1.High Tibial Osteotomy for Medial Compartment Serious Osteoarthritis of the Knee

Objective: High tibial osteotomy treated medial compartment serious osteoarthritis of the knee with varus deformity,and relieving arthritic pain,correcting deformity and normal femoraltibial axial alignment.Methods:From May 1992 to February 1998,18 cases(21 knees) were operated on,We chose fit patients and planed osteotomy angle.Results:All cases were followed up for an average 3.2 years,according to Coventry's rating system,81% of the whole series had a good or excellent results.Conclusion:It increases the medial joint space width and corrects femoraltibial axial alignment,allows for healing of the articular cartilage.Satisfactory results were obtained.
2.Effects of astragalus polysaccharides on the proliferation of bone marrow mesenchymal stem cells from patients with multiple myeloma

BACKGROUND:Bone marrow mesenchymal stem cels can produce a variety of cytokines to provide some support for the growth and proliferation of myeloma cels.
OBJECTIVE:To investigate the effects of astragalus polysaccharides on the proliferation of bone marrow mesenchymal stem cels from patients with multiple myeloma and cytokine secretion.
METHODS:Bone marrow mesenchymal stem cels were isolated by density gradient centrifugation from the bone marrow of a healthy volunteer and a multiple myeloma patient, and were cultured and identified. Passage 3 bone marrow mesenchymal stem cels were colected, and the growth curve was depicted by the enzyme marker method. MTT method and flow cytometry were used to detect the effects of different concentrations (0.5, 1, 2, 4, 6, 8 g/L) of astragalus polysaccharides on the proliferation and cel cycle of bone marrow mesenchymal stem cels. The expression levels of interleukin-1β and interleukin-6 were detected by ELISA method.
RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cels from both the healthy control and multiple myeloma patient showed negative expression of CD34 and positive expression of CD44. The obtained cels from two sources had the characteristics of bone marrow mesenchymal stem cels. Cels from the healthy control had significantly shorter doubling time than those from the myeloma patient. After treated with 1 and 2 g/L astragalus polysaccharide, cel proliferation rate in the multiple myeloma group was significantly higher than that in the healthy control group; after treated with 6 and 8 g/L astragalus polysaccharide, the cel proliferation rate was significantly lower in the multiple myeloma group than in the healthy control group. After treated with 1 g/L astragalus polysaccharide, compared with the healthy control, the proportion of bone marrow mesenchymal stem cels from the multiple myeloma patient in G0/G1 phase was significantly decreased, while the proportions of cels in both S phase and G2/M phase from the myeloma patient were significantly increased. Interleukin-1β and interleukin-6 levels of bone marrow mesenchymal stem cels from the patient with multiple myeloma were significantly higher than those of cels from healthy control. After intervention with 0.5, 1, 2 g/L astragalus polysaccharide, the interleukin-1β and interleukin-6 levels of bone marrow mesenchymal stem cels from the patient with multiple myeloma were al significantly decreased. In conclusion, a certain concentration of astragalus polysaccharides can promote the proliferation of bone marrow mesenchymal stem cels and decrease the levels of interleukin-6 and interleukin-β in patients with multiple myeloma.
3.The angiogenic promoting effect of Morinda officinalis How oligosaccharides in the ischemic myocardium of rats after acute myocardial infarction

Aim To investigate the angiogenic promoting effect of Morinda officinalis How oligosaccharides(MOO) in the ischemic myocardium of rats after acute myocardial infarction(AMI).Methods 40 male Wistar rats were established into AMI model successfully and were randomly divided into 5 groups equally, i. e. the low, medium and high doses of MOO groups, the Shexiangbaoxin group and the model group. They were treated with different doses of the water fraction of the ethanolic extract of Radix morinda officinalis (0.7, 1.4, 2.8 mg·kg~(-1) ·d~(-1)), suspension liquid of Shexiangbaoxin Pill(30 mg·kg~(-1) ·d~(-1)) and distilled water with the same volume respectively.Besides, a sham operated group with 10 rats was set up for control. All rats were sacrificed after 6-week-treatment.The Ⅷ coagulation factor, vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) protein in ischemic myocardium of rats in each group were detected by immunohistochemistry assay.The microvessel density(MVD) was calculated. Gray values of protein expression of VEGF and bFGF in ischemic myocardium were calculated and analyzed by image analysis system.Results The MVD, the gray values of VGF and bFGF were higher in the medium and high doses of MOO groups than those in the model group(P <0.05), but still lower than those in the Shexiangbaoxin group(P <0.05). The MVD and the gray values of VEGF among 3 doses of MOO groups showed significant differences (P <0.05).Significant differences of gray value of bFGF were observed between small and middle doses of MOO groups, also between small and large doses of groups(P <0.05).Conclusion MOO can obviously promote angiogenesis in the ischemic myocardium of the rats after AMI.And up-regulating expressions of VEGF and bFGF protein in the ischemic myocardium may act as one of its angiogenic promoting mechanisms.
4.Effect of Light Emitting Diode Red Light Irradiation on Hyperlipidemia Rats

