1.A clinical observation on efficacy of enema of Shexue Fuyang decoction combined with massage of Hui medicine for treatment of patients with ulcerative colitis

Objective To explore the clinical effect of Chinese medicine enema combined with massage of Hui medicine for treatment of patients with ulcerative colitis (UC).Methods A prospective randomized controlled trial (RCT) was conducted, 390 patients with UC admitted to Department of Traditional Chinese Medicine (TCM) of Ningxia Hui Autonomous Region People's Hospital from January 2013 to December 2014 were enrolled, and they were divided by using random envelope method into the western medicine control group (88 cases), simple TCM enema group (102 cases), simple massage of Hui medicine group (94 cases) and enema combined with massage group (106 cases). The patients in western medicine control group took sulfasalazine orally, once 0.75 g, thrice a day. The simple TCM enema group adopted the self prepared Shexue Fuyang decoction (ingredients: carbonized sanguisorba root 15-20 g, bletilla striata 15 g, patrinia 15-30 g, sculellaria barbata 15-30 g, Chinese goldthread 15 g, radix glycyrrhizae preparata 15 g, Fry terminaliachebuia 10 g, yam 15-30 g, india madder root 15 g) retention enema once a day. The following principles should be followed: firstly, the dose of drug should be large enough (250 mL), secondly the speed of enema should be relatively slow and it finished in 15-20 minutes, thirdly the temperature of decoction should be kept at moderate degree about 39-41 ℃ (a warm bag being around the enema tube), and lastly, the patient's posture should be changed (lying left-side for 20 minutes, lying at chest-abdomen position for 20 minutes, lying right-side for 20 minutes). In massage of Hui medicine group, based on the location and size, depth and seriousness of the lesion, the doctor would selectively use palm or index finger to carry out massage manipulations of pressing, fractioning, rubbing or grasping for different disease situations of patients, once 30 minutes pre day. The patients in enema combined massage group were treated with the same methods as above, and the therapeutic course of the four groups was 15 days; at the end of experiment,the therapeutic effects and the improvement of main symptoms in four groups were observed.Results The clinical total effective rates of Chinese medicine enema group, massage of Hui medicine group, enema combined massage group were obviously higher than the rate of western medicine control group [78.4% (80/102), 85.1% (80/94), 93.4% (99/106) vs. 62.5% (55/88), allP < 0.05]; the symptom integrals, such as abdominal pain, diarrhea, tenesmus, purulent blood stool, fever, etc. were significantly decreased after treatment in the TCM enema, massage and combined treatment groups, and the integrals of the three treatment groups were obviously lower than those in the western medicine control group (abdominal pain: 0.45±0.75, 0.56±0.57, 0.35±0.96 vs. 1.13±0.86, diarrhea: 0.76±0.91, 0.78±0.69, 0.55±0.81 vs. 1.43±0.34, tenesmus: 0.52±0.37, 0.44±0.23, 0.21±0.13 vs. 0.72±0.98, purulent blood stool: 0.43±0.34, 0.50±0.24, 0.52±0.76 vs. 0.68±0.44, fever: 0.32±0.56, 0.35±0.65, 0.26±0.45 vs. 0.49±0.77, allP < 0.05).Conclusion The therapeutic effects of Shexue Fuyang decoction enema combined with massage of Hui medicine for treatment of patients with UC are better than those of each of the following treatment alone: western medicine, Chinese medicine enema or medical massage treatment.
2.Effects of adrenomedullin and proadrenomedullin N-terminal 20 peptide, alone or in combination, on the rat hearts in vitro

Objective: To study the effects of adrenomedullin (ADM) and proadrenomedullin N terminal 20 peptide (PAMP) alone or in combinations on the isolated rat hearts as well as the possible signaling pathways involved in their actions. Methods: In isolated rat hearts, the left ventricular pressure (LVP), LVP?dp/dtmax, coronary fluid (CF) and heart rate(HR) of the hearts infused at different concentrations of ADM and/or PAMP were determined by a 4 cannal physiological recorder, then the cAMP contents were assayed in myocardium. Results: After being infused with ADM from 10 -11 to 10 -8 mol?L -1 or PAMP from 10 -11 to 10 -8 mol?L -1 , the LVP and LVP?dp/dtmax of the isolated hearts decreased gradually in a concentration dependent manner, and at the same concentration, the effects of PAMP were more potent than those of the ADM. When ADM and PAMP were co administrated with both concentrations as low as from 10 -11 to 10 -10 mol?L -1 , the cardiac parameters were decreased more than either ADM or PAMP administrated alone. However, the inhibitory effects of ADM and PAMP were attenuated when they were in combination at higher concentrations as from 10 -9 to 10 -8 mol?L -1 . When the rat hearts were infused with ADM, PAMP,and ADM plus PAMP, the CF were always higher than those of the controls and decreased when co administrated with L NAME, an inhibitor of NOS, but the decreaseddegree of LVP and LVP?dp/dtmax were attenuated by L NAME.The cAMP contents in the left cardiac ventricle were increased significantly by ADM infusions but not changed obviously by PAMP, and were of no statistical difference in rat hearts with ADM administrated alone from those combinated with ADM and PAMP. Conclusion: These results showed that ADM and PAMP infused alone or in combinations inhibited the function of rat hearts in vitro, which might be partly involved with the NOS/NO pathway.
3.PCR-RFLP in gene diagnosis of spinal muscular atrophy

