Zinc is essential for cell proliferation and differentiation, especially for the regulation of DNA synthesis and mitosis. On the molecular level,it is a structural constituent of a great number of proteins including enzymes of cellular signaling pathways and transcription factors.Zinc can modulate cellular signal recognition, second messenger metabolism, protein kinase and protein phosphatase activities. It may stimulate or inhibit activities of transcription factors, and it may as well be used as a second messenger involved in cell signaling regulation.

Objective To investigate the efficacy and safety of sirolimus 0.1% ointment in the treatment of facial angiofibromas in children with tuberous sclerosis complex. Methods Sirolimus 0.1% ointment was prepared. Twenty children with tuberous sclerosis complex who had facial angiofibromas were enrolled in this study. Facial angiofibromas were topically treated with the self?prepared sirolimus 0.1% ointment twice a day for 12 weeks. The facial angiofibroma severity index(FASI)was calculated, the degree of satisfaction with the treatment was evaluated, and adverse reactions were analyzed at weeks 4 and 12. Plasma sirolimus concentrations as well as blood biochemical and immunological parameters were measured, blood coagulation activity was evaluated, and routine blood tests as well as urine tests were performed at baseline and week 12. Results The FASI of patients significantly decreased at weeks 4(4.400 ± 1.284)and 12(2.975 ± 1.543)compared with that at baseline(5.750 ± 1.175, both P<0.000 1), and was significantly lower at week 12 than at week 4(P < 0.000 1). The efficacy index was 49.87% ± 22.08% at week 12, significantly higher than that at week 4(24.43%± 10.18%, t=7.338, P<0.01). The color, size and number of lesions significantly decreased in all the patients, and facial angiofibromas completely disappeared in 2 patients at week 12. At week 4, 10 parents were satisfied with the improvement of erythema, 3 parents with that of lesion volume, and 3 parents with that of lesion area. The degree of parent satisfaction increased at week 12 in all the cases. The blood concentration of sirolimus was lower than 1.0μg/L both before and after the treatment. No severe systemic or local adverse reactions were noted in these patients. Conclusion Sirolimus 0.1%ointment is markedly effective and safe for the treatment of facial angiofibromas in patients with tuberous sclerosis complex.

Objective:To prepare and screen monoclonal antibodies against Herpes simplex virus-1(HSV-1),and develop a double antibody sandwich quantitative enzyme-linked immunosorbent assay( Q-ELISA) for detection of HSV-1 particle. This method was used to control the quality of viral particle in the developing and manufacturing process of HSV-1. Methods: BALB/c mice was immunized with HSV-1 to prepare monoclonal antibodies. A double antibody sandwich Q-ELISA was developed to determine concentration of HSV-1 particle, which was based on the neutralizing monoclonal antibody 1F6 as capture antibody, and 2B1 as HRP-conjugated antibody. The performance of the reagent was evaluated, including specificity, sensitivity, precision, accuracy and linear. And the relation between the amount of virus detected by this method and the virus titer was analyzed by regression analysis method. Results: The Q-ELISA for HSV-1 particle was developed. The quantitation scope was 0. 125-2 μg/ml, the coefficient correlation was 0. 995 5, the limit of detection was 0. 125 μg/ml, the recovery was between 85. 6% and 107. 1%, the variation coefficient was lower than 10%, and the reagent does not react with other samples except HSV-1 antigen. This method has a good correlation with virus titer. Conclusion:The Q-ELISA for HSV-1 particle was successfully developed, which provide a new approach for rapid and quantitative detection of HSV-1 antigen.

AIM:To establish a rapid in vitro method for mast cell degranulation tracing by raster image corre-lation spectroscopy (RICS).METHODS:RBL-2H3, a basophilic granulocyte mast cell line transfected with CD 63-GFP plasmid, was used for evaluating the methods , including β-hexosaminidase ( HEX) colorimetric assay, scanning electron microscopy (SEM) and RICS in the detection of mast cell degranulation .The sensibilities of these methods were com-pared.RESULTS:The sensitivities of β-HEX colorimetric assay and SEM were 5 mg/L and 3.9 ×10 -2 mg/L, respec-tively.RICS detection showed obvious decrease in the diffusion coefficient at dose of 3.9 ×10 -2 mg/L.CONCLUSION:Fluorescent molecular diffusion dynamic measurement can be used for rapid tracing of allergic substances in vitro.Accord-ing to the results, RICS can achieve nearly the same extent of sensitivity as the SEM does and is far more sensitive than β-HEX colorimetric assay.Compared with SEM, RICS has several advantages: it is faster, simpler and cheaper; it can be used in living cells;it is more suitable for rapid in vitro allergenic compounds tracing .Therefore, RICS is applicable in clinic allergic antigen screening and may also be used in pharmaceutical quality control .

Objective The present study was designed to investigate the risk factors affecting bleeding during percutaneous nephrolithotomy. Methods The records of 218 patients with percutaneous nephrolithotomy procedure by a single surgeon were retrospectively reviewed. The mean age was 48 years ( range,19 -70).One hundred and forty six patients had staghore stones, and 7 patients had previous open or percutaneous nephrolithotomy histories.Forty-one patients had concomitant diabetes mellitus, and 89 cases had hypertension. The following factors including age, sex,BMI,diabetes status, hypertension status, stone type, calix of puncture, previous open or percutaneous nephrolithotomy history, number of accesses, size of accesses, operative time, and surgeon experience were analyzed.Univariate analysis and multivariate step regression analysis were used for statistical assessment. Results 207 procedures were successfully performed, and 11 patients failed because of difficulty to establish the accesses.Single-tract was used in 176 cases and multiple-tract was used in 31 cases.163 cases were performed via a 18 F access and 44 cases via a 24 F access. The mean operative time was 78.4 min.The overall blood transfusion rate was 7.7%,and stone type ( P =0.034),diabetes ( P =0.030),number of accesses ( P =0.019 ),size of accesses ( P =0.008) and operative time (P =0.001 ) were the risk factors affecting blood transfusion requirement. The average hemoglobin (Hb) drop after percutaneous nephrolithotomy procedures was 11.2 g/L,and stone type ( P < 0.001 ),diabetes ( P =0.015 ),number of accesses ( P =0.016),size of accesses ( P < 0.001 ) and operative time ( P < 0.001 ) were the risk factors affecting Hb drop. The following covariates including Hb drop:age, sex,BMI,previous open or percutaneous nephrolithotomy history, hypertension status, calix of puncture and surgeon experience were not risk factors affecting blood transfusion requirement and Hb drop.Multivariate stepwise regression analysis showed that diabetes ( OR =1.75 ),stone type ( OR =1.92),number of accesses ( OR =2.45 ),size of accesses ( OR =1.32) and operative time ( OR =1.66) significantly increased risk of bleeding. Conclusions Stone type, diabetes, number of accesses, size of accesses and operative time were the risk factors affecting blood loss during percutaneous nephrolithotomy.