Objective To observe the effect of light emitting diode (LED) red light irradiation on serum lipid in experimental hyperlipidemia rats. Methods 36 healthy male Sprague-Dawley rats were randomly divided into normal control group (n=12) and hyperlipidemic model group (n=24). The normal group was fed with normal diet while the hyperlipidemic model group with fat-rich forage for 6 weeks. The hyperlipidemic model group rats were randomly divided into the hyperlipidemic control group (n=12) and LED treatment group (n=12), and the latter accepted LED red light irradiation for 28 d. The levels of serum lipid including total cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL-C), high-density lipoprotein (HDL-C), and the activities of lipoproteinesterase (LPL) and hepatic lipase (HL) were detected with biochemical assay. The expression of 3-hydroxy-3-methylglutaryl coenzyme A reducase (HMG-CR) of hepatic tissue were measured with immunohistochemical staining. Results Compared with the hyperlipidemic control group, the levels of serum TC, TG and LDL-C decreased while the serum HDL-C increased significantly in the LED treatment group (P<0.01) after treated with LED. The levels of LPL and HL in serum increased (P<0.01) while the activity of HMG-CR decreased (P<0.05). Conclusion LED red light irradiation might play a regulating effect on serum lipid by enhancing the activities of LPL and HL and inhibiting the expression of HMG-CR to interfere the metabolism of TC, TG, LDL-C and HDL-C.
5.Effect of Dermatix Ultra on repair of scar tissue in patients after breast plastic surgery

Objective To explore clinical effect of Dermatix Ultra on repair of scar tissue in patients after breast plastic surgery .Methods 66 patients with breast conserving surgery after breast conserving surgery were selected and divided into two groups.32 cases in the control group were treated with the implementation of breast conserving surgery and breast plastic surgery , 34 cases in the treatment group were treated on the basis of the control group and combined with Dermatix Ultra.Evaluation of patient and observer scar assessment scale ( POSAS ) and visual analogue scale were compared after the treatment and satisfaction degree were compared after 6 months follow-up.Results Compared with the control group, scar evaluation in treatment group were higher(P<0.05), the color, thickness, the degree of pigmentation and the score of the pigment were higher (P<0.05), the color, softness, thickness, pain and itching were higher (P<0.05), and the patients were followed up for 6 months (P<0.05).The color, thickness, roughness and pigmentation score of OSAS in the treatment group were higher (P<0.05), Color, softness, thickness, pain and pruritus in PSAS were higher (P<0.05),after 6 months of follow-up, the satisfaction rate in patients was higher (P<0.05).Conclusion Dermatix Ultra has a good clinical effect in repair of scar tissue after the breast plastic surgery, improve the color, thickness, smoothness and pigmentation of patients after breast augmentation surgery, and improve the prognosis.
6.Determination of Oil-water Partition Coefficients of Rosiglitazone

OBJECTIVE:To determine the oil-water partition coefficients (logP) of rosiglitazone at different pH values by two kinds of methods.METHODS:A shake flask-ultraviolet spectrophotometry method was used to determine plasma concentration of rosiglitazone in water phase and organic phase at variable pH values (0.95,2.0,3.0,4.0,5.07,5.7,6.55,7.06,7.5,8.15,9.21) to calculate logP by using with n-caprylic alcohol-water as simulation system.Above logP values were compared with that calculated by Pallas software.RESULTS:The logP value was lower than 1 when pH value was lower than 4;the logP value was higher than 1 when pH value was ranged from 5 to 9.Measured logP values were in good agreement with that calculated by Pallas software.CONCLUSION:Rosiglitazone exists in the form of hydrochloride salt with good water solubility (pH
7.Effects of happy physical education on physical education learning burnout, body shape and mental health of college students