Objective To study the value of the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in gene diagnosis on spinal muscular atrophy (SMA).Methods PCR-RFLP method was used to detect the homozygous deletion of the exon 7 or exon 8 of SMN gene in 20 SMA patients of Type Ⅰ,Ⅱ,Ⅲ and 15 normal individuals.Results Homozygous deletion of exon 7 and exon 8 of the SMN gene were all identified 7/7 in SMA TypeⅠpatients, and 5/5 and 4/5 respectively in SMA Type Ⅱ patients, but only 1/8 of SMA Type Ⅲ patients, and no homozygous deletion was found in the normal controls.Conclusions PCR-RFLP might be recommended as an effective diagnosis for spinal muscular atrophy Type Ⅰand Ⅱ patients, whereas the method might not be as useful in Type Ⅲ as in Type Ⅰand Ⅱ for the gene diagnosis.
4.Effect of hydrogen sulfide on rat cardiac function in vitro

AIM: To observe the effect of hydrogen sulfide (H_2S) on the rat cardiac function in vitro, to explorer the physiological regulation of endogenous H_2S on myocardial action. METHODS: H_2S concentration and production in the rat myocardial tissues were detected. The expression of CSE (a kind of key enzyme of endogenous H_2S production) mRNA in myocardial tissues was screened by RT-PCR. Langendorff apparatus was used to perfuse the rat heart in vitro. After 20 minutes of stabilization, NaHS (10~-6 -10~-3 mol/L) were added cumulatively in order to the perfusive fluid, and another group applied physiological concentration NaHS (4?10~-4 mol/L), continuously perfusion for 20 min, heart rate (HR), difference of left ventricular pressure (△LVP), left ventricular peak rate of contraction (+LV dp/dt_~max ), peak rate of relaxation (-LV dp/dt_~max ) and coronary perfusive flow (CPF) were measured at the times. Finally, glibenclamide was applied to block the K_~ATP channel of heart, to observe the effect of NaHS at physiological concentration on cardiac function. Aim To study the effects of resveratrol(Res)on the release of soluble adhesion molecules from human umbilical vein endothelial cells(HUVECs)induced by N-formyl-methionyl-leucyl-phenylal-anine(fMLP)and the adhesion between neutrophils and HUVECs.Methods The effects of Res on neutrophils adhesion to human umbilical endothelial cell(HUVECs)triggered by fMLP were examined.The soluble intercellular cell adhesive molecule-1(ICAM-1),the soluble vascular cell adhesive molecule-1(VCAM-1)and E-selectin release from fMLP(10 ?mol?L-1)stimulated HUVECs were determined by ELISA kits.The neutrophils isolated from human vein blood were loaded with Fluo-3,a fluorescent indicator,to detect intracellular free calcium concentration([Ca2+]i),and CLA was used as a chemiluminescent indicator to determine superoxide production in neutrophils.Results Res(1~50 ?mol?L-1)significantly inhibited neutrophil adhesion to fMLP-stimulated HUVECs and also obviously downregulated the levels of ICAM-1,VCAM-1 and E-selectin in supernatant of HUVECs culture stimulated by fMLP in the dose-dependent pattern.Res also suppressed fMLP-activated superoxide generation and[Ca2+]i increase in neutrophils.Conclusions Res involved in neutrophil adhesion to HUVECs intermediated by cell adhesive molecules expression trigged by[Ca2+]i and superoxide production in neutrophils,which means a lot to prevent inflammatory diseases.
6.Intermedin inhibiting cardiac injury induced by ischemic/reperfusion in rat heart

Aim To determine the protective effects of IMD on ischemia/reperfusion(I/R) injury and its possible mechanism.Method Isolated rat hearts were perfused by Langendorff mode,and after 45 min global ischemia and 30 min reperfusion ventricular function was measured on a Power Lab,and adequate amount of ventricular tissues and perfusate were collected for biochemical measurement.Results Treatment with IMD during the reperfusion period significantly attenuated the effects of I/R on cardiac function inhibition and tissue injury.Compared with I/R group,IMD induced increase in △LVP,LV(dp/dtmax,HR and CF,whereas induced decrease in LVDP.Reperfusion with IMD exerted decrease in LDH,total protein,Mb and MDA content compared with I/R group,but increased myocardial cAMP content.All these values were similar to the effects of ADM.Furthermore,I/R induced significant increase in Bmax and Kd value.Conclusion IMD exerted beneficial effects on cardiac injury induced by I/R which might be mediated by cAMP pathway.And the cardioprotective effects of IMD were equal to ADM,a potent cytoprotective factor.
7.Effects of Lokomat Robotic-assisted Gait Training Combined with Drop Foot Stimulator on Gait Function in Patients with Stroke

