1.Predictive value of combined determination of ICA、64KDAb and IAA in the first-degree relatives of type Ⅰ diabetics

Objective To study the predictive value of islet cell antibody (ICA),64KD protein antibody(64KDAb) and insulin auto-antibody (IAA) in the first-degree relatives of type 1 diabetics. Methods The determination of islet autoantibody distribution in type 1 diabetes patients,first-degree relatives without diabetes and normal individuals was made. Results The results revealed that ICA and 64KDAb positive rate in type I diabetes patients and their first-degree relatives was significantly higher than normal individuals. The positive rate of 64KDAb is higher than that of ICA and IAA. Conclusion These results suggest that 64KDAb's sensitivity is higher than ICA and IAA's,so 64KDAb is more valuable as the marker of predicting type 1 diabetes. The combined determination of ICA, 64KDAb and IAA might improve the value for predicting diabetes.
2.Molecular composition of Enzymolyzed Anchovy Soluble Peptide and its of Nutrition Evaluation

The peptides molecular compositions and the amino acids component of the enzymolyzed protein of anchovy soluble peptides hydrolysed by pepsin and trypsin were assayed and determined by gel HPLC and amino acid analytical instrument.The molecular weight of the peptides is less than 7400, of which 1.74 percent is a large peptide chain with composition of 52~58 amino acids and 6600~7400 molecular weight,29.75 percent is a middle large peptide with 20~41 amino acids and 2500~5300 molecular weight , and 50 percent is a oligo-peptide with 2~10 amino acids and less than 1000 molecular weight. The proportion of total nitrogen to rree amino acid nitrogen is 25.9:1 in the hydrolysates. 96 pecent of amino acid exists in form of peptides and 4 percent are free amino acids. The con tent of whole amino acids accounts for 73.98 percent soluble peptides, and the essential amino acid is 32.39 percent or 43.78 percent of the whole amino acids .Comparing whth FAO/WHO , the phenylalanine expresses in the first limited amino acid with the amino acid score of 61. Based on the analysis results, the balance of amino acid composition pf anchovy loluble peptides and the physiological function of loigo-peptide in an animal organism were explored.

Objective To investigate the clinical value of homocysteine (Hcy) ,cystatin C (Cys C) and high‐sensitivity C‐reactive protein (hs‐CRP) in atherosclerosis (AS) .Methods Totally 100 cases of newly diagnosed patients of AS were choosed as the ob‐servation group ,36 cases condition improved markedly of AS after treatment as the treatment group ,and 120 cases of normal medi‐cal groups as the control group ,the Hcy ,Cys C and hs‐CRP in all subjects were detected respectively .Results Hcy ,Cys C and hs‐CRP levels of observation group were significantly higher than those of normal groups ,and the difference was statistically signifi‐cant (P< 0 .05) .While the Cys C and hs‐CRP levels of treatment group were significantly lower than the those of observation group ,the difference was also statistically significant (P< 0 .05) .Conclusion The detection of Serum Hcy ,Cys C and hs‐CRP might be help to the diagnosis ,treatment evaluation and prognosis of AS .

Objective To investigate the C-reactive protein (CRP) concentrations in the pathogenesis of type 2 diabetes mellitus (DM) and its macrovascular complications. Methods Serum CRP levels were assayed by ELISA, which were determined in type 2 DM patients with or without macrovascular complication (88 and 64 cases respectively), non-DM patients presenting with macrovascular disease (72 cases), as well as impaired fasting glucose (IFG) (62 cases) or impaired glucose tolerance (IGT) (70 cases) patients and normal controls (80 subjects). Objective:To study the relationship between HLA-DRB1 gene polymorphisms and macrovascular complication of type 2 diabetics. Methods:HLA-DRB1 alleles in 88 cases of type 2 diabetics, including 52 ones without complication and 36 ones with macrovascular complication, were examined by PCR-SSP. Results:There were at least 11 HLA-DRB1 alleles in type 2 diabetics. The HLA-DRB1 *03 and HLA-DRB1 *09012 gene frequencies of type 2 diabetics with macrovascular complication were significant higher than those without complication.Conclusion:HLA-DRB1 *03 and HLA-DRB1 *09012 alleles or their linkage disequilibrium genes may enhance the risk of the development of type 2 diabetes macrovascular complication.
8.The prevalence of metabolic syndrome in Heilongjiang residents aged 20-74 years

