1.Cloning of ICAT and its anti-cancer function

Objective:To Clone the cDNA of human ICAT,and express it in tumor cell,to investigate its anti-tumor effect.Method:the ICAT cDNA was amplified by RT-PCR,the ukaryotic expression plasmid pcDNA3.1-ICAT was constructed,MCF-7 and MDA-MB231 breast cancer cells were transfected by pcDNA3.1-ICAT using liposome and the cancer cell proliferation was estimated by MTT.Result:The product of RT-PCR was about 260bp,its sequence was the same as that reported,the transfection efficiency for MCF-7 and MDA-MB-23 cells was about 26%,and inhibiting efficiencies on the proliferation of the cancer cell transfected were 18.1% and 16.2% respectively.Conlusion:The cDNA of ICAT was cloned,and its expression in MCF-7 and MDA-MB-23 cells may inhibit the proliferation of these cancer cells.
2.Relationship between serum TNF-α and IL-6 levels and respiratory function in patients with chronic obstructive pulmonary disease

Objective To investigate the relationship between serum TNF-α and IL-6 levels and respiratory function in patients with chronic obstructive pulmonary diseases.Methods 50 cases in our hospital diagnosed as chronic obstructive pulmonary disease from September 2015 to September 2016 were selected as the observation group,and 40 cases of healthy people were enrolled as the control group,serum IL-6,TNF-α were detected by ELISA in those people,correlation analysis was used to explore the relation ship between serum inflammatory factors,grading of disease and respiratory function in patients with chronic obstructive pulmonary disease(COPD).Results In the observation group TNF-α[(4.27±0.34)μg/L],IL-6[(21.54±3.67)ng/L] were significantly higher than those in healthy control group[(1.36±0.29)μg/L,(10.27±1.36)ng/L],the difference were statistically significant(P<0.05).COPD patients in different GOLD stage differed in the serum contents of TNF-α,IL-6 and symptom score(P<0.05).The serum contents of TNF-α,IL-6 and symptom score were higher when the GOLD stage got higher.TNF-α,IL-6 was positively correlated with symptom score,lung function,GOLD classification(P<0.05).Conclusion Serum levels of TNF-α,IL-6 and COPD are positively correlated with the patient′s condition,and positive correlation with pulmonary function.
3.Neurotoxic effects of intrathecal ropivacaine and bupivacaine on the spinal cord in rats

Objective To compare the neurotoxic effects of intratheeal ropivacaine and bupivacaine on the spinal cord in rats.Methods Female SD rats weighing 240-330 g were used in this study.Catheters(PE 10 tubing)were inserted through an incision in the cisternal membrane and advanced caudad until the tip of the catheter reached the lumbar spinal level according to the technique described by Yaksh and Rudy.Fifty-four successfully catheterized rats were randomly assigned tO one of 3 groups(n=18 each):group Ⅰ received normal saline 30μlIT(NS);groupⅡ received 2%bupivacaine 20μl IT(BU)and group Ⅲ 2.7%ropivacaine 20μl IT (RO).Degree of motor block was assessed and scored(0=no block,2=inability to flex the hind limb)before (T1,baseline)and at 10,20,30,60 and 120 min(T2-6)after IT injection.On the 4th day after IT injection specimens were obtained from lumbar spinal cord for microscopic examination.Results The motor block score was significantly higher in group BU and RO than in group NS(P<0.05),and the tissue damage was severe in group BU and RO.The motor block scores were significantly higher at T2 than at T3-6 in beth group BU and RO(P<0.05).There Was no significant difference in motor block score at T2 between groups BU and RO but the motor block scores were significantly lower at T3-5 in group RO than in group BU(P<0.05).The tissue damage was Severer in group RO than in group BU(P<0.05).Conclusion Intrathecal 2.7% ropivacaine is more toxic to the spinal cord than intrathecal 2%bupivacaine in rats.
4.How Essential the Anti-diabetic Medications in Chinese Hospitals?

OBJECTIVE:To analyze the treatment of anti-diabetics medicines and use of essential medicines in Chinese hospi-tal,and provide evidence for developing medicine policy,preventing and treating diabetes. METHODS:Data about annual con-sumption from 2012 to 2014 were extracted from the IMS CHPA database to analyze the object medicines of the target drugs with top 60% consumption volume of oral anti-diabetics and insulins,and the proportion of consumption volume and consumption amount in insulin,its analogues and three generations of insulin by 2 administration mode in Chinese hospital. RESULTS & CON-CLUSIONS:Acarbose tablet manufactured by Beijing Bayer HealthCare Co.,Ltd. and Metformin tablet manufactured by Shanghai Squibb Pharmaceutical Co.,Ltd. had occupied the top 2 of volume consumption in Chinese hospitals,totally accounting for 25%-26%. The 60% volume consumption of insulins and its analogues concentrated on one cheap local animal insulin(volume ac-counting for 13%-16% and amount accounting for 1%)and those imported or joint venture-made the second and third generation of insulins and analogues administrated by pen/cartridge. No matter insulins or oral anti-diabetics,the top consumption was listed as National Essential Medicine System,but none was recommended by WHO. Multi-pharmas dominate the Chinese anti-diabetic mar-ket through introducing new combined formulations and new dosage forms. WHO non-listed anti-diabetics have beautiful sales in Chinese hospitals,while traditional cheap medications for the general public grew slowly and with low profits. Chinese hospitals generally consumed more expensive new generations and original research manufacturers of anti-diabetics. As a lower middle in-come country that just established a universal coverage of basic health insurance,China still has a huge population with very limit-ed benefit package. Efficient use of limited health resources is essential for a successful health reform and a sustainable development of the health system in China.
5.Expression of toll-like receptor 4 in hepatic ischemia-reperfusion injury

