2.Effect of ABCCA gene silencing on cell proliferation and cell cycle in human pancreatic cancer cell lines PANC1 and BxPC-3

Objective To determine the effect of ABCC4 gene silencing on cell proliferation and cell cycle in human pancreatic cancer cell lines PANC1 and BxPC-3.Methods PANC1 and BxPC-3 pancreatic cancer cells were transfected with a lentivirus expressing an ABCCA short hairpin RNA (shRNA).ABCC4 mRNA and protein expression of transfected cells was determined by RT-PCR and Western blot,colony formation ability was measured by colony assay,and cell cycle change was investigated by the flow cytometric analysis.Results Lentivirus expressing an ABCC4 short hairpin RNA (shRNA) was successfully established.After transfection with shRNA lentivirus,ABCC4 mRNA and protein expression were significantly inhibited (0.28 ±0.01 vs 1.00 ±0.03,0.22 ±0.02 vs 1.00 ±0.03,P <0.05).Colony formation ability was significantly decreased (4 vs 65,P <0.05),and cell cycle was significantly blocked at G1 phase [(54.98 ±1.78) % vs (42.93 ± 0.88) %,(68.55 ± 0.75) % vs (54.76 ± 0.29) %].Conclusions ABCC4 gene silencing can significantly inhibit cell proliferation of human pancreatic cancer cell lines PANC1 and BxPC-3,and block the cells at G1 phase.
3.Study of Optimum Process of Microwave Technique to Extract Free Anthraquinones in Rhubarb

Objective To optimize the extraction process of free anthraquinones from Rheum Emodi Wall. with microwave technique. Methods Single factor and orthogonal test design were used to investigante the effects of the four main factors including microwave power, alcohol concentration, extraction time and the ratio of materials and solution on the extraction efficiency of free anthraquinones. Results Using 70% ethanol solution in high-fire and the ratio of materials and solution of 1∶8 for 5 min to extract two times, the extraction rate of Rhubarb anthraquinones was 1.31% and the relative rate was 88.52%. Conclusion Microwave extraction method is high efficient, easy and timesaving for extracting anthraquinones in Rheum Emodi Wall..
4.Developing Pharmacists' Clinical Thinking with Clinical Pharmaceutical Practice as the Core

OBJECTIVE:To promote the development of pharmacists' clinical thinking ability.METHODS:By analyzing the differences between physicians and pharmacists in clinical thinking together with our practical experience,we expounded the key points of pharmacists' clinical thinking,i.e.critical thinking,comprehensive thinking,evidence-based thinking,patient-cored thinking and creative thinking.RESULTS & CONCLUSION:The clinical thinking that is in line with pharmacists' characteristics should be developed with clinical pharmaceutical practice as the core and it can be achieved by enhancing pharmacists' ability to detect and solve the drug related problems.
5.Determination of Capsaicine, Dihydrocapsaicin, Imperatorin and Isoimperatorin in Guanjie Jietong Plas-ters by HPLC

Objective:To develop an HPLC method for the determination of capsaicine, dihydrocapsaicin, imperatorin and isoim-peratorin in Guanjie Jietong plasters. Methods:A CAPCELL PAK C18 column was used as the chromatographic column, and the flow rate was 0. 8 ml·min-1. The mobile phase A consisted of methanol-acetonitrile(1∶1), the mobile phase B was of 1% phosphoric acid. The detection wavelength was 280 nm and 254 nm, respectively. Results:There was a good linear relationship between the con-centrations and the peak areas within the range of 0. 298-5. 956μg(r=0. 9998) for capsaicine, 0. 152-3. 044μg(r=0. 999 6) for di-hydrocapsaicin, 0. 018-0. 352 μg(r=0. 999 5) for imperatorin and 0. 010-0. 204 μg(r=0. 999 3) for isoimperatorin. The average re-covery was 97. 8%(RSD=1. 02%), 97. 0%(RSD=0. 76%), 96. 6%(RSD=0. 65%) and 98. 1%(RSD=1. 35%), respectively. Conclusion:The method is convenient, accurate, sensitive and repeatable, which can be used in the determination of capsaicine, di-hydrocapsaicin, imperatorin and isoimperatorin in Guanjie Jietong plasters.
6.The effects compare analysis of different moist environment for treating severely burned patients

