1.STUDIES ON LUNG TARGETED NIOSOMES OF CARBOPLATIN

AIM To improve the treatmemt efficacy and reduce the side effect of carboplatin (CBP), the antitumor drug was incorporated into niosomes (NS). METHODS Lung targeted niosomes of CBP (CBP-NS) were prepared by the method of hand shaking. The ultraviolet absorption spectrophotometric method was used for the determination of CBP. The method of dynamic dialysis was used for in vitro release of CBP from CBP-NS. The S-180 lung neoplasms models were established by iv cancer cells in mice. The number of pulmonary nodules was examined for evaluating the therapeutic efficacy. RESULTS The data showed that the mean diameter of CBP-NS was 3.72 μm, with a span of 0.66. The entrapment ratio of CBP in CBP-NS was 29.2% and the CBP-NS were stable for three months stored at 3℃-5℃, 15℃-25℃ or 37℃ (relative humidity 75%). The release profile in vitro could be described by a biexpotential equation. The calculated values of the three targeting parameters indicated that CBP-NS showed good targeting efficiency. The results of therapeutic trials showed that the antitumor effects were significantly increased by injection of CBP-NS compared with CBP in the treatment of mice with lung carcinoma. CONCLUSION The results indicated that CBP-NS have good targeting efficiency in vivo, and the biodegradable CBP-NS may decrease the side effects of carboplatin and improve its therapeutic efficacy.
2.Determination of Coumarin in Huoxiang Zhengqi Oral Liquid by UPLC

Objective To establish a UPLC method for detecting content of imperatorin and isoimperatorin in huoxiang zhengqi oral liquid. Methods ACQUITY UPLC BEH C18 (2. 1 mmí50 mm,1. 7 μm) was used and the mobile phase was methanol and water by gradient elution mode. The column temperature was 30 ℃ ,the flow rate was 0. 3 mL·min-1 and the detection wavelength was 248 nm. Results The linear range of imperatorin was 1. 305-13. 050 μg·mL-1 and the regression equation was as follow Y =13 633X+3 976 (r=0. 999 9). The linear range of isoimperatorin was 0. 596-5. 960 μg·mL-1 and the regression equation was Y=10 661X+1 073 (r=0. 999 9). The average recovery was 99. 25% (RSD=0. 74% ) and 98. 94%(RSD = 0. 63% ),respectively. Conclusion The method is accurate, rapid and reliable, and can be used to determine imperatorin and isoimperatorin in huoxiang zhengqi oral liquid.
3.Effect of heterosexual interaction training on rehabilitation in schizophrenic patients

Objective To explore the effect of heterosexual interaction training on rehabilitation in first-episode schizophrenic patients. Methods 246 patients of first-episode schizophrenic were divided into the study group and the control group with 123 cases in each group at random. Both groups were treat-ed with conventional pharmacotherapy under the orders of physicians in hospital. For the study group,both the male patients and the female patients took part in entertainment treatment activities,eating and interaction training together during the day, then they lived separately at night. Two groups were evaluated with BPRS and ITAQ before treatment and at the 2nd, 4th, 6th, and 8th week after treatment. Results The BPRS scores of the study group were significantly lower than the control group after 4th, 6th,and 8th week. The ITAQ scores of the study group were significantly higher than the control group after 4th, 6th,and 8th week. Conclusions The application of heterosexual interaction training in schizophrenic patients can alleviate illness condition, improve treatment compliance and rehabilitation.

5.Effects of pulmonary rehabilitation on patients with stable chronic obstructive pulmonary disease

Objective To observe the effects of puhnonary rehabilitation on the symptoms and quality of life of persons with chronic obstructive pulmonary disease (COPD).Methods COPD patients were recruited from a hospital and community health centers and given pulmonary rehabilitation in the community.The rehabilitation program included health education and nutrition guidance,medication,breathing exercises,and physical exercise.Each patient was followed up every 4 weeks by telephone.The outcome measures included pulmonary function (FEV1%,FEV1/FVC%),St George's questionnaire (SGRQ),the Medical Research Council scale (MRC),Borg scoring,evaluation of anxiety and depression,the six-minute walk test (6MWT),and an evaluation of nutritional status.All of the patients were observed for one year.Results A total of 85 patients completed the program and were included in the final statistical analysis.There was no difference in FEV1 % before and after rehabilitation.Six-minute walking distance improved significantly in statistical terms but the average increase was only from (416.1 ±99.84)m to (437.51 ±116.17)m.The SGRQ scores decreased by an average of 4.8 points after a year of rehabilitation,with significant improvements among the moderately and severely impaired.Incidents of COPD exacerbation also decreased significantly.Patients with a low body mass index had the greatest SGRQ score improvments.Few patients showed depression or anxiety in this research population.Conclusions Pulmonary rehabilitation in the community can improve exercise tolerance and the quality of life for persons with COPD,reducing incidents of acute exacerbation.Patients may benefit from such pulmonary rehabilitation regardless of disease severity.The compliance was good despite the infrequent monitoring.Pulmonary rehabilitation in the community is worthy of more widespread use.
6.Antitumor Responses Induced by Recombinant Vaccinia Viruses Expressing p53 Antigenic Peptide and B7

