The response of murine lymphoreticular tissue with Special reference to regional lymph node after implantation of Ehrlich ascites carcinoma cells was investigated histologically and ultrastructurally. The tumor induced a vigorous histological response in both regional and nonregional lymph nodes that was proliferation of immunoblasts with enlargement of paracortical area in early stage and enlargement, fusion of germinal centers with an increased number of plasma cells in the medullary cords during the late stage of tumor growth.The results suggested that the response of lymph nodes might be mainly related to cell-mediated immune mechanism in the early stage and humoral immunity during the late stage of tumor growth. Regional lymph node gave reaction earlier and more vigorously in participating cell-mediated immunity, nonregional lymph node might be more significantly involved in humoral immunity. It was observed ultrastructurally that sinus histiocyte engulfed tumor cell in which the cooperation with lymphocyte might be necessary. The experimental results indicated that the increased number of immunoblast in paracortex may be considered as an indicator for antitumor cellmediated immunity with an important prognostic significance for host bearing tumor.
2.Screening of High-yield and High-oil Curcuma kwangsiensis

OBJECTIVE:To establish a method for analyzing the volatile oil of high-yield and high-oil Curcuma kwangsiensis, and to provide reference for its breeding. METHODS:GC-MS was performed on the column of HP-5MS quartz elastic capillary column,carrier gas was high purity helium(99.999%),flow rate was 1.0 ml/min,inlet temperature was 250 ℃,the initial tem-perature of column was 50 ℃(temperature programmed),split injection with split ratio of 10:1. Mass spectrometry conditions:ionization mode was the electron impact ion source,ionization energy was 70 eV,the ion source temperature was 230 ℃,the quadrupole temperature was 150 ℃, transmission line temperature was 280 ℃,the electron multiplier voltage was 1588 V,and mass scanning range was m/z 45 to 500. High-yield and high-oil germplasm of were screened from 100 different germplasms,the volatile oil composition of single plant and relative percentage contents of each composition were compared,SPSS 22.0 software was used for cluster analysis. RESULTS:Totally 10 high-yield and high-oil germplasm were screened,54 kinds of compounds were identified,the common compositions of 10 different germplasms were camphor,1-caryophyllene,γ-elemene,curcumene, gemma ketone and new curdione,most germplasms contained borneol,isoborneol,δ-elemene,germacrene and calamine. The C78,C104,purpose 2,volatile oil content and relative percentage contents of active compositions in volatile oil were high. The 10 high-yield and high-oil germplasms can be divided into 3 groups. CONCLUSIONS:The study basically clears the main chemicalcomposition of volatile oil of high-yield and high-oil C. kwangsiensis,C78,C104 and purpose 2 are the more excellent strains in high-yield and high-oil C. Kwangsiensis germplasm.

4.Simultaneous surgery in patients suffering from tumor combined with coronary artery disease

Objective:To investigate the possibility and feasibility of the simultaneous coronary artery bypass grafting (CABG) with tumor excision in patients suffering from tumor combined with coronary artery disease(CAD). Methods:From August 2000 to July 2006, ten patients who were suspected of tumor (four patients suffered from urinary system tumor, two digestive system tumor, and four pulmonary tumor) with coronary artery disease were successfully treated by simultaneous surgeries. Surgical incisions were chose by the different tumor position. All carried on the CABG before tumor excision except one case. The two cases had cardiopulmonary bypass CABG, the rest eight had off-pump CABG. Follow-up was conducted after operation. Results:The combined operations were satisfied. There was no in-hospital mortality, no postoperative hemorrhage, no myocardial ischemia or infarction, with few complications. Postoperative in-hospital stay was 10-34 days, average 22.4 days. All discharged in cure. One patient lost the follow-up. Nine patients were followed up from six months to seven years three months. Two patients died from metastasis, and the others survived with satisfied follow-up without cardiovascular events. Conclusion:The simultaneous coronary artery bypass grafting with tumor excision for patients who suffer from either CAD with benign or malignant tumor is safe and feasible with satisfied short-and long-term survival.
5.Secretory Expression of the Fusion Protein IFN?-HSA in Pichia pastoris

