1.The expression of P-selectin in rats with hepatic ischemia reperfusion injury

Objective To explore the express of P-selectin in liver-tissue,on the model of HIRI in rats,we explored the pathogenesy of HIRI. Methods 32 female Sprague-Dawley rats were divided into four groups randomly ( n = 8 in each group): sham operation group ( group SO ), ischemia 30 min without repe- rfusion group ( group Ⅰ),30 min reperfusion following ischemia 30 min group ( group IR30 min), one hour reperfusion following ischemia 30min group( group IR1h). The model of HIRI was maded by Pringle's hepatic vascular occlusion. The contents of P-selectin in hepatic tissue were measured by kit of SP. Results After ischemia 30 min,the content of P-selectin in hepatic tissue increased less obviously than group SO; After 1 h reperfusion, the content of P-selectin in hepatic tissue vascular endothelial and hepatocyte express more broadly than group SO. Conclusion The content of P-selectin in hepatic tissue was increased gradually in the process of hepatic ischemia reperfusion injury(HIRI) in rats.
2.Effect of natriuretic peptide on vascular endothelial function and inflammatory factors of patients with heart failure complicating myocardial infarction

Objective To investigate the effect of natriuretic peptide on vascular endothelial function and inflammatory factors of patients with heart failure complicating myocardial infarction.Methods Two hundred and twenty-six patients with heart failure complicating myocardial infarction admitted into Hanzhong Central Hospital from March 2012 to March 2016 were randomly divided into experimental group (n =138) and control group (n =128).Patients in control group were treated with conventional drug,and patients in experimental group were treated with natriuretic peptide based on conventional drug treatment.The vascular endothelial function,contents of NO and eddthelin 1,and inflammatory factors including CRP,IL-6,and TNF-α were detected.Results After 2 weeks of treatment,the flow-mediated vasodilation (FMD) and NO in experimental group were remarkably higher than those in control group,and eddthelin 1 was significantly lower than those of control group (P < 0.05).Additionally,CRP,IL-6,and TNF-α in experimental group were significantly lower than those in control group after treatment for 5 d (P < 0.05).Conclusion Natriuretic peptide could improve the vascular endothelial function of patients with heart failure complicating myocardial infarction via increasing the content of NO and decreasing content of eddthelin 1,and was benefit to cardiac function by inhibiting release of inflammatory factors.
3.Repetitive nerve stimulation in amyotrophic lateral sclerosis

ABSTRACT:Objective To evaluate the features and related factors of decremental response in amyotrophic lateral sclerosis (ALS)patients to low-frequency repetitive nerve stimulation (RNS)in proximal nerves.Methods We performed RNS studies in proximal axillary and accessory nerves with recording in deltoid and trapezius mus-cle respectively in 87 ALS patients.Decremental compound muscle action potential (CMAP)and related factors were analyzed prospectively,and abnormal group of decremental response in ALS patients was compared with 39 pa-tients with myasthenia gravis.Results ① Abnormal decremental responses were found in 43.7% and 49.4% of ALS patients with deltoid and trapezius muscle recording respectively.They were found more frequent in trapezius muscle than those of deltoid muscle.② There was no relationship of decremental response with gender,age,onset or course of disease,ALSFRS-r scores,or rate of disease progression in ALS patients.③ There was significant rela-tionship between decremental response and fluctuating muscle weakness.Decremental responses decreased more ob-viously in ALS patients with fluctuating muscle weakness than in those with nonfluctuating muscle weakness.④ Dec-remental responses were greater in patients with myasthenia gravis than that in ALS patients.Conclusion Decre-mental response with proximal muscle recording is not an uncommon feature in ALS patients;therefore,it should not be treated as a criterium to rule out ALS.Abnormal decremental response of trapezius muscle is found more fre-quent than that of deltoid muscle.Decremental response range in patients with myasthenia gravis is significantly lar-ger than that in ALS patients.One should be more careful when diagnosing ALS patients with fluctuating muscle weakness and abnormal decremental response.
4.Influence of TPO-Ab on in vitro fertilization and embryo transfer ending

