1.THE TRANSPOSITION OF PEDICLED MEDIAL LOWER PART OF HUMERUS FLAP WITH VESSELS

Objective:In order to provide a new area of bone flap were supplied for non-unions, osseous defect of upper limbs and chondral defect of elbow joint.Methods:The origin,course,distribu- tion and anastomosis of inferior ulnal collateral artery and ulnal recurrent artery were observed on 40 adult cadaver upper limbs.Based on the results,transposition of pedicled medial lower part of humerus bone flap with vessels was designed.Results:Satisfactary results were achieved in 9 case of patients (non-unions of 3 case and osseous defect of 2 case on medial part of humerus bone;non-unions of 3 case on medial upper part of ulna and radius;chondral defect of 1 case on elbow joint.)Conclusion:This pe- riosteal flap that there are constant vessels and abundant anastomosis with inferior ulnar collateral artery form many area of bone flap were supplied with blood supply for repairing the non-unions,os- seous defect of humerus,ulna,radius and chondral defect of elbow joint.
2.Evaluation of two kinds of chemiluminescence detection of Treponema pallidum antibody-positive samples

Objective To analyze the true‐positive results ( ≥ 95% ) S/CO value of Treponema pallidum specific antibody (anti‐TP) positive samples caused by 2 different chemiluminescence detection assay in comparison with Treponema Pallidum Particle As‐say (TPPA) .Methods We collected the Treponema pallidum specific antibody positive samples of outpatient and hospitalization from October 2014 to January 2016 in Peking union medical college hospital as the research objects .There were 145 positive cases of Abbott laboratories (S/CO value of 1 .02 to 39 .29) ,24 positive cases of Roche (S/CO value of 1 .4 to 33 .07) .The 169 cases of Treponema pallidum specific antibody positive samples were detected with two methods of chemiluminescence detection at the same time ,TPPA was performed as repetition and confirmed test .Gathering and sorting the statistics of the positive predictive value seg‐mented ordered by specimen S/CO value ,to determine 95% or higher S/CO value of true positive results .Results After retested and confirmed by TPPA of the 169 positive cases ,the Abbott positive coincidence rate was 78 .7% ,the Roche positive coincidence rate was 81 .3% .When the S/CO value of Abbott ≥ 8 and the S/CO value of Roche ≥ 14 ,the positive predictive value was 100% . Conclusion When the S/CO value of Abbott ≥ 8 and the S/CO value of Roche ≥ 14 ,the S/CO value can be used as the true posi‐tive results( ≥ 95% ) .Abbott laboratories results S/CO value ≥ 8 ,Roche test results S/CO value ≥ 13 ,it is a 95% or higher S/CO limit of true positive results .
4.Down-regulation of FTO in human gastric cancer and its effect on cell line MGC-803 function

Objective To investigate the expression of FTO in gastric cancer tissues and the functional significance of FTO in MGC-803 cell line.Methods The FTO mRNA was detected by RT-qPCR in 54 cases of gastric cancer samples and their paired adjacent normal control tissues.The effect of FTO overexpression in MGC-803 on cell proliferation,cell migration and invasion were detected by CCK-8,wound heal and ranswell assays,respectively.Results The expression of FTO mRNA was significantly lower than that of adjacent tissues(P<O.05).Furthermore,overexpression of FTO in MGC-803 cells inhibited cell proliferation,cell migration and invasion.Conclusions FTO is low expressed in gastric cancer tissues and inhibits gastric cancer cell line MGC-803 proliferation,migration and invasion.FTO is closely associated with the development of gastric cancer.
5.Study on Quality Standards for Stem Leaf ofAstragali Radix

Objective To establish quality standards for stem leaf ofAstragali Radix.Methods A method of TLC identification was used for qualitative discrimination. The moisture, total ash and extracts contents of the stem leaf of Astragali Radix were determined by pharmacopoeia method. Total contents of astragaloside and astragalus polysaccharide were measured by colorimetry. The astragalosideⅣ and calycosin-7-O-β-D-glucoside contents were detected by HPLC.Results The spots of TLC were round and clear with good repeatability. The moisture contents of the stem leaf ofAstragali Radix were between 6.24%–12.24%; the total ash was between 8.21%–10.55%; the water- solubility extracts were between 12.12%–27.30%; alcohol-solubility extracts were between 6.89%–10.28%; the total contents of astragaloside were between 23.74–26.52 mg/g; astragalus polysaccharide were between 23.31–45.70 mg/g; the astragalosideⅣ contents were between 0.047%–0.18%; the calycosin-7-O-β-D-glucoside were 0.21%–0.26%. Conclusion The method is convenient, fast and repeatable, and the results are accurate and reliable, which can be used to control the quality of the stem leaf ofAstragali Radix effectively, and as the main index of the quality standard.
6.An analysis of results of the elderly colorectal cancer screening study

