1.Application of residual grey model(1,1) to the prediction of tuberculosis prevalence

Objective:To discuss the application of residual Grey Model(1,1) in the time series data to set up the prediction model for Tuberculosis prevalence.Methods:Grey Model(1,1)was set up by using the material on Tuberculosis prevalence collected by Tuberculosis Prevention and Treatment Institute of Chongqing city from 1993 to 2003,and then used to predict the Tuberculosis prevalence of 2004.Results:The residual Grey Model(1,1)is ■(1)(k+1)=89.5776e0.0392k-85.0913+?(k-1)1.6435e-0.3068k,the relative error values of predictive value for 1993 to 2003 were all less than 0.05,and the degree of grey incidence was bigger than 0.6,and the little error probability was bigger than 0.8;The predictive prevalence value in 2004 was 5.2942 per 100 thousands,the relative error was 0.0461 and the modified one was 0.0121.Conclusion:The residual grey model(1,1) had a good fitness and can be used to predict tuberculosis prevalence with high prediction precision in short-term time series.
2.Applications of decision tree model in classification of axillary Ⅲ lymph nodes metastasis of breast cancer

Objective:Todiscuss the application of Decision Tree model and fit classification model of maxillary Ⅲ lymph nodes of breast cancer,which can abstract diagnostic knowledge used for computer-based diagnosis system.Methods:Four classification models of Decision Tree with the arithmetic of C5,CR,QUEST and CHAID were applied for classifying maxillary Ⅲ lymph nodes in 223 cases of breast cancers,confirmed by with 11 diagnostic indexes in the first affiliated hospital of chongqing medical university from 1993 to 2003.Results:CHAID classification model was better than other three models,according to the reliability of diagnostic knowledge and the accuracy of the test samples,the accuracy being 83.79%.Conclusion:The diagnostic knowledge of CHAID classification model automatically from the cases can be used to classify maxillary Ⅲ lymph nodes of breast cancer and other diseases.It is worth of widely practical use.
3.Soluble expression of human carboxypeptidase A1 active center in pichia pastoris

(Objective)To express the human carboxypeptidase A1 in soluble form in pichia pastoris.(Methods)Human carboxypeptidase A1 gene was amplified from pGEX-CPA1 recombinant plasmid and subcloned into pPIC9K yeast expression vector.Recombinant vector pPIC9K-CPA1 was transformed into pichia SMD1168 by electroporation.Positive recombinant yeast strains were screened through nutrition defect and G418 resistance.Expression of the integrated multicopy recombinant yeast strains were induced.Results The yeast recombinant expression vector pPIC9K-CPA1 was constructed.Recombinant yeast strains which integrated over 16 recombinant vector copies and soluble human carboxypeptidase A1 protein were obtained.(Conclusion)The human carboxypeptidase A1 can be expressed successfully in pichia pastoris,which will be useful for further research on ADEPT of prospate.
5.Expression of HLA-G protein in placental tissues and its influence on Th1/Th2 cytokines in peripheral blood in patients with intrahepatic cholestasis of pregnancy

Objective To investigate the changes of human leukocyte antigen-G (HLA-G) protein expression and Th1/Th2 type cytokines in intrahepatic cholestasis of pregnancy (ICP) and their relativity to the etiology of ICP. Methods Peripheral blood and placental tissues were obtained from 26 ICP patients (the ICP group) and 22 normal pregnant women (the NP group) in the operation room for Cesarean birth. Immunohistochemistry was used to detect the expression of HLA-G protein in the placental tissues. Meanwhile we tested the concentrations of tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) by enzyme-linked immunosorbent assay (ELISA) in the peripheral blood and checked the levels of TBA in the serum.Results TBA level in the ICP group was (27.05±6.08) μmol/L, significant higher than that in the NP group (4.35±2.68)μmol/L (P<0.01). The positive expression of HLA-G protein in extravillous trophoblast in the ICP group was significantly lower than that in the NP group (P<0.01). The mean optical density (MOD) of positive expression of HLA-G protein in the placenta tissues in the ICP group (52.91±7.19) was significantly lower than that in the NP group (69.26±7.72) (P<0.01). The concentration of TNF-α was significantly higher in the ICP group (101.31±19.30) pg/mL than that in the NP group (54.51±23.72) pg/mL (P<0.01). The concentration of IL-4 was lower in the ICP group (22.16±6.55) pg/mL than that in the NP group (31.69±8.25) pg/mL (P<0.01). The ratio of TNF-α/IL-4 was higher in the ICP group (4.52±1.91) than that in the NP group (1.72±0.61) (P<0.01). There was a negative correlation between the MOD of HLA-G protein and TNF-α (r=-0.98, P<0.01) in the ICP group. No correlation with IL-4 and TNF-α/IL-4 was seen (P>0.05). There was a positive correlation between TBA and TNF-α (r=0.99, P<0.01), and a negative correlation between TBA and the MOD of HLA-G protein (r=-1.00, P<0.01) in the ICP group. No correlation with IL-4 and TNF-α/IL-4 was seen (P>0.05). Conclusion There is an imbalance of Th1/Th2 cytokines to the Th1 type in the peripheral blood of ICP patients. The expression of HLA-G protein in the placenta of ICP patients decreases, leading to an increase of Th1 type cytokines that may be one of the reasons for liver destroy in ICP.
6.Warm Needling-based Specificity of Acupoints and Its Relationship with Mast Cell Function in Adjuvant Arthritis Rats

