5.Study of cognitive dysfunction and its features in patients with age-related cerebral white matter changes
Chinese Journal of Geriatrics 2010;29(11):912-915
Objective To explore the relationship between age-related cerebral white matter changes (ARWMC) and cognitive function, observe the features of cognitive impairment in ARWMC patients, and provide evidences for preventing and treating cognitive impairment in elderly people. Methods Fifty patients with ARWMC were divided into mild-to-moderate group (n= 26, ARWMC score 1-10) and severe group (n= 24, ARWMC score> 10) based on the ARWMC scale of CT/MRI scans. Twenty healthy elderly people, who underwent physical examination in hospital, were selected as control group. The cognitive function was evaluated using Montreal Cognitive Assessment (MoCA,Beijing version). Cognitive functions were compared between patients with ARWMC at different extents and the controls. The correlation between severity of ARWMC and cognitive function, the characteristics of cognitive impairment in patients with ARWMC were analyzed. Results The cognitive function in patients with ARWMC was impaired significantly (the MoCA scores were 26.60±1.23 in control group, 23. 19±2. 62 in mild-to-moderate group and 19.83±3. 09 in sever group, F =39. 930, P = 0. 000). The severity of ARWMC was negatively related with MoCA score (r =-0. 476, P=0. 000). The patients showed cognitive impairment especially in the visuoconstructional and executive functions (F= 13. 189, P<0.05), delayed recall (F=32.340, P<0.05) and orientation (F= 15.813, P<0. 05). Conclusions ARWMC is related with cognitive impairment. The severity of white matter changes is correlated with worse cognitive function.

7.Allogeneicversus heterogeneous bone marrow mesenchymal stem cells for laryngeal cartilage repair
Chinese Journal of Tissue Engineering Research 2016;20(7):966-971
BACKGROUND:Because chondrocytes have no regeneration ability, to select suitable seed cels is the primary problem to repair cartilage defects.
OBJECTIVE:To investigate the effect of alogeneicversus heterologous bone marrow mesenchymal stem cels (BMSCs) in repairing laryngeal cartilage defects after chondrogenic induction.
METHODS:BMSCs from human and rabbits were isolated and cultured. Passage 3 cels were cultured in chondrogenic induction medium containing transforming transforming growth factor beta 1 and bone morphogenetic protein, and then were dropped onto a poly(lactic-co-glycolic acid) (PLGA) scaffold. Thirty New Zealand rabbits were randomly assigned into three groups: blank control group, human BMSCs group, rabbit BMSCs group. Animal models of laryngeal cartilage defects were made in the three groups. After modeling, saline-soaked PLGA scaffold, PLAG scaffold with human BMSCs or with rabbit BMSCs were implanted respectively into the rabbits in the normal blank, human BMSCs and rabbit BMSCs groups. The expression of type II colagen in the larynx and its surrounding tissues was detected by immunohistochemistry at 4 and 8 weeks postoperatively.
RESULTS AND CONCLUSION: The animals in each group breathed normaly with no presence of wheezing, and their eating and activity were good. Moreover, there was no purulency or infection in the three groups. At 4 and 8 weeks after operation, the positive rates of type II colagen in the two BMSCs groups were significantly higher than that in the blank control group (P < 0.05). There was no significant difference between two BMSCs groups (P > 0.05). These results show that both alogeneic and heterologous BMSCs have good therapeutic effects on the repair of laryngeal cartilage defects in rabbits.
8.Effect of Continuous Positive Airway Pressure in the Treatment of Obstructive Sleep Apnea Hypopnea Syn-drome Complicated with Coronary Heart Disease
China Pharmacist 2016;19(6):1105-1107
Objective:To explore the application value of continuous positive airway pressure ( CPAP) in the treatment of obstruc-tive sleep apnea hypopnea syndrome (OSAHS ) complicated with coronary heart disease (CHD).Methods:Totally 50 patients with OSAHS complicated with CHD from June 2012 to June 2014 in our hospital were randomly divided into the control group and CPAP group with 25 ones in each .The patients in the control group were treated with the conventional drug therapy for coronary heart disease , while those in CPAP group were treated with CPAP additionally .After 6-month treatment, PSG index, clinical symptoms, arrhythmia and ischemia electrocardiogram changes of the patients in the two groups were compared .Results:After the 6-month treatment , AHI and oxygen desaturation index in CPAP group were significantly decreased when compared with those before the treatment , and the min-imum value of SaO2 and the average value of SaO 2 were higher than those before the treatment (P<0.01), and the above indices were better than those in the control group (P<0.01).The incidence rate of chest pain and chest tightness , arrhythmia, times of myocardi-al ischemia in a day and time of myocardial ischemia in a day were decreased after the treatment (P<0.01), and the decrease in CPAP group was more notable than that in the control group (P<0.01).Conclusion: CPAP combined with the conventional drug therapy for coronary heart disease can improve the oxygen concentration and reduce the incidence of arrhythmia and myocardial ischemia in the patients with OSAHS with CHD , which can be widely used in clinics .
9.The regulation of Cathepsin B on cell apoptosis induced by SAHA in breast cancer cell line MCF-7
Chinese Pharmacological Bulletin 2016;32(8):1086-1090
Aim To clarify the regulation role of ca-thepsin B ( Cat B ) in cell proliferation and apoptosis induced by SAHA in ER-positive breast cancer cell line MCF-7.Methods MTT was used to screen the optimal concentration and treatment time of SAHA . The expression levels of related proteins were deter-mined by ELISA , and the morphological changes were observed through time-lapse live cell imaging acquisi-tion.Cell viability and apoptosis assay in MCF-7 cells were assessed by Muse Cell Analyzer with SAHA and /or Cystatin C treatment .Results MTT assay showed that the anti-tumor efficacy of SAHA was significant . The optimal concentration and treatment time were 10μmol? L-1 and 24 h respectively . ELISA assay showed that SAHA could induce expression of Cat B in MCF-7 cells.Real-time live-cell imaging experiments demonstrated that the combination treatment of Cystatin C and SAHA significantly resumed the inhibitory effect caused by SAHA alone .Cytology test showed that SA-HA alone obviously depressed the cell viability and in-duced apoptosis . However , the effect was reversed with the combination of Cystatin C .Conclusion Cat B plays an important role in apoptosis induced by SAHA in ER +breast cancer cells MCF-7.
