1.HPLC Simultaneous Determination of Sodium Danshensu, Hesperidin and Salvianolic ;Acid B in Niaosaitong Capsules

Objectives To establish an HPLC method for determination of sodium Danshensu, hesperidin and salvianolic acid B in Niaosaitong capsules. Methods Three components were separated by C18 chromatographic column with a gradient mobile phase consisting of acetonitrile and 0.1%phosphoric acid solution at a flow rate of 1.0 mL/min, column temperature of 30 ℃, and detection wavelength was set at 286 nm. Results Sodium Danshensu, hesperidin and salvianolic acid B had good linearity in the range of 0.038 44-1.922 μg, 0.000 207 052 8-4.044 μg, 0.034 28-1.714 μg respectively, and the average recovery rate was 90.9%-107.1%, RSD was 2.1%-3.2%. Conclusion The methods is simple and accurate, and can be used for the quality control of Niaosaitong capsules.
2.Protective effect of Anisodamine on ionizing radiation damage of cochlea

Objective To explore protective effect of Anisodamine against injure of cochlea hair cells induced by ionizing radiation. Methods 50 healthy guinea pigs were randomly divided into 3groups: radiation group( n = 20) , anisodamine protective group( n = 20) and control group( n = 10). 20ears were observed in each group. Tempora of guinea pigs in anisodamine protective group and radiation group were exposed to fractional doses of 6 MeV electron beam produced by linear accelerators (2 Gy/d)with a total dose of 60 Gy. Animals were injected intramuscularly with anisodamine (20 mg/kg) in protection group and equal saline solution in irradiated group, respectively, 30 min pre-irradiation.Morphological and functional changes in the guinea pig cochlea were observed. Results After radiation,the average ABR threshold value of the radiation group (52. 27 ± 2.42 dB peSPL) was significantly higher than that of the anisodamine protection group (37.65 ± 1.92 dB peSPL, t = 2.01, P< 0.05 ), and outer fair cells of cochlea reduced greatly in quantity by observing the stretched preparations of cochlea basilar membrane ( F=135. 362, P< 0.05 ). Under transmission electron microscope, outer fair cells of cochlea in the radiation group had obscure boundaries and swelled to lose their original shapes. Meanwhile, cell implement and the nucleus were obviously obnormal after radiation. In the anisodamine protection group,the boundaries of cochlea outer fair cells were very clear and the cells swelled a little. Cell implement and nuclear envelope were complete. Cochlea outer fair cells in the control group were found to line up in order with no collapse and reduction in number under scanning electron microscope, but those in the radiation group collapsed obviously, reduced in number and aligned in a state of chaos, while the outer fair cells in the anisodamine protection group regularly posed on the whole and the collapse was occasionally found.Conclusions The 60 Gy radiation at fractioned dose on the tempuses of the guinea pigs could cause damage to the cochlea hair cells. Anisodamine might have a protective effect against radiation damage.
3.Treatment of senile diseases from spleen in TCM

It is discussed in this paper the origins and pathogenesis of treating senile disease from spleen in TCM,and the relationship between various methods of invigorating the spleen and senile deficiency syndromes.It has been proposed that the therapy principles of invigorating qi and spleen,stimulating the appetite to awake the splee,primering blood to go back to the spleen,dehumidification to activate spleen-energy,warming the kidney and spleen,dispersing the depressed liver-energy to belifit the spleen,going on dieting to belifit the spleen and clearing away heart-fire to support spleen etc.are used to prevent and cure senile diseases.It also listed the corresponding treatment prescriptions for the elderly patients.It provides a theoretical basis for treatment of senile diseases from the governance of spleen.And it provided a realistic basis and the implementation of the method for the prevention and treatment of elderly patient's deficiency aging.
4.Effect of percutaneous coronary Intervention on coronary artery disease with complicated lesions

