1.Clinical study on moxibustion combined with pressing beans on auricular point in the trentment of elderly patients with chronic diarrhea

Objective To observe the efficacy of the combination of moxibustion and pressing beans on auricular in the treatment of elderly patients with chronic diarrhea.Methods 45 elderly patients met the diagnostic criteria for diarrhea were divided into two groups,the treatment group (23 cases) received the treatment of combined moxibustion with ear pressure beans,the control group (22 cases) received traditional Chinese medicine treatment.The efficacy was observed and compared after two weeks.Results The effective rate reached 95.6% in the treatment group,81.8% in the control group (statistically significant difference,odds ratio 1.16).Conclusion Combined ear pressure beans with moxibustion treatment will receive a significant effect for the elderly patients with chronic diarrhea.The therapy is worth clinical application.
2.Influence of aerobic exercise on blood pressure in patients with white coat hypertension

Objective: To observe influence of aerobic exercise on blood pressure in patients with white coat hypertension (WCH). Methods: A total of 51 newly diagnostic WCH patients were randomly divided into exercise group (n=26) and WCH control group (n=25). Patients in exercise group received aerobic exercise training (walking on flat ground by constant speed, twice a day, 50~80 min each time for three months); The WCH control group did not exercise. Office blood pressure (OBP) and 24h ambulatory blood pressure monitoring (ABPM) were measured in both groups before and three months after treatment. Results: There were no significant difference in indexes of OBP and ABPM between two groups before treatment (P>0.05). After three months, compared with before treatment, indexes of OBP and ABPM significantly decreased in exercise group and they were significantly lower than those of WCH control group in OBP [OSBP: (132.54±17.60) mmHg vs. (143.68±20.35) mmHg, ODBP: (89.12±18.39) mmHg vs. (92.75±17.24) mmHg] and ABPM [ mSBP:(121.57±20.86) mmHg vs. (128.06±19.45) mmHg, mDBP:(76.24±15.28) mmHg vs. (84.55±17.31) mmHg; dSBP:(124.27±22.50) mmHg vs. (130.85±24.61) mmHg, dDBP:(77.95±18.42) mmHg vs. (82.36±17.59) mmHg], P<0.05~0.01. Conclusions: Aerobic exercise can significantly improve blood pressure in patients with white coat hypertension.
4.Effects of estradiol on the proliferation and metastasis of adenoid cystic carcinoma SAcc83 cells

Objective:To investigate the effects of 17-? estradiol (E2) on the proliferation and metastasis of adenoid cystic carcinoma SAcc83 cells.Methods:The effects of E2 on the proliferation of SAcc83 cells were studied by MTT assay and flow cytometry.The invasion potential of the cells was investigated with chicken embryo heart invasion assay.The in vivo growth and metastasis of SAcc83 cell induced tumor were studied in ovary-resected female nude mice. Results:E2 at 10 -7 ,10 -8 and 10 -9 mol/L promoted the proliferation of SAcc83 cells. E2 at 10 -7 mol/L showed the strongest growth stimulation effects and increased the cell proliferation by 34.4%.With exposure time of 1,2 and 3 d it increased the proliferation index by 33.3%,40.9% and 17.8% respectively;with the exposure time of 12,18 and 24 h,it increased S phase cells in cell cycle by 12.3%,3.3% and 9.7% respectively. Neoplasm developed in all nude mice after subcutaneous transplantation of 106 cells.In the mice treated with 0.2 ml of E2 at 9.55 ?10 -5 mol/L every day for 15 days neoplasm volume doubling time was 61.5 h,that in control mice 76.5 h.In lung metastasis trial, 42 days after tail injection of the cells the metastasis foci in E2 treated mice inreased by 41.2%.Conclusion:E2 may increase the proliferation and metastasis of adenoid cystic carcinoma cells.
5.Effects of Iodine Deficiency and Hypothyroidism during Pregnancy on the Protein Expressions of cAMP Response Element-Binding Protein in the Entorhinal Cortex of Pups

Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of CREB in the entorhinal cortex of pups.Methods Twenty-eight female Wistar clean rats after pregnancy were randomly divided into control group,hypothyroid 1,2 group and iodine deficient group(7 in each group).From GD6 till PN28,iodine-deficient group was administered with iodine-deficient diet [iodine content:(14.11 ?1.96) ng/g] and tap water;hypothyroid 1 and 2 group were administered with 5 mg/L and 15 mg/L PTU in the drinking water and fed with normal diet [iodine content:(470.50?46.52) ng/g].Control group received tap water and normal diet during the experiment.Five pups from each group were sacrificed and intracardiac perfused at PN7,PN14,PN21,PN28 and PN42.Brains were removed,fixed and sectioned coronally.All sections were observed and were analyzed for the protein expression of CREB by immunohistochemistry in the entorhinal cortex.Results At PN7,there was no significant difference in the expression of Ng in all four groups.At PN14 and PN21,the expression of CREB in the entorhinal cortex in hypothyroid 1,2 group and iodine deficient group were significantly lower than those of controls(P
6.THE STUDY ON QUASISPECIES OF HEPATITIS B VIRUS:REVERSE TRANSCRIPTASE REGION IN POLYMERASE GENE AS AN EXAMPLE

