1.Processing of realgar and re-evaluation on medicinal efficacy and toxicity

Objective: To prepare the processing solution of realgar by bacteria and to explore the change of pharmacodynamic action and toxicity. Methods: The processing solutions of realgar were prepared using the strain thiobacillus ferroxidans, the realgar powder processed by normal method was selected as control. The effects on RPMI8226 cell proliferation of them were investigated by methyl thiazolyl terazolium (MTT),cell cycle distribution was determined by FCM in RPMI8226 cell. The distribution of arsenic concentration in blood and tissues including heart, liver, spleen, lung, and kidney in the samples were determined by Inductively Coupled Plasma-atomic Emission Spectrometry (ICP-AES). Results: Compared with control group, the solubility rate and the induced apoptotic rate increased, and dosage significantly reduced, while arsenic was hardly found in the tissue in processing group by bacteria. Conclusions: The processing group by bacteria may be enhanced medicinal efficacy and inhibited the toxicity in realgar. It’s intending to provide new approaches for scientific processing realgar.
2.Pharmacokinetics of bacteria bioleaching solution of realgar in rat.

The paper is to report the preparation of realgar bioleaching solution (RBS) by bacteria and the comparison of pharmacokinetics of RBS and H3AsO3 (ATO), and the study of its possible change of absorption and distribution of soluble arsenic in rat. The experiment was carried out on Wistar rats given peritoneal injection of RBS at a dose of 0.3 mg x kg(-1) (soluble arsenic content, 0.3 mg x kg(-1)), and rats given ATO at the dose of 0.3 mg x kg(-1) (soluble arsenic content, 0.3 mg x kg(-1)). The arsenic concentrations in many tissues including heart, liver, spleen, lung, renal and brain were determined. The changes of pharmacokinetic parameters and arsenic distribution in different tissues were detected and compared in these two groups of rats. The pharmacokinetic parameters of RBS and ATO are very similar. There is very few distribution of arsenic in the tissues in RBS group, compared with the ATO group. There is significant difference in the content of arsenic between two groups statistically (P < 0.01). It is feasible that we select the bacteria bioleaching solution as a candidate drug, which may be employed for primary change of arsenic compounds including dissolved inorganic arsenic and organic arsenic, in order to improve bioavailability and decrease the amount of arsenic accumulation in animal tissues. In addition, there is significant difference in the change of arsenic compounds between two groups. It can be concluded that, the investigation on application of microbial technology may provide a basis for exploratory research of realgar.
3.Expression of cyclin D1 and p16 protein in vulvar white lesion

4.DETECTION OF PRE-S2 PROTEIN IN THE KIDNEY IN PATIENTS WITH GLOMERU-LONEPHRITIS

The presence of pre-S2 protein in the kidney was investigated in 30 patients with biopsy-proved glomerulonephritis (GN) using indirect immunoperoxidase technique and monoclonal antibody. HBsAg and HBeAg in the kidney were detected using the same techniques and HBsAg, HBeAg, anti-HBs, anti-HBe and anti-HBc in serum were assayed at the same time.It was shown that pre-S2 prtein was present in the kidney in 7/22 patients with primary GN and 6/8 with lupus nephritis. It was deposited along the glomerular capillary walls or within the mes-angium, depending on the type of GN as well as the deposition of immunoglobulins and complement components in the kidney. Deposition of pre-S2 protein in renal tissue was correlated with HBsAg deposition in the glomeruli, but not HBeAg deposition. In membranous GN, there was a good correlation between the presence of pre-S2 protein in the kidney and virus antigetiemia. It is concluded that pre-S2 protein detection in the kidney is of value in evaluating the hepatitis B virus associated GN.
5.Clinical Effect of Electrochemical Therapy on Hemangiomas of Liver.