Objective To assess the clinical efficiency of percutaneous coronary intervention (PCI) on cor-onary artery disease patients with complicated lesions (CAD-CL). Methods A total of 252 patients of CAD-CL treated with PCI from August,2002 to September,2009 were enrolled in the study. Clinical data on prognosis e. g, success rate, complication occurrence and major adverse cardiac events were observed. Results Two hundred and forty-four patients with CAD-CL(including chronic obstruction, bifurcation lesion, long lesion and restenosis) were implanted stent successfully(96.83%), with no residual stenosis or residual stenosis less than 20%. There were col-lateral obstructions in 32 patients and artery secret compartment in 48 patients, and restenosis in 15 patients. One patient of acute anterior myocardial infarction complicated by cardingenic shock was in low blood pressure status after two anterior descending artery stent placement, and died after temporary pacemaker implantation and other positive rescue measures. One patient occurred subacute stent thrombosis on the third day, and was cured after occlusive vas-cular recanalization through emergency PCI. Conclusions Percutaneous coronary intervention for coronary artery disease patients with comphcated lesion is safe and effective.
5.Effect of Ammonium Metavanadate on Liver Ultramicro-Structure in Rats

Objective To understand the effect of vanadium to liver ultramicro structure in SD rats.Methods One hundred and fifty generaMevel SD rats of 2 months were randomly divided into control groups,30 in each,the males and females were equivalent,treated with ammonium metavanadate at doses of 0,10,20,40,60 mg/L through drinking water.After 2,6 and 10 weeks of treatment,6 rats from each group were weighed and the changes of ultramicro-structure in the liver were examined with electron microscope.Results As the increasing of dose,the increase in body weight showed a downward trend,but the coefficient of liver showed a increasing trend,the consistent performance was seen in male and female rat.After two weeks of exposure,the rats acted,ate and drank less,hair roughing and no shining,often getting together were seen.After 10 weeks,the rats had not the basic activities,the hair was disheveled and loose,some died,the structure of mitochondria and endoplasmic reticulum was normal in the 10,20,40 mg/L group,the number of endoplasmic reticulum increased,in 60 mg/L group,the rough endoplasmic reticulum vanished,the liver cytoplasm had a swelling phenomenon,the mitochondria reduced.Conclusion Vanadium have little effect on the liver cells of rats in low exposure level,but high level exposure may damage the liver cells in rats.
6.Decreased insulin sensitivity in rat hepatocytes with intrauterine growth retardation and establishment of insulin resistance cell model in vitro

Objective:To explore the hepatocyte insulin sensitivity of intrauterine growth retardation ( IUGR) rats and establish an insulin resistance cell model in vitro.Methods: An IUGR animal model was established by protein malnutrition during the mother pregnancy .On 60 d and 90 d after birth , the offspring rats were fasted for 12 hours and then their angular vein blood was collected to measure the fasting plasma glucose and fasting serum insulin level , then the insulin resistance index ( HOMA-IR) and insulin sensitivity index ( ISI) were calculated .The insulin sensitivity was evaluated by HOMA-IR and ISI.Primary hepatocytes from each group were respectively isolated by two-step perfusion with collage-nase and were defined as normal hepatocytes group and IUGR hepatocytes group .The normal hepatocyte group was divided into two groups: control group and insulin induction group .Insulin induction group was established by primary cultures of normal hepatocyte incubated with varying dilutions of insulin . CCK-8 was used to detect the viability of the cultured hepatocytes .Glucose oxidase-peroxidase method kit was used to measure glucose consumption of the hepatocytes .Results:HOMA-IR was significantly higher in IUGR rats than in the normal rats at the age of 60 days ( t=-17 .02 , P<0 .05 ) and 90 days ( t=-12.52, P<0.05).ISI was significantly lower than in the normal rats aged 60 days (t=5.61, P<0.05) and 90 days (t=12.42, P<0.05).There were no significant differences in hepatocyte viability among the control group , IUGR group and insulin induction group after incubation of 48 h on day 60 (F=1.34, P=0.29) and day 90 (F=0.22, P=0.81).The glucose consumption of the IUGR group and insulin induction group were significantly decreased compared with the control group on day 60 ( F=9.28, P=0.002) and day 90 (F=56.60, P<0.001), while there was no significant difference be-tween the IUGR group and insulin induction group (P=0.08, P=0.10).Conclusion:The insulin sen-sitivity of hepatocytes of IUGR rats decreased from adolescence to adulthood .High-dilution insulin may induce insulin resistance cell model in vitro.
7.Silence of Wnt5a gene inhibits migration and invasion of lung squamous carcinoma cells

