1.Detection methods and clinical significance of chimerism after hematopoieticstem cell transplation

Sequential monitoring of chimerism after hematopoietic stem cell transplantation is important for disease prognosis, improving transplantation success rate, and reducing relapses of the original diseases. The application of molecular genetic markers in combination with quantitative PCR to the assessment of chimerism,in particularly the dynamic quantitative analysis of lineage-specific chimerism, can provide important guidance for clinical practice.
2.Expression of nitric oxide in uterine junctional zone of patients with adenomyosis

Objective To investigate the relationship between nitric oxide and contractile disfunction of junctional zone of smooth muscle by detecting the expression of nitric oxide in uterine junctional zone in patients with adenomyosis.Methods From February 2012 to July 2012,23 patients with adenomyosis undergoing hysterectomy as adenomyosis group matched as 16 patients with cervical carcinoma or ovarian cancer were enrolled as control group in Center of Minimally Invasive Gynecology,Beijing Obstetrics and Gynecology Hospital.Location of nitric oxide synthase (inducible NOS and endothelial NOS) in uterine junctional zone was analyzed by immunohistochemistry.Smooth muscle cells of junctional zone were cultured and cellular mRNA levels of nitric oxide synthase were detected by real-time polymerase chain reaction.Furthermore,concentration of nitrites in lysates of fresh junctional zone tissues and cultured cells were determined by Griess reaction.Results (1) The expression of nitric oxide synthase proteins in smooth muscle cells of junctional zone tissues:the staining of iNOS was intensive in smooth muscle cells of junctional zone in patients with adenomyosis,while staining of eNOS was scant.Both staining of iNOS and eNOS were not observed in control group.(2) The expression of NOS genes in cultured smooth muscle cells from adenomyosis:mRNA level of iNOS was elevated by 1.5 times (P <0.01) while eNOS level was reduced by 2.6 times when compared with those in control groups (P < 0.05).(3) Nitrites concentration in lysates of junctional zone tissues and cultured cells:concentrations of nitrites in lysates of 100 mg tissues were respectively (37 ± 8) μmol/L and (28 ± 5) μmol/L in adenomyosis and control group (P > 0.05).While concentrations of nitrites in 1 × 106 cells were respectively (11.8 ± 0.6) μmol/L and (10.8 ± 1.5) μmol/Lin adenomyosis and control group,which did not reach statistical difference (P > 0.05).Conclusion Upregulation of nitric oxide might be associated with contractile disfunction of smooth muscle cells in uterine junctional zone of patients with adenomyosis.
3.Research on Clinical Application of Vinpocetine Injection

Objective:To investigate the clinical application and adverse events of vinpocetine injection. Methods:The application of vinpocetine injection in the patients in neurology department during December 2013 and December 2014 in a hospital was statistically analyzed. The adverse reactions of the injection reported in the professional literatures and relevant documents were also retrieved and statistically analyzed. Results:The infusion concentration of vinpocetine injection for 363 patients was more than 0. 06 mg·ml -1 ,and 3 cases of adverse reactions appeared with the main symptoms of rash and drug fever. Among 28 published literatures,19 articles were with the infusion concentration of vinpocetine injection above 0. 06 mg·ml-1 and 8 articles reported adverse reactions in varying degrees. Conclusion:Clinicians should pay attention to the instructions in the clinical course of medication in order to improve the safe and rational use of drugs.
4.Effects of Oxidative Challenge on Defensive Enzyme and Cofactors Level in Alkalibacterium sp.F26

