1.Apoptotic effects of bisphosphonate inosteosarcoma MG-63 cells in vitro

Objective To observe the effects of bisphosphonate on the inhibit proliferation and the apoptosis effect in osteo‐sarcoma MG‐63 cells in vitro ,explore the phosphonic acid salt of bone sarcoma cells ,induce apoptosis and its possible mechanism . Methods Sixty three osteosarcoma MG‐63 cells were cultured in vitro .After treated with bisphosphonate 400 μg/mL ,without bi‐sphosphonate but normal saline ,they were incubated 72 h after the application of the two group cell immunofluorescence test ;then observe the expression of apoptosis factors Caspase 3 and Fas ;Flow cytometry detection line was used to detect the osteosarcoma cell line MG‐63 cells apoptosis rate of each group .Results 72 h after treatment with bisphosphonate ,the expression of apoptosis factor of Caspase‐3 and Fas in osteosarcoma MG‐63 cells were strongly expressed ,and it was observed by immunofluorescent assay , while in blank control group ,we could barely see the expression of apoptosis factors Caspase‐3 and Fas ;Flow cytometry test results showed that two phosphonic acid salt 400 μg/mL intervention group cell apoptosis rate was 54 .00% ,far more than normal saline blank control group ,of which the apoptosis rate was 3 .10% ,the difference was statistically significant (P<0 .05) ,there is an obvi‐ous phenomenon of induced apoptosis .Conclusion Bisphosphonate has a strong apoptotic effects of bisphosphonate in osteosarcoma MG‐63 cells in vitro .Bisphosphonate can inhibit osteolysis of osteosarcoma MG‐63 cells via regulating the expression of Caspase‐3 , Fas in osteosarcoma MG‐63 cells .Bisphosphonate may serve as a potential therapeutic agent for treatment of osteosarcoma .
2.Influence of bisphosphonates and cisplatin on MG-63 cell lines culturedin vitro

BACKGROUND:Studies have shown that bisphosphonates can act on osteosarcoma cels, but the comparative analysis of bisphosphonates and cisplatin, a traditional first-line chemotherapy drug, is rarely reported. OBJECTIVE:To investigate the differences between bisphosphonates and cisplatin to inhibitin vitroproliferation of osteosarcoma cels and induce cel apoptosis. METHODS: Subcultured MG-63 cel lines were intervened with different concentrations of bisphosphonates and cisplatin, respectively, and cels with no treatment served as negative controls (blank group). Cel inhibition rate was detected using MTT method, and cel morphology was observed using fluorescent staining. RESULTS AND CONCLUSION:After intervention with different concentrations of bisphosphonates and cisplatin for different time, the growth inhibition rate of MG-63 cels was significantly higher as compared with the blank group (P < 0.05). But there was no difference in the growth inhibition rate between the bisphosphonates and cisplatin groups at 24, 48 and 72 hours (P > 0.05). These findings indicate that bisphosphonates has obvious inhibitory effect on the growth of osteosarcoma celsin vitro, and its inhibitory effect is similar to that of cisplatin.
3.Application of Valsalva experiment combined with improved eustachian tube MRI tomography tilt in nasopharyngeal carcinoma

Objective To discuss the value of Valsalva experiment combined with improved MRI tilt tomography applying on the pharyngeal ostium of eustachian tube opening function of patients with nasopharyngeal carcinoma.Methods 1 7 cases with patholog-ically proved NPC underwent conventional MRI plain and enhanced scan.In the eustachian tube area oblique transection T2 WI and T1 WI scanning was done before and after the Valsalva experiment.The invasion of eustachian tube and its surrounding structures was analyzed and the eustachian tube function was evaluated.Results Valsalva experiments combined with improved MRI trans-verse oblique scan could clearly display the whole structure of the eustachian tube and surrounding tissue invasion compared with conventional scanning MR scan.And it could better evaluate the eustachian tube opening function for patients suffered nasopharynge-al carcinoma.Conclusion Valsalva experiments with improved MRI oblique transverse imaging could achieve individualized scan of the eustachian tube,and could be used for evaluating the pharyngeal opening function of eustachian tube in patients with nasopharyn-geal carcinoma.
4.Finite element model establishment of thoracolumbar hemangioma and biomechanical analysis

BACKGROUND: Most of studies focus on the biomechanical characteristics of thoracic spine neoplasm, but there is little report on the fracture risk in the patients with vertebral hemangioma through finite element analysis.OBJECTIVE: To establish a finite element model of vertebral hemangioma, and to analyze its biomechanical characteristics, and assess the risk of vertebral fracture.METHODS: Three-dimensional finite element models of T12-L2 vertebrae from normal individuals, the patients with vertebral hemangioma (hemangioma accounting for 20%, 40%, 60%, 80% of the vertebral cancellous bone) and bone cement filling were established, respectively, and then the mechanical characteristics were analyzed. The stress distribution and characters of each model were determined under a vertical static pressure of 600 N.RESULTS AND CONCLUSION: (1) Three-dimensional finite element models of T12-L2 vertebrae were established successfully. Under static pressure, the stress distribution of L1 cortical bone showed no significant difference among models, and the maximum stress all occurred at the base of pedicle, zygapophysial joint and isthmus. (2) The stress distribution did not differ significantly between vertebral hemangioma accounting for 20%-40% of vertebral cancellous bone with complete cortical bone and normal ones, but which differed significantly in hemangioma accounting for 60%-80%. (3) To conclude, the established thoracolumbar three-dimensional model is available. Additionally,biomechanical tests manifest that the completeness of cortical bone and destruction ratio of cancellous bone destruction are key factors for the fracture risk of vertebral hemangioma.
5.Sonographic detection for calcification in thyroid nodule