Overlapping PCR technology was employed to splice IFN?and HSA genes in vitro. The spliced gene was inserted into Pichia pastoris secretory vector pPIC9K. The IFN?-HSA gene was designed for secretory expression under the control of promoter AOX1 and Mat a signal peptide in pPIC9K. The recombinant plasmid pPIC9K/IFN?-HSA was linearized by restriction enzyme SalI and transformed into Pichia pastoris KM71 by electroporation. The recombinant strains identified by G418 selection and confirmed by PCR analysis were induced by methanol to express fusion protein IFNp-HSA. SDS-PAGE and Western blot analysis of the fusion protein showed that the expressed fusion protein IFNp-HSA with an apparent 90kDa molecular weight had the antigenicity of HSA. The specific activity of culture supernatant was about 640IU/ml assayed by the standard amiviral activity test on WISH cells challenged with VSV virus.
7.STUDIES OF THE ULTRASTRUCTURAL CHANGES OF HUMAN NASO-PHARYNGEAL CANCER CELLS INVADED INTO ORGAN IN ORGAN CULTURE BY FREEZE CRACKING TECHNIQUE AND SCANNING ELECTRON MICROSCOPY

The present experiment applied the freeze cracking technique and scanning electron microscopy for further studies of ultrastructural changes of cancer cells after they invaded into the organ. It was found that, after two days of cultivation, there were a lot of cancer ceils which adhered on to the surface of organ fragments. The cancer cells migrated on the surface of the organ and at the same time penetrated into the organ by stretching out their pseudopodia. After 3 days of cultivation, some cancer cells had already invaded into the organ. After invasion they connected target cells of the organ by different ways, but their filopodium is the important organelle for the connection and communication with those target cells.The cancer cells caught the projecting portion of surface of target cells by branches of filopodia forming a mosaic pattern with them. Sometimes the filopodia of cancer cells fused with the opposed surface of target cells by inserting into it. Some filopodia became short and thick inside the organ due probably to the limited distance between cancer cells and target cells when they were in close contact to each other. Once the tumor cell nests were formed in the organ, the tumor cells connected each other by stretching out pseudopodia from both poles, but kept a definite space between cells. It is postulated that these inter-cellular spaces were formed probably by the repelling force of the identical electric charge of the tumor cell membranes.
8.THE OBSERVATION OF THE CHANGED PROCESS OF SPECIFIC SURFACE STRUCTURE OF HUMAN NASOPHARYNGEAL CARCINOMA CELL LINE(CNE)IN VARIOUS TIMES OF CULTIVATION BY SCA NNING ELECTRON MICROSCOPE

CNE cells were cultured on cover glasses placed in the culture flasks, and were examined under scanning electron microscope 6,24, 48,72, and 96 hours after cultivation. The results were as following: The surface structures of tumor cells changed in different intervals of cultivation. Evenly distributed zeiotic blebs were found on the surface of tumor cell after 24 hours of cultivation and most of these blebs were seen at marginal areas of tumor cells after 2 days of cultivation, and finally disappeared after 3 days of cultivation. In addition to the blebs some microvilli which had increased since the second day of cultivation were seen over the margin of tumor cells. On the third day of cultivation a large number of microvilli were seen distributed over the entire surface of tumor cells. Besides microvilli and zeiotie blebs, lamellipodia and ruffles were also observed after 24 hours, but vanished after 48 hours of cultivation. The filopodia were seen over the whole process of cultivation. They usually divided into 2、3 branches and the expanded ends looked like suckers. All these data indicated that the surface structure of tumor cells were variable in different periods of cultivation.
9.Expression of tumor necrosis factor-alpha and receptor I (P55) in pterygium