Objective To evaluate the detection rates of colorectal polyps and cancer in the elderly and characterize the pathological features of the colorectal neoplasm in DongLi District in Tianjin.Method Totally,46 743 subjects aged 60-74 were recruited from the national colorectal cancer screening program,including subjects registered from November 2012 to April 2013 in DongLi District in Tianjin.Each subject completed a questionnaire and a fecal occult blood test.Colonoscopies were conducted in 434 subjects with high risk for colorectal cancer.Results Among all the subjects,2 880 were at high risk for colorectal cancer which accounts for 6.16% (2 880/46 743) of the total subjects.Among the subjects who underwent the colonoscopy,165 (men:105,women:60) were colorectal polyps accounting for 5.73% (165/2 880)in the total risk,with 51 subjects (1.77%) advanced neoplasms and 4 subjects (0.14%) cancer.The detection rates of colorectal polyps is 0.35% (165/46 743),and colorectal cancer is 8.6/105(4/46 743).More polyps were found on the left colon compared with the right.More tubular adenoma was found compared with the other pathological types.Most of the polyps were smaller than 0.5 cm.Colorectal polyps and advanced neoplasms were more frequent in men than women [45.45% (105/231) vs 29.56% (60/203),16.02% (37/231) vs 6.90% (14/203),all P < 0.01].Multiple polyps (P < 0.05) and polyps with diameter larger than 1.0 cm (P < 0.01) were more often in men than in women.Men tended to have more colorectal polyps in right colon and higher detection rate for moderate or sever atypical hyperplasia than women(all P < 0.05).Conclusion There is a higher incidence of colorectal polyps in the elderly.It is important to establish the national sequential colorectal cancer screening program for the early diagnoses of colorectal polyps and cancer in the elderly.
7.Application value of sequential colorectal screening program in health check-ups

Objective To explore the application value of the sequential colorectal cancer screening program in population completed health check-ups .Methods The healthy individuals were enrolled from November 2013 to July 2014 .Questionnaire survey of risk factors quantity assessment and fecal occult blood test (FOBT) were used for initial screening for colorectal cancer .According to the results of initial screening ,subjects were divided into high-risk group and low-risk group .The detection rates of polyps and cancers of two groups were compared .Chi-square test was performed for statistic alanalysis .Results A total of 525 individuals completed the colonoscopy .There were 187 individuals in high-risk group and 338 individuals in low-risk group .The results of colonscope examination showed that 70 cases of high-risk group were detected with polyps or colorectal cancer ,including 7 (10 .0% ) cases of colorectal cancers , 8(11 .4% ) cases of advanced adenoma ,30(42 .9% ) cases of non-advanced adenoma and 25(35 .7% ) cases of inflammatory polyps . In low-risk group , 63 individuals had polyps or colorectal cancer , including 5(7 .9% ) cases of advanced adenoma ,38(60 .3% ) cases of non-advanced adenoma and 20(31 .7% ) cases of inflammatory polyps .The differences of polyps and colorectal cancer detection rates between two groups were statistically significant (χ2=3 .905 and 10 .136 ,P=0 .048 and 0 .001) .Conclusion The sequential colorectal screening program including colorectal cancer risk factors quantity assessment questionnaire and FOBT seems to be a valuable tool in clinical practice .
8.Research of HIF-1α,ALDH1 and Hedgehog signaling pathway cooperation involved in activation of cancer stem cell in triple negative breast cancer

