1.The changes of oxygen free radicals in isoproterenol-induced myocardial infarction and its significance

The MDA contents, SOD and. GSH-Px activities of the heart, liver, brain, kidney and serum were observed systematically in isoprotercnol-induced myocardial infarction. The results suggested that the MDA contents, SOD and GSH-Px activities were increased in ischemic myocardium. Oxygen free radicals exerted detrimental effects on vitals organs, and also was accompanied by a compensatory increase of SOD and GSH-Px activities.
2.An experimental study of myocardial membrane injury in isoproterenol-induced myocardial infarction and the modulative effects of Svate

Changes in lipids, enzymes and free-radical system of myocardial membranes were investigated in isoproterenol-induced myocardial infarction so as to study the modulative effects of Svate on myocardial membrane injury. Animals were killed 24h after ISP(85mg/kg) injection. The results suggested that disturbances in lipids metabolism, enzymes and free-radical system in ischemic myocardium, indicating the existence of myocardial membrane injury. In the Svate(0.25U/kg)-treated group, a beneficial effect of Svate exerted on membrane injury, as indicated by increased activities of GSH-Px, SOD and Na~+K~+-ATPase, and decreased Ch/PL ratio, MDA contents and Ca~(2+)-Mg~(2+)-ATPase activities.
3.Rat mesenchymal stem cells differentiate into neuron-like cells with adrenaline hormones

AIM:To investigate the differentiation from rat mesenchymal stem cells (rMSC) into neuron-like cells. METHODS:rMSC were separated from femur marrow and expanded in L-DMEM culture medium supplemented with 10% FSC. rMSC were induced to differentiate into neurons with L-DMEM/adrenaline,L-DMEM/noradrenaline and L-DMEM/isoprenaline, respectively. Meanwhile, rMSC were cultured in L-DMEM in control group. Nestin, neuron-specific enclose (NSE), glial fibrillary acidic protein (GFAP) were detected by immunocytochemistry. RESULTS: rMSC were expanded as undifferentiated cells in culture from 5 to 22 passages, indicating their differentiated capacity. Simple method induced rMSC to exhibit a neuronal phenotype, expressing positive NSE,nestin, and GFAP, at 5 hours in all group. The undifferentiating cells (control group 53.1%?4.3%), and differentiating cells (treated group: adrenaline 74.7%?2.6%; noradrenaline 75.9%?2.4%; isoprenaline 72.1%?4.4%), expressed characteristics of various neuronal cells, from 5 hours to 6 days. There were neuron-like cells in rMSC cultured in L-DMEM/10%FBS from 7 to 13 passage(66.5%?6.4%). CONCLUSION: It suggests that rat neural stem cells (rNSC) exist in bone marrow, rMSC can be differentiated into various neural cells with adrenaline hormones in vitro.
4.Determination of astragloside Ⅳ in Danqi Injection by HPLC-ELSD

AIM: To establish a method for determination of astragloside Ⅳ in Danqi Injection (Radix Salviae Miltiorrhizae,Radix Astragali). METHODS:HPLC-ELSD with Hypersil C_(18) column was used.The mobile phase was composed of acetonitrile-water(38∶62).The flow rate was 0.85 mL?min~(-1).An evaporative light-scattering detector(ELSD) model 2000 was used as detector. RESULTS:For astragloside Ⅳ,the linear range was 1.074-(3.408 ?g),and the average recovery was 100.71%,RSD was 1.79%. CONCLUSION:The method is simple,accurate,reproducible and can be applied to the quality control of the preparation. anqi

