2.Research progress on negative regulation of adaptive immune response by NK cells

Natural killer ( NK) cells are important innate effector cells and play a vital role in maintaining homeostasis through potent cytotoxic activity and cytokine production. Recent findings show that NK cells can also shape adaptive immune responses by in-fluencing a variety of immune cells. In addition to direct interactions with other immune cells,NK cells can indirectly stimulate or inhibit adaptive immune response via influencing infected cells and pathogen load. Abundant studies have highlighted the positive regulatory functions of NK cells, while their negative regulatory functions have increasingly attracted attention in recent years. Here, we review recent findings on negative regulation of adaptive immune response by NK cells, discussing the involved effector cells and function mechanism,and demonstrate how this negative regulation influences the overall outcome of adaptive immunity in infection and tumor disease.
3.The electrophysiological characteristics of ventralis intermedius nucleus and its role in localizations of Vim-thalamotomy for Parkinson's disease patients

Objective To investigate the electrophysiological characteristics of the ventralis intermedius nucleus (Vim) in order to find an easy and safe way to confirm the target in Vim-thalamotomy. Method In microelectrode-guided selective Vim-thalamotomy for 23 Parkinson's disease patients, the background activity, amplitude and discharge frequencies of Vim were compared with its surrounding structures. The response of kinesthetic neuron and tremor cell to microstimulation was also compared. Result There were differences in backgroud activity and discharge amplitude for Vim, ventralis lateralis nucleus (VL), ventralis caudalis nucleus (VC), and internal capsule. Based on the response to active or passive movement of contralateral limb tremor cells were divided into two subgroups, which were different in localization. Contralateral tremor showed different response when the two subgroups of tremor cells were mircrostimulated. Conclusion The anterior border of Vim was easily found by microrecording. Only by combining microstimulation with microrecording could the posterior border of Vim and its interior and lateral ordination of target were identified exactly and safely. Kinesthetic neurons and tremor cells which responded to the movement of contralateral limbs should be destroyed.

5.Closed establishment of pneumoperitoneum in patients with peritoneal adhesion in laparoscopic cholecystectomy

Objective To investigate the way of closed establishment of pneumoperitoneum(CEPP) in patients with peritoneal cvity adhesion in laparoscopy cholecystectomy(LC). Methods CEPP experiences of 1046 patients in 6600 cases LC in our hospital from September 1991 to September 1999 were retrospectively analysed. The difficulty in establishing pneumoperitoneum was divided into two kinds: real establishment pneumoperitoneal difficulty(REPPD) and false establishment pneumoperitoneal difficulty(FEPPD). REPPD was due to Veress needle penetrating into visceral or extensive adhesion in peritoneal cavity resulting in CO2 flowing into difficulty. FEPPD was due to Veress needle pentrating in the fat out of peritoneum, in round hepatic ligament or in greater omentum. The formal situation needed to open laparotomy as a change, and the latter situation could establish pneumoperitoneum successfully by regulating the Veress needle penetrating direction or depth in the second penetration. Results Of the 1046 patients, 1028 cases had been establishedpneumoperitoneum successfully though CEPP; 6 cases of REPPD and 18 of FEPPD were required opening laparotomy as a change. The successful rate of CEPP was 98.3%. Conclusions CEPP is a safe and feasible method in patients with peritoneal adhesion in LC. It is the main reason for CEPP failure regarding REPPED as FEPPD made by the deficiency in LC experience and loss confidence in laparoscopist.
6.Helicobacter priori induces changes in the degranulation of mast ceils and the permeability of gastric mucosa in BALB/c mice

Objective To investigate the effect of Helicobacter pylori (Hp) on the degranulation of mast cells and the permeability of gastric mucosa in BALB/c mice, in order to provide further evidence for the important role of Hp in the pathogenesis of chronic urticaria (CU). Methods Seventy healthy BALB/c mice were randomly classified into 3 different groups, namely, Hp group (n = 30), fed with Hp liquid once every 3 days for a total of 7 times, and two control groups, alcohol group (n = 20) fed with 45 percent alcohol solution, and bovine serum albumin (BSA) group (n = 20) fed with BSA 2% solution. Alteration in the blood sucrose level was used to assess the permeability of gastric mucosa. Toluidine blue staining of gastric mucosa was used to assess the total number of mast cells, the amount of degranulated mast cells and the per-centage of degranulated cells after feeding. Results There was a statistically significant difference between the total number of mast cells (F = 207.59, P < 0.01 ) and the percentage of degranulated mast cells (F = 108.16, P <0.01 ) among the three groups, with the highest number and percentage observed in the Hp group followed by the alcohol group. In both groups, the concentration of blood sucrose was increased after feed-ing, with a higher increase in the alcohol group; no significant change occurred in the concentration of blood sucrose in the BSA group (F = -4.06, P > 0.05 ), which differed significantly from the other two groups (F =277.03, P< 0.01 ). Conclusions Hp infection could increase the number and degranulation of mast cells in gastric mucosa, resulting in the release of histamines and vasoactive mediators, which may be related to the manifestation of urticaria.
7.Mechanism underlying intrauterine growth retardation induced by caffeine and its research advance