Objective: To explore antitumor responses induced by recombinant vaccinia viruses expressing a p53 antigenic peptide (rVV p53 M) and enhanced effect of recombinant vaccinia viruses expressing costimulatory molecule B7 (rVV B7). Methods: A 135 Cys to Tyr point mutation p53 transduced P815 mastocytoma (P815 mp53) was used as an experimental tumor and the p53 protein as the model of tumor associated antigen. rVV p53 M and rVV B7 were used as vaccine to test their induction of CTL and antitumor immunity. Results: Immunization of BABL/c mice with rVV p53 M could elicit specific CTLs, which could specifically lyse P815 mp53 cells. Immunization of mice with rVV p53 M could survive a part of mice challenged with 1?10 6 P815 mp53. Treatment with rVV p53 M could significantly prolong the survival of tumor bearing mice. Admixture at 1∶1 ratio of rVV p53 M and rVV B7 could enhance antitumor responses induced by rVV p53 M. Conclusion: Immunization with recombinant vaccinia virus expressing antigenic peptide is a useful alternative to peptide based vaccine. Costimulatory molecule B7 can enhance antigenic peptide to induce antitumor responses.
7.Literature Research of Compatibility Laws of Homologous Chinese Medicine of Medicine and Food in Prevention and Treatment of Alzheimer Disease

Objective To analyze the compatibility laws of homologous Chinese medicine of medicine and food in prevention and treatment of Alzheimer disease (AD). Methods We screened the clinical literature for the treatment of AD during 1988-2012 in TCM Clinical Diagnosis and Treatment Database, CNKI, VIP and Wanfang database. The nature, flavor, channel tropism, high frequency drug pair and compatibility laws of Chinese medicine and homologous Chinese medicine of medicine and food in prevention and treatment of AD were analyzed by frequency statistics and association rules. Results Two hundred and twenty-two clinical literatures were included. The top five frequently-used homologous Chinese medicines of medicine and food were Poria, Fructus Lycii, Fructus Alpiniae Oxyphyllae, Semen Persicae, and Rhizoma Dioscoreae. The top five frequently-used medicine were Rhizoma Acori Tatarinowii, Radix Polygalae, Rhizoma Ligustici Chuanxiong, Radix Salviae Miltiorrhizae, and Radix Angelicae Sinensis. The top five frequently-used drug pairs of homologous Chinese medicine of medicine and food were Rhizoma Dioscoreae→Poria, Poria→Pericarpium Citri Reticulatae, Rhizoma Dioscoreae→Fructus Lycii, Fructus Lycii→Fructus Alpiniae Oxyphyllae, and Fructus Lycii→Poria. The top five frequently-used drug pairs were Rhizoma Acori Tatarinowii→Radix Polygalae, Radix Salviae Miltiorrhizae→Rhizoma Acori Tatarinowii, Rhizoma Ligustici Chuanxiong→Rhizoma Acori Tatarinowii, Radix Salviae Miltiorrhizae→Rhizoma Ligustici Chuanxiong, and Rhizoma Acori Tatarinowii→Radix Astragali Seu Hedysari. Conclusion The commonly used homologous Chinese medicine of medicine and food in preventing and treating AD are tonic herbs with mild nature.

10.In vitro release and gastrointestinal absorption of novel pyridostigmine bromide nanoemulsion

Aim To prepare the novel pyridostigmine bromide nanoemusion(PPNE)and study its release in vitro, and to investigate the intestinal absorption. Methods Pyridostigmine bromide (PB)and PPNE were tested by HPLC in pH 1 .2 HCl,pH 6.8,pH 7.4,pH 7.8 PBS.Rat single pass intestinal perfusion method was employed to investigate the absorption mechanism of PB and PPNE.Results PB release rate was faster than PB in the four release media;the intes-tinal absorption rate constant(Ka )and apparent perme-ability coefficient(Papp)of PPNE were increased in the duodenum,jejunum,ileum and colon segments.PB and PPNE had significant difference in the duodenum, jejunum,ileum and colon segments by t test (P <0.05).Conclusions PPNE can improve the bioavail-ability of drugs,increase the drugs permeability,sig-nificantly improve the absorption of the drugs in the in-testinal segments. PPNE has obviously sustained effects.
