf~c subtypes in mainland Chinese.
2.Determination of prim-o-glucasylcimifugin,5-o-methylvisammiosode and gardenoside in Huoxiang Qingwei Granules by HPLC

AIM: To establish a method of determing prim-o-glucasylcimifugin,5-o-methylvisammiosode,gardenoside quantitatively in Huoxiang Gingwei Granules(Herba Pogostemonis,Fructus Gardeniae,Radix Saposhnikoviae,etc.). METHODS: Three components were determined by HPLC in the same chromatographic condition(Column: Shimadzu VP-ODS 250 mm?4.60 mm,5 ?m;mobile phase: acetonitrile-0.1% phosphoric acid gradient elution;flow rate: 1.0 mL/min: column temperature: at 30 ℃). RESULTS: Prim-o-glucasylcimifugin showed a good linear relationship at the range of 26.0-390 ng,r=(0.999 9),the average recovery was 101.7% and RSD was 1.1%.5-o-methylvisammiosode showed a good linear relationship at the range of 21.5-322.2 ng,r=(0.999 9),the average recovery was 101.9% and RSD was 1.1%.Gardenoside showed a good linear relationship at the range of 47.1-706.5 ng,r=(0.999 9),the average recovery was 101.6% and RSD was 1.1%.(CONCLUSION):() The established method can provide the content determination of 3 ingriedents at the same test condition and be used for the quality control of Huoxiang Qingwei Granules.
3.Unconserved C terminal of pUL76 in human ctomegalovirus elicits aggresome formation

Objective To define the nuclear agrresome formation is determinated by which part of the UL76 of
HCMV. Full-length, conserved N terminal and unconserved C terminal of pUL76 were constructed to eukaryotic ex-pression plasmid pEGFP-N1 . Methods Primers were designed to amplify full-length and different part of pUL76 according to standard sequence of HCMV AD169 which had been submitted to GenBank(FJ527563. 1). These frag-ments were constructed to eukaryotic expression plasmid pEGFP-N1 . The recombinant plasmids were designated pEGFP-UL76,pEGFP-UL76N, pEGFP-UL76C respectively. Double digestion and sequencing were performed to verify the accuracy of recombinant plasmids construction. Empty vector and three recombinant plasmids were transi-ent transfected to HELF and HepG-2 cells respectively. Reverse transcriptation PCR and Western blot were per-formed to detect the RNA and protein expression level respectively. Different nuclear aggresome formations were visualized with an Olympus fluorescence microscopy. Results pEGFP-N1 and pEGFP-UL76N were unable to in-duce nuclear aggresome formation, whereas pEGFP-UL76 and pEGFP-UL76C were able to elicit nuclear aggresome formation. Conclusion The unconserved C terminal of pUL76 is sufficient to induce nuclear aggresome formation.
4.Comparative analysis on the performance evaluation of hospitals between China and some countries

This study aimed at comparing the United States, Britain, Australia, the Netherlands and China’s national performance evaluation, and sum up the experience to provide a theoretical basis for the China’s development of hospital performance evaluation system. The study found that China needs to consider the patient's perspective, to establish a fixed third-party performance evaluation agencies, establish an effective incentive mechanism and feedback mechanism and combine a variety of assessment methods in the development of hospital performance evaluation index system.
5.Study of carbapenem-resistant genes in a strain of Myroides odoratimimus and clinical therapy for its infection

Objective To study carbapenem-resistant genes in a Myroides odoratimimus strain and clinical therapy.Methods A strain of Myroides odoratimimus was isolated from nephrostomy drainage fluid of a patient with urinary tract infection in Xinhua Hospital in May 2013.MicroflexTM MALDI-TOF MS and 16S rDNA sequencing were performed for strain identification.Vitek-2 Compact combined with E-test method was used for antibiotic susceptibility test.Modified Hodge test and imipenem/imipenem-inhibitor (IP/IPI)E-test were performed for drug-resistant phenotype screening.Carbapenemase genes blaMUS-1,blaVIM-1,blaVIM-2,blaIMP-1,blaIMP-2,blaNDM-1,blaOXA-48 and blaGESwere amplified by PCR,and the positive products were sequenced and analyzed.Results The isolated strain was identified as Myroides odoratimimus.The strain was resistant to 17 antibacterial agents,and the minimum inhibitory concentration (MIC) of imipenem was ≥ 16 μg/mL,while it was sensitive to minocycline (MIC =0.38 μg/mL) and intermediate to meropenem (MIC =6 μg/mL).It was negative in modified Hodge test but positive in IP/IPI E-test (IP/IPI≥ 8).blaMUS-1 gene was detected by PCR,and further confirmed by sequencing.Meropenem combined with minocycline was effective for the patient.Conclusion Carbapenem-resistance in this Myroides odoratimimus strain may be related to blaMUS-1 gene,and appropriate therapy for its infection should be based on the result of antibiotic susceptibility test.
6.In vitro susceptibility of Geotrichum silvicola to five antifungal agents and electron microscopic observation of terbinafine-treated Geotrichum silvicola