BACKGROUND: By targeting inducing differentiation in vitro,mesenchymal.stem cells(MSCs) transform into osteoblasts,lipocytes,chondrocytes,muscular cells,neuronal cells,etc. Whether Chinese herbs act on induced differentiation of MSCs in rats or not?OBJECTIVE: To study the amplification of MSCs cultured in vitro in SD rats and efficacy of Chinese herbs on targeting inducing differentiation of neuron-like cells.DESIGN: Exploring study with repeated observation and measurement based on cells.SETTING: Department of pathophysiology in a medical college.MATERIALS: Experimental marrow collected from SD male tested-healthy rats.METHODS: By adhesion method,MSCs in rats were isolated for amplifying culture in vitro. Flow-type cell instrument was applied for the determination of its surface antigen expression. Various Chinese herbal components were used for the targeting inducing differentiation of MSCs into neuron-like cells. The cellular morphology was observed under optical microscope. and the specific antigen label of neuronal cells was determined with immunocyto-chemical method.MAIN OUTCOME MEASURES:①Results of MSCs isolation and amplification;②Results of identification of MSCs surface antigen and neuon-like cells.RESULTS: By adhesion method,MSCs in rats were isolated successfully and amplified in a large amount in vitro. It was indicated in the results determined by flow-type cell instrument that CD14,CD1 1α,CD34,CD38,CD45,CD80 and CD86 presented negative,and CD29,CD44,CD90,CD105 and CD166 presented positive. By induced with various kinds of Chinese herbs,like huangqi(Radix Astragali seu Hedysari),tianma(Rhizoma Gastrodiae),renshen (Radix Ginseng),danggui(Radix Angelicae Sinensis),naoxinshu,renshen fengwangjian for 1 to 3 hours,most MSCs transformed into neuron-like cells,presenting soma and neurite. With immunocyto-chemical staining,neuron-specific enolase(NSE) and nestin displayed positive and glial fibrillary acidic protein negative.CONCLUSION: MSCs in SD rats have the potential of multi-targeting differentiation,presenting a strong capacity of amplification and self-replacement. In a suitable inducing condition,MSCs may differentiate into neuron-like cells.
6. Application of valvuloplasty in mitral valve insufficiency in infants and small children

Objective:
To explore the efficacy of valvuloplasty on mitral valve regurgitation (MR) in infants and small children, in order to provide evidence for clinical treatment.
Methods:
Retrospective analysis was conducted in patients aged less than 3 years old who have undergone mitral repair surgeries at Department of Cardiovascular Surgery, General Hospital of Western Theater Command from January 2015 to December 2017.Cardiopulmonary bypass (CPB) surgery was performed under general anesthesia.The corresponding repair operations were selected for various MR types.All the children were examined by cardiac ultrasound before discharge.Patients were followed up after surgery and mitral regurgitation was recorded.
Results:
A total of 69 patients were enrolled, including 30 boys and 39 girls, with an average age of (19.3±11.6) months and an average weight of (9.6±2.1) kg.There were 32 cases of Carpentier type Ⅰ, 30 cases of type Ⅱ and 7 cases of type Ⅲ.Twenty-nine patients were diagnosed as simple MR, while the other 40 patients were combined with other cardiac malformations.One patient died at postoperative day 7.One patient still had severe MR after surgery, and then second operation was performed on the 10th day after first surgery.Sixty-five patients were followed up from 1 month to 3 years[(15.9±10.9) months]. During the follow-up period, no patient died or had reoperation.Aggravated MR was observed in 4 patients.
Conclusions
Mitral valvuloplasty is a safe and effective surgical method for treating infants and younger children with MR.The key to the success of surgery is to choose the appropriate individualized repair operation.At the same time, the growth potential of mitral valve should be fully considered to avoid restrained development.
7.Expression of autophagy-associated protein light chain 3B in human pulpitis and periapical periodontitis

BACKGROUND: Role of autophagy in pulpitis, periapical periodontitis and alveolar bone destruction and the underling mechanisms have not been fully elucidated. OBJECTIVE: To investigate the expression of autophagy-related protein light chain 3B in human pulpitis and periapical periodontitis, in order to explore the rule of autophagy. METHODS: The healthy first premolar or surgically removed third premolar was selected from the patients undergoing orthodontics after their consent informs. The dental pulp cells were treated with 1 mg/L lipopolysaccharide for 0, 6, 12, 24 and 48 hours to induce inflammation. The expression of light chain 3B in normal and lipopolysaccharide-stimulated dental pulp cells was detected by western blot assay. The dotted accumulation of light chain 3B was observed by immunofluorescence. The expression level of light chain 3B in specimens of human chronic periapical periodontitis tissues was tested by real-time RT-PCR. RESULTS AND CONCLUSION: The expression of light chain 3B in dental pulp cells stimulated by lipopolysaccharide for 6, 12, 24 and 48 hours was higher than that in normal dental pulp cells. The expression of light chain 3B increased with time, and peaked at 24 hours. Dotted accumulation of light chain 3B increased with lipopolysaccharide simulation time prolonged. The expression level of light chain 3B in specimens of human chronic periapical periodontitis tissues was higher than that in normal periapical tissues. These results indicate that the expression of light chain 3B is increased in human pulpitis and periapical periodontitis, and morphology also shows an increase of autophagy, suggesting that autophagy may be associated with the occurrence and development of pulpitis and periapical periodontitis.
8. Related factors analysis for lymph node metastasis in papillary thyroid carcinoma: a series of 2 073 patients

