1.Recurrent implantation failure: gamete and embryo factors.

Despite the immense achievement in the field of IVF in recent years, many patients still suffer from recurrent implantation failure. Therefore, much attention has been drawn to its etiology and treatment. Chromosomal abnormality, sperm DNA damage, zona hardening, and inappropriate culture conditions are important factors that lead to recurrent implantation failure. Results of studies suggest that preimplantation genetic screening does not improve the rate of implantation or live birth. Comparative genomic hybridization array and single nucleotide polymorphism array could offer a more comprehensive screening of chromosomes. Assisted hatching may help to solve the problem of zona hardening in some situations. Co-culture and blastocyst transfer could be conducive to the improvement of the rates of implantation and pregnancy. Cytoplasmic transfer may give a solution to ooplasmic composition anomalies.

2. "Sweating" of traditional Chinese medicinal materials during primary processing and its mechanisms of enzymatic reaction and chemical conversion

Primary processing of traditional Chinese medicinal materials (TCMM) is an important link which closely relates to the quality of products in TCMM. The traditional primary processing method and technology systems were derived from the long-term practices and experiences, which are distinctive, colorful, diverse, and scientific. The method of "sweating" is a critical step for processing the rhizome, root, cortex, and sclerotium and helpful for distributing moisture from inside to outside of TCMM in drying process, regulating and promoting the enzymatic reaction and chemical conversion by enzyme and germs, and starting or accelerating the biotransformation and chemical conversion of primary/secondary metabolites, which could directly affect the quality of TCMM. In this paper, the authors discuss the origins and development, purpose, and significance of "sweating" processing and the mechanisms of enzymatic reaction and chemical conversion of chemical compositions during the primary processing of "sweating". These data may provide the foundation and support for processing in normalization and standardization and formulating the Standard Operating Procedure (SOP) of primary processing of TCMM.
3. Establishment and practice for evaluating model of best harvest time of traditional Chinese medicinal materials

The harvest time of traditional Chinese medicinal materials (TCMM) is a very essential part for production of TCMM, and affects their quality and yield. The research on the best harvest time of TCMM has been a key problem taken into account more and more with practicing GAP of TCMM. Based on thoughts and methods of resource chemistry, this paper established an evaluated mode to objectively determine the best harvest time of Angelica sinensis through system investigating from different producing areas and harvest times, and characterize the phenology and multi-index components comprehensive criteria. The author firstly proposed the suitable methods of the best harvest time of TCMM, which was a basis of theory and methods of resource chemistry of TCMM, and including the time-space relationships and laws of substances dynamic accumulation. These data could provide the guidance for standardized production and guaranteed quality and yield of TCMM.
4.Relationship of blood pressure variability and heart rate variability with prostatic volume in patients with benign prostatic hyperplasia

Objective To investigate the relationship of blood pressure variability(BPV) and heart rate variability(HRV) with prostatic volume (PV) in patients with benign prostatic hyperplasia (BPH).Methods A total of 133 patients admitted to our department between January 2011 to April 2013 were analyzed retrospectively.The patients were divided into BPH group and non-BPH group according to the PV value.The ambulatory blood pressure monitoring was used to measure the blood pressure parameters,including 24-hour systolic blood pressure (SBP),24-hour diastolic blood pressure (DBP),day time systolic blood pressure(DSBP),day time diastolic blood pressure (DDBP),night systolic blood pressure (NSBP),night diastolic blood pressure(NDBP),the standard deviation of DSBP(DSBPSD),DDBPSD,NSBPSD,and NDBPSD.At the same time,heart rate variability was measured by 24 hours Holter monitor.The ambulatory electrocardiogram was used for calculating normal to normal intervals (SDNN),standard deviation of the averages of R-R internals in all 5-minute segments(SDANN),rate mean square of the differences of successive RR intervals(RMSSD),percentage of RR intervals differing >50 ms (PNN50%).The differences of the ambulatory blood pressure parameters or heart rate variability were compared between the two groups.Results The levels of 24-hour SBP,DSBP,and DSBP-SD were higher in the BPH group than in the non-BPH group[(126.03±14.66)mmHg (1 mmHg=0.133 kPa) vs.(118.88 ±10.79) mmHg,(126.97±14.46)mmHg vs.(119.94±11.24)mmHg,(12.52±3.85)mmHg vs.(11.04±2.44)mmHg,all P<0.05].All the parameters of HRV were significantly different between the two groups [(97.22±38.14)mmHg vs.(119.23±36.16)mmHg、(90.11±34.4)mmHg vs.(107.1144.4)mmHg、(19.11±2.76)mmHg vs.(31.96±21.10)mmHg、(2.31±2.87)% vs.(5.02±4.88)%,all P<0.05]Conclusions The BPV and HRV are the important influencing factors for PV in BHP patients.