A set of specific primers was synthesized according to DNA sequence of HBV found in China, the reverse transcriptase (RT) region in polymerase gene was amplified by PCR method from the serum of 3 patients with chronic HBV infection, and then the PCR products were subcloned into pGEM Teasy vectors. 13 clones were sequenced. Sequence comparison of the selected clones was made to look for the difference. After being compared, 13 sequences of RT were found different. Besides 2 clones with long sequence deletion, the different rate of RT coding nucleic acid sequences, RT and HBsAg amino acid sequences of the 11 clones is 5 1%, 4 9% and 7 5%, respectively. Many mutation types, including point substitution and deletion mutation, were found in this region. There is quasispecies population and defective HBV genome in patients with chronic HBV infection.
7.CLONING AND ANALYSIS OF THE CODING SEQUENCE FOR HEPATITIS B VIRUS PRES1 BINDING PROTEIN BY T7 cDNA PHAGE DISPLAY SYSTEM

T7 cDNA phage display method was employed to find the binding protein of PreSl protein. PreSl protein was coated in a 96-well ELISA plate, and then T7 cDNA library phages were bound to the target protein. Phages which did not bind to garget protein were washed away and the binding phages were eluted. Insertions from different clones were sequenced, and the deduced amino acid sequences were analyzed by Vector 6.0 software. Using BLAST software in GenBank, whole length of amino acid sequence of binding protein was obtained. After 4 rounds of biopanning, recombinant T7 phages with binding ablity were amplifed by infection to E. coli. One piece of amino acid sequence was found to be amino terminal of product of glioma tumor suppressor candidate region gene 2 (GLTSCR2). There was a binding domain KxPxKSGxxxL in these clones. T7 cDNA phage display technique can be used bo find the ligand. GLTSCR2 coding protein may be the binding protein to preSl protein of HBV.\;
8.Preliminary study on hepatitis B virus nuclei acid vaccine with interleukin-18 as co-stimulator

Objective To construct plasmid pVR1012 M as nuclei acid vaccine for hepatitis B,was constructed to immunize mice with or without plasmid pcDNA 3.1 - IL 18 to identify the effect of Interleukin 18(IL 18). Methods Polymerase chain reaction method was used to amplify the PreS2 and S region of HBV and reconstruct plasmid pVR1012 M as nuclei acid vaccine. Plasmid pcDNA 3.1 - IL 18 was used as a co stimulator. Twenty five Balb/c mice were divided into 3 groups, group 1 immunized with 100 ?g plasmid pVR1012 group 2 pVR1012 M,Group 3,pVR1012 M with pcDNA 3.1 - IL 18, respectively, every 2 weeks for 3 times. Anti HBs were detected in serum 2 weeks after each injection. Lactated ehydrogenase (LDH) cytotoxicity assay was done to analyze the cytotoxic T lymphocytes funciton. Results The positive rate and the antibody titer of serum from mice injected pVR1012 M increasing gradually with the increasing frequency of inoculation, while those from mice injected pVR1012 M and pcDNA 3.1 - IL 18(joint group) were lower than those injected with pVR1012 M alone (Difference after 3rd inoculation was significant, P
9.Cloning gene of hepatitis B virus PreS1 binding protein by phage display system

Objective T7 cDNA phage display system and bioinformatics methods were employed to find the binding protein to the PreS1 protein of hepatitis B virus (HBV). Methods PreS1 protein was coated in ELISA plate as the target protein, and then T7 cDNA library phage display system was used to scan the binding protein or peptide. A piece of cDNA was found to have the function to bind the PreS1 protein, and the product was named as PreS1 binding protein (PreS1BP). Using BLAST in GenBank, the amino acid sequence of PreS1BP was compared in the protein sequence database. Results The amino acid sequence of PreS1BP was identified as a piece of glioma tumor suppressor candidate region gene 2 (GLTSCR2), and the length of cDNA of PreS1BP was proved to be 1436 nt. The gene was located at chromosome 19q arm (19q13.3) with a length of 11445 base pair between 10403483 and 10414989, containing 13 exons and 12 introns. Conclusion HBV PreS1BP gene could be obtained by T7 cDNA phage display system in combination with bioinformatics methods.
10.New drug delivery systems and the function of camptothecin

Camptothecin is known to possess a broad spectrum of anticancer activities, however due to its high toxicity and low solubility in water, camptothecin is severely limited in clinic. To optimize the effects of camptothecin in the treatment of malignancies, besides the structural modification and reconstruction of camptothecin, the establishment of new drug delivery systems becomes a hot topic in the camptothecin research. This paper aims to summarize the advanced progress in the delivery system and function of camptothecin at recent years. The advances in improving the poor solubility, lactone ring instability, controlled release systems and targeting delivery systems of camptothecin are also reviewed. As the related methods and technologies are improved and developed, camptothecin will be a group of prospective anticancer drugs.
