1.Changes of microcirculation in congested flap

Objective To explore the change patterns of microcirculation in congested flap. Methods A congested flap model for long-term microeirculation observation was set up, and the mi-crocireulation of the congested tissues was observed carefully. Results Following congestion, the number of the capillaries in the congested flap were decreasing, the velocity of the bloodstreams were getting slower, and the aggregation of the red blood cells and the formation of the white thrombus were becoming more serious. About 72 hours later, the original microcirculation system of the flap was nearly exhausted. About 36 hours later, the neogenetie capillaries could be observed, with clear neogenetie capillaries observed at 48 hours. The congested tissues with nascent capillaries survived, while the capillaries without nascent formation suffered from necrosis in the end. Conclusions If the cause is not removed, the microcirculation status of the congested flap would get worse irreversibly, and the nascent capillaries would play an important role in the survival of congested tissues.
2.Different gene expressions in human periodontal cells after being treated with Emdogain

Objective:To study the different gene expressions in human periodontal cells after being treated with Emdogain(EMD). Methods: The fourth passage periodontal cells were cultured. Cells in experimental group were cultured with 100 μg/ml EMD in non-serum DMEM for 3 days. Cells in control group were cultured in non-serum DMEM for 3 days. The gene expressions in human periodontal cells were screened with gene chip technique. Results: There were 112 differently expressed genes, 11 down-regulated genes and 101 up-regulated genes. Conclusion: After treated with Emdogain, expression of genes related to cell growth, cell proliferation, biosynthesis of extracellular matrix, angiogenesis and osteogenesis is up-regulated dramatically. Genes related to lipid metabolization and keratinocyte growth is down-regulated in human periodontal cells.
3.Interleukin-8 content in GCF and its relationship with subgingival Porphyromo nas gingivails in patients with chronic periodontitis

Objective:To investigate the levels of IL-8 i n GCF in patients with chronic periodontitis and their relationship with subgingi val Porphyromonas gingivalis(Pg).Methods:10 GCF samples from periodontal healthy volunteers, 60 from the teeth with chronic periodontiti s in patients and 10 from the periodontal healthy teeth in the patients with chr onic periodontitis were collected. Subgingival plaque samples and clinical data on GI, PPD and CAL were obtained. A sensitive ELISA was used to measure IL-8 in GCF and a PCR method was employed to measure the amount of Pg.Resul ts:①Higher total amount of IL-8 in GCF was found in chronic periodonti tis subjects than that in healthy volunteers(P0.05). ②There was positive correlation between the total amount of IL-8 and clinical parameters (GI, PPD, CAL).③There was no correlation between the amount of Pg and the concentration or total amount of IL-8. Conclusions:I L-8 increases with the increace of GCF in the patients with chronic periodontit is, but is not related to Pg in subgingival plaque.

5.Inhibitory effect of antisense human telomerase RNA(hTR) gene on hepatocellular carcinoma cells

Objective: To study the inhibitory effect of retrovirus-mediated antisense human telomerase RNA (hTR) gene on hepatocelluar carcinoma, so as to explore an effective way to inhibit telomrerase activity in the treatment of hepatocellular carcinoma. Methods: Sense and antisense hTR gene were transfected into the packaging cell line PT67 by electroporation, and the stablely transfected cells were selected with G418. The recombinant retroviral supernatant was collected and transfected into HepG2 cells. After G418 selection, PCR was used to verify the integration of the hTR gene. Cell growth curves were drawn using MTT assay and the expression of PCNA was determined by immunofluorescence. TRAP-PCR-ELISA was adopted to detect the telomerase activity; cell cycle and apoptosis were evaluated by flow cytometry (FCM).Results: The expression of hTR gene could be amplified in HepG2-hTR-EcoRⅠ and HepG2-hTR-BamHⅠ cells, but not in untransfected HepG2 cells. The antisense hTR complementary to the template region of telomerase inhibited growth and proliferation of HepG2 cells. The expression of neutrophil proliferating cell nuclear antigen (PCNA) was decreased. Objective: To investigate the relationship between TIMP-1 gene polymorphisms and susceptibility of chronic periodontitis(CP) in Han Cantonese. Methods: Buccal swabs from 70 CP patients and 74 periodontal healthy controls were collected. DNA was extracted from these buccal swabs by using Chelex-100. TIMP-1 +372T/C (rs4898)、TIMP-1+533C/T (rs1062849) polymorphisms were tested by polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP). Allele distribution and genotypes frequencies in the patients and controls were analyzed. Results: Frequency variance of allele T and C at site of TIMP-1 +372 in patients and controls showed no statistical difference. TIMP-1+533C/T polymorphism of TIMP-1 hadn't been found in the present study. Conclusion: There is no relationship between TIMP-1 +372T/C polymorphism and susceptibility of chronic periodontitis, and TIMP-1 +533C/T polymorphism doesn' t exist among Han Cantonese.
7.The application of the Delphi method in screening indicator system of assessing sub-health

