Ever from FDA approved the Rituxan in 1997,an Anti-CD-20 chimeric antibody,there have been more than 20 antibodies approved and they have got into more than 56 countries.About half approvied antibodies are curing kinds of cancers and seven of them have got a excellent sale targets to be "Blockbuster Drug".As the cell culture technology improving,the large scale cell expression titer comes from 1g/L to 5g/L.At the same time,the culture scale is coming form 12,000L to 15,000L,even 20000L and it can run several bioreactors at the same time.All these are make the improvement of the antibody purification.The new edition of media,system,chromatography technology and filtration technology can product the protein to several tens kilogram antibody per cycle and tons per year,although the productivity is only several kilograms per cycle many years ago.Also the new combined technology makes a two-step chromatography production protocol come into been.This will highly improve the antibody productivity and save the investment cost.

Objective To analyze the relationship between life style and chronic diseases.Method 2 886 people were selected randomly from three districts of Tianjin and surveyed with a questionnaire on their healthy condition and physical examination.2 622 people were taken a diet survey by 3-days foods intakes weighing and 24-hours diet recall,then the relative risk factors between life style and coronary heart disease,cerebrovascular disease,diabetes,hypertension,overweight and obesity were analyzed.Result The standardized prevalence rates of coronary heart disease,cerebrevascular disease,diabetes,hypertension, overweight and obesity of residents were 9.24%,4.33%,6.74%,34.05%,and 42.62%,respectively.The more the intake of energy, the more the risk for people to suffer from overweight and obesity,OR value being 1.00-1.70.With intake of more than 20 g dietary fiber per day,the risk for suffering from overweight and obesity could be reduced,OR value being 0.68.Logistic analysis showed that heavy salty taste could increase the risk for suffering from hypertension by 42%.Watching TV more than 3 hours per day increased the risk for suffering from cerebrovascular disease and diabetes,OR value being 1.802 and 1.703 respectively.Sleep less than 7 hours per day was a dangerous factor for hypertension,coronary heart disease and cerebrovasular disease,OR value being 1.517, 1.833 and 2.041 respectively.Conclusion The life style such as dietary pattern,sedentary occupation and sleep hours have strong connection with chronic diseases,such as hypertension,diabetes etc.Reasonable diet,suitable exercises,reducing sedentary position and increasing sleep hours will be good for health,and also for the prevention and control of chronic diseases.

Retinitis pigmentosa (RP) encompasses many different hereditary retinal degenerations that are caused by a vast array of different gene mutations and have highly variable disease presentations and severities.Work over the past 25 years has resulted in the identification of genes responsible for about 50％ of the RP cases,and it's predicted that most of the remaining disease-causing genes will be identified by the year 2020 or most likelysooner.This marked acceleration is the result of dramatic improvements in DNA-sequencing technologies and the associated analysis.The advent of two recent innovations, induced pluripotent stem cells (iPSCs) and clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated nuclease-9 (Cas-9) mediated genome editing,are changing the landscape of RP research, with causative genes being identified at an accelerated rate withgreat potential to translate these discoveries into personalized therapeutic strategies.

Objective To evaluate the safety and clinical efficacy of high intensity focused ultrasound (HIFU) for patients with solid tumors under sedation and analgesia. Methods Sedation and analgesia were induced with i.v. fentanyl(1?g/kg) and midazolam (0.03mg/kg) in 81 patients with benign or malignant solid tumors. High intensity focused ultrasound (HIFU) ablation treatment was then performed. The short-term effect and adverse events of sedatives and analgesics, as well as the effect of HIFU ablation treatment were observed. Results One hundred and twelve HIFU courses were performed on 81 patients, among them 23 patients with benign solid tumors received 26 courses of treatment, and 58 patients with malignant solid tumors, received 86 courses of treatment. A total of 153 foci were targeted. It was estimated that in 81% of all the patients, the tumor size was decreased by 50%, including 72.2%(52/72)of malignant tumors and 97.4%(37/38)of benign tumors. Tumor markers of malignant tumors decreased by more than 50% in 81.3%(13/16) of patients, and remission rate of tumor-related symptoms reached 77.9% (30/39). The adverse events during sedation and analgesia were nausea, reduction in the respiratory rate, visual hallucination, and that of HIFU were pain and swelling. No comblications with severity surpassing grade Ⅲ were found as a result of either sedatives, analgesics or HIFU treatment. Conclusion Sedation and analgesia are relatively safe, effective and feasible during HIFU treatment for solid tumor.

