1.Expression and clinical significance of miR-96-5p in primary hepatocellular carcinoma at early recurrence after radical surgery

Objective To investigate the expression and clinical significance of miR-96-5p in primary hepatocellular carcinoma (HCC) at early recurrence after radical surgery. Methods 61 HCC eryopreservation tissue samples from the liver carcinoma specimens data obtained after radical surgery and banked in our hospital were divided into 2 groups: early recurrence group (33 cases) and non-early recurrence group (28 cases). Aquantitative real-time polymerase chain reaction (qRT-PCR) was performed to detect the expression of miR-96-5p. Results Compared with the non-early recurrence group , the expression of miR-96-5p was observably down-regulated [(0.634 ± 0.783) vs (5.182 ± 11.321), P = 0.043]. The expression of miR-96-5p was correlated to tumor diameter, early recurrence and vascular invasion (P<0.05). Conclusions miR-96-5p are significantly related to early liver cancer recurrence and metastasis. miR-96-5p may be a molecular marker of HCC at early recurrence as well as a target for targeted therapy of liver cancer in future.
2.Effects of hypertonic saline on TNF-α、IL-1β、S-100β following focal cerebral ischemia-reperfusion injury in rats

Objective To investigate the effect of hypertonic saline on TNF-а, IL-1β, S-100β level following focal cerebral ischemia-reperfusion injury in rats and explore the mechanisms involved. Methods Ninety rats were randomized equally into 4 groups,namely the pseudo-operated group (A group),untreated IR injury group (B group),C group (4.2% hypertonic saline) and D group (7.5% hypertonic saline). Cerebral ischemia was induced by middle cerebral artery occlusion for 2 h followed by administration of the corresponding treatments in group C and D. After 22 h of reperfusion , TNF-а, IL-1β, S-100βexpression in the ischemic brain tissue were measured by enzyme-linked immunosorbent assay (ELISA). Results After hypcrtonic saline treatment, the two saline-treated groups showed significant reduction in TNF-а、IL-1β、S-100β levels , but were still higher than pseudo-operated group(A group). Compared with B group, significant difference can be seen among C and D group. Conclusion Hypcrtonic saline can reduce cerebral TNF-а、IL-1β、S-100βlevel of isehemia-reperfusion injury.
3.Controllable Abdominal Breath in treatment of Migraine

Objective:To study the effect of controllable abdominal breath in treatment of migraine.Method:20 patients(10 male,10 female,aged 20-30 years)with migraine according to international diagnostic standard published 1990 were randomly divided into study group and control group.The study group received training of controllable abdominal breath.All subjects were followed for 3 months.Changes of symptoms and hemorhelogy were recorded.Result:Study group improved in both symptom and index of hemorheology after treatment,and the results were better than that of control.Conclusion:Controllable abdominal breath is effective in treatment of migraine.

5.Investigate of correlation between non-lacunar infarcts in the white matter of brain and intracranial or extracranial vessels stenosis

Objective To investigate the correlation between non-lacunar infarcts in the white matter of brain and intracranial or extracranial vessels stenosis.Methods Brain magnetic resonance imaging(MRI) and digital subtraction angiography(DSA) were performed in 30 patients with non-lacunar infarcts in the white matter of brain and the results were analyzed.Results Of 12 cases with focal non-lacunar infarcts in the area of basal ganglion,9 cases were shown occlusion or highly stenosis of proximal segments of unilateral internal carotid artery(ICA) in DSA,2 cases were shown highly stenosis of M1 segment of middle cerebral artery(MCA),and 1 case was normal.Of 6 cases with focal non-lacunar infarcts in the area of basal ganglion and lateral periventricular white matter,3 cases were shown occlusion or highly stenosis of proximal segments of unilateral ICA in DSA,1 case was shown occlusion of C5 segment of unilateral ICA,and 2 cases were shown occlusion of M1 segments of MCA.Of 4 cases with focal non-lacunar infarcts in periventricular white matter or centrum ovale,1 case was shown occlusion of C5 segment of unilateral ICA,2 cases were shown highly stenosis of M1 segments of MCA,and the other one was normal.Of 8 cases with subcortical superior border infarction(SSBI) or subcortical lateral border infarction(SLBI),6 cases were shown occlusion or highly stenosis of proximal segments of unilateral ICA,1 case was shown occlusion of proximal segments of bilateral ICA,and 1 case was shown occlusion of M1 segment of MCA.Conclusion There are strong correlations between focal non-lacunar infarcts in the areas of basal ganglion or lateral periventricular white matter and the stenosis or occlusion of large arteries of ICA system.
6.Relationship of tumor necrosis factor gene polymorphism and acute graft-versus-host disease after unrelated ailogeneic hematopoietic stem cell transplantation

