1.Clinical Application of Tidal Flow-volume Measurement and Its Impact Factors

Objective To study the clinical application and its impact factors of tidal flow - volume measurement in infants with bronchiolitis and pneumonia. Methods Thirty infants with bronchiolitis and thirty infants with pneumonia were studied. Tidal breathing flow- volume loops were obtained after the patients were sedated. The tidal volume per kilogram(VT/kg) ,the fraction of tidal volume ex-heled at peak tidal expiratory flow (PTEF) to total tidal volume (VPTEF/VE), and the fraction of exhaled time at PTEF to total expirato-ry time (TPTEF/TE) were measured. Results VT/kg was not significant different between the bronehiolitis group and the pneumonia group. VPTEF/VE and TPTEF/TE were lower in the bronehiolitis group than those in the pneumonia group. Conclusion The measure-ment of tidal breathing flow - volume loops is a good technology, which would be helpful to evaluate airway obstruction in wheezing in-fants. The impact factors should be controlled during the measurement. Otherwise, the results would be affected.
2.Factors Influencing Agrobacterium-mediated Transformation of Foxtail Millet (Setaria italica)

An efficient and repeatable approach in transforming the foxtail millet (Setaria italica ) using Agrobacterium LBA4404 horboring the plasmid pBI121 was established. Factors affecting transformation efficiency were investigated including the genotype, explants, the density of bacteria, the duration for inoculation and co-cultivation, and the concentration of acetosyringone in the medium. The maximum transformation conditions were: the callus induced from inflorescence was used as explant; the duration for inoculation with Agrobacterium at low cell density was 30-40 min; for the co-cultivation , the suitable concentration of acetosyringone in the medium was 0.1 mmol/L, and the duration was 2 days.
3.Detection of TTV DNA and analysis of partial TTV gene sequence in different population of Guizhou area.

OBJECTIVETo investigate the prevalence of transfusion-transmitted virus (TTV) infection and analyze partial nucleotide sequence of TTV isolated from Guizhou area.
METHODSA nested polymerase chain reaction (nPCR) assay with two-set of primers deduced from the first read frame of TTV genome was established, the prevalence of TTV DNA were detected in 395 sera collected from different population in Guizhou area. The partial genes of TTV DNA fragments amplified from two liver disease patients and one hemodialysis patient were analysed and then compared with gene from Japan strain.
RESULTSThe positive rates of TTV DNA in 62 normal adults, 37 blood donors, 50 hemodialysis patients, 107 intravenous drug users (IVDUs), 139 patients with liver diseases were 6.45% (4/62), 8.10%(3/37), 26.00% (13/50), 25.23% (27/107) and 16.54% (23/139), respectively. The homology of TTV DNA sequence was from 99% to 100% within three Guizhou strains. As compared with published sequence, the nucleotide homology is 98% between all three Guizhou strains and Japan strain.
CONCLUSIONSThere are TT virus infection in Guizhou area, the infection rates were higher in hemodialysis patients and IVDUs. The nucleotide homology was high between Guizhou strain and Japan strain.

5.Infrared spectroscopic study on water processing of kusnezoff monkshood root

Objective Infrared spectroscopy was used to study on both raw material and different degree water processing varieties extract of Radix Aconiti kusnezoffii,to observe the changes of main toxic components in processing,and thus to improve the quality of Radix Aconiti kusnezoffii processed products.Methods Fu Liye transform infrared spectroscopy was adopted to study the infrared spectrum characteristic of raw materials and different degree water processing varieties of.Radix Aconiti kusnezoffii.Results ①Aconitine and hypaconitine infrared spectrum showed that 1717 cm-1,1727 cm-1 and 1711 cm-1 is the C=O stretching vibration peak.It is a diester alkaloids characteristic peaks; ② although absorption peak of all vesicular samples had a certain change,it still existed diester alkaloids absorption peak,indicating the incomplete hydrolysis; ③ boiled aconite processing methods demonstrated diester alkaloids absorption peak shift in the water sample.Conclusion Diester alkaloids in Radix Aconiti kusnezoffii shows a positive relation with its time soaked in water.
6.Expressions of octamer-binding transcription factor 4 and phosphorylated-protein kinase B in colorectal cancer and their clinical significances

Objective To investigate the expressions of octamer-binding transcription factor 4 (Oct4) and phosphorylated-protein kinase B (p-Akt) proteins in colorectal cancer and their clinical significances, in order to explore the roles of Oct4 and p-Akt in the staging and grading of colorectal cancer. Methods Immunohistochemical technique was used to examine the expression of Oct4 and p-Akt proteins in 78 cases of colorectal cancer in Wuxi Second Hospital of Traditional Chinese Medicine and Wuxi First People's Hospital from January 2011 to December 2016. Relationship between expressions of Oct4 and p-Akt proteins and clinicopathological parameters were analyzed. Results The positive rate of Oct4 in tumors was 74.36 % (58/78), which was obviously higher than that in normal tissues [35.90 % (20/78), χ2= 23.32, P < 0.01]; and the positive rate of p-Akt in tumors was 67.95 % (53/78), which was obviously higher than that in normal tissues[28.21 %(25/78),χ2=24.68,P <0.01].The double positive and negative expression rate of these two proteins accounted for 80.8 %(63/78), with a linear positive correlation (r= 0.455, P < 0.000 1). In 78 cases of colorectal cancer, the expression of Oct4 protein was correlated with histological grade, lymph node metastasis, and Duke staging (all P < 0.05), and the expression of p-Akt protein was correlated with histological grade and lymph node metastasis (both P < 0.05). The multivariate logistic regression analysis showed that the expression of Oct4 protein was related to histological grade and Duke staging(both P<0.05),and the expression of p-Akt protein was only related to lymph node metastasis (P<0.05). Conclusion The combined detection of Oct4 protein and p-Akt protein has reliable and important clinical significance for judging the histological grade,lymph node metastasis and Duke staging of colorectal cancer.
7. The role of DFO in Al (mal) 3-induced ferroptosis in PC12 cells

Objective:
To investigate the mechanism of Al (mal) 3-induced ferroptosis in rat adrenal pheochromocytoma cells (PC12), to explore the effect of deferoxamine (DFO) .
Methods:
Taken PC12 cells growing at logarithmic phase and divided into 6 groups: control group, 200 μmol/L Al (mal) 3 group, 0.5% DMSO group, 200 μmol/L DFO group, Al (mal) 3+DMSO group, Al (mal) 3+DFO group. DMSO and DFO were added to the DMSO group and the Al (mal) 3+DMSO group, the DFO group and the Al (mal) 3+DFO group for 2 h, respectively, Al (mal) 3 was then added to the Al (mal) 3 group, Al (mal) 3+DMSO group, and the Al (mal) 3+DFO group to a final concentration of 200 μmol/L. The cell viability was detected by CCK8, the morphology and ROS levels of PC12 cells was observed by inverted microscope, the cell proliferation toxicity and intracellular iron ion content were detected by colorimetry, the GSH content and GSH-PX activity were detected by biochemical method.
Results:
Al (mal) 3 exposure significantly inhibited the growth of PC12 cells and destroyed the cell morphological structure, resulting in increased LDH activity and intracellular iron ion content in PC12 cells, decreased GSH content and GSH-PX activity, increased ROS levels; the combined treatment of Al (mal) 3+DFO can significantly improve the cell viability of PC12 cells, improved cell morphology, decreased cell LDH activity and intracellular iron ion content (