In this paper,a slight halophilic alkaliphile swain,Alkalibacterium sp.F26,which produced high level of intracellular CAT observed in previous research,was selected as a model microbial to explain the responses of this bacterium to oxidative stress.The results indicated that Alkalibacterium sp.F26 had obvious responses to higher concentration (>1 mmol/L) of H2O2 than that to lower H2O2 (<1 mmol/L) challenge from the aspects of defensive enzyme synthesis and cofactors level variation.As for catalase production,the activity increased up to 106.54 U/rag protein which was 1.76 fold of the control when cells were challenged by 3 mmol/L H2O2,but its activity only was 1.13 fold when H2P2 was 100 μmol/L.As far as energy state was concerned,ATP production and NAD+ generation were significantly inhibited from 20.55 μmol/L to 17.80 μmol/L and 69.89 μmol/L to 31.77 μmol/L,respectively,leading to the drop of energy charge from 0.77 to 0.68 and the increase of the portion of NADH/NAD+ from 0.08 to 0.41 in the former case.However,these effects were less distinct under lower concentration of H2O2.Except of the condition of 100 μmol/L H2O2,under which the activation of defensive mechanism resulted in an increase of ATE the level of ATP dropped from 22.69 μmol/L of the control to 22.38 μmol/L and 13.70 μmol/L when challenged by 50 μmol/L and 500 μmol/L H2O2.Besides,the concentration of NADH fluctuated and the NAD+ gradually reduced when H2O2 below 1 mmol/L.
5.Expression of RhoA and Rho kinase in junctional zone of human adenomyosis and its relationship with dysmenorrheal

Objective To investigate the expression of RhoA and Rho kinase in junctional zone (JZ) of adenomyosis and normal myometrium and explore its relationship with severity of dysmenorrheal.Methods From Mar.to Dec.2012,32 cases with adenomyosis undergoing hysterectomy were enrolled as adenomyosis group including 18 cases with proliferative endometrium and 14 cases with secretory endometrium matched with 29 cases with hysterectomy due to cervical disease and ovarian tumor as control group including 12 cases with proliferative endometrium and 17 cases with secretory endometrium in Beijing Obstetrics and Gynecology Hospital Affiliated to Capital Medical University.JZ smooth muscle cells were isolated and cultured immediately after the operation.The expression of mRNA and protein of RhoA and ROCK Ⅰ in JZ in two groups were measured by real-time fluorescence quantitative RT-PCR and western blot.Results (1) The mRNA expression of RhoA and ROCK Ⅰ in JZ of adenomyosis group did not show cyclic change.In proliferative phase,the expression of RhoA and ROCK Ⅰ (1.41 ±0.16,1.05 ±0.15) was not significantly higher than that in secretory phase (1.17 ± 0.25,0.98 ± 0.10) (P > 0.05).While JZ in control group,it showed obviously cyclic change.The expression level of them in proliferative phase(0.93 ±0.10,1.00 ± 0.18) was significantly higher than that in secretory phase (0.48 ± 0.03,0.55 ± 0.05) (P <0.05) ; It also showed that expressions of RhoA and ROCK Ⅰ in adenomysis group were significant higher than those in the control (P < 0.05).(2) The mRNA and protein expression of RhoA and ROCK Ⅰ was positively correlated in each of two groups (r =0.48,P < 0.01 ; r =0.67,P < 0.01).(3) The expression of RhoA and ROCK Ⅰ were 1.66 ±0.19,1.32 ±0.11 in severe dysmenorrheal,1.28 ±0.12,1.09 ±0.08 in moderate dysmenorrheal and 0.93 ± 0.09,0.81 ± 0.06 in mild dysmenorrheal,it all reached statistical difference when compared the other group.(All P < 0.05).Conclusions The expressions of RhoA and ROCK Ⅰ in JZ in adenomyosis group were higher than those in control group,and positively correlated with the severity of dysmenorrheal in adenomysis group,but it does not change with the menstrual cycle.High expression of RhoA and ROCK Ⅰ might be involved in abnormal contraction of uterine myometrium and correlated with the dysmenorrheal in adenomysis.
6.Clinical significance of cyclooxygenase-2 and BCRP expressions in non-Hodgkin lymphoma

Objective To investigate the correlation and significance of cyclooxygenase-2 (COX-2)and the breast cancer resistance protein (BCRP) expressions in non-Hodgkin lymphoma(NHL). Methods Ten patients with reactive lymph nodes (RLN) were considered as a control group. Compared with β-actin as internal control, the BCRP mRNA expressions of 61 NHL samples and 10 lymph tissues in control group were detected by semi-quantitative revere transcription polymerase chain reaction (RT-PCR) assay, while the expressions of COX-2 protein of the above specimens were detected by SP immunohistochemistry. Results The positive rates of COX-2 and BCRP in NHL were 50.8 %(31/61) and 45.8 % (28/61), respectively , and were higher than those in the control group (P <0.05). The expression of COX-2 was statistically positive correlated to that of BCRP (X2 =8.795, r=0.355, P<0.05). The expressions of COX-2 and BCRP were not correlated to clinical and pathological factors, such as age, sex, IPI, LDH, β2-microglobulin level and Ann Arbor stage, however, the expression of BCRP was statistically correlated to chemotherapy efficacy.Conclusion BCRP may be involved in multi-drug resistance (MDR) of NHL, so it may contribute to the assessment of chemotherapy and prediction of NHL. Since there is a strong correlation between COX-2 expression and MDR in NHL, the application of COX-2 inhibitors may enhance sensitivity of chemotherapy.
7.Application of nucleic acid test in voluntary blood donors in Hefei area

