1.lnhibition effects of Bevacizumab on migration of human Tenon capsule fibroblasts

Abstract? AlM: To investigate the inhibition effect of bevacizumab on human Tenon capsule fibroblasts ( HTFs ) and discuss the countermeasures of bleb scarringscarring in glaucoma surgery countermeasures.? METHODS: Adopted cell recovery method and followed the aseptic principles, we performed the culture of HTFs which came from the Central Laboratory of Shaanxi People's Hospital cell library. Wound Healing assay:We scraped a cell-free zone on the cell surface when the cells reached confluence at 80%. The control group was added to serum-free DMEM medium. The HTFs of bevacizumab group were stimulated with 1mg/mL concentrations without DEME for 0, 24, 48, and 72h. The scratch width was observed and measured.? RESULTS: HTFs were long fusiform shape under microscope, the nucleus is in the center of the cell with larger nucleus, abundant cytoplasm, were arranged in a whorled growth out of shape, strong ability to proliferate, conform to general forms of fibroblast. The morphological and biological characteristics of cells after cryopreservation and resuscitation remain unchanged. Wound healing assay: 0h, equal to the initial width of the two groups, 24h when the migration distance of the two groups of cells are basically the same, 48h when the control group cell migration distance is greater than that in the bevacizumab processing group, 72h when the control group scratches basichealing, bevacizumab treated cells migrate closer than 48h no significant change, and a lot of cells died.?CONCLUSlON:Cell recovery method can successfully cultured HTFs, which was stability on morphology and biological properties, laying the cellular basis for experimental research. Fibroblast itself has a strong ability to migrate, outside - derived bevacizumab can inhibit HTFs migration evidently and it will cause excessive cell death when. Bevacizumab has certain extent inhibitory effect on HTFs migration, and it is likely to become one of the important drugs for creating bleb scarring after glaucoma surgery in the future.
2.Effect of Mingjing decoction with argon laser on plasma VEGF in patients with diabetic retinopathy

AIM: To investigate the effect and mechanism of Mingjing decoction combining with argon laser on the levels of plasma vascular endothelial growth factor (VEGF) in patients with diabetic retinopathy(DR).
METHODS:The 66 patients(118 eyes)with DR at Ⅲ ~ Ⅴstage were randomly divided into two groups (treatment group and control group). The control group (33 patients with 56 eyes ) was treated with argon laser, and the treatment group (33 patients with 62 eyes) was treated with mingjing decoction combining with argon laser, and a blank group(30 eyes) was set up. The levels of plasma VEGF were detected by enzyme linked immunosorbent assay (ELISA). The results of best corrected visual acuity and fundus fluorescein angiography(FFA) were analyzed, as well as detected glycosylated hemoglobin ( HbA1c), blood coagulation function and the function of liver and kidney.
RESULTS: Patients with DR had significantly risen plasma VEGF before treatment when compared with blank control group ( P < 0. 05 ). After 3mo of combined treatment, the levels of plasma VEGF significantly reduced and HbA1c decreased in patients treated with mingjing decoction and argon laser, there were statistically significant difference compared to control group ( P < 0. 05 ). In the treatment group, the best corrected visual acuity and FFA were significantly improved in patients treated with mingjing decoction combining with argon laser compared to patients treated with argon laser alone after 3mo of combined treatment. CONCLUSION: Mingjing decoction combining with argon laser for DR can effectively reduce the level of plasma VEGF, stabilize blood sugar levels, improve the function of retina, and delay DR progresses.
3.Determination of Loganin in Productions of Liuweidihuang by HPLC

Objective To determine loganin in productions of Liuweidihuang. Methods The chromatographic column was shim-packvp ODS-C18 (4.6 mm?150 mm,5 ?m),the mobile phase was THF-acetonitrile-methanol-0.05%H4PO3 (1∶8∶4∶87),the detection wavelength was 236 nm,the temperature of column was 40 ℃ and the flow rate was 1.0 mL/min. Results A good linearity of loganin was in the range of 4.42?10-2~22.1?10-2 ?g,r =0.999 7,and the average recovery rate was 98.96% (n =6,RSD=1.42%). Conclusion The method provided a reference for evaluating and controlling quality of series productions of Liuweidihuang.
4.Correlated Factors of Compliance in Patients with Depression in Polyclinic