Objective To explore the clinical effect of electrochemical therapy (EchT) on hemangiomas of liver.Method 186 cases of hemangiomas of liver with,male 97 and female 89 were selected for the study.The age ranged from 31~55 years old.The electrochemical therapy instrument of model ZAY-B and the electrode made of platinum were used.Suitable manipulating technique and treating quantity of electricity were investigated for the patients.Electrodes were inserted into the tumor through a tracer directly by opening the abdominal cavity.Normal tissue of liver was protected by the spaghetti and the electrodes were connected to EchT device,then the treatment began.The therapeutic voltage was 6~8V,electic current 120~160mA.The quantity of electricity was 100~150 Coulombs per 1.0cm diameter of the tumor.The treatment could be terminated when the tumor became from soft to stiff.Result The therapeutic efficacy was evaluated 6 months after EchT.Through the follow-up of 1~5 years,103 of total 186 cases of hemangiomas of liver completely disappeared(72.5%);31 cases partially relieved (21.8%) and 8 cases were ineffective (5.6%).Conclusion EchT to treat hemangiomas of liver is safe and efficacy,with less traumatic and a quick recovery.It can be served as a new method for treating hemangiomas of liver.

8.Clinical study of osteoporosis in patients with ankylosing spondylitis

Objective To investigate the incidence of osteoporosis (OP) in patients with ankylosing spondylitis and the relationship between OP and the clinical data. Methods Serum levels of bone alkaline phosphatase (BALP) and tartrate resistant acid phosphatase 5b (TRACP5b) were detected by enzyme linked immunosorbent assay ( ELISA ) in 60 cases with ankylosing spondylitis, and it was compared with normal controls. Bone mineral density (BMD) was measured through dual-energy X-ray absorptiometry ( DXA), including lumbar ( L2 - L4), bilateral femoral neck and greater trochanter. Some clinical data was collected and analyzed at the same time. Results The incidence of OP in AS patients was 35%, and the incidence of OP in the femoral proximal end was higher than that in lumbar. Compared with normal controls[ ( 1.06 ±0. 18 )U/L ], the levels of serum TRACP5b in AS[ (1.31 ± 0. 82 )U/L] patients was significantly higher ( P <0. 05 ). The levels of serum BLAP in OP combined AS group[ ( 21.65 ± 5.41 ) U/L]were significantly lower than non-OP group[ (32. 37 ± 16. 5 ) U/L] ( P <0. 05 ). The disease duration was negatively correlated with the BMD of femoral neck ( P < 0. 01 ). Conclusions There was higher incidence of OP in AS patients, which were related with the abnormality of bone metabolism and the disease duration.Multiple factors participated in the regulation of bone metabolism of AS.
9.Optimization of flash type extraction methods forDanzhuye granules by orthogonal test

Objective To optimize the flash type extraction ofDanzhuye Granules.Methods With the total flavonoids and extract yield as indices, the orthogonal test was adopted to optimize the extraction times, the ethanol concentration, solid-liquid ratio, and the duration of extraction forDanzhuye Granules.Results The factors that infuenced the extract efficiency from high to low were the extraction times, volume fraction of ethanol, the ratio of liquid to solid, and the extraction time. The optimum extracting condition included that the origin was extrated by the ethanol volume fraction 80% with the material to liquid ratio 30:1, total of 3 times, with each time of 3 minutes. Validation experiments of the potimal condition showed that the extraction of total flavonoids was 29.1 mg/g, and the extract yield was 23.6%.Conclusions The flash type extraction method is suitable for extraction, which provides a new method for the development and utilization ofDanzhuye Granules.
10.Roles of interleukin-17 and RORγt in a murine model with paraquat- induced acute lung injury

Objective To explore the roles of Th17 cells in paraquat-induced acute lung injury(ALI)in a murine models through investigating the expression of Th17 type cytokine IL-17 and the specific transcription factor of Th17 cells-RORγt mRNA. Methods Male Kunming mice were randomly divided into the normal control group and the poisoned group. The ALI model induced by PQ poisoning was produced by intraperitoneal injection PQ solu-tion (20 mg/kg),and the normal control group was given normal saline solution with the same volume. After treatment for 48 hours,the animals were sacrificed. H&E staining and TUNEL staining were done to measure histo-logical changes and apoptosis. Serum level of Th17 related-cytokines was assayed by ELISA. The mRNA ex-pression of RORγt and the protein expression of IL-17 in the lung tissue were detected by real time-PCR assay and western blot,respectively. Results Compared with the normal control group,the lung inflammation and apopto-sis were significantly higher in the poisoned group. The serum level of IL-17A was significantly increased (P <0.05) and the lung parenchyma RORγt mRNA and IL-17 protein levels were significantly increased. Conclu-sion IL-17 plays an important role in the pathogenesis of ALI induced by paraquat poisoning.