Objective:
To investigate the related factors for lymph node metastasis (LNM), especially for high volume LNM (>5 metastatic lymph nodes) in papillary thyroid carcinoma (PTC).
Methods:
The medical records of 2 073 consecutive PTC patients who underwent lobectomy, near-total thyroidectomy or total thyroidectomy with ipsilateral or bilateral central lymph node dissection in Department of General Surgery, Peking Union Medical College Hospital from November 2013 to October 2014 were reviewed. Clinical and pathological features were collected. Univariate and multivariate analysis were performed to identify the related factors for LNM/high volume LNM.
Results:
In all 2 073 patients, LNM and high volume LNM were confirmed in 936 (45.15%) cases and 254 (12.25%) cases respectively. In univariate analysis, large tumor size, young patients (<40 years), male were associated with both LNM and high volume LNM. In multivariate analysis, tumor size >2.0 cm, young patients (<40 years), male were independent related factors of LNM (

Objective To summarize clinicopathologic features of papillary thyroid carcinoma (PTC) coexistent with chronic lymphocytic thyroiditis (CLT) and investigate risk factors for lymph node metastasis.Methods The medical records of 4 264 consecutive papillary thyroid carcinoma patients who received surgical treatment from Oct 2013 to Oct 2015 in Peking Union Medical College Hospital were reviewed.The diagnoses was confirmed by histopathological tests.Univariate analysis was performed to identify specific clinicopathologic features of PTC with CLT.Univariate and multivariate analysis were performed to determine whether each clinicopathologic feature was an independent risk factor for lymph node metastasis.Results In all 4 265 cases,there were 3 059 papillary thyroid microcarcinoma (PTMC) (71.7%),1 010 PTC patients (23.7%) with CLT.909 female patients (90%),624 cases with multifocal lesions (61.8%),422 cases with extra-thyroid extension (41.8%),429 cases with lymph node metastasis (42.5%),and 133 cases with metastatic lymph nodes(LNs) ≥6 (13.2%).The median age was 43 years old and median tumor size was 0.8 cm.Patients with CLT were more females (90.0% vs.70.2%;P < 0.001),younger median age (43 vs.44 years;P =0.001),and lower incidence of lymph node metastasis (42.5% vs.50.9%;P <0.001).CLT was not associated with tumor size,multifocal lesions,extra-thyroid extension and metastatic LNs≥6 (0.8 cm vs.0.7 cm,61.8% vs.62.9%,41.8% vs.42.1% and 13.2% vs.14.8%,respectively,all P > 0.05).In multivariate analysis,CLT was an independent protective factor for lymph node metastasis (OR =0.713,95% CI 0.609-0.835,P <0.001).In PTC patients with lymph node metastasis,CLT was not associated with lymph node metastasis number (3 vs.3,P =0.300).Conclusions Chronic lymphocytic thyroiditis was an independent protective factor for papillary thyroid carcinoma patients with lymph node metastasis.But in patients with lymph node metastasis,the metastatic number didn't decrease.
10.Retrospective study of the diagnosis and treatment of culture-negative aortic infective endocarditis

Objective To discuss the diagnosis and treatment of culture-negative aortic infective endocarditis. Methods The clinical data of 73 patients with infective endocarditis of the aortic valve whose results of bacteria culture were negative from January 2013 to January 2018 were retrospectively analyzed, including 59 males and 14 females aged 14-71 (39.2±14.8) years. Results Sixty seven (91.8%) patients received aortic valve replacement, 2 (2.7%) patients received the second operation in hospital, and 12 (16.4%) patients had concomitant mitral valvuloplasty. In-hospital death occurred in 8 (11.0%) patients. Postoperatively, 11 (20.7%) patients had a low cardiac output and 4 (11.0%) patients had heart block, and 1 patient required implantation of a permanent pacemaker. The 1- and 5- year survival rates were 92.3%±2.3% and 84.5%±4.5%, respectively. Conclusion There are difficulties in the diagnosis and treatment of culture-negative infective endocarditis. Most of the affected patients are in a healed status, which could be a cause of negative culture results. In-hospital mortality in the patients is associated with a history of previous cardiac surgery, whereas the long-term survival rate is good for the patients after surgery.