Objective To evaluate in vitro antifungal activities of five clinically common antifungal agents against Geotrichum silvicola,and to observe morphological changes of Geotrichum silvicola after treatment with terbinafine.Methods Five antifungal agents commonly used in clinic,including terbinafine (TER),amphotericin B (AmB),flucytosine (FC),fluconazol (FCZ) and itraconazole (ICZ),were tested in this study.According to the National Committee for Clinical Laboratory Standards (NCCLS) M27-A2 document and relevant literatures,a susceptibility test was performed to determine the minimal inhibitory concentration (MIC) values of the five antifungal agents against G eotrichum silvicola.Scanning electron microscopy (SEM) was used to observe morphological changes of Geotrichum silvicola treated with different concentrations of TER.Results The MIC values against Geotrichum silvicola were as follows:TER,0.01 μg/ml;AmB,0.4 μg/ml;FC,2 μg/ml;FCZ,2.69 μg/ml;ICZ,0.25 μg/ml.Geotrichum silvicola was sensitive to TER,AmB,FC and FCZ,and sensitive to ICZ in a dose-dependent manner.SEM revealed that after TER treatment,the surface of Geotrichum silvicola became rough and shrinking with irregular defects and holes,and even gave a fragment-like appearance.Conclusions Among the five tested antifungal agents,TER shows the strongest activity against Geotrichum silvicola.Within a certain range,the higher the concentration of TER,the more serious the damage to Geotrichum silvicola.
7.Preliminary study on prevalence and transmission route of Klebsiella pneumoniae carbapenemases-producing Klebsiella pneumoniae

Objective To investigate the prevalence and transmission route of Klebsiella pneumoniae carbapenemases ( KPC)-producing Klebsiella pneumoniae in Xinhua Hospital Affiliated to Shanghaijiao Tong University School of Medicine .Methods Carbapenem-resistant Klebsiella pneumoniae isolates from clinical samples were collected from Xinhua Hospital during January 2010 and December 2013.Vitek 2 Compact and disc diffusion method ( Kirby-Bauer method ) were used for identification of the strains and antibiotic susceptibility test . Modified Hodge test was performed for drug-resistant phenotype screening . Carbapenemase gene blaKPC was amplified by PCR, and the positive products were sequenced and analyzed . Enterobacterial repetitive intergenic consensus ( ERIC )-PCR was used to analyze molecular epidemiology of the KPC-producing strains .And clinical information of these isolates was analyzed .Results There were 77 carbapenem-resistant Klebsiella pneumoniae isolates in total , and 71 of them were positive in modified Hodge test.Sixty-nine isolates were identified carrying blaKPC-2 gene.All of the KPC-2-producing isolates were classified as the same genotype by ERIC-PCR. Among 12 patients with KPC-2-producing Klebsiella pneumoniae infection who were first identified in each departments , 7 were transferred from other departments, and 4 were treated in surgery intensive care unit (SICU).Conclusions Most of carbpenem-resistant Klebsiella pneumoniae strains isolated from Xinhua Hospital are KPC-2-producing .The outbreak of carbpenem-resistant Klebsiella pneumoniae infection in the hospital may be associated with the interdepartmental transfer of patients .
8.Ribosomal protein S7 affects apoptosis of HeLa cervical cancer cells

Objective To preliminarily investigate the effect of ribosomal protein S7 on apoptosis of HeLa cervical cancer cells.Methods The previously constructed recombinant plasmid pIRES2-EGFP-RPS7 was transfected into HeLa cells,the empty vector pIRES2-EGFP transfected cells as control.Enhanced green fluorescent protein(EGFP)expressing cells were quantified by flow cytometry,and RPS7 protein level was also determined by Western blotting.Cell apoptosis of both RPS7 over-expression cells and knockdown cells were evaluated by flow cytometry after staining using allophycocyanin labeled Annexin-V.Results Apoptotic cell level in the obtained RPS7 transient over-expression HeLa cells was significantly higher than that of vector con-trol cells [(1 0.00 ±0.60)% vs (5.73 ±0.61 )%],with a statistic difference (t =8.63,P =0.001 ). Moreover,the apoptotic level in RPS7 knockdown cells was lower than that in control cells [(3.08 ± 0.49)% vs (5.97 ±0.63)%],with a statistic difference (t =6.40,P =0.003).Conclusion Up-regula-tion of RPS7 may promote apoptosis,while down-regulation of RPS7 may inhibit apoptosis of HeLa cells,indi-cating that RPS7 may play roles in regulating cell apoptosis.
9.A Research on SQL Server Association Rules in Data Mining forZhizichi Decoction Analogous Prescriptions

Objective To discuss the specific role of association rules in the compatibility of herbs and the relationship between herbs and symptoms by analyzing ancient analogous prescriptions Zhizichi Decoction with SQL Server software.Methods The prescriptions which contain Gardenia and Semen Sojae Preparatum were searched in the database of “Ancient Prescriptions for Epidemic Disease”, and they were imported to software with the standardized data of their symptoms and ingredients, then the proper association rules were found. The laws between parameter setting and rule generation were searched through parameter adjustment.Results Many laws of the drug compatibility and relationship between herbs and symptoms were obtained effectively in data mining by adjusting the parameters:The support setting was good for finding common used drug compatibility, and the confidence was good for finding specific drug compatibility and relationship between herbs and symptoms;the item set which had high frequency, sometimes made the rule’s importance low.Conclusion The laws of compatibility of herbs and the relationship between herbs and symptoms for data mining of analogous prescriptions are discovered by analyzing the support, confidence and importance of the association-rules and the clinical magnificence between symptoms and herbs.