Objective To explore the relationship between tumor necrosis factor (TNF) gene polymorphisms in donors and recipients and the incidence and severity of acute graft-versus-host diseases (aGVHD) after unrelated allogeneic hematopoietic stem cell transplantation (alIo-HSCT). Methods Single nucleotide polymorphisms (SNPs) of TNFα-238 (G/A), TNFα-857 (C/T), TNFα-863 (C/A), TNFα-1031 (T/C), TNFβ + 252 (A/G) were analyzed by Multiplex SNaPshot analysis in 76 pairs of donors and recipients. Results Transplantation involving donors with TNFα-857 CC genotype resulted in a higher incidence of grade Ⅱ-Ⅳ aGVHD than donors with CT genotype (91.3% vs 8. 7% , P =0. 039). In the 23 patients with grade Ⅱ-Ⅳ aGVHD, no patients had TNFβ +252 AA genotype, 19 (82.6%) had GA genotype and 4 (17.4%) had GG genotype. There was a significant difference in the distribution pattern of the TNFβ +252 (AA, GA and GG) genotypes in these patients (P =0.03). There was no significant association of TNFα-238 (G/A), TNFα-863 (C/A) and TNFα-1031 (T/C) polymorphisms with the risk of aGVHD. Conclusion These results suggest donor TNFα-857 CC genotype is related to a higher incidence of grade Ⅱ -Ⅳ aGVHD, and patients with TNFβ +252 AA genotype have protection against the risk of grade Ⅱ -Ⅳ aGVHD.
7.REPORT OF HLA DISTRIBUTION IN 2315 VOLUNTEER DONORS OF CHINESE BONE MARROW BANK FROM NORTHWEST CHINA

Objective To report the HLA data of volunteer donors of Chinese bank from Northwest China and characterize the distribution of HLA genes in Northwest China. Methods HLA-A, B antigens of 2315 volunteer donors were examined by the method of microlymphocytetoxicity (MLT) test .The antigen frequencies(AF) were assessed by directly counting; and based on that gene frequencies(GF) were calculated. HLA data from other population were collected and distribution characteristics were compared. With the raw data, Hardy-Weinberg equilibrium, statistical parameters of forensic medicine interest for HLA were computed. Results A total of 18 specific antigens were detected in HLA-A and the most frequent antigen was A2 . AF and GF were 0.5136 and 0.3026, respectively. A total of 42 specific antigens were detected in HLA-B and the most frequent antigen was A13. Its AF and GF were 0.1978 and 0.1044, respectively. The heterozygosity(H), polymorphism information content(PIC), discrimination power(DP) and probability of paternity exclusion (PPE) of HLA-A were 0.8215, 0.8212, 0.9356 and 0.7798 accordingly; while H,PIC, DP and PPE of HLA-B were 0.9322, 0.9322, 0.9878 and 0.9528. Conclusion The polymorphism of HLA-A,B genes is characteristic in Chinese. In this research, the genetic trait of HLA in 2315 volunteers is consistent with Northern Han population.

9.Reticulate pigmented anomaly of the flexures: a family survey and mutation analysis of KRT5 gene

ObjectiveTo observe clinical features and identify causative genes of reticulate pigmented anomaly of the flexures in a pedigree.Methods A survey was conducted in a pedigree with reticulate pigmented anomaly of the flexures.Clinical manifestations were recorded in details for each patient in this pedigree.Tissue specimen was obtained from the proband for histopathological examination and ultrastructural observation.Mutation scanning was carried out by PCR and direct sequencing in 3 patients in the family.ResultsAll the patients in this pedigree presented with reticular pigmentation of the flexures and idiopathic guttate hypomelanosis on the abdomen and back.Histopathological and ultrastructural study revealed epidermal hyperpigmentation with an increase in melanin content in epidermal keratinocytes but no changes in the number of melanocytes.No mutation was found in the KRT5 gene in this family.ConclusionsThis is the first case report of reticulate pigmented anomaly of the flexures associated with idiopathic guttate hypomelanosis.No mutation is identified in the KRT5 gene of patients with reticulate pigmented anomaly of the flexures in this family,indicating the existence of other causative genes.
10.Open reduction and internal fixation for fractured condylar neck and mandible ramus via intraoral approach

Objective To discuss the way of open reduction and internal fixation for fractured condylar neck and mandible ramus via intraoral approach aiming to avoid the facial incision. Methods Fifteen cases (17 sides) with mandibular condyle fractures underwent open reduction and osteosynthesis with plates and screws. After the mandibular ramus was under vertical osteotomy with an oscillating saw, the posterior border bone block of mandibular ramus as well as the free condyle neck were taken out. The fractured condyle neck and the posterior border bone block of mandibular ramus were fixated with a titanium miniplate in vitro. The reunion bone was implanted and reposited in the mouth incision. Of all, two cases suffered fracture of the condyle neck in the other hospital when they received esthetic surgery and resection of prominent mandible angle (PMA). Results Anatomic reduction was achieved in all cases, without damage to facial nerve and major auricular nerve or salivary fistula. There were slight bony resorption and good temporomandibular joint function one year after surgery, with range of mouth opening for 25-40 mm (mean 35.8 mm). Two cases regained their occlusion before their PMA operation. One case had premature contact of the buccal teeth,with 1 mm diverging to medline of the incisor teeth. Intraoral approach not only could avoid large facial scars and facial nerve injury, but also allow visualization of the occlusion during the procedure. Conclusions As more and more consideration is taken to cosmetology, the transoral approach is a reliable surgical alternative for fractures of the condyle neck, without leaving extensive visible scars or damaging facial nerve. The disadvantage is vertical osteotomy of the posterior border of the mandibular ramus.