Objective To discuss the role of nucleic acid test(NAT)in blood screening to provide the scientific basis for the se-lection of blood screening strategies.Methods The serological detection and NAT were simultaneously performed on 68 662 speci-mens of voluntary blood donation from January 2012 to April 2013;29 samples of HBV DNA positive with serological HBsAg nega-tive were performed the serum HBV markers detection.Partial samples of anti-HIV antibody positive were sent to CDC for conduc-ting the confirmation test.Results Among 68 662 samples,120 cases of single NAT positive were detected out,the residual risk of blood transfusion was 0.175%.In the serum HBV markers detection,the mode of HBcAb was predominant.11 cases of HIV posi-tive were confirmed and all were the NAT positive samples.Conclusion NAT can reduce the residual risk of blood transfusion,en-sure the safety of blood transfusion.NAT and the serological detection are mutual complementation and not replaced by each other. The suitable screening strategy shold be selected.
8.Effect of Gemcitabine on viability and apoptosis of non-small cell lung cancer HCC827 in vitro

Objective:To observe the effect of Gemcitabine ( GEM) on the viability and apoptosis of non-small cell lung cancer HCC827 in vitro. Methods:The cell viability,apoptosis and cell cycle of HCC827 cells induced by Gemcitabine were detected with cell counting kit-8 assay (CCK-8),Annexin V-FITC/PI staining and flow cytometry. The expression of Bcl-2 protein of cells treated with GEM was examined by Western blot assay. Results: There was significant inhibition effect on HCC827 cells treated with 0. 1-1 000 ng/ml of GEM,which can promote the occurrence of HCC827 cell apoptosis and arrest cell in the S phrase. The apoptosis induced by GEM was accompanied with the down regulation of Bcl-2 protein. Conclusion: GEM can inhibit the cell viability and induce the HCC827 cell apoptosis and S phrase arrest. Its cell dead type was apoptosis,which was related with the expression of Bcl-2 protein.

Time to positivity(TTP) is a new parameter in blood culture field.The article shows us the concepts of TTP,differential time to positivity (DTTP),and introduces their relation with bloodstream infection (BSI),catheter-related bloodstream infection (CRBSI).Besides,it stresses TTP' s clinical application,including determining the severity of disease;identifying the isolates whether pollution or not; identification of isolate strains ; detection of the drug resistance of isolates ; evaluating the effect of antibiotic ; helping to adjust the therapeutic drug; diagnosing or excluding CRBSI by means of DTTP;deciding whether the catheter is the source of infection in patients with candidemia; understanding the epidemiological distribution of strain.At the same time,the article also describes the shortcomings and domestic current status.
10.Analysis on HIV detection and epidemiological status of voluntary blood donors in Hefei area

Objective To understand the HIV infection status and epidemic characteristics among blood donors in Hefei area to provide the basis for recruiting the blood donors and establishing the blood detection scheme.Methods Two kinds of serological re-agent were adopted for screening the blood donors.HRV RNA in the detection samples of the nucleic acid test system and positive serological samples were simultaneously sent to the municipal Center for Disease Control for conducting the confirmation detection from 2012.Results 495 279 blood donors were performed the serological screening,44 cases were confirmed HIV positive with the detection positive rate of 0.9;112 940 samples were performed the NAT detection,20 cases were confirmed HIV positive,all were the serological dual reagent positive samples.Conclusion The HIV infection among unpaid blood donors in Hefei area shows the increasing trend year by year.Effective measures must be taken to ensure the safety of blood.