Objective To investigate the effects of Lokomat robotic-assisted gait training combined with drop foot stimulator on gait function in patients with stroke. Methods Thirty-six stroke patients were randomly divided into control group (n=18) and observation group (n=18). Both groups received routine rehabilitation training. The control group accepted Lokomat robotic-assisted gait training, while the ob-servation group worn drop foot stimulator in addition. The training intensity was 30 minutes every day, five days per week for 6 weeks. Fugl-Meyer Assessment-Lower Limb (FMA-LL), Functional Ambulation Category (FAC) and footprint analysis were used to evaluate the motor ability of lower limbs, walking ability and gait before and after training. Results There was no significant difference in the scores of FMA-LL, FAC and gait parameters (walking speed, stride width, stride length difference) before training (t<0.765, Z=0, P>0.05). The scores of FMA-LL, FAC and walking speed improved, and the stride width and stride length difference decreased after training (t>2.190, Z>3.630, P<0.05). After training, the scores of FMA-LL, FAC, walking speed and stride length difference were better in the observation group than in the control group (t>2.030, Z=-2.560, P<0.05), however, there was no significant difference in stride width between two groups (t=0.570, P>0.05). Conclusion Lokomat robotic-assisted gait training combined with foot drop stimulator could improve the lower limb motor func-tion and gait ability in patients with stroke, and the effect might be better than using Lokomat robotic-assisted gait training only.
8.Clinical analysis of 314 children with chest pain

Objective To understand the cause of chest pain (precordium indisposition) in children and explore the thread of thought in its analysis.Methods Clinical data of 314 children with chest pain who visited the cardiovascular department for children of our hospital were collected and analyzed to study its causes.Results The cause could be attributed to organic cardiac diseases in 43 of 314 children (13.7%) with chest pain,to non-organic cardiac diseases in 108 (34.4%),to psychiatric-psychological factors in 60 (19.1%) and to unknown cause in 41 (13.1%).Average age of children with organic cardiac diseases was younger than that of children with other causes (P<0.05).Symptoms accompanied simultaneously in children with organic cardiac diseases were much more frequent significantly than those in them with non-organic cardiac chest pain and unknown causes (P<0.05).Conclusions Chest pain in children could be caused by a few causes,which was non-specific symptom for organic cardiac diseases,so, more should be taken into account in analyzing the cause of chest pain in children.
9.Effect of Conditioned Medium from Endothelial Cells on Cancer Stem Cell Phenotype of Hepatoma Cells.

In this study, we aimed to investigate the influences of conditioned medium from human umbilical vein endothelial cells (HUVEC) on cancer stem cell phenotype of human hepatoma cells. HUVEC and human hepatoma cells (MHCC97H) were cultured, respectively, and then the MHCC97H cells were co-cultured with conditioned medium from HUVEC (EC-CM) with Transwell system. Anti-cancer drug sensitivity, colony-formation, migration/invasion ability, expression of cancer stem cell marker and sphere formation were performed to determine the cancer stem cell phenotype in MHCC97H cells. We found that MHCC97H cells co-cultured with EC-CM exhibited significantly higher colony-formation ability and lower sensitivity of anti-cancer drugs 5-FU and Cis. Transwell assay showed that treatment with EC-CM obviously increased migration and invasion of MHCC97H cells. Moreover, increased sphere forming capability and expression of CD133 in MHCC97H cells were observed after co-cultured with EC-CM. These results suggested that EC-CM could promote cancer stem cell phenotype of hepatoma cells.

10.STUDIES ON XYLANASE PRODUCTION BY ASPERGILLUS CLAVATUS 22

A high xylanase producing strain 22 of Aspergillus clavatus was screened from 105 strains of molds and yeasts. The suitable medium consisted of (g/L): bagasse hemicellulose 30, NH_4NO_3 5, yeast extract 5, wheat bran 10, Tween 80 1 and a small quantity of other minerals; initial pH 5.5. Theoptimalsporeinoculumwas4.9X10~6spores/ml (final concentration). Theactivity of xylanase was as high as 335.9 U/ml in shake-flask experiment at 28℃ for 72 h. The optimal temperature and pH for xylanase reaction were 50℃ and pH 4.8. 72.6% of its original activity was remained after incubation at 50℃ for 1 h, and 90% of the enzym activity was observed upon storage at 8℃ for 9 days . Sugars. Na~+. Ca~(2+). and Zn~(2+) increased its activity wherease Co~(2+)and Cu~(2+) inhibited it.