Objective To screen the indicator system of sub-health.Methods Three rounds delphi consultations were conducted for selected indicators of sub-health.30 experts were selected for the three rounds' consultations.Results The experts who had been invited are from about 10 different majors and have been engaged in their current profession for 21.75 years on average.The enthusiasm coefficient of specialists in pretest and two rounds' formal consultations were 90.00%, 93.33% and 89.29% respectively.The coordination coefficients Kendall' W of the experts' opinion were 0.327,0.185 and 0.249 respectively.The weight of the first grade indexes of the sub-health namely physical, mental and social sub-health indicator dimension were 40.61%, 33.34%and 26.05% respectively.After two rounds' formal consultation,64 assessing indicators were remained in the final.Conclusion It is practical,efficient and successful to use Delphi method in screening indicator system of assessing sub-health.
8.Clinical Application of Detecting Lung Cancer based on PET/CT

Objective To research the validity of PET/CT in clinic for detecting lung cancer. Method There were 20 subjects who may have lung cancer according to the result form other detecting way, including 15 males and 5 females with an average age from 54 to 86. Results Malignant lesions were found in 15 cases and benign lesions in 5 cases. Discussion The examination of chest with PET/CT is suit for early diagnosis of lung cancer. The number of patient of false postive and false negative is decreased by controlling some factor.
9.Study on bioavailability of domestic acyclovir chewable tablets

Aim To compare the relative bioavailabilities of domestic acyclovir chewabletablets and normal tablets. Methods A single oral dose of 800 mg acyclovir chewable tablets or normal tablets was given to 8 volunteers respectively in a randomized, cross-over study, acyclovir serum concentration was determined by high performance liquid chromatography. Results The AUC of chewable and normal tablets was respectively 0.42 and 0.40 μg · ml · h-1, and the relative bioavailability of a cyclovir chewable tablets was (105.8 + 13.1) %. Conclusion The chewable tablets and reference tablets is bioequivalent in AUC.

Objective To compare the performance of Lung Imaging Reporting and Date System (Lung-RADS)with National Lung Screening Trail (NLST)in pulmonary nodules screening.Methods 1 5 3 patients with pulmonary nodules were analyzed retro-spectively,including,baseline screening and after baseline screening.We compared the sensitivity,false-positive result rate,positive predictive value,and negative predictive value in two methods of screening.Results In baseline screening of all patients with pulmo-nary nodules,the sensitivity,false positive rate and positive predictive value and negative predictive value of Lung-RADS and NLST were 83.9%,13.2%,6.5%,99.7% and 92.5%,26.9%,3.8%,99.8% respectively.In after baseline screening,the sensitivity, false positive rate and positive predictive value and negative predictive value were 79.6%,5.5%,11.5%,99.8% and 93.5%,22.8%, 3.5%,99.9% respectively.Conclusion Lung-RADS classification can reduce the false positive rate in screening of pulmonary nod-ules,which reduced the screening of lower risk nodules.