Objective To analyze the expression and the role of Toll-like receptor 4 (TLR4) and the effect of N-acetylcysteine (NAC) on hepatic ischemia- reperfusion (I/R) injury.Methods The rat model of 45 min hepatic ischemia followed by 24 h reperfusion was used. Animals were divided into 3 groups: the pseudoperated control group (P), I/R group and treated group with NAC at a dose of 300 mg/kg at 5 min before reperfusion (I/R-NAC). Plasma concentrations of ALT, AST and lipopolysaccharide (LPS) at various time points were measured. The expression of TLR4 protein and mRNA was detected in liver tissue by using immunohistochemistry and RT-PCR.Results The TLR4 expression, plasma ALT, AST and LPS levels were markedly increased during 45 min hepatic ischemia and 24 h reperfusion, but they were lower in I/R-NAC group and no alteration almost in group P. The plasma concentrations showed a significant correlation between ALT, AST and LPS at different time points in I/R group and I/R-NAC group.Conclusion LPS and TLR4 play a prominent role in the pathophysiological processes of I/R and NAC can be of therapeutic potential for reducing the expression of TLR4 mRNA and alleviating endotoxemia derived from gut.
6. Effect of borneol on absorption of chlorogenic acid by two absorption models and its correlation of these absorption models

OBJECTIVE: To investigate the enhancing effect of borneol on the absorption of chlorogenic acid (CGA) with two different biological models and to evaluate the correlation of these two models. METHODS: The in situ rat perfusion model was used to investigate the enhancing effect of borneol on the absorption of CGA. The in vivo pharmacokinetics study was carried out to evaluate the correlation of the two-compartment open model. RESULTS: With the in situ rat perfusion model, the absorption rate constant (Ka) of CGA was 0.083 4 h-1, which was up to 1.58-fold higher by borneol. The results of in vivo pharmacokinetics study in rats suggested that 0.04% borneol could increase the relative bioavailability by 116.02% and maximum plasma concentration increased slightly. The consistent result of these two different absorption models was provided (r=0.941). CONCLUSION: Borneol could promote the oral absorption of CGA.
7.Significance of serum homocysteine in patients with diabetes mellitus complicating proliferative retinopathy

Objective To evaluate the role of serum homocysteine (HCY) in diabetes mellitus(DM ) complicating proliferative diabetic retinopathy (PDR) .Methods 174 patients with DM were selected and divided into PDR ,non‐PDR(NPDR) and non‐dia‐betic retinopathy (NDR) groups .The serum HCY level was detected .Results Serum HCY levels in the PDR ,NPDR and NDR groups were (18 .20 ± 5 .60) ,(14 .40 ± 6 .70) and ,(12 .10 ± 6 .80)μmol/L respectively ,which was highest in the PDR group (P<0 .05) .Conclusion HCY could result in PDR generation possibly by injuring the retinal blood vessels .
8.Progress in laboratory diagnosis of heparin induced thrombocytopenia

Heparin induced thrombocytopenia ( HIT ) is a severe side effect of heparin with antibody-mediation.Laboratory assays can be divided into two major categories, about functional assays and HIT antibodiesdetection.Thefunctional assays, such as the serotonin release assay ( SRA ) and heparin-induced platelet activationassay( HIPA) , are sensitive and specific for HIT.They arethe reference standard assays generally, but have thedeficiencies of complicated operation and time-consuming, and cannot be used as a routine examination. TheHIT antibodiesdetections, such as ELISA, immune turbidimetry assay, chemiluminescent assay and lateral flow immunoassay, have high diagnostic sensitivityandareavailable at routine laboratories.They can exclude the diagnosis of HIT or beused to diagnose HIT effectively combined with the pre-test probability score(4Ts score) of HIT.
9.Protective effect of neurogenesis in cerebral ischemia

In recent years,researchers have carried out a lot of studies about endogenous and exogenous protective mechanisms of ischemic brain injury and have found that cerebral ischemia can stimulate cell proliferation and neurogenesis.This article reviews the cerebral ischemia induced neural stem cell proliferation,the protection mechanisms of directional migration and differentiation,as well as the promoting effects of the application of exogenous factors on neurogenesis after ischemia.
10.The Change Feature of the Signal of Fusiform before and after the Vocabulary Learning

Objective To explore the function of fusiform in learning process according to the change of its activation volume and intensity.Methods Ten Chinese normal university students with Guangdong dialect performed the verb generation.fMRI was used to scan the activities of brain.Deconvolution and ANOVA were used to deal with the data of individuals and groups respectively.Results After practice,the activations of the most brain areas were weakened in different degree of great quantities along with practice and the activation of fusiform disappeared.Conclusion Learning leads to the change of brain activation.The function of fusiform is not specific,but general neural activation during learning process.
