1.Clinical application of two kinds of connecting tubes in hemodialysis combined with perfusion

Objective To explore the clinical effect of two different connecting tubes in hemodialysis combined with hemoperfusion. Methods A total of 25 patients were selected and divided into observation group and control group by self- control method. In the observation group, the hemodialysis combined with hemoperfusion was used 8 times, and the control group was treated with hemodialysis combined with hemoperfusion 8 times using conventional connecting tube. The time of the unloading of the perfusate and the amount of physiological saline required were compared between the two groups. There was no blood spillover during the unloading of the perfusate, the number of cases of allergic reaction during the treatment, and the coagulation of the dialyzer and the pipeline after the treatment. Results There was no allergic reaction in both groups. There was no significant difference between the two groups in the amount of saline needed to return blood (t=46.412, P=0.307). In the control group, there was 15 cases of blood spillover, while the observation group did not show blood spillover. There was significant difference between the two groups in unloading perfusion time(4.43±0.14)min vs. (3.02±0.11) min (t=10.784, P=0.003). The level Ⅰ and Ⅱ blood coagulation of dialyzer and pipeline was 12, 2 cases in the control group after the treatment and 5, 0 case in the observation group, no level Ⅲ blood coagulation cases, and there was a significant difference between the two groups (χ2=10.667, P<0.01). Conclusion In the hemodialysis combined with hemoperfusion therapy, multi-function group of the application effect is superior to conventional piping, is worthy of clinical application.
2.Analysis of glomerular complement C5b-9 deposits and synthesis of NO, TNF? in the model of the rats with anti-thymocyte serum nephritis

AIM: To explore the localization and semi-quantification o f the glo merular complement C5b-9 complexes and synthesis of some inflammatory mediators or cytokines such as nitric oxide (NO) and tumor necrosis factor ?(TNF?) in the ra ts with anti-thymocyte serum nephritis(ATSN). METHODS: The animal mo del of rat AT SN was reproduced by a single intravenous injection of anti-thymocyte serum (ATS ). Then, the deposits of glomerular C5b-9 complexes were localized and quantifie d by immunohistochemical staining and microscopic image scanning separately. And the glomerular mesangial cells (MC) surrounded by C5b-9 complexes were counted under microscope. In addition, the expression of glomerular MC inducible NO synt hase(iNOS) mRNA and excretion of urinary NO metabolite ( NO - 2/NO - 3 ) and TNF ? in the rats with ATSN were detected. RESULTS: The MC in t h e rats with ATSN emerged necrosis followed by a rapid proliferation. In the early time of MC injury, the C5b-9 complexes were mainly seen in glomerular mesangium and MC sur fac e. But with the progression of ATSN, the MC enclosed by C5b-9 appeared gr adual decrease. Moreover, the expression of MC iNOS mRNA in early stage of ATSN obviously increased and the excretion of urinary NO - 2/NO - 3 and TNF ? also significantly increased. However, the changes of parameters mentioned abov e in ATSN proliferative stage (after 7 days) alleviated gradually. CONCLUSION: The second ary lysis of MC has relation to the deposition of C5b-9 complexes and synthesis and release of NO and TNF ? in rats with ATSN.
3.Effects of Combination of Wax Therapy and Occupational Therapy on Spastic Cerebral Palsy with Thumb Adduction

ObjectiveTo observe effects of combination of wax therapy and occupational therapy on spastic cerebral palsy with thumb adduction.Methods100 cases of children with spastic cerebral palsy were randomly divided into observation group(n=50) and control group(n=50). The observation group received occupational therapy and wax therapy, while the control group received only occupational therapy. The effect was compared between the two groups 3 months after treatment.ResultsThe two groups improved significantly 3 months after treatment, while the observation group was better than the control group(P<0.001).ConclusionWax therapy combined with occupational therapy have better effects on thumb adduction in spastic cerebral palsy than simple occupational therapy.
4.Effects of Occupational Therapy in Group on Cognitive Function of Children with Cerebral Palsy