Objective To compare the effects of different moist environment for treating severely burned patients.Methods From November 2009 to December 2014 in the Burn Unit of Hainan General Hospital, 170 hospitalized patients with severe burns were randomly equally divided into observation group and control group.The patients of control group were treated with the traditional wet therapy treatment, while of the observation group were given the closed moist therapy treatment.The wound healing, pain relief and adverse reactions were observed.Results All patients completed the treatment,and the cure rates at the 7th and 14th day of treatment in the treatment group were 80.0% (68/85) and 97.6% (83/85), significantly higher than those of the control group (52.9% (45/85), 89.4% (76/85), x2 =8.344,4.111, P < 0.05).The number of dressing changes((7.44±2.39) times), cumulative dressing time ((45.98 ± 13.29) min) and wound healing time ((14.98±4.33)d) in the observation group were significantly less than those in the control group((11.76 ±3.14) times, (87.03 ± 14.99) min), (18.87 ± 5.13) d)), the differences were statistically significant (t =6.349,6.540,3.111, P<0.05).After treatment, the pain scores were showed clear downward trend, with had significant difference compared before treatment (P<0.05), while the pain scores at the 7th day((3.10± 1.34) scores) and 14th day((1.76±0.67) scores) of treatment in the treatment group were significantly lower than the control group((4.09±0.89) scores, (3.11±0.56) scores, P<0.05).The incidence of adverse events during treatment in both groups were 5.9%, obviously improved after symptomatic treatment.Conclusion The moist environment conducive to the treatment of severely burned patients, and safety.Conclusion to traditional wet therapy, closed moist therapy has better cure rate of severe burns, can promote wound healing, relieve pain and shorten the dressing cycle time.
7.The experimental progress on arsenic trioxide for non-APL and solid tumor

Arsenic trioxide(As2O3)effectively triggers apoptosis in acute promyelocytic leukemia (APL) cells and induces complete remission and cure in most APL patients ,if they can refrain from early mortality .In the last few years,it has been under investigation as a potential treatment for non -APL leukemia and a variety of sol-id tumors.In the meantime,it has been found that As2O3 also inhibits proliferation of non -APL leukemia cell and several solid tumor cell such as liver ,esophageal ,and gastric cancer cell in vitro or in vivo in the more and more studies .The molecular mechanisms that As 2 O3 induces cellular apoptosis in non APL leukemia cell and solid tumor cells have been discussed ,which also includes building new targeting preparation in order to obtain much more satisfying therapeutic effect and relieve its cellular toxic and side effect and so on.Here, we give a summary which is connected with the research progress above .
8.Prevention and treatment of chylous leakage after Whipple operation

Objective To study the prevention and treatment of chylous leakage after Whipple operation.Methods Clinical data of 381 patients underwent Whipple operation from Jan 2010 to march 2013 were retrospectively analyzed.Results Among 381 patients,23 patients had chylous leakage,the incidence was 6.04%.While in 89 patients,in which intraoperative precautionary physical ligation or mattress suturing of the lymphatic vessels were undertaken,no chyle leakage occurred ; Most of chyle leakage occurred in malignant tumor after radical resection(21 patients),most chylous leakage was found 5-8 days after surgery,with daily volume of 260-450 ml.All patients of chylous leakage were cured by conservative treatment.Conclusions Chylous leakage are common in PD.Intraoperative preventive measures such as lymphatic duct ligation or safe suturing can effectively prevent chyle leakage.Conservative therapy heals most chylous leakage after Whipple operation.
9.Effect of Paecilomyces cicadae polysaccharide on duplication of hepatitis B virus in HepG2.2.15 cell strain

AIM: To investigative the inhibitory effects of Paecilomyces cicadae polysaccharide (PcPS) against HBV in vitro, and the effects on the Toll like receptor 4 (TLR4) mRNA expression in HepG2.2.15 cell strain. METHODS: HepG2.2.15 cell strain was co-cultured in vitro with PcPS in different concentrations, and lamivudine (LMV) was applied as positive control. MTT assay was employed to detect the cytotoxicities of PcPS in vitro, when the HepG2.2.15 cells was used as target cells. The effects of PcPS on the secretion of HBsAg and HBeAg were assayed by ELISA method. Fluorescence quantitative-PCR (FQ-PCR) was used to detect the inhibitory effects of PcPS on the content of HBV-DNA and TLR4 mRNA in HepG2.2.15 cells. RESULTS: The inhibitory effects of PcPS on the HBsAg and HBeAg were observed and the maximum inhibitory ratio up to 44.8% and 31.0%, respectively. The same inhibitory effects of PcPS on the HBV-DNA replication and TLR4 mRNA expression in HepG2.2.15 cells were also found. CONCLUSION: A certain concentration of PcPS significantly inhibits HBV replication in vitro.