AIM:To observe the expression of tumor necrosis factor-alpha ( TNF- α) and its receptor I ( P55 ) in different pterygium and discuss the role of TNF-α and receptor I (P55) in pterygium.
METHODS: Immunohistochemistical staining method ( PV) was adopted to detect the expression of TNF-α and receptor I in pterygium ( 72 eyes ) and para-pterygium conjunctival tissue ( 30 eyes ) . The relationship between the expression and clinical-pathological parameters was also analyzed.
RESULTS:The positive rates of TNF-αwere 65. 3% (47/72), 26. 7% (8/30) in pterygium and para-pterygium conjunctival tissue. The positive expression of TNF-α had statistic difference between the two groups (χ2=12. 706, P<0. 01). The positive rates of TNF-α receptor I were 56. 9% (41/72), 16. 7% (5/30) in pterygium and para-pterygium conjunctival tissue. The positive expression of P55 had statistic difference between the two groups (χ2=13. 875, P<0. 01). The positive rate of TNF-αin recurrent pterygium group was higher than primary pterygium group (χ2=6. 547, P=0. 011). There had no statistically significance of the expression intensity between the two groups (F=1. 288, P=0. 393); the positive rate in advanced pterygium group was higher than quiescent pterygium group (χ2=4. 082, P=0. 043). The expression intensity had no statistically significance between the two groups (F=0. 489, P=0. 708). The positive rate of P55 in recurrent pterygium group was higher than primary pterygium group (χ2 =9. 907, P= 0. 002). There had no statistically significance of the two group's expression intensity ( F = 1. 175, P = 0. 424 ); the positive rate in advanced pterygium group was higher than in quiescent pterygium group (χ2=11. 140, P=0. 001). The expression intensity had no statistically significance between the two groups (F=0. 665, P=0. 621).
CONCLUSION:The expression of TNF-α and P55 are changing according to the development of clinical staging and onset. The expression of TNF-α and P55 may be related to clinical classification, staging and patient's working conditions of pterygium. There has no significant difference expression intensity of TNF - α and P55 in clinical staging and onset of pterygium.
10.THE BEHAVIOR OF LYMPHATIC METASTASIS OF WALKER CARCINOSARCOMA 256 IN RATS

Walker carcinosarcoma 256 cells were injected intradermally into the medialaspect of right footpad of rats.After injection,the drained lymph nodes and overthe area of route of entry were examined microscopically in different period,inorder to determine the positive rate of appearance of cancer cells and metastasis inlymph nodes as well as other pathological changes.In the local area of inoculation cancer cells were noticed passing through spacesbetween the endothelial cells by ameboid movement into capillary lymph-vessels ormoved along with the lymph stream and get in to the capillary lymph-vessels fromtheir open ends.Individual or clumped cancer cells were seen in lymph-vesselsunder the epithelium and in the afferent lymph-vessels of lymph nodes.One hourafter the injection individual cancer cells were found first in the popliteal lymph nodes,then in the lumbar or renal lymph nodes on the third day.These cancer cellsappeared in the subcapsular sinuses at first,then in the intermediate and medullarysinuses subsequently and might be passed from one lymph node to another throughthe efferent lymph-vessels,for instance,cancer cells in popliteal nodes might migrateto lumbal or renal lymph nodes.When the cancer cells in lymph nodes became amass of a certain size associating with mitotic figures,a metastatic focus was thenestablished.Within twelve days after the inoculation,the total positive rates ofcancer cells and metastasis present in the popliteal lymph nodes were 59.5% and thetotal positive rate of multiple metastasis present simultaneously in several lymphnodes was 18.9 per sent.In addition to metastasis,these lymph nodes showed also themselves some im-mune changes,as including proliferation and enlargement of lymph follicles in cortex;widening of the paracortical area;ameboid movement present in different forms ofsmall lymphocytes and reticulosis,transformation of plasmacytes as well as increaseof immunoblasts.