Objective To explore the effect of Occupational Therapy in Group on cognitive function of children with March to cerebral palsy, and to observe the evaluation results of P300 of event-related potentials on cognitive function. Methods 40 children with spastic cere-bral palsy from March to September, 2015, were divided into control group (n=20) and observation group (n=20). Both groups received rou-tine rehabilitation training, and the observation group received Occupational Therapy in Group additionally, for 3 months. They were as-sessed with Wechsler Preschool and Primary Scale of Intelligence (WPPSI) and P300 of event-related potentials before and after treatment. Results The latency of P300 shortened after treatment (t>7.778, P<0.001), and the amplitude of P300 was larger after treatment (t>5.808, P<0.001) in both groups, especially in the observation group (t>0.461, P<0.05). The score of WPPSI increased after treatment in both groups (t>5.944, P<0.001), and was higher in the observation group than in the control group (t=-3.472, P=0.001). Conclusion Occupational Ther-apy in Group could further improve the cognitive function of children with spastic cerebral palsy. P300 can be used as an assessment tool for cognitive function.
5.Isoflavonoid Glycosides from the Pericarps of Sophora japonica

AIM Five Isoflavonoid glycosides were isolated from the pericarps of Sophora japonica L. and their structures were elucidated by chemical and spectral method as genistein 7,4′-di-O-β-D-glucoside(Ⅰ), sophorabio-side(Ⅱ), prunetin 4′-O-β-D-glucoside(Ⅲ), sophororicoside(Ⅳ) and genistin(Ⅴ). Among them, compounds Ⅰ, Ⅲ and Ⅴ were isolated from in S. japonica for the first time.

Objective To investigate the effect of octreotide on inhibition of growth of subcutaneously implantedtumor with human gall bladder cancer cells in nude mice,and to explore the mechanisms.Methods We established subcutaneous implanted tumor model in nude mice by using human gallbladder carcinomacell line GBC-SD.A total of 18 male nude mice bearing xenografts of the cell line were randomly divided into therapy and control groups,with 9 in each group.Octreotide was administered intraperitoneally at a dose of 100 ?g/(kg?d) to the therapy group and isovolumic normal saline was administered to the controlgroup for 6 weeks.All mice were put to death,and the weight and volume of the tumors were assayed.Flow cytometry was used to examine apoptosis of tumor cells.Immunohistochemical staining was used to examin the expression of p53,bcl-2,and Ki-67.Results The weight of implanted tumors in nude mice in the therapy group[(0.99?0.54)g] was lower than that in control group [(1.58?0.51)g,P
7.Effects of Hand-arm Bimanual Intensive Training in Group on Functional Independence and Quality of Life of Children with Hemiplegic Cerebral Palsy

Objective To explore the effects of hand-arm bimanual intensive training (HABIT) in group on functional independence and quality of life of children with hemiplegic cerebral palsy. Methods From November, 2015 to September, 2016, 20 children with hemiplegic cerebral palsy were randomly divided into control group (n=10) and experimental group (n=10). Both groups received routine occupational therapy, and the experimental group received HABIT in group one hour per day additionally, for eight weeks. They were assessed with the Carroll Upper Extremities Functional Test (UEFT), Functional Independence Measure Children's Edition (WeeFIM) and Chinese Version Pe-diatric Quality of Life Inventory (PedsQLTM) 3.0 Cerebral Palsy Module respectively before and after treatment. Results After treatment, the scores of UEFT, WeeFIM and the PedsQLTM3.0 improved in both groups (t>3.970, P<0.05), and were higher in the experimental group than in the control group (t>2.306, P<0.05). Conclusion HABIT in group could improve the fine function of upper extremity and hands coordina-tion of children with hemiplegic cerebral palsy, and enhance the functional independence and quality of life.
8.Expression and significance of B lymphocyte stimulator and BAFF-receptor in peripheral blood mononuclear cells of patients with systemic lupus erythematosus.

