Chinese Pharmacological Bulletin 2003;0(07):-
Pro-rich antimicrobial peptides are a group of linear peptides isolated from animals. These peptides have antibacterial activities and play an important role in innate immunity. Pro-rich antimicrobial peptides are devided into two classes:Pro-rich antimicrobial peptides from mammals and Pro-rich antimicrobial peptides from insects and other invertebrates.Members of Pro-rich antimicrobial peptides are all characterised by a high content of proline residues and predominantly active against Gram-negative bacterial species which they kill by a non-lytic mechanism,at variance with the majority of the known antimicrobial peptides. Evidence is accumulating that the Pro-rich peptides enter the cells without membrane lysis and, once in the cytoplasm, bind to,and inhibit the activity of DnaK protein that is essential to bacterial growth, thereby causing responsive bacteria death. This mode of action makes these peptides suitable for drug development efforts. As one of the best characterized Pro-rich peptides,PR-39 plays an important role in a number of biological processes. It has been found that PR-39 can induce the syndecan expression in mesenchymal cells, inhibit the NADPH oxidase activity of neutrophils and block the degradation of 1?B? and HIF-1?. These findings show that PR-39 can provide novel means in the control of inflammation, wound repair, ischemia-reperfusion injury, and angiogenesis for therapeutic purposes.

Chinese Pharmacological Bulletin 1987;0(01):-
Many species of animals contain antimicrobial peptides as components of their immune systems. Many antimicrobial peptides have a broad spectrum of activity not only against gram negative and gram positive bacteria but also against antibiotic resistant bacteria, fungi, viruses, and parasites. Such peptides can act in synergy with other host molecules to kill pathogens. It has been found that certain peptides have a high affinity for bacterial products, allowing them to modulate the host inflammatory response to sepsis. They have also been found that they have the ability to recruit inflammatory cells. These properties as well as some other functions of antimicrobial peptides enable them to have an important impact in innate immunity.

Obejective To evaluate the accuracies of ten commercial total 25-hydroxyvitamin D [25(OH) D] immnoassays. Methods NIST 25 (OH) D reference material SRM 972a,which consisted of four vials of frozen serum with different concentration levels of different 25 (OH) D types, and two human serumsamples provided by our lab (BIMT),which had different concentration levels of 25 (OH) D3,were analyzed by ten total 25-hydroxyvitamin D immnoassays from Biomerieux,Mindray,Maccura,Chemclin,Abbott (2),Siemens,SNIBE (2),Roche,and by isotope-dilution liquid chromatography-tandem mass spectrometry(ID-LC/MS/MS) founded by BIMT.For the measurements of SRM 972a,the biases between tested values and certified values were calculated as a evaluating indicator, meanwhile the test biases between immnoassays and ID-LC/MS/MS were used as a evaluating indicator for the measurements of BIMT 25(OH) D3 serum samples.Test bias lower than 10% was deemed acceptible. Results The ID-LC/MS/MS exhibited low biases at (1.6%-2.8%) in the measurements of all levels of SRM 972a.8 immnoassays showed low biases at(1.5%-3.5%) in the measurements of level 1 of SRM 972a,which had a high 25(OH) D3 concentration level, but only 2 immnoassays gave low biases at (3.6%-3.7%)in the measurements of high 25 (OH)D2 concentration sample (level 3).While,5 immnoassays gave low biases at (-0.3%-9.0%) in the measurements of high 3-epi-25 (OH) D3 concentrationsample (level 4).It seems that, when SRM 972a were analyzed, only one of the ten commercial total 25 (OH)D immnoassays were in good accuracy and analytical specificity agrements with ID-LC/MS/MS.When two human serum25(OH) D3samples from BIMT were tested, most immunoassays were overall in relative good agreement with ID-LC/MS/MS at high 25 (OH) D3concentration level. Conclusion The test biases in the total 25 (OH) D measurements are differences between differentimmnoassays and ID-LC/MS/MS,and the specificities of current commercial total 25 (OH) D immnoassays should be improved, especially the performance on the equivalent recognition of 25 (OH) D2 and 25 (OH) D3.

Objective To evaluate the clinical results of one stage ante rior and posterior approach in the treatment of severe lower cervical injury. Me thod 8 cases of severe lower cervical injury underwent reduction, decompression and fixation through one stage anterior and posterior approach. 6 cases had fra cture and dislocation of cervical spine, and 2 cases cervical spine burst fractu re with cervical stenosis. Their clinical and radiographic records were reviewed retrospectively. Results Follow-ups of a mean period of 14 months showed that satisfactory recovery of neurological function was achieved. The Frankel Grade w as A in 7 cases and B in 1 case before operation, but D in 2 cases, C in 3 cases , and B in 1 case after operation except in 2 cases who had no improvement and r emained A. X-ray film demonstrated satisfactory stabilization of the fracture a nd dislocation, complete fusion of autograft, and preservation of the normal int ervertebral height and cervical lordosis. Conclusion One stage anterior and post erior approach operation is essential, safe and effective when radiographs demon strate that lower cervical canal is compressed from both anterior and posterior directions and clinical symptoms show severe neurological defects.

Chinese Journal of Biochemical Pharmaceutics 2001;22(3):109-111
Purpose The aim is to obtain the cDNA sequence of encoding extramembrane human FL gene with high level expression in E.coli. Methods The primers were designed based on the known FL cDNA sequence. The total RNA was isolated from fetal liver cells , and then RT-PCR was performed. The fragment was cloned into pUC-18T vector, and further sequenced by automatic sequence analyzer. The gene was inserted into GST fusion expression vector between BamH Ⅰ and EcoR Ⅰ sites. The recombinant plasmid was transformed into E.coli strain DH5 α and induced with 1mmol/L IPTG.Results The 546bp DNA fragment was amplified by RT-PCR method from fetal liver cells and its sequence was identical to the published sequence encoding human FL. The expressed fusion protein, with molecular weight of about 22kD, was about 10% of the total bacteria protein by SDS-PAGE and densitometry analysis. Conclusion cDNA was cloned successfully. This study provided a basis for the further fundamental research and clinical application of FL.

Objective To investigate the clinical pathologic significance of lymphangiogenesis in in colorectal cancer. Methods New lymphatic-specific markers D2-40 was used immunohistochemically to detect the lymphatic vessel density(LVD) in the intratumoural and peritumoral areas, and in normal tissue from 96 cases of colorectal cancer, which were analyzed with clinical pathologic parameters of those colorectal cancer. Results Significandy higher LVD was found in the intratumoural area(14.5±2.4), when compared with normal(5.9±1.1)and peritumoural areas(6.7±1.2) (P<0.01). LVD of the peritumoural area was higher than normal area (P< 0.01). However, peritumoural LVD was associated with both depth of invasion and liver metastasis (r=0.71,0.78 P<0.05), but not associated with tumour size, macroscopic type and lymph-node metastasis (P>0.05). Intratu-moural LVD was not correlated with tumour size, macroscopic type, the depth of invasion,lymph-node metastasis, and liver metastasis(P>0.05). Conclusion Lymphangiogenesis in the peritumoural area may be helpful in evalution of liver metastasis and prognosis.