Objective:To study the influence of Wnt5a gene silence on the proliferation,migration and invasion of lung squamous carcinoma cells.Methods:A recombinant plasmid pHI-siRNA~(Wnt5a-)was constructed and used to deliver small interference RNA (siRNA)targeting Wnt5a in SK-MES-1 cells;the transfected cells were screened to establish a stable transgenic cell line.MTT,cell cycle and Transwell assays were employed to evaluate the effect of Wnt5a gene silence on the proliferation,migration and invasion of lung squamous carcinoma cells.Results:Western blotting assay revealed that Wnt5a was lowly expressed in SK-MES-1~(Wnt5a-)(13.6%).The proliferation index(PI)of transgenic cell line was slightly lower than that of the control cell line([28.3?3.8]% vs[30.5?5.2]%). The migration and invasion capabilities of SK-MES-1~(Wnt5a-)cells were(47.3?9.2)% and(39.7?11.7)% of the control cells, respectively.Conclusion:Low Wnt5a expression can significantly inhibit the migration and invasion capabilities of SK-MES-1 cells, indicating that Wnt5a might be a potential target for gene therapy of lung squamous carcinoma.

The first research on the NS1 protein as the non-structural protein of the type A influenza virus was emphasized on the considering of its depressant effect on the composition of the protein of host cell.And now,with deep research,the evolution of its genes and the antigenicity of its protein have been explained.The NS1 protein of influenza virus has the association with the apoptosis induced by the influenza virus,the regulating function of apoptosis has the direct correlation with possibility of producing interference and the cell line infected by influenza virus.The NS1 protein restrained to producing the interferon by infected cell.The NS1 protein plays an important role on the host anti-viral cytokine responses,it possesses nagitive regulation for interferon's antiviral activity,most observation indicated nagitive regulation might associate with the virulence of influenza virus.Furthermore NS1 protein as an inspection antigen to differentiate and diagnose the poultry which was immunized or naturally infected has a very wide prospect,because the traditional vaccine was used extensively.
9.Roles of phosphatidylinositol 3-kinase/protein kinase B signaling pathway in skeletal muscle, peroxisome proliferator-activated receptor γ and phosphatase and tension homologue deleted on chromosome 10 in regulating insulin sensitivity of rats with fetal growth restriction