Objective To Study the effect of thyroid peroxidase-Ab (TPO-Ab) on in vitro fertilization and embryo transfer (IVF-ET) ending. Methods From March 2011 to October 2013, a total of 378 assisted reproduction cycles in Reproductive Medical Center of Qinghai Provincial People ’s Hospital were retrospectively analyzed in. Experiment group included 69 cycles from patients with positive thyroid peroxidase antibody and normal thyroid function and control group 309 cycles from those with negative thyroid peroxidase antibody and normal thyroid function. Results IVF fertility rate (79.76%) and high quality embryo rate (46.59%) in experiment group were significantly lower than those in control group (84.08%, 54.49% respectively) and the difference was statistically significant (P < 0.05); abortion rate (33.33%) in experiment group was obviously higher than that in control group (16.11%), which was statistically significant (P < 0.05). Conclusion Positive thyroid peroxidase antibody exerts adverse effects on IVF-ET ending since TPO-Ab reduces IVF fertilization rate and high quality embryo rate and increases the rate of abortion.
5.Effect of oxymatrine on the expression of p-STAT3 and PIAS3 in human mesangial cells

Objective To study the effect of oxymatrine on p-STAT3 and PIAS3 signaling molecule and it's mRNA expression in the proliferation of the human mesangial cells (HMCs) induced by lipopolysaccharide(LPS) and to explore their relationship. Methods HMCs were divided into three groups: control group, LPS group and oxymatrine group. HMC proliferation was detected by methyl thiazolyl tetrazolium (MTT) assay. Type Ⅳ collagen in the supernatants of the cultured HMCs was detected by ELISA at 12, 24, 48 hours respectively. At the same time, the protein and mRNA expressions of p-STAT3 and PIAS3 were measured by Western blot and real-time quantitative RT-PCR. Results The cell proliferation, the mRNA and protein expression of type Ⅳ collagen, p-STAT3 in LPS group were increased compared with the control group (P<0.01), but they were decreased in oxymatrine group (P < 0.01). The expressions of protein and mRNA of PIAS3 in LPS group were decreased significantly compared with control group (P<0.01), but they were increased in oxymatrine group (P<0.01). Conclusion Oxymatrine can down-regulate the expression of p-STAT3 and up-regulate the expression of PIAS3, which plays an important role in the process of LPS-induced HMCs proliferation.
6.The impact of combined laparoscopic and choledochoscopic treatment on biliary leak in hepatic hydatid disease in a high attitude region

Objective To explore the impact of combined laparoscopic and choledochoscopic treatment on bile leak in hepatic hydatid disease at a high attitude region.Methods The clinical data of 50 patients who underwent combined laparoscopic and choledochoscopic treatment were compared with 50 patients who underwent laparoscopic surgery alone for hepatic hydatid internal capsule excision surgery.Results In the combined laparoscopic and choledochoscopic group,the incidence of biliary fistula after surgery was 15%,the incision infection rate was 13%,the length of hospitalization was (11.6 ± 3.8) days and the postoperative time to take off drain was (14.3 ± 7.9) days.These were significantly better than those in the laparoscopic alone group.The operation time in the former group was (108.2 ± 28.4)min,which was slightly longer than the laparoscopy group.Conclusion The combined laparoscopic and choledochoscopic treatment is a safer therapeutic option in treating hepatic hydatid disease with less trauma,faster recovery and shorter hospital stay than laparoscopic surgery alone in a high attitude region.
7.Oxymatrine suppresses p-STAT1/PIAS1 signaling in LPS-induced human mesangial cells proliferation

Objective To observe the effect of oxymatrine(OM) on the expressions of p-STAT1 and PIAS1 signaling molecules at protein and mRNA levels in the proliferation of the human mesangial cells(HMC) induced by lipopolysaccharide(LPS) and explore the relationship between them.Methods HMCs were primarily cultured from a 4-month-old aborted human fetus(with informed consent and approved by the Ethics Committee of Lanzhou University),and then divided into 3 groups,that is,control group,LPS group(10 ng/ml) and OM group(LPS 10 ng/ml and OM 320 mg/L).After cultured for 12,24 and 48 h respectively,HMC proliferation were analyzed by methyl thiazolyl tetrazolium(MTT) assay and type Ⅳ collagen in the supernatants were detected by ELISA.At the same time points,the cells lysates were collected for the mRNA and protein expressions of p-STAT1 and PIAS1 by real-time quantitative RT-PCR and Western blot analysis.Results The cell proliferation of LPS group was faster and the type Ⅳ collagen protein was increased more than the control group(P
8.Space Mutation Effects of Rhodobacter Sphaeroides