Objective To investigate the analgesic effect of warm needling and explore its relationship with the function of mast cells in the acupoint area and the specificity of acupoints.Method Sixty SD rats were randomized into blank (C), model (M), warm needling Zusanli (Z), contralateral warm needling (O), manual acupuncture (A), warm needling Futu (F), warm needling Yanglingquan(Y), warm needling Weizhong (W), disodium cromoglycate + warm needling (DSCG+Z) and saline + warm needling (Saline+Z) groups. A rat with adjuvant arthritis was used as a model of inflammatory pain (AA model). Hindpaw withdrawal latency was measured in therats during warm needling at Zusanli (ST36), Yanglingquan (SP9), Futu (ST32) and Weizhong (BL40). Pre-treatment and post-treatment rates of mast cell degranulation in the acupoint area were compared and the effect of acupoint area injection of disodium cromoglycate on the degranulation was observed by acupoint tissue section staining.Result Obvious degranulation of mast cells in the acupoint area appeared after warm needling (P>0.05). The analgesic effect of warm needling was better than that of manualacupuncture (P>0.05). After treatment, pain threshold was significantly higher in the warm needling Zusanli group than in the warm needling Futu, warm needling Yanglingquan and warm needling Weizhong groups (P>0.05). Acupuncture-induced mast cell degranulation could be prevented by disodium cromoglycate (P>0.05).Conclusion Warm needling at point Zusanli can produce an analgesic effect in adjuvant arthritis rats. Its analgesic effect is better than that of manual acupuncture. The rate of mast cell degranulation in the acupoint area is higher in the warm needling Zusanli group than in the manual acupuncture group. The analgesic effect of warm needling is reduced after pretreatment with disodium cromoglycate, a mast cell stabilizer. In an AA model, the analgesic effect of warm needling at point Zusanli is better than those of warm needling at points Yanglingquan, Futu and Weizhong.
7.The clinical therapeutic effect of acupuncture combined with Vitalstim electric stimulation therapy on dysphagia after ischemic stroke

Objective To investigate the therapeutic effect of acupuncture combined with Vitalstim electric stimulation therapy for treatment of patients with dysphagia after ischemic stroke.Methods Ninety-two patients with dysphagia after ischemic stroke in accord with the criteria of enrollment in this study were admitted to the Department of Acupuncture of the First Affiliated Hospital of Tianjin Traditional Chinese Medicine University from May 2014 to November 2016, and they were divided into a control group and an observation group by random number table, 46 cases in each group. Both groups were treated with stroke conventional drug therapy and feeding swallowing rehabilitation training, in addition, Vitalstim swallowing therapeutic electrical stimulation therapy was used in the control group, and acupuncture combined with Vitalstim stimulation were applied in observation group. The changes of swallowing ability score, video fluoroscopic swallowing study (VFSS) score and adverse reactions were observed before and after treatment in the two groups, and the curative effect of combined method was evaluated.Results Compared with those before treatment, the swallowing ability and VFSS score of two groups after treatment were obviously higher, and the degrees of changesin observation group were more significant than those in control group (swallowing score: 8.12±1.13 vs. 7.45±1.04, VFSS score: 7.97±1.05 vs. 5.67±0.88, allP < 0.05); the total efficiency in the observation group was markedly higher than that in the control group [95.7% (44/46) vs. 78.3%(36/46)], the adverse reactions (wrong aspiration, pulmonary infection) in the observation group were significantly lower than those in the control group [10.9% (5/46) vs. 28.3% (13/46)], and there were statistically significant differences between the two groups (bothP < 0.05).Conclusions Acupuncture combined with Vitalstim electric stimulation therapy can improve the swallowing ability in patients with dysphagia after ischemic stroke, and the therapy has less adverse reactions; compared with the simple Vitalstim electric stimulation therapy, in the aspect of improving swallowing ability, the therapeutic effect of combined therapy is more significant.

9.Progress in the application of statins in Kawasaki disease

Kawasaki disease is an autoimmune vasculitis disease, mainly involving small and medium-sized arteries, especially coronary arteries.The severity of coronary artery lesions is closely related to the prognosis of Kawasaki disease.Statins have been proved effective in the treatment of cardiovascular diseases.Recent studies have demonstrated the critical role of statins in the treatment of Kawasaki disease and their potential value in preventing the formation of coronary artery aneurysms in the acute and convalescent stages of Kawasaki disease.In this article, the research progress of statins in Kawasaki disease was reviewed.
10.Smooth muscle progenitor cells and cardiovascular disease

It is possible for bone marrow cells,peripheral blood cells,myocardial cells,muscle cells and embryonic stem cells differentiating into smooth muscle progenitor cells(SPCs),which can express special marks of myosin heavy chain.SPCs remained quiescent,until mobilized in response to injury or disease.With various growth factors,cytokines,SPCs are mobilized following physiological stress and subsequently home to site of vascular damage,where they contribute to the remodeling process.Recently,various studies have found that SPCs from different sources contributed to the remodeling process of vessels and development in atherosclerosis.It is generally accepted that SPCs contribute to pathological changes,leads to restenosis.But several recent papers reported findings that injection of SPCs increased collagen and SMC content and reduced macrophage content,consistent with a more stable plaque.SPCs may secrete cytokines and growth factors both locally and systemically that enhance plaque stability.It remains to be determined whether smooth progenitor cells,which are less well studied than their endothelial counterparts,can likewise be manipulated to achieve therapeutic genes benefit.This review has elucidated the origin and distribution of SPCs,mobilization and recruitment,and their roles in cardiac vascular diseases.