Objective To investigate the roles of phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT) signaling pathway and its regulatory protein peroxisome proliferator-activated receptor γ (PPAR γ) and phosphatase and tension homologue deleted on chromosome 10 (PTEN) in regulating insulin sensitivity in rats with fetal growth restriction (FGR).Methods Sixteen pregnant rats were randomly divided into two groups including FGR and control groups on the 12th day of pregnancy (eight in each group).The FGR group was given low protein diet (8% of casein) and restriction diet to establish the neonatal rat model of FGR.All maternal rats after delivery and newborn rats after weaning on 21 days after born were fed with normal diet.Each time blood samples were collected from eight newborn rats of each group to measure levels of fasting plasma glucose (FPG) and fasting insulin(FINS) at the time points of 21 days,two and four months after birth.Then insulin resistance index (IRI) and insulin sensitivity index (ISI) were calculated to evaluate insulin sensitivity.Expression of PI3K,AKT,PPAR γγ,PTEN and glucose transporters 4 (GLUT4) in skeletal muscle at mRNA and protein levels were measured at 21 days,two and four months after birth with real time fluorescence polymerase chain reaction and Western blot,respectively.Relationships between the expression of key molecules of PI3K/AKT signaling pathway and insulin sensitivity were analyzed.T-test,and Pearson's correlation analysis were used for statistical analysis.Results (1) The average birth weight of newborn rats in the FGR group was lower than that of the control group [(4.37± 0.69) vs (7.03±0.55) g,t=-20.75,P<0.05].The incidence of FGR in the FGR group was 93.33% (70/75).(2) Compared with normal offspring,those in the FGR group showed significantly increased FPG [two months after birth:(5.53± 0.58) vs (7.49 ± 0.38) mmol/L,t=8.08;four months afterbirth:(6.35±0.66) vs (8.94±0.90) mmol/L,t=6.58],FINS [two months afterbirth:(9.18±0.66) vs (14.67± 1.90) mU/L,t=7.71;four months after birth:(33.08±2.76) vs (56.33±2.81) mU/L,t=16.71] and IR1 (two months after birth:2.25±0.31 vs 4.90±0.81,t=8.63;four months after birth:9.30±0.90 vs 22.44±3.10,t=1 1.51),but decreased ISI (two months after birth:0.020 ± 0.002 vs 0.009± 0.001,t=-10.1 4;four months after birth:0.005±0.000 vs 0.002 ±0.000,t=-14.91) at two and four months after birth (all P<0.05).(3) Compared with normal offspring,those in the FGR group showed decreased expression of PI3K (21 days after birth:0.082±0.028 vs 0.019±0.004,t=-6.29;two months after birth:0.020±0.003 vs 0.010±0.005,t=-4.78;four months after birth:0.014±0.004 vs 0.003±0.001,t=-7.87) and GLUT4 (21 days after birth:0.132±0.057 vs 0.041 ±0.019,t=-4.32;two months after birth:0.183±0.084 vs 0.069±0.017,t=-3.74;four months after birth:0.248±0.069 vs 0.113±0.040,t=-4.74) at mRNA level at 21 days,two and four months after birth (all P<0.05).Compared with normal offspring,decreased expression of PPAR γ (two months after birth:0.028±0.002 vs 0.012±0.005,t=-3.70;four months after birth:0.030±0.008 vs 0.012±0.005,t=-3.80) and increased expression of PTEN (two months after birth:0.020±0.004 vs 0.045±0.014,t=5.09;four months after birth:0.023±0.007 vs 0.034±0.009,t=2.57) at mRNA level were observed in offspring of the FGR group at two and four months after birth (all P<0.05).(4) Compared with normal offspring,expression of PI3K protein (21 days after birth:0.22±0.01 vs 0.17±0.02,t=-6.62;two months after birth:0.27±0.03 vs 0.16±0.02,t=-7.25;four months after birth:0.18±0.01 vs 0.09±0.02,t=-9.79) and GLUT4 protein (21 days after birth:0.21 ±0.01 vs 0.03±0.01,t=-27.29;two months after birth:0.10±0.01 vs 0.06t±0.01,t=-3.90;four months after birth:0.13 ±0.01 vs 0.08± 0.02,t=-8.10) decreased in offspring in the FGR group at 21 days,two and four months after birth (all P<0.05).Compared with normal offspring,those in the FGR group showed decreased expression of PPAR γ protein (two months after birth:0.10 ± 0.01 vs 0.07± 0.01,t =-7.29;four months after birth:0.09±0.01 vs 0.08±0.01,t=-2.83),but increased expression of PTEN at protein level (two months after birth:0.10±0.01 vs 0.15±0.02,t=6.01;four months after birth:0.09±±0.01 vs 0.13±0.02,t=5.51) at two and four months after birth (all P<0.05).(5) The IRI levels in offsprings in the FGR group were negatively correlated with the expression of PI3K,GLUT4 and PPAR γ at protein level (two months after birth:r=-0.90,-0.92 and-0.79;four months after birth:r=-0.92,-0.75 and-0.73,all P<0.05),but positively correlated with the expression of PTEN at protein level (r=0.87 and 0.86,both P<0.05) at two and four months after birth.Conclusions The abnormal expression of the key molecules of PI3K/AKT signaling pathway precedes the decrease of insulin sensitivity in newborn rats with FGR and the expression regulatory protein PPAR γ and PTEN are also changed,suggesting that these molecules may induce the impairment of insulin sensitivity in rats with FGR and be involved in the development of insulin resistance.
10.Errors analysis of prone position in intensity modulated radiation therapy of cervical cancer

Objective To study the spatial distribution of set-up errors for cervical cancer with intensity modulated radiation therapy (IMRT) and to provide referential safety margin out of clinical tumor volume (CTV) during treatment plan design. Methods Six patients with cervical cancer were treated with IMRT in prone position, belly board and thermoplastic cast was used for immobilization. Measurement were made on a daily basis setup under five consecutive treatments with electron portal images device (EPID).Portal films from two projection (one anter-posterior and one opposite lateral)were taken. Sixty portal films were analyzed. The translational and rotational deviations were analyzed by registering and comparing the bony structures of EPID and digitally reconstructed radiographs (DRR). Results The translational deviations were (3.1 ±1.8) mm, (3.9 ±3.3) mm, (4.2 ±2.6) mm in medi-lateral, cranio-caudal and anterior-posterior directions, the rotational deviations were in coronal plane (0.8±0.9)° and sagittal plane (1.2±1)°. Conclusion For the patients with cervical cancer undergoing IMRT, the margins between the CTV and PTV should be 7.1 mm in lateral direction, 10.4 mm in cranio-caudal and 10.8 mm in anterior-posterior directions. The sign on patients body can help to reduce the setup errors.
