1.The Notch signaling pathway is involved in intestinal adaptation in a massive small bowel resection rat model

Objective To investigate the role of Notch signaling in massive small bowel resection(MSBR)-associated crypt epi-thelial cell proliferation.Methods Male Sprague-Dawley rats were subj ected to sham operation(bowel transection and reanastomo-sis)or 70 % mid-SBR.Intestinal tissue samples were collected at 7 d after operation.The expression of Notch pathway mRNAs and proteins were analyzed by RT-PCR and Western blot.The expression of the Notch pathway proteins Jagged-1,NICD and Hes-1 also determined through immunohistochemical.Furthermore,IEC-6 cells were used to examine the function of the Jagged-1 signaling system.Results SBR increased crypt epithelial cell proliferation and increased expression of Jagged-1 and Hes-1 mRNA and protein along with cleaved Notch-1.Immunohistochemical staining showed that Jagged-1,cleaved Notch-1 and Hes-1 colocalized in the same crypt epithelial cell population.Recombinant Jagged-1 significantly stimulated the proliferation of IEC-6 cells.Conclusion The Jag-ged-1/Notch-1/Hes-1 signaling pathway is involve in intestinal adaptation through increase crypt epithelial cell proliferation.
2.The effect of letrozole plus metformin on infertility of polycystic ovary syndrome

Objective To study the effect of letrozole plus metformin on infertility of polycystic ovary syndrome(PCOS).Methods According to the digital table,90 patients with infertility of PCOS were randomly divided into the control group(n =45 cases) and the treatment group(n =45 cases).The patients of the control group were treated though clomiphene,while the patients of the treatment group were treated though letrozole plus metformin.The ovulation rate,pregnancy rate,sex hormones and insulin resistance were detected.Results The homeostasis model assessment of insulin resistance index (HOMA-IR) was (5.72 ± 2.37) before treatment and (3.16 ± 1.39) after treatment in the treatment group.HOMA-IR was (5.69 ± 2.40) before treatment and (4.81 ± 2.06) after treatment in the control group.There was a significantly decrease in the treatment group (t =3.7246,P < 0.05).The ovulation rate was 80% in the treatment group,while 60% in the control group,the difference between the two groups was significant(x2 =4.06,P <0.05).The pregnancy rate was 48.9% in the treatment group,while 28.9% in the control group,the difference between the two groups was significant (x2 =5.77,P < 0.05).Conclusion Letrozole plus metformin could promote the ovulation and decrease insulin resistance.
3.Cyclosporine inhibits apoptosis in experimental murine xerophthalamia conjunctival epithelium.

This study examined the inhibitory effect of topical cyclosporine (CsA) treatment on conjunctiva epithelial apoptosis in a murine model of xerophthalamia. Dry eye was induced in 3 groups of C57BL6 mice by subcutaneous injection of scopolamine (t.i.d) and exposure to an air draft and low-humidity environment for 16 h each day for 12 days. The dry eye control group received no topical treatment; another group received 1 microL of 0.05% CsA topically (t.i.d, dry eye+CsA); and the third group received 1 microL of the castor oil vehicle of CsA topically (t.i.d, dry eye + vehicle). Normal mice were used as untreated controls. Twelve days later, the mice were killed, and their conjunctivas were excised. The number of the conjunctival goblet cells was counted in tissue sections stained with periodic acid Schiff (PAS) reagent. Their conjunctiva epithelium had been investigated by immuno-histochemical staining to detect the goblet cells and the expression of Caspase-3, Bax and bcl-2. Our results showed that compared with dry eye control and dry eye mice + vehicle groups, the number of conjunctival epithelial goblet cells was significantly greater in the untreated controls and dry eye mice receiving CsA (P < 0.01 for both groups). There was no significant difference in the number of conjunctival epithelial goblet cells between the dry eye control and dry eye+vehicle group. It was also true of the number of conjunctival epithelial goblet cells when comparison was made between the normal group and the dry eye+CsA group. Expressions of Caspas-3 and Bax were increased and ex-pression of bcl-2 was decreased in conjunctival epithelial cells in dry eye control and dry eye mice+vehicle groups. There was a significant positive correlation between goblet cell number and the number of cells that expressed bcl-2, and a negative correlation between goblet cells and Caspase-3 and Bax expression. It is concluded that the topical use of CsA could significantly reduce conjunctival epithelial apoptosis and protect goblet cell against the loss in experimental murine xerophathalamia. Inhibition of apoptosis appears to be a key mechanism responsible for the therapeutic effect of CsA on xerophthalamia.
5.Expression and protective effect of induced heat shock protein 70 in retinal neurons and M?ller cells

