1.Up-regulation of tumor necrosis factor-? on c-jun in cultured human retinal pigment epithelial cells

Objective:To investigate the effect oftumor necrosis factor (TNF)-? on the expression ofc-jun in the cultured human retinal pigment epithelial(RPE)cells.Methods:RPE cells were exposed to 40 ng/ml TNF-? for 0,10,20,40 and 40 minutes,respectively.Immunofluorescence staining and reverse transcription polymerase chain reaction(RT-PCR)were used to detect the levels of c-jun protein and mRNA in RPE cells.Results:Untreated RPE cells revealed faint or none green fluorescence of c-jun in the cytoplasm and some nuclei.With exposure to TNF-?,c-jun increased obviously with stronger green fluorescence.With treatment of TNF-?,c-jun mRNA increased gradually and enhanced to the peak at 40 min.Conclusion:TNF-? can up-regulate c-jun protein and mRNA in cultured human RPE cells and induce its nuclear transposition,which suggests the activation of c-jun.

3.Application of continuous blood purification machine in treatment of multiple organs dysfunction syndromes in children

Objective:To observe the application effect of continuous blood purification(CBP) machine in treatment of multiple organs dysfunction syndromes(MODS) in children.Methods: 26 cases of children with MODS implemented CBP therapy were choosed as the research objects. All the patient’s treatment time, replacement liquid dosage, the quantity of liquid ultrafiltration were recorded. All the patient’s blood pressure and heart rate were monitored before and after CBP therapy. The changes in the renal function indices such as BUN and SCr, the blood gas indexes such as pH, PaO2, PaCO2, HCO3-and BE, the electrolyte indicators such as Na+, K+, Cl- and osmotic pressure were observed and compared before and after CBP therapy.Results: (1)The average period of treatment of 26 cases in children with MODS was (77±53)h. The average amount of fluid replacement was (18.4±15.8)L/d, and the average quantity of liquid of ultrafiltration was (2630±1760)ml/d. The vital signs of all the children were stable in the process of CBP therapy, and the blood pressure and heart rate had no obvious changes before and after treatment. (2)The renal function indices such as BUN and SCr decreased obviously after treatment. The differences were statistically significant (t=50.392,t=96.115;P<0.05). The blood gas indexes such as PH, PaO2/FiO2, HCO3- and BE were improved significantly after treatment (t=4.450,t=20.105,t=11.209,t=54.840;P<0.05). The electrolyte indicators such as Na+, K+, Cl- and osmotic pressure had no obvious change after treatment (P>0.05).Conclusion: CBP therapy can effectively reduce various inflammatory mediators and toxin, improve renal function, help to stabilize the environment in the body and it is an effective, safe and reliable therapeutic method in the treatment of children with MODS.
4.Effect of batroxobin on vascular smooth muscle cell proliferation induced by balloon angioplasty in rat aorta

A model of vascular smooth muscle cell proliferation was estabolished by balloon angioplas-ty in rat's aorta. We found that treatment with batroxobin decreased plasma fibrinogen level after angio-plasty, and inhibited the increase in smooth muscle cell count of injured vascular wall and incorporation rate of H-TdR into vascular tissues effectively. The results suggest that batroxobin may have clinically prospective significance in prevention and therapy of angioplasty-induced restenosis.
5.Antagonistic Effects of Ganoderma Spore on Mutation Induced by Sodium Nitrite in Drosophila melanogaster

Objective To study the antagonistic effects of Ganoderma spore on mutation induced by NaNO2 in Drosophila melanogaster. Methods After 24 hours of starvation,male Drosophila melanogaster were transferred to the culture bottles with medium with NaNO2 of 0,0.001,0.01,0.05 and 0.1 g/L for 24 hours and then cultured with female Drosophila melanogaster for 7 days. Male Drosophila melanogaster were treated with NaNO2 at the doses of 0,0.01,0.001 g/L for 24 hours and then transferred to the medium with Ganoderma spores at the dose of 0.2%,1%,2%,cultured with female Drosophila melanogaster for 7 days. The distilled water was used as the negative control. The amount of F1 generation and the mutation of the F1 generation were calculated. Results The mutation Drosophila melanogaster were mainly vestigial,some had malformation with white body and strange eyes. Compared with the negative control group,the count of offspring decreased,the rate of mutation proved positively correlated to the concentration of NaNO2. High dose of sporoderm-unbroken spores could cause a decrease of number of offspring. 6.Effects of Spironolactone on Vascular Remodeling After Angioplasty of Rabbit Iliac Artery

Objective To explore the effect of spironolactone on vascular remodeling of injuried rabbit artery. Methods 20 male New Zealand rabbits were randomly divided into two groups. The rabbit models of iliac artery angioplasty were set up by BC. After injury, control group was fed with standard forage, and experimental group was fed with standard forage and spironolactone(20mg?kg~ -1?d~ -1). The rabbits were killed four weeks after injury. The injuried segments of iliac artery were removed, and processed for histological and morphological analysis by imumohistochemistry and TD 2000 analysis system of pathology image. Results AUIEM and AUEEM in the experimental group significantly increased as compared with the control group(P0.05). Conclusion Spironolactone could improve vascular remodeling after angioploasty of iliac artery in rabbits.
7.Effects of cytokines on the expression of syndecan-1 in cultured human retinal pigment epithelial cells

Objective To investigate the effects of cytokines on the expression of syndecan-1 in cultured human retinal pigment epithelial (RPE) cells and the signal transduction pathway. Methods Reverse transcription polymerase chain reaction and immunofluorescence staining were used to detect the expression of syndecan-1 mRNA and protein in normal RPE cells. The expression of syndecan-1 in RPE cells stimulated by different cytokines was detected and quantitatively analyzed by image process of immunofluorescence. The stimulation included 7 and 35 ng/ml tumor necrosis factor (TNF)-? for 24 hours, 1 and 6 ?g/ml lipopolysaccharide (LPS) for 11 hours, 7 ng/ml TNF-? for 0 to 24 hours (once per 2 hours, and 13 times in total), and 30% supernatant of monocyte/macrophage strain (THP-1 cells) for 3, 14 and 43 hours. The effect of 30% supernatant of THP-1 cells was assayed after pretreated by PD098059 [the specific inhibitor of extracellular signal regulated kinase(ERK) 1/2] for 2 hours. After exposed to 30% supernatant of THP-1 cells for 3 hours and treated by 0.25% trypsin for 5 minutes, RPE cells attaching was evaluated by methyl thiazolyl tetrazolium assay. Results In normal human RPE cells, expressions of syndecan-1 mRNA and protein were detected, and strong syndecan-1 positive yellowish green fluorescence was found in the cell membrane and cytoplasm while light green fluorescence was in the nucleus. The contents of gallic acid, catechin, albiflorin, paeoniflorin, benzoic acid and paeonol extracted in different growth years, collecting season and of different parts of Paeonia lactiflora were determined. The results showed that the contents of catechin and paeoniflorin in Paeonia lactiflora collected in autumn are the highest, and the contents of benzoic acid was lower than that of those collected at other time. The longer is the age of Paeonia lactiflora, the higher is the contents of catechin and paeoniflorin. The contents of catechin and paeoniflorin in the root of Paeonia lactiflora were higher than those in other parts of the plant. There is a certain content of paeoniflorin in the leaves of Paeonia lactiflora. Judging from the result, paeoniflorin is synthesized in the leaf and then transported to the root. Catechin is not synthesized in the leaf, but mainly in the root. Paeonia lactiflora should be collected in autumn, and immature plant should not be collected.