Objective:
To explore the effects of happy physical education based on constructivism on physical education learning burnout, body shape and mental health of college students, so as to provide a reference for the reform of physical education teaching in universities.
Methods:
By using cluster sampling method, students from two teaching classes of the 2022 physical badminton elective course were enrolled as the research objects at Shanghai Institute of Commerce and Foreign Languages from September to December 2022. They were randomly divided into experimental group ( n =38) and control group ( n =40) by drawing lots. The control group received routine physical education teaching for 16 weeks, with 2 class hours weekly and 45 minutes per class hour. While the experimental group was given happy physical education based on constructivism, with a teaching process of building a framework, independent exploration, collaborative learning, and effect evaluation. The idea was to integrate Happy Sports into badminton technology and tactical training courses, emphasizing the fun of sports activities, and drawing on various forms of sports games when designing sports activities. Two groups of students were tested for body shape, surveyed with the Physical Education Learning Burnout Scale and Symptom Check List-90 (SCL-90). The scores of Physical Education Learning Burnout Scale, body mass index (BMI), waist to hip ratio, body fat rate and SCL-90 were compared between the two groups by t-test, Chi-square test and Fisher s exact probability test.
Results:
At the end of the semester, BMI [(21.58±1.59,22.30±1.46)kg/m 2], waist to hip ratio (0.75±0.08, 0.79 ±0.07) and body fat rate [(19.77±2.50)%,(21.02±2.46)%] in experimental group were lower than those in control group ( t= -2.09, -2.35, -2.23, P <0.05). The scores of depression (15.10±3.92,17.24±4.18), improper behaviors (13.60±3.05,15.48± 3.52 ) and low sense of achievement (5.52±1.36,6.54±1.42) and total score of Physical Education Learning Burnout Scale ( 34.22± 7.55,39.26±7.86) in experimental group were lower than those in control group ( t=-2.33, -2.52, -3.24, -2.89, P <0.05). The scores of somatization (16.27±3.75,18.57±4.12), interpersonal sensitivity (14.30±4.57,16.85±4.21), depression (17.02±3.89,19.36±4.28) and anxiety (12.68±3.96,15.32±3.87), and total scores of SCL-90 (127.87±22.54,140.54±25.83) in experimental group were lower than those in control group ( t=-2.57, -2.57, -2.52, -3.00, -2.30, P <0.05).
Conclusions
The happy physical education mode based on constructivism can alleviate learning burnout, improve mental health and optimize body shape of college students. Appropriate reforms should be made to the teaching mode of physical education in universities to promote the physical and mental health development of college students.
8.Angiogenesis promoting effect of Morinda officinalis oligosaccharides on chicken embryo chorioallantoic membrane.

OBJECTIVETo study the angiogenesis promoting effect of Morinda officinalis oligosaccharides(MOO) on chick embryo chorioallantoic membrane (CAM).
METHODRats blood serum containing low, medium and high doses of MOO was prepared using Chinese herbs serum pharmacology method. 60 chick embryoes were randomly divided into low, medium and high doses of MOO groups, as well as NS group, blank serum group and bFGF group. Each group included 10 embryoes. CAM model was prepared after 7 days incubation. Then NS, blank serum, bFGF (2500 U x mL(-1)), three doses of serum containing MOO were added respectively onto the carriers on the CAM. CAM sample was prepared after 3 days incubation. The state of angiogenesis was observed and the number of new blood vessels was counted.
RESULTCompared with blank serum and NS group, a more specific CAM angiogenesis appearance could be observed in each MOO group and bFGF group. Compared with blank serum group, the number of new blood vessels in each MOO group increased significantly (P < 0.05). But the drug had a lower efficacy than bFGF (P < 0.05). Compared with low dose group, the number of new blood vessels increased significantly in medium and high doses groups (P < 0.05). But there was no significant difference between the latter two groups. The number of new blood vessels showed no significant difference between NS group and blank serum group.
CONCLUSIONMOO can obviously promote angiogenesis of CAM.

9.Comparison of three approaches to establishing Balb/c mouse models of hind-limb ischemia.

OBJECTIVETo compared 3 approaches to establishing Balb/c mouse models of hind-limb ischemia suitable for studies of regenerative medicine.
METHODSThirty eight-week-old male Balb/c mice were randomly divided into 3 groups for establishing models of hind-limb ischemia by both ligating and transecting the femoral artery proximal to the origin of the deep femoral artery branch, ligating the femoral artery proximal to the origin of the deep femoral artery branch, or ligating the femoral artery distal to the femoral artery trunk. The mice were continuously observed for 5 days after the operation, and hind-limb blood perfusion was monitored by laser Doppler imaging on the second postoperative day.
RESULTSAll the 3 surgical approaches caused blood flow block in the hind-limbs of the mice. In mice with ligation and transaction of the femoral artery proximal to the origin of the deep femoral artery branch, dry gangrene occurred in the whole hind-limb. Dry gangrene also occurred in the legs and paws of mice with ligation of the femoral artery proximal to the origin of the deep femoral artery branch. In mice with femoral artery ligation distal to the origin of the deep femoral artery branch, the sole of the foot was dry with scaling skin and necrosis of several digits; microscopically examination revealed a large quantity of necrotic muscles accompanied by muscle regeneration.
CONCLUSIONLigating the femoral artery distal to the origin of the deep femoral artery branch can be optimal for establishing Balb/c mouse models of hind-limb ischemia.

10.Effects of Senescence on Mechanical Response of Osteocyte

Bone is a dynamic organ, and the morphology, structure and function of bone can vary with the size, direction and form of mechanical stimulation. Appropriate mechanical stimulation is the key to maintain the dynamic balance of bone formation and bone resorption. However, with aging, the senescence of bone tissues causes a series of changes, including bone microenvironment, osteocyte morphology, signaling pathways in the osteocyte, etc., which weakens its mechanical response ability and then leads to osteoporosis and other diseases. Therefore, it is of great significance to study how aging affects the mechanical response of osteocyte. This review mainly discusses the influence of aging on mechanical response of the osteocyte.