Anterior cervical plating has been widely used to treat cervical spine trauma, degeneration, deformity and tumor due to the advantages such as augmentation of cervical stability and promotion of bone graft fusion. This paper introduced the classification and evolution of plate-screw internal fixation system, analyzed the biomechanics of anterior cervical plate, and discussed its clinical application. Although the therapeutic effect of anterior cervical plating is favorable, there are still various complications postoperatively. Therefore, it is necessary to improve the anterior cervical plate from different aspects to expand its clinical application.

Objective To explore the effect of sealing tube with regular urokinase for the malfunction of central venous catheter(CVC) in patients with hemodialysis (HD).Methods This was a prospective randomized controlled study,132 HD patients with cuff CVC were studied.In the early stage,52 H D patients without 80 patients with regular catheter-locking solution of urokinase.To analyze effect of cuff CVC malfunction,blood flow(BF),dialysis adequacy,anemia and serum albumin (ALB) with regular catheter-locking solution of urokinase.The incidence of catheter malfunction was calculated based on the catheter dysfunction in the first 3 months.Clinical index was measured,recorded BF and calculated urea clearance rate (Kt/V).Results HD patients catheter-locking solution with regular urokinase could reduce the incidence of dialysis catheter malfunction ( 26.3％ vs 32.7％,x2 =32.727,P ＜0.01 ),increase the BF,Kt/V,levels of hemoglobin(HGB) and ALB(P＜0.05 or P ＜0.01).Conclusion Regular catheter-locking solution with urokinase is effective in reducing HD patients incidence of dialysis CVC malfunction,increasing patients BF,dialysis adequacy and the level of ALB,improving anemia.

Objective To quantify the morphometric characteristics of the pedicles of the infantile thoracolumbar spine and to determine the feasibility and safety of pedicle screw fixation in very young children. Methods A total of 52 patients younger than three years of age underwent standard spiral computed tomography (CT) were enrolled in this study. The patients were grouped according to age, group 1 (0 ～ 18months of age) and group 2 (18 ～ 36 months of age). Images were reformatted, and multiplanar reconstructions were used to attain images of thoracic and lumbar pedicles on sagittal, coronal and transverse planes.The measurements included the inner and outer pedicle diameters on the transverse plane, pedicle sagittal diameter, pedicle length and the pedicle angle on the transverse. Characteristics of the pedicles of the immature thoracic and lumbar spine were studied. Results Pedicle diameters on the transverse plane decreased gradually from T1 to T5 and increased gradually from T6 to L5. The shortest transverse diameter of the thoracic pedicle was T5 or T6. The inner diameter of the segment of L4 in infant (younger than eighteen months) was less than 3.0 mm and the external diameter less than 3.5 mm from T3 to T9. The inner diameter of the segment from T5 to T8 in infant (more than eighteen months) was less than 3.0 mm and the external diameter from T4 to T7 was less than 3.5 mm. The sagittal diameter was significantly more than the transverse diameter. Pedicle diameters on the transverse plane, the sagittal diameter, and the length of the pedicle changed significantly with age, but the pedicle angle did not changed significantly with age. Conclusions The pedicles in the segment from T3 to T9 of less than eighteen months and from T4 to T7 of more than eighteen months were too small, there was no feasibility and safety of pedicle screw fixation. So pedicle screw fixation was feasible and safe for below T9 segment in the infants who are younger than three years old.

AIM:To observe the situation of rat retinal tissue DNA damage at early diabetic period, discuss the role of the blood glucose fluctuations, and provide a new method for studying the pathogenesis of diabetic retinopathy ( DR) .
METHODS: SD rats were randomly divided into four groups:normal control group (NC), normal fluctuation group ( NF ) , diabetes group ( DM ) and diabetes fluctuation group ( DF ) . Diabetic models were established through intraperitoneal injection of STZ. A certain amount of glucose was injected in the rats of group NF and DF in an intraperitoneal mode three times a day after the model was established, thereby causing blood glucose fluctuations. Rats were killed and the retinal tissues were taken in the 8th week. Single cell gel electrophoresis ( SCGE ) technique was adopted for detecting DNA injury extent in the retina tissue.
RESULTS:Groups NF and DF showed significant and regular fluctuations. The curve of blood glucose fluctuations was relatively stable. All values of MBG, SDBG, LAGE and M were significantly increased compared with group NC. Group DF was increased more significantly. It was statistically significant (P<0. 01). SCGE showed that there were DNA damages in different levels in the cells of group NF, DM and DF. Indicators of cells such as TL, TDNA %, TM, OTM were higher than that in group NC. It was statistically significant ( P<0-01). The comparison difference between two groups was also significant (P<0. 01).
CONCLUSION: Rat retinal tissues have DNA injury during early diabetic period. DNA injury is gradually aggravated with blood glucose fluctuation. It indicates that high blood glucose and blood glucose fluctuation are involved in the mechanism of cell DNA injury, and they may be one of DR early event, have played a certain role in the incidence of DR.