Objective To investigate the expression of induced heat shock protein (HSP) 70 in rat′s retinal neurons (RNs) and M?ller cells, and evaluate the protective effect of HSP 70 on RNs injured with glucose deprivation and glutamate. Methods Rat′s RNs and M?ller cells cultured in vitro were treated with heat shock (42℃ for 1 hour), and duration of the expression of HSP70 was detected by immunocytochemical techniques. Viability of the cells was measured by methyl thiazolyl tetrazolium (MTT) chromatometry after incitant toxic injury with glucose deprivation (0.56 mmol/L glucose for 6 hours) and glutamate (100 ?mol/L for 6 hours). Simultaneously, the expression was interdicted by HSP70. Results Hypereffective expression of HSP70 was found in cultured RNs and M?ller cells after heat shock. The viability of RNs pretreated by heat shock after injured with glucose deprivation and glutamate significantly increased which could be interdicted by HSP70 antibody. Conclusion Hypereffective expression of HSP 70 may be induced by heat shock, which enhances the ability of tolerance of RNs to the incitant toxic injury by glucose deprivation and exitotoxicity.
6.Distribution and Antibiotic Resistance of Escherichia coli in Hospital

OBJECTIVE To understand the distribution and antibiotic resistance of E.coli isolated from our hospital and offer suggestions for clinic.METHODS The antibiotic susceptibility to 21 kinds of antibiotics were tested by K-B method and analyzed by Whonet software.RESULTS The antibiotic resistance of 301 E.coli strains to ampicillin,quinolones,piperacillin,gentamicin,cefazolin,cefuroxime,ceftriaxone,ceftazidime,aztreonam,cefepime, ampicillin/sulbactam were 83.39%,69.44%,62.79%,44.19%,42.86%,36.88%,35.22%,31.23%,29.90%,29.57% and 26.91%,respectively.CONCLUSIONS E.coli isolated from our hospital show high resistance to several kinds of antibiotics.The drugs should be choosed reasonably according to their antibiotic suscepitibity results.

Ezrin,a member of the ERM(Ezrin/Radixin/Moesin)protein family,fuctions as a molecular linker between the cytoskeleton and the plasma membrane.Recently,it has been found to be expressed abnormally in many malignant tumors,which indicats that it plays indicative of an important role in tumor infiltration and metastasis.Ezrin influences tumor metastasis in many ways,aspects including altering tumor cell movement,regulating cell-cell adhesion,participating in signal transduction in tumor cells,inhibiting cell apoptosis and regulating the function of phagocytes.
8.Making Monoclonal Antibodies by Phage Antibody Libraries

The emergence of repertoire cloning using phage antibody libraries has revolutionized the antibody engineering technology. We prepared phage antibody libraries from both human breast cancer cell immunized murine spleen cells and human peripheral lymphocytes from volunteer received HBsAg vaccination. Mouse anti-human breast cancer cell antibodies and human anti-HBsAg antibodies were successfully cloned by panning of the phage antibody libraries against breast cancer cells and immobilized HBsAg. The resulting Fab fragments were expressed in E.coli cells and the specificity, affinity and DNA sequences were analyzed. The results showed the potential of the phage display system in antibody preparation.
9.Allogeneic hematopoietic stem cell transplantation using reduced-intensity Bu/Cy conditioning for patients with hematologic malignancies

Objective To explore the efficacy of allogeneic hematopoietic stem cell transplantation using reduced-intensity Bu/Cy conditioning for patients with hematologic malignancies.Methods Five patients with hematologic malignancies were treated by allogeneic hematopoietic stem cell transplantation using reduced-intensity Bu/Cy conditioning,which consisted of busufan3~4 mg/(kg?d) for 3 days,cyclophosphamide50 mg/(kg?d) for 2 days, Ara-C 2 g/(m~2?d) for 1 or 2 days,and CsA 3 mg/(kg?d) and mycophenolate mofetil 1 g/d 7 days before the transplantation.Results Five patients established successful engraftment and no severe complications occured.After a follow-up of median 10.5(3-22)months,five patients still survived without diseases.Conclusion Reduced-intensity Bu/Cy conditioning may reduce transplantation-related toxicities.Allogeneic hematopoietic stem cell transplantation using reduced-intensity conditioning is a safe and effective option for the patients with hematologic malignancies.
10.Expression of Id3,VEGF and C-erbB-2 and clinical significance in patients with breast cancer

Objective To investigate the correlation between clinicopathological parameters and the expressions of inhibitor of differentiation/DNA binding 3 (Id3), vascular endothelial growth factor (VEGF) and human epidermal growth factor receptor-2 (HER-2/C-erbB-2) in patients with breast cancer. Methods The protein expressions of Id3, VEGF and C-erbB-2 were detected by SABC immunohistochemistry in 70 spceimens of breast cancer, 40 specimens of tissue adjacent to breast cancer, 20 specimens of fibroadenoma and 20 specimens of normal breast tissue.