ObjectiveTo investigate compliance with drug, the factors influencing compliance in patients with depression, main dropout time and the characteristics. MethodsA retrospective study was carried out with a total of 154 patients with depression. The correlated factors were studied and dropout time and characteristics were explored. ResultsThe depression patients with bad compliance were 95 (61.7%). The factors influence the drug compliance was various. Different dropout reasons occur at different time. The different dropout rate of depression were 25.3%,18.2%,7.8%,5.8%,4.5% within the 1st, 1st~3rd, 3rd~6th ,6th~9th,after 9th month respectively. ConclusionBad compliance with antidepressant is popular with various reason. Dropout of antidepression is mainly in the 1st month after treatment. Deficiency of associated knowledge is the main reason to stop treating.

Objective To investigate the clinical effect of 153 Sm-EDTMP treatment on pain palliation in the patients with bone metastasis of lung cancer, breast cancer or prostate cancer. Methods 166 patients with bone metastasis of cancers were intravenously injected 18.5~37 MBq/kg (0.5~1.0 mCi/kg ) body weight of 153 Sm-EDTMP, once a month or two months. Results The effective rate of pain palliation of 153 Sm-EDTMP treating bone metastasis of lung cancer, preast cancer and prostate cancer were 68.5%, 89.3% and 100%, respectively. 9. Effect of carvedilol on cardiac metalloproteinases and tissue inhibitors of metalloproteinases after myocardial infarction in rats

Objective: To investigate the effect of carvedilol on expression of cardiac matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) after myocardial infarction in rats. Methods: An animal model of acute myocardial infarction (AMI) was established by descending left coronary artery ligation in 24 rats and they were divided into carvedilol (10 mg • kg-1 • d-1) group (n=12) and normal saline group (n=12). Sham-operated group (n=9) received the same procedure but with no ligation. All animals were treated for 6 weeks via a gastric lavage. Heart function and hemodynamic parameters were determined after 6 weeks. The protein expression of cardiac MMP-2, MMP-9 and TIMP-2 was detected by immunohistochemical analysis in AMI groups, and the MMPs activities were assessed by zymography. Gene expression of myocardial MMPs/TIMPs (MMP-2,-9 and TIMP-1, -2) and cytokines (TNF-α, IL-1β) were measured by real-time quantitative PCR. Results: Compared with Sham-operated group, carvedilol group had significantly higher left ventricular end-diastolic pressure (LV-EDP) and lower LV upstroke velocity (+dp/dtmax) and LV descent velocity (- dp/dtmax) (P<0.01). Activities of MMP-2 and MMP-9, protein expression of MMP-2, MMP-9 and TIMP-2, and mRNA expression of MMP-2, MMP-9, TIMP-1, TIMP-2, IL-1β, and TNF-α were all higher in carvedilol group compared with sham-operated group (P<0.05). Objective To determine the effects of EGCG on lipopolysaccharide ( LPS)-induced neuroinflamma-tion and investigate the role of neuroprotection mediated by EGCG .Methods Primary cultures of rat gliacyte were used as an in vitro model to examine the effects of EGCG on LPS-induced neuronal damage .The intracellu-lar Glu andγ-GABA were quantified via HPLC .Then the protein level of TNF-α,IL-1βand IL-8 was determined by ELISA and Western blot assay .Results Compared with the control group , LPS apparently induced the pro-duction of intracellular ROS ( P<0.05 ) and released the TNF-α, IL-1βand IL-8 in the primary cultures super-natant (P<0.05).Compared with the LPS group,EGCG significantly attenuated the release of TNF-α, IL-1βand IL-8 ( P<0.05 ) and the level of iNOS protein ( P<0.05 ) .LPS apparently induced the production of intra-cellular ROS( P<0.05 ) and released the TNF-α, IL-1βand IL-8 in the primary cultures supernatant ( P <0.05 ) .EGCG significantly attenuated the release of TNF-α, IL-1βand IL-8 ( P<0.05 ) and the level of iNOS protein(P<0.05), and rugulated the concentration of Glu/γ-GABA(P<0.05).Conclusions EGCG is effective in protecting hosts against LPS-induced neuroinflammation through anti-inflammatory effects and regulating extracel-lular Amino acid levels .