Intrauterine growth retardation (IUGR) is one of the most commonly encountered developmental toxicity, which could lead to perinatal morbidity and mortality, be also extended from the fetus to adulthood, and seriously affect the quality of the population. Caffeine widely exists in a variety of daily beverages and some drugs. Its consumption is increasing year by year. Caffeine intake during pregnancy is one of the risk factors for IUGR. However, its mechanism of adverse outcome based on embryonic research is still unclear. In this paper, the possible mechanisms of caffeine-induced IUGR focusing on 3 important factors-the mother, placenta and fetus were explored. Caffeine's impact on the mother is the chronic activation of renin-angiotensin system; on the placenta, caffeine induces cell damage or the failure of the cell proliferation/apoptosis balance, leading to blockage of blood supply to the placenta; caffeine is also capable of directly affecting fetal development through interfering its neuroendocrine.
8.The relationship between serum ANA expression and clinicopathologic characteristics and prognosis in colorectal cancer

Objective To investigate the correlation between serum antinuclear antibody (ANA) expression and clinicopathologic characteristics as well as the prognosis in colorectal cancer patients.Methods Analysis was performed for 198 cases with histologically proven adenocarcinoma of colorectal cancer who underwent curative resection from January 2009 to December 2010.There were 113 males and 85 females,average age of 63.9 (22 to 92) years.Serum ANA expression was detected by indirect immunofluorescence.Chi-square test was used to analyze the relationship between clinicopathological features and serum ANA expression,Kaplan-Meier analyses were used to compare DFS between ANA positive and negative group,multivariate analysis was used to measure the impact of confounding variables on survival.Results Positive serum ANA was detected in 51 (25.76%) of the 198 patients,higher than that of the control group 8% (4/50).By univariate analysis,we found that serum ANA positive was related to age < 65 years (x2 =4.87,P =0.027 3),tumor size ≥5 cm (x2 =5.56,P =0.018 3),invasion depth (x2 =325.89,P <0.000 1) and tumor course≥5 months (x2 =5.43,P =0.019 8) respectively,and not related to gender,tumor location,tumor differentiation,lymph node and distant metastasis (P > 0.05).Multivariate analysis identified that only invasion depth of tumor led to serum ANA expression significantly.Conclusions Tumor invasion depth impacts on serum ANA expression significantly,while the expression has no correlation with the prognosis of colorectal cancer.
9.Relationship between MiR-195 and DLL4 expression and clinical features of colorectal cancer

Objective:To observe the expression patterns of miR-195 and DLL4 in colorectal cancer, and to explore the relationship between miR-195 and DLL4 and clinicopathological parameters and prognosis of patients with colorectal cancer.Methods:The relative expression of miR-195 in 56 colorectal cancer tissues was detected by real-time fluorescent quantitative PCR the expression of DLL4 protein and tumor microvessel density (MVD) were detected by bloting and immunohistochemistry. Colon cancer cell line SW480 was treated with miR-195. The expression of DLL4, Jagged1, (the intracellular domain of notch, NICD), CyclinD1, Hes1, Bcl-2 and NF-kB were detected by bloting.Results:The expression level of DLL4 protein in colorectal cancer tissues was significantly higher than that in normal intestinal mucosa (30/56 vs.16/56, t=5.323, P=0.018). The expression level of miR-195 was significantly lower than that in normal intestinal mucosa (36/56 vs.20/56, t=2.371, P=0.008). The expression of DLL4 was negatively correlated with the expression of miR-195 ( r=- 0.881, P=0.015) , which was closely related to the differentiation degree, lymph node metastasis and TNM stage . The prognosis of patients with high expression of DLL4 was significantly worse than that with low expression of DLL4 ( P=0.013). The prognosis of patients with low expression of miR-195 was worse than that with high expression of miR-195 ( P=0.009) . Conclusion:The antagonistic expression of miR-195 and Notch might be closely related to the occurrence and development of colorectal cancer, which can be used as a new reference index for prognosis of colorectal cancer.
