Objective:To study the effects of 17 beta-estradiol (E2) on the proliferation and metastasis potential of salivary gland mucoepidermoid carcinoma M_3SP_4 cells. Methods:The effects of E2 on the potential of proliferation and metastasis of M_3SP_4 cells were investigated by cell counting, MTT assay, clonegenetic assay, in vivo tumour growth and metastasis assay in nude mice. The expression of VEGF,c-erbB-2,Ki-67 and P16 in M_3SP_4 cells was examined by immunohistochemistry assay. Results:E2 increased the proliferation of M_3SP_4 cells at 10~-10-10~-5 mol/L. E2 at 10~-7 mol/L showed the strongest effects. After treatment with E2 at 10~-7 mol/L,the population doubling time of M_3SP_4 cells decreased by 10.8%,clonegenecity increased by 225%, VEGF,c-erbB-2 and Ki-67 expression increased,P16 decreased. In the in vivo assay, the tumour growth increased by 65% and metastasis increased by 609% in nude mice treated with E2 at 0.005 2 mg/d. Conclusion:E2 may stimulate the proliferation and metastasis potential of M_3SP_4 cells.

Objective:To establish a culture method of human salivary gland epithelial cells and to study their growth characteristics in vitro. Methods:Tissue explant technic was employed to culture human salivary gland epithelial cells in serum free medium (SFM),1∶1 DMEM/F12 and 1∶1 DMEM/F12 containing 25 ml/L fetal boven serum (FBS) respectively. The morphology of the cultured cells was observed by phase contract microscope. The cell growth was studied by cell counting. The cells were identified by HE staining,PAS staining and SP staining. Results:Growth of human salivary gland epithelial cells was observed in primary culture in the three kinds of medium in all 10 cases. The cultured cells were epidermoid, positive for cytokeratin, negative for Vimentin and positive for PAS staining. The cells in SFM could be subcultured for five passages, while only for one passage in the other two kinds of medium. Conclusion:SFM is superior to serum free 1∶1 DMEM/F12 or 1∶1 DMEM/F12 containing 25 ml/L FBS for the culture of human salivary gland epithelial cells.

Chinese Journal of Tissue Engineering Research 2014;(47):7638-7642
BACKGROUND:In the treatment of early caries, fluoride can be used for remineralization of white spot lesions. Varnish XT (durable fluoride-releasing coating) and ICON penetration resins are two new materials. Varnish XT as a new type of resin reinforced glass ionomer can be selected as mineralized material. ICON penetration resin is a high-permeability resin with good liquidity that can infiltrate by capilary action into the pores created by enamel demineralization. Low-viscosity resin is used to replace the lost hard tissue due to demineralization and occupy the micropores, so as to fil the region of enamel demineralization and prevent further development of lesions. OBJECTIVE:To study the effects of two minimaly invasive surgical treatment materials, Varnish XT and ICON penetrating resin, on the microhardness of enamel caries white spot. METHODS:Totaly 100 incisors of cows were selected, embedded with ethoxyline resin and polished. The lip side facing down served as the observation side. An area of at least 6 mm×10 mm on the enamel face was exposed, and there were five regions from incisal to dental cervix, A, B, C, D, E. After demineralization liquid for artificial caries, no treatment was adopted in region A, treatment with Varnish XT was for region B, treatment with ICON penetrating resin for region C, treatment with fluoride for region D, and region E was sealed with antacid nail. Surface micro-hardness was detected. RESULTS AND CONCLUSION: After demineralization, surface micro-hardness of regions A, B, C, D decreases remarkably as compared with region E (P < 0.05). Surface micro-hardness of regions B, C, D was higher than that of region A, and ranged as folows: C > B and D for surface micro-hardness with statistical significance (P < 0.05). There was no statistical significance of surface micro-hardness between regions B and D (P > 0.05). ICON resin infiltration, Varnish XT and fluoride have obvious improvement effects on surface micro-hardness, and ICON resin infiltration is superior to Varnish XT and fluoride.

OBJECTIVETo explore the clinical effects of minor bone anchors and palmaris longus tendon graft in treating chronic mallet fingers deformity.
METHODSFrom January 2008 to June 2013, 26 patients with chronic mallet fingers deformity were treated with minor bone anchors and palmaris longus tendon graft. There were 18 males and 8 females, aged from 18 to 52 years old with an average of (32.0±1.3) years. Among them, 8 cases caused by machine injury, 6 cases by fall injury, 6 cases by sprain from fight, 4 cases by tendon spontaneous rupture, 2 cases by knife trauma. There was no tendon attachment of extensor tendon check in 16 cases, and with 0.3 to 0.5 cm tendon attachment in 10 cases. All patients had the flexion deformity and the disability of dorsiflexion activity. During operation, the distal interphalangeal joint was fixed in 10° to 20° dorsiflexion by a Kirshner wire, the minor bone anchor was used to reconstruct the extensor tendon insertion, the palmaris longus tendon slice was transplanted the decayed area of extensor tendon insertion. Four weeks postoperatively, the Kirshner wire was removed and the plaster external fixation was used, and the patient began function exercises. Postoperative complications were observed and fingers functions were assessed according to Dargan standard.
RESULTSThe patients were followed up from 6 to 14 months with an average of (5.0±0.3) months. Wound superficial infection occurred in 2 cases, the skin pressure ulcer in 2 cases, joint activities disability in 1 case; these symptoms got improvement after symptomatic treatment. Traumatic arthritis occurred in 2 cases, 1 case was improved after treatment, and 1 case had chronic pain for a long time. No internal fixation loosening or breakage and tendon rupture were found. According to Dargan standard to evaluate the finger function, 17 cases got excellent results, 8 good, and 1 poor.
CONCLUSIONIt is an effective way to treat the chronic mallet finger deformity using minor bone anchors and palmaris longus tendon graft, and the method has advantages of reliable fixation, easy operation, satisfactory effect and less complication.

Chinese Journal of Clinical Pharmacology and Therapeutics 2007;12(8):935-938
AIM: To establish a sensitive HPLC method for determining the concentrations of paeonol in human plasma and to evaluate its pharmacokinetic characteristics. METHODS: A single oral dose of 160 mg paeonol capsules was given to 24 Chinese healthy volunteers. Paeonol was separated on a XB-C18 column with tetrahydrofuran-methanol-water-phosphonic acid (6∶60∶34∶0.1, V∶V) as mobile phase. The plasma concentrations of paeonol were determined and its pharmacokinetic parameters were calculated and evaluated using DAS 2.0. RESULTS: The linear range of the paeonol was 10-500 ng/mL and the determination limit was 10 ng/mL. The main pharmacokinetic parameters, as Cmax, tmax, t1/2,AUC0-3, AUC0-∞ after a single dose of paeonol capsules were (116±46)ng/mL,(1.02±0.13) h,(1.03±0.35) h, (174±45) ng/mL, (217±56) ng/mL,respectively. CONCLUSION: The HPLC method for determining paeonol concentration in plasma is rapid, sensitive and suitable for pharmacokinetic studies.