Objective To investigate the expression and significance of B lymphocyte stimulator (Blys) and its receptor BAFF (BAFF-R) in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE). Methods The expression of Blys and BAFF-R was measured by flow cytometry in 90 pa-tients with SLE,which was compared with that of 45 healthy controls. The relationships between the expression of Blys, BAFF-R and other laboratory parameters as well as disease activity were analyzed. Results The expression of Blys and BAFF-R in PBMCs from patients with SLE was significantly elevated compared to healthy controls (P <0.001), so did the active group (P < 0.001) and inactive group (P < 0.001 and P < 0.01). The expression of Blys in PBMCs from active SLE patients was higher than that of inactive patients (P <0.05). However,there was no statisti-cal difference of BAFF-R between the two groups. The expression of Blys in PBMCs was positively related to SLEDAI (r =0.728,P <0.001) ,IgG and IgM(r=0.691,P<0.001 and r =0.453,P<0.01) ,but negatively related to C3 and CA (r = -0.510, P < 0.001 and r = -0.312, P < 0.05). The expression of Blys in dsDNA positive group was higher than those of dsDNA negative group (P < 0.01). The expression of Blys and BAFF-R in Cl qAb positive group was higher than those of ClqAb negative group as well (P <0.01). Conclusion The expression of Blys and its receptor BAFF-R in PBMCs from SLE is elevated ,which may reflect the disease activity and is related to the pro-duction of autoantibody. They might be involved in the pathogenesis of SLE.
9.Using FRET to Study The Interaction Domain of TLR4 Binding to MD-2 in Living Cells

TLR4-MD-2 complex plays a key role in LPS recognition and its signal transduction. These steps are the vital elements of the host's defensive reaction. Studying the functional domain of TLR4 and MD-2 is very important to further understand the mechanism of LPS signal transduction. It was studied the interaction domain of TLR4 and MD-2 in living cells based on gene mutation, gene transfection and fluorescence resonance energy tramsfer(FRET) which is considered as one of the best methods used for intracellular protein-protein interaction study. CY-15P which was fused by CFP and YFP through 15 neutral amino acids was used as positive control, while co-expressed CFP and YFP proteins were used as negative control. The results showed that the ability of TLR4 binding to MD-2 decreased dramatically after the deletion of Glu~(24) ~ Met~(41) in N terminal of TLR4. Aggregation of TLR4 to LPS stimulation was observed, however, TLR4 without the Glu~(24)~ Met~(41) mutation did not aggregate. All these results indicated that TLR4 Glu~(24)~ Met~(41) might be the interaction domain of TLR4 binding to MD-2 and participate in the aggregation effect of TLR4 upon LPS stimulation.
10.Expression and significance of Th17 cells in peripheral blood of patients with rheumatoid arthritis

ObjectiveTo investigate the expression and significance of Thl7 cells in peripheral blood of patients with Rheumatoid Arthritis (RA).MethodsThirty-three active RA and 27 stable RA patients and 30 healthy individuals were enrolled in the study.Intracellular flow cytometry detection of TH17 cells in peripheral blood was established.The correlations between Th17 cells and RA disease activity (DAS28 scale) as well as bone erosion were analyzed.ResultsThe expressions of Th17 cells in the patients with RA were significantly higher than those of healthy controls [ ( 1.60 ± 0.51 ) % vs (0.81 ± 0.21 ) %,t =10.30,P < 0.001 ].The Th17 levels in active RA patients were also higher than that of stable RA patients and healthy subjects [ ( 1.94 ±0.38 ) % vs ( 1.19 ± 0.27) %,t =8.85,14.60,P < 0.01 ].And the Th1 7 levels in stable RA patients were also higher than that of healthy subjects (t =5.88,P <0.01 ).Active RA patients with bone erosion ( 19 cases) also had higher levels of Th17 cells than those without bone erosion ( 14 cases) [ ( 2.09 ± 0.39 ) % vs.( 1.74 ±0.29 ) %,t =2.83,P < 0.01 ].The expression of Th17 cells was positively correlated with the DAS28 score ( r =0.86,P < 0.01 ).ConclusionThere is a high expression of Th17 cells in peripheral blood of RA patients,which is related to disease activity and bone erosion.Th17 cells might play an important role in the pathogenesis of RA.