Objective:To study the characters of perioral muscle p re ssure of the adult patients with skeletal crossbite. Methods:20 patients were diagnosed to be with skeletal crossbite. The perioral muscle press ures on upper and lower central incisors, first molars and cuspids were measured buccally and lingually at rest position. 10 health adults were served as the co ntrols. Results:The perioral force in skeletal crossbite group was larger than that in the control group on the correspondence area(P0.05). In the skeletal cross bite group, the perioral force on the mandibular area was larger than that on th e correspondence maxillary area(P0.05). 6.Construction and identification of mcpr1 gene eukaryotic expressing vector

Objective: To construct a high effective eukaryotic expre ss ing vector containing mcpr1 gene. Methods: mcpr1 gene w as amplified by PCR from the plasmid T-easy/ mcpr1, then PCR product was in serted into eukaryotic expressing vector pcDNA3.1/V5-His B. The positive recomb inant was identified by PCR analysis, HindIII and BamHI restriction analysis and Sequence analysis. Results: A 400 bp DNA fragment was amplified from the recombinant. Sequence analysis and restriction digest demonstrated tha t the mcpr1 gene was successfully inserted into pcDNA3.1/V5-His B plasmid. Conclusion: The eukaryotic expressed vector pcDNA3.1/V5-His B/ mcpr1 has been successfully reconstructed.
7.Influence of rebonded orthodontic brackets and different treatment methods on shear bond strength

Objective:To compare shear bond strength(SBS)of bonded and rebonded orthodontic brackets and influence of rebonded orthodontic brackets using different treatment methods on SBS.Methods:60 extracted premolars were evenly divided into three groups.Brackets were debonded with conventional techniques and then removed of residual adhesives from the base mesh using three different treatments.In groupⅠ,rebond a new bracket at the second,third bonding times.In group Ⅱ,remove residual adhesives from the base mesh with fire at the second,third times,then rebond the same bracket.In group Ⅲ,remove that using a tungsten carbide bur at the second,third times,then rebond the same bracket.Shear test was performed to compare the SBS and adhesive remnant indexes(ARI)among different groups.Results:The SBS of three groups at the first bonding sequence were(10.00?2.95)MPa,(9.57?2.45)MPa and(9.09?2.58)MPa.The SBS at the second,third debonding sequence in group I,group II,and group III were(9.55?2.84)MPa and(10.32?2.59)MPa,(7.30?2.15)MPa and(7.14?1.93)MPa,(12.13?2.93)MPa and(12.86?3.08)MPa,respectively.In group Ⅱ,the SBS was significantly lower,however,in group Ⅲ,the SBS was significantly higher,and the results showed significant differences(P
8.Construction of psm-negative mutans of S.mutans UA159

Objective:To knock the phospho-sugar mutase gene (psm) out of the genome of S. mutans strain UA159 and to construct the mutant will lay a foundation for the study of the function of the psm. Methods:Two upstream and downstream DNA sequences of the psm were selected and cloned respectively into multiple cloning sites I and II of suicide plasmid pFW5 to construct the recombinant plasmid which was confirmed by enzyme digesting and sequencing. The recombinant plasmid was transformed into S. mutans UA159 by natural transformation and antibiotic was used to screen the positive transformants. According to the principle of homologous recombination, allelic exchange between the recombinant plasmid and S. mutans UA159 was achieved. Results:By PCR analysis and sequencing, it was confirmed that the psm of S. mutans UA159 was substituted for resistant gene of spectinomycin. Conclusion: The psm-negative mutants of S. mutans UA159 is successfully constructed.
9.Effect of Copper and Cu/Zn Ratio in Blood on Blood-fat in Copper Deficiency Rats

Objective To study the relation between copper,copper-zinc ratio in blood and blood-fat only in the copper intake lower than the normal acceptable daily intake,and to provide a unified dosage evidence for copper multi-ways exposure.Methods Forty-two SD rats were divided into 7 groups randomly.The rats in one group among them as the normal control group were fed on the normal fodder and water,the other 6 groups were fed on special fodder without copper and deionized water and i.g.copper gluconate at doses of 0ADI(the acceptable daily intake of rat),1/625ADI(0.000 128 mg/ml),1/125ADI(0.000 64 mg/ml),1/25ADI(0.003 2 mg/ml),1/5ADI(0.016 mg/ml) and ADI(0.8 mg/ml),for 30 days.The activity of ceruloplasmin(CP),the content of copper,zinc in the blood and the level of total cholesterol(TC),triglyceride(TG),high-density lipoprotein(HDL) and low density-lipoprotein(LDL) were determined.Results In the case of copper intake lower than the normal acceptable daily intake,the activity of CP in the blood of all treated rats were lower than the normal level(P