Objective To investigate the significance of sonographic patterns of thyroid calcification in diagnosis of thyroid nodule.Methods 235 patients with 355 thyroid nodules were retrospectively analyzed.Nodule calcification size,distribution and pattern were observed.Results The incidence of calcification in benign and malignant nodules was 30.3% (89/294)and 73.7% (45/61) respectively.The difference had statistical significance(x2 =24.3,P <0.01).The rate of microcalcification in cancer was 47.5% (29/61),higher than that in benign one 3.1% (9/294)(x2 =99.1,P < 0.01).Coarse calcification in benign nodules and malignant lesion was 27.2% (80/294)and 26.2% (16/61)respectively.The difference had no statistical significance(x2 =0.42,P >0.05).Conclusions Microcalcification of thyroid nodules is a specific index for thyroid carcinoma.Any type of sonographically detected calcification represents risk of malignancy.Not only microcalcification,these cases should raise the suspicion of malignancy in coarse calcification,especially involving a solitary nodule and irregular shape.
6.Effect of pioglitazone on PPARγ mRNA expression in rats with acute necrotizing pancreatitis

Objective To observe the effect of pioglitazone on peroxisome proliferators activated receptors (PPARγ) mRNA expression in rats with acute necrotizing pancreatitis (ANP). Methods 54 rats were randomly divided into sham group, ANP group, and pioglitazone group. ANP was induced by the retrograde injection of sodium taurocholate into pancreatic duct. The levels of serum amylase at 3 h, 6 h, and 12 h after ANP induction were determined and pancreatic pathological scores were measured, and the expression of pancreatic PPARγ mRNA was determined by RT-PCR. Results The levels of serum amylase and pancreatic pathological scores at 6 h in ANP group were (7171 ± 1636) U/L and 13.00 ±2.36; which were significantly higher than those in sham group [ (523 ± 166)U/L and 1.67 ± 234, P < 0. 01 ]. While the expressions of PPARγ mRNA were weakly expressed in ANP gyoup and sharm group(0. 18 ± 0.05 vs 0. 22 ±0. 03, P > 0. 05). The levels of serum amylase and pancreatic pathological scores at 6 h in pioglitazone group were (4504 ± 1901 )U/L and 9.00 ±0.89, which were significantly lower than those in ANP group (P <0. 05).While the expressions of PPARγ mRNA was higher than that in ANP group (0.56 ±0.05 vs 0. 18 ±0.05, P <0.05). Conclusions The protective role of pioglitazone on ANP rats was closely correlated with up-regulation of PPARγgene expression.

Objective To obtain the peptide mass fingerprintings(PMF)of serums of the patients with bone metastasis and with-out metastasis and to filtrate and identify the differential serum proteins of patients with bone metastasis after radical mastectomy . To establish diagnostic models for diagnosis of bone metastasis after breast cancer operation .Methods Two groups of serum sam-ples were analyzed by ClinprotTM MALDI-TOF MS and gain PMF ,18 samples from patients with merely bone metastasis and 18 samples from patients without metastasis .Characteristic protein peaks were analyzed and selected by analyses software within Clin-prot system .Every group was repeated 2 times .Results All serum samples were repeated after 5 days and the fingerprintings were similar to the former .4 protein peaks were selected randomly to compute coefficient of variation which are less than 30% .The Clin-prot system has excellent repeatability .No differential protein was detected by analyzing PMF (P>0 .05) .Conclusion No differen-tial serum protein exists in patients with bone metastasis and without metastasis and detecting differential protein in peripheral blood may be impossible .
8.Influence of rehabilitation training on heart rate of stroke patients in the early stage

ObjectiveTo observe the influence of rehabilitation training on heart rate of stroke patients in the early stage.MethodsThe heart rate (HR) of 30 patients within one week after the onset of stroke was evaluated by FUKUDA DS 880A teleelectrocardiograph and the rating of perceived exertion (RPE) was collected during practicing bridging, rolling and moving the arms.ResultsThe HR of 30 patients was only increased 7.57, 6.23 and 6.57 beats per minute during practicing bridging, rolling and moving the arms. RPE of all patients was less than 11.ConclusionAppropriate rehabilitation training is safety and unable to increase the loading of heart in the early stage after the onset of stroke.

Objective To explore the clinical regularity of drugs on epidemic encephalitis B with text mining technique. Methods The data set on epidemic encephalitis B was downloaded from CBM database. The regularities of Chinese herbal medicine, Chinese patent medicine (CPM), western medicine and the combination of CPM and western medicine on epidemic encephalitis B were mined by data slicing algorithm. The results are showed visually with Cytoscape 2.8 software. Results The main function of herbal pieces was focused on clearing heat and removing toxicity. The herbal pieces such as Rehmanniae Radix Isatidis Raxis, Isatids Folium, Paeoniae Radis rubra and Scutellariae Radix were most often used. Angong Niuhuang Wan and Qingkailing Injection were usually used as CPM. For western medicine, antiviral drugs such as interferon and ribavirin were often used, and they were often used together with brain cells nutrition medicine, hormone, immunopotentiator and so on, and they were also frequently used together with CPM such as Angong Niuhuang Wan and Qingkailing Injection. Conclusion Text mining approach provides an important method in exploring the medication regularity for diseas, and provide refenrence for clinical use.