A cross-sectional study was conducted in Heilongjiang in a representative sample of 2 875Chinese adults aged from 20 to 74 years. Questionnaire was conducted. Their height, weight, waist circumference,blood pressure, blood glucose, serum insulin, lipids, and other indexes were determined. The criterion of International Diabetes Federation was applied for metabolic syndrome (MS) diagnosis. The results showed that the preliminary prevalence rate of MS was 19.34% and the age-adjusted prevalence rate of MS was 21.92% ( male 20.41% , female 23.11% ). The prevalence rate of MS increased with the age. The prevalence rate in the urban area was higher than that in the town and rural area ( both P < 0.05 ). The result of multiple factor logistic regression analysis showed that body fat content, insulin resistance, age, residential difference, drinking habit,daily staple food quantity, urine albumin, education, family history of obesity, and gender were related with MS.
9.Clinical Observation of Alteplase Intravenous Thrombolysis in the Treatment of Acute Cerebral Infarction

OBJECTIVE:To observe the bleeding,therapeutic efficacy and ADR of patients with acute cerebral infarction (ACI) treated by alteplase intravenous thrombolysis. METHODS:140 ACI patients were randomly divided into group A and B with 70 cases in each group. Group A was give alteplase 0.6 mg/kg for intravenous thrombolytic therapy,and group B was given al-teplase 0.9 mg/kg for intravenous thrombolytic therapy. The bleeding after thrombolysis,bleeding time,therapeutic efficacy and ADR were compared between 2 groups. RESULTS:The incidence of Subcutaneous ecchymosis,gingival bleeding,bleeding of di-gestive tract and intracranial hemorrhage of group A were 4.29%,2.86%,2.86% and 2.86%;those of group B were 14.29%, 12.86%,11.43% and 11.43%;the bleeding time of those symptomsin 2 groups were(6.04±0.75)and(7.22±0.56)h、(24.63± 10.24)and(35.22±9.87)min、(3.04±0.11)and(4.08±0.25)h、(3.12±0.48)和(4.53±0.66)h respectirely,with statistical signifi-cance(P<0.05). There was no statistical significance in therapeutic efficacy and ADR between 2 groups after treatment(P>0.05). CONCLUSIONS:The bleeding should be monitored strictly when ACI patients receive alteplase intravenous thrombolysis,and low-dose alteplase thrombolysis can ultimately reduce the incidence of bleeding.
10.Mechanism of Wendan decoction in preventing obesity by regulating multiple signal pathway networks based on gene promoter methylation

Objective:
To investigate the potential mechanism of Wendan decoction in obesity by screening target genes with promoter region methylation changes and constructing a multiple signaling pathways network based on promoter methylation.
Methods:
The methylation degree of Itgad, Col8a1, Adra2b, Jund, Rab2a, Wnt8b, Fzd9, B4galt7, Pik3cd, Creb1, Stard8, and Mmp1 in the abdominal adipose tissue of obese rats was determined using the Agena MassARRAY system. Western blot was performed to assess protein expression levels. Target genes were identified based on the methylation degree in the promoter region and protein expression. Enrichment analysis of signaling pathways was conducted to identify relevant target genes and obtain a multiple signaling pathway network associated with obesity. Core and terminal effector molecules in the pathway networks were selected as research targets for reverse transcription-polymerase chain reaction (RT-PCR) analysis.
Results:
Four genes (Adra2b, Creb1, Itgad, and Pik3cd) showed a degree of promoter methylation consistent with their respective protein expression levels. Among them, Adra2b, Creb1, and Pik3cd expression increased, while that of Itgad decreased. Enrichment analysis revealed that Creb1 and Pik3cd were involved in 6 signaling pathways related to obesity: tumor necrosis factor (TNF) signaling pathway, growth hormone synthesis/secretion and action, adenosine 5′-monophosphate-activated protein kinase (AMPK) signaling pathway, relaxin signaling pathway, cyclic nucleotide (cAMP) signaling pathway, and phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway. Subsequently, a multiple signaling pathways network was constructed based on promoter methylation. Key molecules including protein kinase B (AKT), mechanistic target of rapamycin complex 1 (mTORC1), and unc-51 like autophagy activating kinase 1 (ULK1), as well as terminal effector molecules interleukin-1β (IL-1β), interleukin-6 (IL-6), and chemokine (C-X-C motif) ligand 2 (CXCL2) were selected as research targets. Wendan decoction decreased the expressions of AKT, mTORC1, IL-1β, IL-6, and CXCL2 while up-regulating ULK1 expression.
Conclusion
The mechanism of Wendan decoction in preventing obesity involves the regulation of multiple signaling pathways through the control of Creb1 and Pik3cd gene promoter methylation. However, the associated multi-path gene regulation mechanism in preventing obesity is complex. Thus, further exploration is needed to elucidate the role of methylation changes in this mechanism.