Objective:To explore the cooperation and clinical significance of HIF-1α,ALDH1 and Hedgehog signaling pathway in the activation of cancer stem cell(CSC) in triple negative breast cancer(TNBC).Methods: ALDH1+(Aldehyde dehydrogenase1)breast cancer stem cells and ALDH1-breast cancer cells were selected from MDA-MB-231 cells by magnetic activated cell sorting system(MACS),qRT-PCR method was employed to analyze the expression differences of HIF-1α and Hedgehog signaling molecules Sonic hedgehog(SHH),patched1(PTCH1),Smoothened(SMO) and Glioma-associated oncogene homoglog1(GLI1) in ALDH1+ breast cancer stem cells and ALDH1-breast cancer cells.Immunohistochemical method was applied to study the expressions of HIF-1α and ALDH1 and the relationships among HIF-1α,ALDH1 and Hedgehog signaling molecules in TNBC.Results: The expressions of HIF-1α mRNA,SMO mRNA and GLI1 mRNA in ALDH1+ breast cancer stem cell were higher than those in ALDH1-breast cancer cell(P all<0.05).The positive expression rates of HIF-1α were 90.0% and 70.0%,and the positive rates of ALDH1 were 93.3 % and 66.7 % in TNBC and non-TNBC,respectively(P all<0.05).Spearman rank correlation analysis showed that the expression of HIF-1α was positively related with that of ALDH1 in TNBC(r=0.53,P<0.01).HIF-1α expression was correlated with lymph node metastasis and TNM stage(P all<0.05),ALDH1 expression was correlated with histological grade and TNM stage(P all<0.05).In addition,the expression of HIF-1α was positively related with that of Hedgehog signaling molecules SHH(r=0.584,P<0.01),SMO(r=0.467,P<0.01) and GLI1(r=0.439,P<0.05),the expression of ALDH1 was positively related with that of SHH(r=0.426,P<0.05) and GLI1(r=0.394,P<0.05).Conclusion: HIF-1α and Hedgehog signaling pathway were activated in ALDH1+ breast cancer stem cell.HIF-1α,ALDH1 and Hedgehog molecules may cooperate with each other to activate breast CSC to promote the malignant progression of TNBC.
9.Expressions of HGF and its receptor c-met in non-small-cell lung cancer and its association with lymphangiogenesis

Objective To test the expression of HGF and its receptor c-met and to evaluate their relationship to lymphangiogenesis in NSCLC. Methods Immunohistochemical study was used to determine the lymphatic microvessel density (LMVD) and the expression of HGF and its receptor c-met in 113 patients with NSCLC and 20 normal lung tissue samples.Results The levels of expression of HGF (67.3 %) and c-met (74.3 %) in NSCLC tissues were significantly higher than that in normal lung tissues (20.0 %, 25.0 %respectively), (u=4.008, 4.342; both P <0.05).The expressions of HGF and c-met in NSCLC tissues had no correlation to age, gender, tumor size and differentiation grade.However, the expression of HGF and c-met was positively correlated to TNM stage and lymph node metastasis.In NSCLC samples, HGF and c-met were significantly associated with LMVD contrast to normal sample (HGF: 16.3051±5.3753 vs 10.9934±4.9668,t =4.58; c-met: 15.6692±5.5386 vs 11.3700±5.3875, t =3.97, both P < 0.05).Conclusion The high expression of HGF and its receptor c-met is closely correlated with LMVD and metastasis of lymph nodes, which indicates that HGF and c-met may play an important role in lymphangiogenesis and lymphatic metastasis in NSCLC.
10.Biological behaviors of two novel syngeneic human osteosarcoma cell lines

Objective To characterize and compare the different biological behaviors of two novel human osteosarcoma cell lines, Zos and Zos-M, established respectively from the primary site and the skip metastasis of an osteosarcoma patient. Methods Two novel human osteosarcoma cell lines, Zos and Zos-M,were developed using tissue plant culture method. The vitro examinations included observations of morphology, analysis of karyotype and cell cycle, calculation of doubling time and growth curve, detection of osteoblastic markers and matrigel invasion assay. Subcutaneous, intratibial and intravenous inoculations into nude mice were performed to study the in vivo tumorigenicity and metastatic potentials of both cell lines.MTT were used to detect sensitivity of the cell lines to chemotherapeutic drugs. RT-PCR was performed to assess the expression of and some metastasis-related genes. Results Both cell lines proliferated actively and remained stable for more than 100 passages in vitro without interruption. The morphology and expression of osteoblastic markers of Zos and Zos-M were conformed to the characteristic of osteosarcoma. The karyotype analysis displayed aneuploidy and various structural abnormalities. The population doubling time of Zos and Zos-M were 33.65 h and 31.58 h respectively. Both cell lines were less sensitive to the current chemotherapy protocols compared to U-2OS. Zos and Zos-M were 100% tumorigenic by subcutaneous and othotopic injection. 37.5% of nude mice injected Zos-M and none of nude mice injected Zos developed lung metastasis after intravenous injection. The comparison of the expression patterns of some metastasis-related genes revealed that the decreased expression of cadherin-11 in Zos-M may correlate with a high potential of metastases. Conclusion The two novel established human osteosarcoma cell lines, Zos and Zos-M and related animal models could serve as models for the study of drug resistance and screening of new therapeutics for osteosarcoma. In addition, the study also provide tools for the study of metastasis because the same genetic background and different potential of metastasis of Zos and Zos-M.