Objective To study mutagenic effects of spaceflight on physiological and biochemical parameters of Rhodobacter sphaeroides and select high-yield mutants in co-enzyme Q10(CoQ10) production for providing the experimental and theoretical basis for industrial production through mutagenic effects of spaceflight.Methods Variations in stress resistance and CoQ10 production of isolated strains were studied,the strain Rhodobacter sphaeroides was taken back by a recoverable satellite after 15 d flight in space.Results Compared to the control,the strain was characterized by highter NaCl tolerance and higher stress resistance,as well as with broader scope in growth temperature and pH value after spaceflight.The mutant colonies appeared white or pink which was different from their original red.The CoQ10 production of mutant 10 strain was increased by 73.13% much higher than that of control.Conclusion Spaceflight mutagenic effects on Rhodobacter sphaeroides shows to be multi-factor compared to the traditional single-factor mutagenesis methods.It can enhance stress resistance and increase CoQ10 production of isolated strains,and can be utilized in industrial microbial mutagenesis and breeding in the future.
9.Disease onset regions and spreading patterns in sporadic amyotrophic lateral sclerosis and related influencing factors

Objective To evaluate the disease onset regions and spreading patterns in sporadic amyotrophic lateral sclerosis (ALS)patients and related influencing factors.Methods We performed a prospective analysis of 1 58 ALS patients.The disease-onset was confirmed according to the patients’self-reports,neurological examination results and electromyogram study.We followed up 1 5 1 patients with the second or other affected body regions during the disease progression.Data were analyzed according to the different groups of onset regions.Results 1.In 94.3% (149/1 58)of the patients,the early motor manifestations were focally in the bulbar,upper or lower limbs.2.The region of onset was associated with signs of lower motor neuron (LMN)and upper motor neuron (UMN)involvement (P = 0.000 ).The LMN involvement was more distinctive in patients with bulbar onset (65.4%,1 7/26 )group.Patients with cervical onset more frequently showed pure LMN (47.9%,45/94 )or concomitant UMN (52.1%,49/94)signs in the affected limbs.The highest proportion of UMN and LMN signs in the affected lower limb was found in the lumbar onset (83.8%,31/37 )group.3.Spreading patterns:Rostral to caudal spreading pattern was more frequent in bulbar onset patients.For patients with limb onset,there were significant differences between spreading patterns and disease-onset regions (P =0.04).Circular (31.5%,28/89),horizontal (30.3%,31/89)and vertical (21.3%,1 9/89)spreading patterns were more frequent in cervical onset patients whereas circular (47.2%,1 7/36)spreading patterns were more frequent in lumbar onset patients.4.There was a strong association between the rate of progression and age of disease onset (P =0.01 1).Patients aged over 60 had a faster progression.Conclusion ALS is a focal process at motor axis along the spinal cord and cerebral cortex.Different disease-onset can cause different distribution of UMN and LMN signs.Therefore,special attention should be paid to the signs of disease-onset clinically.ALS does start focally and spreads to adjacent regions.Elder patients have a faster disease progression.
10.Effects of Matrine on STAT1, STAT3, CTGF, PDGF expression in fetal glomerular mesangial cells under lipopolysaccharide

Objectives To determine the effects of Matrine (Ma) on signal transducers and activators of transcription (STAT) 1, 3, connective tissue growth factor (CTGF) and platelet -derived growth factor (PDGF) in glomerular mesangial cells (MC) induced by lipopolysaccharide (LPS), and to explore the protective mechanisms of Ma. Methods Ma were added to cultured glomerular mesangial cells which were exposed to LPS for 12, 24 and 48 hours. Real time polymerase chain reaction were used to determine STAT1, 3, CTGF, PDGF mRNA, The protein expression of STATI, 3 and p-STAT1, 3 were observed by Western blot. Results Compared with the control group, MC proliferation of the LPS group (10μg/ml) significantly increased, which were suppressed in the Ma (320 μg/ml) group (P < 0.01) at 12, 24, 48 hours. The expression of STAT1, 3, CTGF, PDGF mRNA and the levels of p-STAT1 , p-STAT3 protein was significantly increased in MC under LPS medium at 12, 24, 48 hours, which were obviously lower in Ma group than those of the LPS group, especially at 24, 48 hours. Conclusions The protective mechanisms of Matrine is considered to down-regulate the expression of STAT1, 3, CTGF and PDGF.
